Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide. The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry. Both SARS-CoV-2 and SARS-CoV share this mechanism. Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies. The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however, the cross-neutralizing activity was much weaker, indicating that there are distinct antigenic features in the RBDs of the two viruses. This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall, our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable. | Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun | 2020 | Cellular & Molecular Immunology2020,17,6: | 13
Involvement of C2H2 zinc finger proteins in the regulation of epidermal cell fate determination in Arabidopsis显示文摘Cell fate determination is a basic developmental process during the growth of multicellular organisms.Trichomes and root hairs of Arabidopsis are both readily accessible structures originating from the epidermal cells of the aerial tissues and roots respectively, and they serve as excellent models for understanding the molecular mechanisms controlling cell fate determination and cell morphogenesis. The regulation of trichome and root hair formationis a complex program that consists of the integration of hormonal signals with a large number of transcriptional factors, including MYB and b HLH transcriptional factors.Studies during recent years have uncovered an important role of C2H2 type zinc finger proteins in the regulation of epidermal cell fate determination. Here in this minireview we briefly summarize the involvement of C2H2 zinc finger proteins in the control of trichome and root hair formation in Arabidopsis. | An Yan Minjie Wu Yongqin Zhao Aidong Zhang Bohan Liu John Schiefelbein Yinbo Gan | 2014 | Journal of Integrative Plant Biology2014,56,12: | 7
Corridor use by Asian elephants显示文摘There are 18 km of Kunming–Bangkok Highway passing through the Mengyang Nature Reserve of Xishuangbanna National Nature Reserve in Yunnan Province,China. From September 2005 to September 2006 the impact of this highway on movement of wild Asian elephants between the eastern and western part of the nature reserve was studied using track transecting, rural surveys and direct monitoring. Our results showed that the number of cross-road corridors used by Asian elephants diminished from 28 to 23 following the construction of the highway. In some areas, the elephant activity diminished or even disappeared, which indicated a change in their home ranges. The utilization rate of artificial corridors was 44%.We also found that elephants prefered artificial corridors that were placed along their original corridors.During the research, wild elephants revealed their adaptation to the highway. They were found walking across the highway road surface many times and for different reasons. We suggest that the highway management bureau should revise their management strategies to mitigate the potential risks caused by elephants on the road for the safety of the public and to protect this endangered species from harm.It is also very important to protect and maintain current Asian elephants corridors in this region. | Wenjing PAN Liu LIN Aidong LUO Li ZHANG | 2009 | Integrative Zoology2009,4,2: | 4
Simultaneous determination of ivabradine and N-desmethylivabradine in human plasma and urine using a LC-MS/MS method: application to a pharmacokinetic study显示文摘A sensitive and specific liquid-chromatography tandem mass spectrometry(LC-MS/MS)assay has been developed and validated for the simultaneous quantification of ivabradine and its active metabolite N-desmethylivabradine in human plasma and urine. The assay employed a single liquid–liquid extraction of the analytes from plasma and urine samples, and diazepam was used as internal standard(IS).The chromatographic separation was achieved on a Diamonsil C18 column(150 mm4.6 mm,5 mm,Dikma)using a mixture of methanol and aqueous 5 mM ammonium acetate buffer containing 0.2%formic acid(80:20,v/v)as mobile phase. The assay for ivabradine and N-desmethylivabradine in plasma showed good linearity(r≥0.99)over the ranges 0.1013–101.3 ng/mL and 0.085–25.5 ng/mL,respectively.The assay for ivabradine and N-desmethylivabradine in urine showed good linearity(r≥0.99)over the ranges 10.13–6078 ng/mL and 8.5–850 ng/mL,respectively.The intra-and inter-day accuracy and precision values were found to be within the assay variability limits(RSD<15%)in accordance with FDA guidelines. The methods were successfully used for evaluating the pharmacokinetic properties of ivabradine and N-desmethylivabradine in human plasma and urine in Chinese healthy volunteers. | Chengtao Lu Yanyan Jia Jing Yang Xin Jin Ying Songa Wenxing Liu Yi Ding Xiaoli Sun Aidong Wen | 2012 | Acta Pharmaceutica Sinica B2012,2,2: | 3
