1Morphological study on colonic pathology in ulcerative colitis treated by moxibustion显示文摘AIM To observe the therapeutic effect ofmoxibustion on ulcerative colitis and itsinfluence on the colonic mucosal morphology.METHODS Forty-six patients with ulcerativecolitis were randomly divided into themoxibustion with herbal medicine underneathgroup and the western medicine group.Thirtypatients were treated with the abovemoxibustion and 16 patients with Salicylayefapyridine(SASP).The colonic mucosa of 13patients in the moxibustion group was observedby colonoscopy before and after the treatment.Mucin was also analyzed by H.E and AB-PASstaining.RESULTS Seventeen patients were clinicallycured,12 were improved and 1 unchanged in themoxibustion group.In the control group,5patients were clinically cured,7 improved and 4unchanged.Thirteen patients with active UCwere taken as the subjects for histopathologicanalysis in this study.The colonic mucosallesions were remarkably improved and thecharacteristic of the mucin also changed.Inmost sections,the chronic inflammation ofmucosa was geatly ameliorated(P<0.01).Theinflammatory cell infiltratation much decreased and neutrophils,disapeared in most sections(P<0.001).The goblet cells significantlyincreased(P<0.001);crypt paracrypt abscessor mucosal ulceration was seen(P<0.001).CONCLUSION The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. 2Soil Erosion Changes over the Past Five Decades in the Red Soil Region of Southern China显示文摘This paper reports the dynamic changes of soil and water loss in the red soil region of Southern China since the 1950s. The red soil region covers eight provinces: Jiangxi, Zhejiang, Fujian, Anhui, Hubei, Hunan, Guangdong and Hainan. From the 1950s to 1986, the annual rate of soil erosion increased by 3.4%. From 1986 to 1996 and from 1996 to 2000, the annual rates of soil erosion decreased by 2.0% and 0.32%, respectively. Field surveys showed that from 2000 to 2005, the area of soil and water loss decreased annually by 1.2%. This decrease was a result of large-scale erosion control activities across China. Although the eroded soil has been restored, the restoration process is very slow and full restoration will take a long time. 3Recent advances in nano scaffolds for bone repair显示文摘Biomedical applications of nanomaterials are exponentially increasing every year due to analogy to various cell receptors, ligands, structural proteins, and genetic materials(that is, DNA). In bone tissue, nanoscale materials can provide scaffold for excellent tissue repair via mechanical stimulation, releasing of various loaded drugs and mediators, 3D scaffold for cell growth and differentiation of bone marrow stem cells to osteocytes. 4Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. 5Photoperiodism dynamics during the domestication and improvement of soybean显示文摘Soybean(Glycine max) is a facultative short-day plant with a sensitive photoperiod perception and reaction system, which allows it to adjust its physiological state and gene regulatory networks to seasonal and diurnal changes in environmental conditions. In the past few decades, soybean cultivation has spread from East Asia to areas throughout the world. Biologists and breeders must now confront the challenge of understanding the molecular mechanism of soybean photoperiodism and improving agronomic traits to enable this important crop to adapt to geographical and environmental changes. In this review, we summarize the genetic regulatory network underlying photoperiodic responses in soybean. Genomic and genetic studies have revealed that the circadian clock, in conjunction with the light perception pathways, regulates photoperiodic flowering. Here, we provide an annotated list of 844 candidate flowering genes in soybean, with their putative biological functions. Many photoperiod-related genes have been intensively selected during domestication and crop improvement. 8Photodegradation of 17α-ethynylestradiol in dissolved humic substances solution:Kinetics,mechanism and estrogenicity variation显示文摘17α-Ethynylestradiol(EE2) in natural waters may cause adverse effects on organisms due to its high estrogenic potency. Laboratory studies were performed to study the effects of a local humic acid(LHA), fulvic acid(LFA) and Aldrich humic acid(AHA) on the photochemical behavior and estrogenic potency of EE2. Here photolytic experiments demonstrated that pure aqueous EE2 could undergo direct and self-sensitized photodegradation at a global rate of 0.0068 hr^(-1).Photodegradation rate of EE2 in 5.0 mg/L dissolved humic substances(DHS) was determined to be 0.0274, 0.0296 and 0.0254 hr^(-1) for LHA, LFA and AHA, respectively. Reactive oxygen species(ROS) and triplet dissolved humic substances(~3DHS*) scavenging experiments indicated that the promotion effect of DHS on EE2 photodegradation was mainly aroused by the reactions of HOU(35%–50%),~1O_2(<10%) andDHS*(22%–34%). However, the photodegradation of EE2 could also be inhibited when DHS exceeded the threshold of 10 mg/L. Three hydroxylation products of EE2 were identified using GC–MS and their formation pathways were also proposed. In vitro estrogenicity tests showed that EE2 was transformed into chemicals without estrogenic potency. 