| 2 | Rapid quantification of hepatitis B virus DNA by real-time PCR using efficient TaqMan probe and extraction of virus DNA显示文摘AIM: To rapidly quantify hepatitis B virus (HBV) DNA by real-time PCR using efficient TaqMan probe and extraction methods of virus DNA. METHODS: Three standards were prepared by cloning PCR products which targeted S, C and X region of HBV genome into pGEM-T vector respectively. A pair of primers and matched TaqMan probe were selected by comparing the copy number and the Ct values of HBV serum samples derived from the three different standard curves using certain serum DNA. Then the efficiency of six HBV DNA extraction methods including guanidinium isothiocyanate, proteinase K, NaI, NaOH lysis, alkaline lysis and simple boiling was analyzed in sample A, B and C by real-time PCR. Meanwhile, 8 clinical HBV serum samples were quantified. RESULTS: The copy number of the same HBV serum sample originated from the standard curve of S, C and X regions was 5.7 × 104/mL, 6.3 × 102/mL and 1.6 × 103/ mL respectively. The relative Ct value was 26.6, 31.8 and 29.5 respectively. Therefore, primers and matched probe from S region were chosen for further optimization of six extraction methods. The copy number of HBV serum samples A, B and C was 3.49 × 109/mL, 2.08 × 106/mL and 4.40 × 107/mL respectively, the relative Ct valuewas 19.9, 30 and 26.2 in the method of NaOH lysis, which was the efficientest among six methods. Simple boiling showed a slightly lower efficiency than NaOH lysis. Guanidinium isothiocyanate, proteinase K and NaI displayed that the copy number of HBV serum sample A, B and C was around 105/mL, meanwhile the Ct value was about 30. Alkaline failed to quantify the copy number of three HBV serum samples. Standard deviation (SD) and coefficient variation (CV) were very low in all 8 clinical HBV serum samples, showing that quantification of HBV DNA in triplicate was reliable and accurate. | 3 | Two-step method for creating a gastric tube during laparoscopic-thoracoscopic Ivor-Lewis esophagectomy显示文摘AIM To introduce a two-step method for creating a gastric tube during laparoscopic-thoracoscopic Ivor-Lewis esophagectomy and assess its clinical application.METHODS One hundred and twenty-two patients with middle or lower esophageal cancer who underwent laparoscopicthoracoscopic Ivor-Lewis esophagectomy at Liaoning Cancer Hospital and Institute from March 2014 to March 2016 were included in this study,and divided into two groups based on the procedure used for creating a gastric tube. One group used a two-step method for creating a gastric tube,and the other group used the conventional method. The two groups were compared regarding the operating time,surgical complications,and number of stapler cartridges used.RESULTS The mean operating time was significantly shorter in the two-step method group than in the conventional method group [238(179-293) min vs 272(189-347) min,P < 0.01]. No postoperative death occurred in either group. | 5 | Effect of^3B Group in Rings on Special Dispersion of B2O3-SiO2-ZrO2-Ta2O5-Na2O System Glass显示文摘Borosilicate glasses,with x B_2O_3-(60-x)SiO_2-8ZrO_2-8Ta_2O_5-24Na_2O(7≤x≤59 mol%)composition,were fabricated by melt-quenching technique.NMR,UV-Vis,Raman and IR spectroscopic studies were utilized to investigate the structure of fabricated glasses.The NMR spectrum was deconvoluted into five Gaussian bands,assigned to ~4B(0B,4Si),~4B(1B,3Si),~4B(2B,2Si),~3B(rings)and ~3B(loose),to get their quantitative information.The relative dispersion deviation?P_(g,F) was attributed to the relative quantity of ~3B(rings)but not all ~3B groups,and B_2O_3 existed mainly as[BO_3]in rings firstly,and then as[BO_3]in loose condition.The UV-Vis spectra revealed that the quantity of non-bridging oxygen increased firstly and then decreased with increasing concentration of B_2O_3.Acting as complementary techniques,Raman and IR measurements revealed that four-coordinated boron and silica mainly existed as ~4B-O-B,and Si-O-Si in Q^2,respectively,as chain structure but not framework structure,and[B?_4]units prefered connections with borate rather than with silicate entities of the glass network in these studied glasses.In addition,the conclusion also certified that ~3B in'loose'condition located in lower wavenumbers between 1 200-1 600 cm^(-1) in Raman spectra. | 许晓典 WANG Yanhang ZU Chengkui ZHOU Peng | 2018 | Journal of Wuhan University of Technology(Materials Science)2018,33,5: | 2 |