A novel visual attention method for target detection from SAR images显示文摘Synthetic Aperture Radar(SAR) imaging systems have been widely used in civil and military fields due to their all-weather and all-day abilities and various other advantages. However, due to image data exponentially increasing, there is a need for novel automatic target detection and recognition technologies. In recent years, the visual attention mechanism in the visual system has helped humans effectively deal with complex visual signals. In particular, biologically inspired top-down attention models have garnered much attention recently. This paper presents a visual attention model for SAR target detection, comprising a bottom-up stage and top-down process. In the bottom-up step, the Itti model is improved based on the difference between SAR and optical images. The top-down step fully utilizes prior information to further detect targets. Extensive detection experiments carried out on the benchmark Moving and Stationary Target Acquisition and Recognition(MSTAR) dataset show that, compared with typical visual models and other popular detection methods, our model has increased ability and robustness for SAR target detection, under a range of Signal to Clutter Ratio(SCR) conditions and scenes. In addition, results obtained using only the bottom-up stage are inferior to those of the proposed method, further demonstrating the effectiveness and rationality of a top-down strategy. In summary, our proposed visual attention method can be considered a potential benchmark resource for the SAR research community. | Fei GAO Aidong LIU Kai LIU Erfu YANG Amir HUSSAIN | 2019 | Chinese Journal of Aeronautics2019,32,8: | 3
Highly stretchable and sensitive strain sensor based on silver nanowires/carbon nanotubes on hair band for human motion detection显示文摘In this work,a highly stretchable and sensitive strain sensor using silver nanowires(Ag NWs)/multi-walled carbon nanotubes(MWCNTs)on hair band is proposed. The fabrication process of such sensors enabled scalable and facile production with low cost by dipping hair band into Ag NWs/MWCNTs composites suspension. The sensing performance of the sensors was optimized by tuning the Ag NWs/MWCNTs mass ratio. The hair band based strain sensor with Ag NWs/MWCNTs mass ratio of 30 exhibited the best performance with high sensitivity of 416.0 and good stretchability up to 70%.Moreover,great repeatability and long-term stability could be achieved over 7500 cycles of stretch-release.This Ag NWs/MWCNTs-hair band sensor can be used for monitoring human motions and swallowing action. | Yanqiang Gao Tiancheng Lai Furui Teng Ghang Liu Aidong Li | 2021 | Progress in Natural Science:Materials International2021,31,3: | 2
Safety and efficacy of meplazumab in healthy volunteers and COVID-19 patients: a randomized phase 1 and an exploratory phase 2 trial显示文摘Recent evidence suggests that CD147 serves as a novel receptor for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection.Blocking CD147 via anti-CD147 antibody could suppress the in vitro SARS-CoV-2 replication.Meplazumab is a humanized anti-CD147 IgG_(2) monoclonal antibody, which may effectively prevent SARS-CoV-2 infection in coronavirus disease 2019(COVID-19)patients.Here,we conducted a randomized,double-blinded,placebo-controlled phase 1 trial to evaluate the safety, tolerability, and pharmacokinetics of meplazumab in healthy subjects, and an open-labeled,concurrent controlled add-on exploratory phase 2 study to determine the efficacy in COVID-19 patients. In phase 1 study,59 subjects were enrolled and assigned to eight cohorts, and no serious treatment-emergent adverse event(TEAE)or TEAE grade≥3 was observed. The serum and peripheral blood Cmax and area under the curve showed non-linear pharmacokinetic characteristics. No obvious relation between the incidence or titer of positive anti-drug antibody and dosage was observed in each