1Anti-disturbance control of hypersonic flight vehicles with input saturation using disturbance observer显示文摘This paper proposes an anti-disturbance control scheme for the near space vehicle(NSV) based on terminal sliding mode(TSM) technique and disturbance observer method. To tackle the system uncertainty and the time-varying unknown external disturbance of the NSV, a disturbance observer based on TSM technique is designed which can render the disturbance estimate error convergent in finite time. Furthermore, an auxiliary design system is introduced to analyze the input saturation effect. Based on the developed disturbance observer and the auxiliary design system, an anti-disturbance attitude control scheme is developed for the NSV using the TSM technique to speed up the convergence of all signals in closed-loop system. For the closed-loop system,the stability is rigorously proved by using the Lyapunov method and we guarantee the finite time convergence of all closed-loop system signals in the presence of the integrated affection of the system uncertainty, the input saturation, and the unknown external disturbance. 2FGF1^(△HBS) prevents diabetic cardiomyopathy by maintaining mitochondrial homeostasis and reducing oxidative stress via AMPK/Nur77 suppression显示文摘As a classically known mitogen,fibroblast growth factor 1(FGF1)has been found to exert other pleiotropic functions such as metabolic regulation and myocardial protection.Here,we show that serum levels of FGF1 were decreased and positively correlated with fraction shortening in diabetic cardiomyopathy(DCM)patients,indicating that FGF1 is a potential therapeutic target for DCM.We found that treatment with a FGF1 variant(FGF1^(△HBS))with reduced proliferative potency prevented diabetes-induced cardiac injury and remodeling and restored cardiac function.RNA-Seq results obtained from the cardiac tissues of db/db mice showed significant increase in the expression levels of anti-oxidative genes and decrease of Nur77 by FGF1AHBS treatment.Both in vivo and in vitro studies indicate that FGF1^(△HBS) exerted these beneficial effects by markedly reducing mitochondrial fragmentation,reactive oxygen species(ROS)generation and cytochrome c leakage and enhancing mitochondrial respiration rate and β-oxidation in a 5;AMP-activated protein kinase(AMPK)/Nur77-dependent manner,all of which were not observed in the AMPK null mice.The favorable metabolic activity and reduced proliferative properties of FGF1^(△HBS) testify to its promising potential for use in the treatment of DCM and other metabolic disorders.Dezhong Wang Yuan Yin Shuyi Wang Tianyang Zhao Fanghua Gong Yushuo Zhao Beibei Wang Yuli Huang Zizhao Cheng Guanghui Zhu Zengshou Wang Yang Wang Jun Ren Guang Liang Xiaokun Li Zhifeng Huang 2021Signal Transduction and Targeted Therapy2021,6,4:6
3Unraveling a genetic roadmap for improved taste in the domesticated apple显示文摘Although taste is an important aspect of fruit quality, an understanding of its genetic control remains elusive in apple and other fruit crops. In this study, we conducted genomic sequence analysis of 497 Malus accessions and revealed erosion of genetic diversity caused by apple breeding and possible independent domestication events of dessert and cider apples. Signatures of selection for fruit acidity and size, but not for fruit sugar content, were detected during the processes of both domestication and improvement. Furthermore, we found that single mutations in major genes affecting fruit taste, including Ma1, MdTDT, and MdSOT2, dramatically decrease malate, citrate, and sorbitol accumulation, respectively, and correspond to important domestication events. Interestingly, Ma1 was identified to have pleiotropic effects on both organic acid content and sugar:acid ratio, suggesting that it plays a vital role in determining fruit taste. Fruit taste is unlikely to have been negatively affected by linkage drag associated with selection for larger fruit that resulted from the pyramiding of multiple genes with minor effects on fruit size. 14Hierarchical loading of CuO on SiO_(2) aerogel@high crystalline TiO_(2) nanofibers for efficiently photocatalytic reduction of CO_(2) without sacrificial agent显示文摘Silica aerogel was prepared by a sol-gel method with combination of freeze drying.The aerogel was filled with TiCl4 in autoclave and used to fabricate a hierarchical structure of TiO_(2) nanofiber shell and SiO_(2) aerogel core(SiO_(2)@TiO_(2)).The TiO_(2) nanofibers with a diameter of 10-15 nm were highly crystalline and mainly grew along the(101)or(001)planes,favoring charge migration along the growth axis of the fibers.The photoluminescence(PL)emission spectra show that the TiO_(2) nanofibers exhibited much lower PL intensity than P25.The free standing TiO2 nanofibers loaded with CuO had a band gap of 3.04 eV.When CuO was hierarchically loaded on the nanofiber surface and into the aerogel core(SiO_(2)/CuO@TiO_(2)/CuO),the absorption edge significantly red shifted,and the band gap was further narrowed to 2.66 eV.Meanwhile,Fe^(3+)implanted TiO_(2) nanofibers on the aerogel surface(SiO_(2)@Fe-TiO_(2))were also fabricated in the same strategy.The CuO loaded nanofibers(SiO_(2)/CuO@Fe-TiO_(2)/CuO)had a band gap of 2.62 eV.The photocatalytic reduction of CO_(2) was performed under light irradiation by a 300 W Xe-lamp for 4 h.The methanol yield over the SiO_(2)/CuO@Fe-TiO_(2)/CuO reached~2,400 μmol·gcat^(-1) in the absence of sacrificial agent.Man Wen Beibei Ren Xiangzhi Ye Jianxiong He Hong Jiang Chunrong Xiong 2022Nano Research2022,15,5:0