| 18 | Prediction of synthesis cross sections of new moscovium isotopes in fusion‑evaporation reactions显示文摘In the framework of the dinuclear system model,the synthesis mechanism of the superheavy nuclides with atomic numbers Z=112,114,115 in the reactions of projectiles 40,^(48)Ca bombarding on targets^(238)U,^(242)Pu,and^(243)Am within a wide interval of incident energy has been investigated systematically.Based on the available experimental excitation functions,the dependence of calculated synthesis cross-sections on collision orientations has been studied thoroughly.The total kinetic energy(TKE)of these collisions with fixed collision orientation shows orientation dependence,which can be used to predict the tendency of kinetic energy diffusion.The TKE is dependent on incident energies,as discussed in this paper.We applied the method based on the Coulomb barrier distribution function in our calculations.This allowed us to approximately consider all the collision orientations from tip-tip to side-side.The calculations of excitation functions of^(48)Ca+^(238)U,^(48)Ca+242Pu,and^(48)Ca+^(243)Am are in good agreement with the available experimental data.The isospin effect of projectiles on production cross-sections of moscovium isotopes and the influence of the entrance channel effect on the synthesis cross-sections of superheavy nuclei are also discussed in this paper.The synthesis cross-section of new moscovium isotopes 278−286 Mc was predicted to be as large as hundreds of pb in the fusion-evaporation reactions of^(35,37)Cl+^(248)Cf,^(38,40)Ar+^(247)Bk,^(39,41)K+247 Cm,^(40,42,44,46)Ca+^(243)Am,45 Sc+^(244)Pu,and^(46,48,50)Ti+237Np,51 V+^(238)U at some typical excitation energies. | Peng‑Hui Chen Hao Wu Zu‑Xing Yang Xiang‑Hua Zeng Zhao‑Qing Feng | 2023 | Nuclear Science and Techniques2023,34,1: | 0 |

| 20 | Single-cell RNA sequencing reveals the cell landscape of a radiation-induced liver injury mouse model显示文摘The mechanisms of radiation-induced liver injury(RILI)has not been fully elucidated so far.In the present study,a RILI mouse model was constructed by exposing the liver to a single dose of 30 Gy X-rays.Liver injuries consisting of liver function damage and histopathological variations were confirmed after 2 weeks.And then the cellular atlas of RILI liver was generated by profiling 9,641 cells isolated from X-ray irradiated mice livers and control ones from RILI mice model using single-cellRNAsequencing(scRNA-seq).Seven cell types were identified,including B cells,natural killer cells,T cells,macrophages/Küpffer cells/Dendritic cells(DC),neutrophils,endothelial cells,and hepatocyte.Although there was no significant difference of overall cell typing was observed between the Control and RILI groups,hepatocytes and macro/Küpffer/DC cell types were chosen for further functional exploration.Gene expression profiles and bioinformatics analysis of hepatocytes revealed that multiple metabolic related pathways were enriched in livers exposed to IR.These scRNA-seq data were confirmed in RILI liver samples via adipose staining.Besides,obviously varied M1-/M2-macrophages polarization was observed in RILI liver,which was in accordance with the enzyme linked immunosorbent assay(ELISA)results of IR-induced M2 to pro-inflammatory M1 macrophages transformation in mouse macrophage cell line Raw264.7.In addition,we predicted that several genes were found to differentially expressed during the process of macrophage polarization from M2 to M1 subtype.Overall,our study provides a cellular landscape of RILI at single-cell resolution that indicates the characteristics of hepatocytes and macrophages,which will contribute to investigate the novel therapeutic or preventive management for RILI. | Ying Xu Shuang Feng Qiliang Peng Wei Zhu Qidong Zu Xiaohua Yao Qi Zhang Jianping Cao Yang Jiao | 2021 | Radiation Medicine and Protection2021,2,4: | 0 |