cohort. The biodistribution study indicated that meplazumab reached lung tissue and maintained>14 days stable with the lung tissue/cardiac blood-pool ratio ranging from 0.41 to 0.32.In the exploratory phase 2 study,17 COVID-19 patients were enrolled, and 11 hospitalized patients were involved as concurrent control. The meplazumab treatment significantly improved the discharged(P=0.005)and case severity(P=0.021),and reduced the time to virus negative(P=0.045)in comparison to the control group. These results show a sound safety and tolerance of meplazumab in healthy volunteers and suggest that meplazumab could accelerate the recovery of patients from COVID-19 pneumonia with a favorable safety profile. | Huijie Bian Zhao-Hui Zheng Ding Wei Aidong Wen Zheng Zhang Jian-Qi Lian Wen-Zhen Kang Chun-Qiu Hao Jing Wang Rong-Hua Xie Ke Dong Jie-Lai Xia Jin-Lin Miao Wen Kang Guoquan Li Di Zhang Mingru Zhang Xiu-Xuan Sun Likun Ding Kui Zhang Junfeng Jia Jin Ding Zhiqin Li Yanyan Jia Lin-Na Liu Zhe Zhang Zhao-Wei Gao Hong Du Na Yao Qing Wang Ke Wang Jie-Jie Geng Bin Wang Ting Guo Ruo Chen Yu-Meng Zhu Li-Juan Wang Qian He Rui-Rui Yao Ying Shi Xiang-Min Yang Jian-Sheng Zhou Yi-Nan Ma Ya-Tao Wang Xue Liang Fei Huo Zhe Wang Yang Zhang Xu Yang Ye Zhang Lu-Hua Gao Ling Wang Xiao-Chun Chen Hao Tang Shuang-Shuang Liu Qing-Yi Wang Zhi-Nan Chen Ping Zhu | 2021 | Signal Transduction and Targeted Therapy2021,6,6: | 2

A nuclear localized protein ZCCHC9 is expressed in cerebral cortex and suppresses the MAPK signal pathway显示文摘The CCHC-type zinc finger motif has numerous biological activities (such as DNA binding and RNA binding) and can also mediate protein-protein interaction. This article gives a primary report about the human ZCCHC9 gene. Protein ZCCHC9 contains four CCHC motifs and is highly conserved in humans, mice, and rats. The whole cDNA sequence of the ZCCHC9 gene has been amplified by PCR and a number of plasmids have been constructed for further study. The results show that ZCCHC9 is localized in the nucleus, and especially concentrated in the nucleolus. It is highly expressed in the brain and testicles of the mouse. This has been confirmed by real-time reverse transcription polymerase chain reaction (RT-PCR). In situ hybridization of the mouse brain indicates that ZCCHC9 is mainly expressed in the cerebral cortex. Reporter gene assay shows that ZCCHC9 suppresses the transcription activities of NF-kappa B and SRE, and may play roles in the Mitogen-Activated Protein Kinase (MAPK) signaling transduction pathway. | Aidong Zhou Jianlin Zhou Liping Yang Mingjun Liu Hong Li Su Xu Mei Han Jian Zhang | 2008 | Journal of Genetics and Genomics2008,35,8: | 1
Challenges in Atomic-Scale Characterization of High-k Dielectrics and Metal Gate Electrodes for Advanced CMOS Gate Stacks显示文摘The decreasing feature sizes in complementary metal-oxide semiconductor (CMOS) transistor technology will require the replacement of SiO2 with gate dielectrics that have a high dielectric constant (high-k) because as the SiO2 gate thickness is reduced below 1.4 nm, electron tunnelling effects and high leakage currents occur in SiO2, which present serious obstacles to future device reliability. In recent years significant progress has been made on the screening and selection of high-k gate dielectrics, understanding their physical properties, and their integration into CMOS technology. Now the family of hafnium oxide-based materials has emerged as the leading candidate for high-k gate dielectrics due to their excellent physical properties. It is also realized that the high-k oxides must be implemented in conjunction with metal gate electrodes to get suffcient potential for CMOS continue scaling. In the advanced nanoscale Si-based CMOS devices, the composition and thickness of interfacial layers in the gate stacks determine the critical performance of devices. Therefore, detailed atomic-scale understandings of the microstructures and interfacial structures built in the advanced CMOS gate stacks, are highly required. In this paper, several high-resolution electron, ion, and photon-based techniques currently used to characterize the high-k gate dielectrics and interfaces at atomic-scale, are reviewed. Particularly, we critically review the research progress on the characterization of interface behavior and structural evolution in the high-k gate dielectrics by high-resolution