15Generation and metabolomic characterization of functional ductal organoids with biliary tree networks in decellularized liver scaffolds显示文摘Developing functional ductal organoids(FDOs)is essential for liver regenerative medicine.We aimed to construct FDOs with biliary tree networks in rat decellularized liver scaffolds(DLSs)with primary cholangiocytes isolated from mouse bile ducts.The developed FDOs were dynamically characterized by functional assays and metabolomics for bioprocess clarification.FDOs were reconstructed in DLSs retaining native structure and bioactive factors with mouse primary cholangiocytes expressing enriched biomarkers.Morphological assessment showed that biliary tree-like structures gradually formed from day 3 to day 14.The cholangiocytes in FDOs maintained high viability and expressed 11 specific biomarkers.Basal-apical polarity was observed at day 14 with immunostaining for E-cadherin and acetylatedα-tubulin.The rhodamine 123 transport assay and active collection of cholyl-lysyl-fluorescein exhibited the specific functions of bile secretion and transportation at day 14 compared to those in monolayer and hydrogel culture systems.The metabolomics analysis with 1075 peak pairs showed that serotonin,as a key molecule of the tryptophan metabolism pathway linked to biliary tree reconstruction,was specifically expressed in FDOs during the whole period of culture.Such FDOs with biliary tree networks and serotonin expression may be applied for disease modeling and drug screening,which paves the way for future clinical therapeutic applications.Jiaxian Chen Shiwen Ma Hui Yang Xi Liang Heng Yao Beibei Guo Deying Chen Jing Jiang Dongyan Shi Jiaojiao Xin Keke Ren Xingping Zhou Yun Li Lei Geng Jun Li 2023Bioactive Materials2023,,8:0
16SOX2 heterozygous mutations cause multiple extraocular phenotypes in boys显示文摘To the Editor:The SRY-related high-mobility-group-box protein-2(SOX2)is most notably expressed in the eye,placodes,forebrain,and hypothalamus-pituitary and is involved in early embryonic development.[1]Loss-of-function mutations or deletions in SOX2 could lead to uni-or bi-lateral anophthalmia/microphthalmia(A/M)as well as other related disorders,such as anophthalmia/esophageal-genital syndrome.An increasing number of studies have found that SOX2 mutations can cause variable extraocular symptoms,including growth retardation,sensorineural hearing loss,mental retardation,no pubertal signs,and male genitourinary tract malformations(micropenis,cryptorchidism,and hypospadias).Indeed,cases with SOX2 pathogenic mutations but no or minor ocular symptoms have been reported less frequently.Several studies found that SOX2 heterozygous mutations cause typical signs of complete hypogonadism without major ocular malformations in men or women,such as isolated hypogonadotropic hypogonadism(HH),but no other HH pathogenic gene was identified.[2,3]Therefore,our study is the first to report the cases of three Chinese patients with SOX2 mutations referred due to micropenis and/or cryptorchidism combined with craniofacial deformities or intellectual disability.Yi Wang Lijun Fan Xiaoya Ren Yanning Song Beibei Zhang Chunxiu Gong 2022Chinese Medical Journal2022,,4:0

18High Strength and Excellent Ductility of AZ80 Magnesium Alloy Cabin Component Developed by W-Shaped Channel Extrusion and Subsequent T6 Heat Treatment显示文摘The AZ80 magnesium(Mg)alloy cabin component with high strength and excellent ductility was developed by W-shaped channel extrusion(WCE)at 350℃ and subsequent T6 heat treatment.The effect of WCE process on the microstructure and mechanical properties of the alloy was experimentally investigated,and the age-hardening behavior with microstructure evolution during heat treatment was revealed.Due to the introduction of multi-stage asymmetric extrusion and severe shear deformation along the annular channel,the average grain size of the WCE extruded alloy could be effectively refined to 4.7μm.Besides,theβphase particles were dynamically precipitated from the fine grain boundaries during extrusion,which hindered the grain growth,but worsen the material plasticity.After T6 treatment,the properties of component were eventually improved to a yield strength(YS)of 218 MPa and ultimate tensile strength(UTS)of 344 MPa with elongation(EL)of 14.5%.It was revealed that the rod/lath-and needle-shaped continuousβphase(CP)with finer size precipitated after T6 treatment was more effective in hindering the movement of dislocations and strengthened the alloy than lamellar discontinuousβphase(DP).The dispersed phase precipitated in the grains,the annihilation of dislocations,the uniformly distributed grains and the re-dissolution ofβphase particles at initial grain boundaries after T6 treatment greatly contributed to the ductility of alloy.Moreover,the T6 treatment also promoted the basal plane of most grains which were re-arranged to the extrusion direction,which promoted the possibility of non-basal slip activation and further improved the elongation of the alloy.As a result,the UTS and YS of the final component increased by 10%and the EL increased by 7%,respectively.Yuxing Zhang Zhen Wang Shuchang Li Xi Zhao Zhimin Zhang Yaojin Wu Xianwei Ren Fafa Yan Beibei Dong 2023Acta Metallurgica Sinica(English Letters)2023,36,5:0