transmission electron microscopy (HRTEM) and the related techniques based on scanning transmission electron microscopy (STEM), including high-angle annular dark-field (HAADF) imaging (also known as Z-contrast imaging), electron energy-loss spectroscopy (EELS), and energy dispersive X-ray spectroscopy (EDS), due to that HRTEM and STEM have become essential metrology tools for characterizing the dielectric gate stacks in the present and future generations of CMOS devices. In Section 1 of this review, the working principles of each technique are brieffy introduced and their key features are outlined. In Section 2, microstructural characterizations of high-k gate dielectrics at atomic-scale by electron microscopy are critically reviewed by citing some recent results reported on high-k gate dielectrics. In Section 3, metal gate electrodes and the interfacial structures between high-k dielectrics and metal gates are discussed. The electron beam damage effects in high-k gate stacks are also evaluated, and their origins and prevention are described in Section 4. Finally, we end this review with personal perspectives towards the future challenges of atomic-scale material characterization in advanced CMOS gate stacks. | Xinhua Zhu Jian-min Zhu Aidong Li Zhiguo Liu Naiben Ming | 2009 | Journal of Materials Science & Technology2009,25,3: | 1
Sulfur vs. tellurium: the heteroatom effects on the nonfullerene acceptors显示文摘The effect of chalcogen heteroatom variation on donor materials has been systematically investigated. However, this effect on acceptors has rarely been explored. Herein, nonfullerene acceptors BFPSP and BFPTP were reported by simply changing the chalcogen atoms from S to Te. The differences between BFPSP and BFPTP in light absorption, energy levels, excited-state lifetimes, energy loss, charge mobilities, morphology, and photovoltaic properties were systematically investigated to understand the heteroatom effects. More importantly, the electroluminescence spectra, external quantum efficiency of photovoltaics and TDDFTcalculations revealed that the triplet excited state(T1) in energy of BFPTP equals to the charge transfer(CT) state in PBDBT:BFPTP, which allows T1 excitons, generated by intersystem crossing, to split into free charges to contribute to the efficiency. This contribution provides a strategy for tuning the photophysical properties of nonfullerene acceptors and designing high performance triplet materials for OSCs. | Lei Yang Linqing Qin Yunxiao Xu Huotian Zhang Lei Lv Kepeng Chen Xinyu Sui Yangguang Zhong Yuan Guo Feng Gao Jianzhang Zhao Yuhao Li Xinfeng Liu Yuanping Yi Xinhui Lu Aidong Peng Hui Huang | 2019 | Science China Chemistry2019,62,7: | 1

To Construct High Level Secure Communication System: CTMI Is Not Enough显示文摘Public key cryptographic(PKC) algorithms, such as the RSA, elliptic curve digital signature algorithm(ECDSA) etc., are widely used in the secure communication systems, such as OpenSSL, and a variety of information security systems. If designer do not securely implement them, the secret key will be easily extracted by side-channel attacks(SCAs) or combinational SCA thus mitigating the security of the entire communication system. Previous countermeasures of PKC implementations focused on the core part of the algorithms and ignored the modular inversion which is widely used in various PKC schemes. Many researchers believe that instead of straightforward implementation, constant time modular inversion(CTMI) is enough to resist the attack of simple power analysis combined with lattice analysis. However, we find that the CTMI security can be reduced to a hidden t-bit multiplier problem. Based on this feature, we firstly obtain Hamming weight of intermediate data through side-channel leakage. Then, we propose a heuristic algorithm to solve the problem by revealing the secret(partial and full) base of CTMI. Comparing previous nec-essary input message for masking filtering, our procedure need not any information about the secret base of the inversion. To our knowledge, this is the first time for evaluating the practical security of CTMI and experimental results show the fact that CTMI is not enough for high-level secure communication systems. | Sen Xu Xiangjun Lu Aidong Chen Haifeng Zhang Haihua Gu Dawu Gu Kaiyu Zhang Zheng Guo Junrong Liu | 2018 | China Communications2018,15,11: | 0