9The Hippo pathway regulates stem cell proliferation,self-renewal,and differentiation显示文摘Stem cells and progenitor cells are the cells of origin for multi-cellular organisms and organs.They play key roles during development and their dysregulation gives rise to human diseases such as cancer.The recent de-velopment of induced pluripotent stem cell(iPSC)technology which converts somatic cells to stem-like cells holds great promise for regenerative medicine.Nevertheless,the understanding of proliferation,dif-ferentiation,and self-renewal of stem cells and or-gan-specific progenitor cells is far from clear.Recently,the Hippo pathway was demonstrated to play important roles in these processes.The Hippo pathway is a newly established signaling pathway with critical functions in limiting organ size and suppressing tumorigenesis.This pathway was first found to inhibit cell proliferation and promote apoptosis,therefore regulating cell num-ber and organ size in both Drosophila and mammals.However,in several organs,disturbance of the pathway leads to specific expansion of the progenitor cell com-partment and manipulation of the pathway in embryonic stem cells strongly affects their self-renewal and dif-ferentiation.In this review,we summarize current ob-servations on roles of the Hippo pathway in different types of stem cells and discuss how these findings changed our view on the Hippo pathway in organ de-velopment and tumorigenesis.Huan Liu Dandan Jiang Fangtao Chi Bin Zhao 2012Protein & Cell2012,3,4:8
10Effect of Water Level Fluctuations on Wintering Goose Abundance in Poyang Lake Wetlands of China显示文摘The Poyang Lake is a Ramsar site and is the important over-wintering site for migratory waterbirds along the East Asian–Australasian Fly way. Examining the effects of water level fluctuations on waterbird abundance and analyzing the influencing mechanism is critical to waterbird protection in the context of hydrological alteration. In this study, the effect of water level regime on wintering goose abundance was examined and the influencing mechanism was interpreted. Synchronous waterbirds survey data, hydrological data, Moderate Resolution Imaging Spectroradiometer-Normalized Difference Vegetation Index(MODIS-NDVI) data and habitat data derived from Landsat TM/ETM data and HJ/CCD data were combined. The satellite-derived Green Wave Index(GWI) based on MODIS-NDVI dataset was applied to detect changes in goose food resources. It was found that habitat size and vegetation conditions are key factors determining goose abundance. Geese numbers were positively correlated with habitat area, while intermediate range of vegetation productivity might benefit the goose abundance. Water level affects goose abundance by changing available habitat areas and vegetation conditions. We suggested that matching hydrological regime and exposed meadows time to wintering geese dynamics was crucial in the Poyang Lake wetlands. 11Low WT1 transcript levels at diagnosis predicted poor outcomes of acute myeloid leukemia patients with t(8;21) who received chemotherapy or allogeneic hematopoietic stem cell transplantation显示文摘Background:Acute myeloid leukemia(AML) with t(8;21) is a heterogeneous disease.Identifying AML patients with t(8;21) who have a poor prognosis despite achieving remission is important for determining the best subsequent therapy.This study aimed to evaluate the impact of Wilm tumor gene-1(WT1) transcript levels and cellular homolog of the viral oncogene v-KIT receptor tyrosine kinase(C-KIT) mutations at diagnosis,and RUNXTRUNX1T1 transcript levels after the second consolidation chemotherapy cycle on outcomes.Methods:Eighty-eight AML patients with t(8;21) who received chemotherapy only or allogeneic hematopoietic stem cell transplantation(allo-HSCT) were included.Patients who achieved remission,received two or more cycles of consolidation chemotherapy,and had a positive measureable residual disease(MRD) test result(defined as <3-log reduction in RUNX1-RUNX1T1 transcript levels compared to baseline) after 2-8 cycles of consolidation chemotherapy were recommended to receive allo-HSCT.Patients who had a negative MRD test result were recommended to receive further chemotherapy up to only 8 cycles.WT1 transcript levels and C-KIT mutations at diagnosis,and RUNX1-RUNX1T1 transcript levels after the second consolidation chemotherapy cycle were tested.Results:Patients who had a C-KIT mutation had significantly lower WTl transcript levels than patients who did not have a C-KIT mutation(6.7%± 10.6%vs.19.5%± 19.9%,P < 0.001).Low WTl transcript levels(<5.0%) but not C-KIT mutation at diagnosis,a positive MRD test result after the second cycle of consolidation chemotherapy,and receiving only chemotherapy were independently associated with high cumulative incidence of relapse in all patients(hazard ratio[HR]= 3.53,2.30,and 11.49;95%confidence interval[CI]1.64-7.62,1.82-7.56,and 4.43-29.82;P = 0.002,0.034,and <0.001,respectively);these conditions were also independently associated with low leukemia-free survival(HR =3.71,2.33,and 5.85;95%CI 1.82-7.56,1.17-4.64,and 2.75-12.44;P < 0.001,0.016,and <0.001,respectively) and overall survival(HR = 3.50,2.32,and 4.34;95%CI 1.56-7.82,1.09-4.97,and 1.98-9.53;P = 0.002,0.030,and <0.001,respectively) in all patients.Conclusions:Testing for WTl transcript levels at diagnosis in patients with AML and t(8;21) may predict outcomes in those who achieve remission.A randomized study is warranted to determine whether allo-HSCT can improve prognosis in these patients.Ya-Zhen Qin Yu Wang Hong-Hu Zhu Robert Peter Gale Mei-Jie Zhang Qian Jiang Hao Jiang Lan-Ping Xu Huan Chen Xiao-Hui Zhang Yan-Rong Liu Yue-Yun Lai Bin Jiang Kai-Yan Liu Xiao-Jun Huang 2016Chinese Journal of Cancer2016,35,7:7
12Substrate stiffness modulates bone marrow-derived macrophage polarization through NF-κB signaling pathway显示文摘The stiffness of the extracellular matrix(ECM)plays an important role in regulating the cellular programming.However,the mechanical characteristics of ECM affecting cell differentiation are still under investigated.Herein,we aimed to study the effect of ECM substrate stiffness on macrophage polarization.We prepared polyacrylamide hydrogels with different substrate stiffness,respectively.After the hydrogels were confirmed to have a good biocompatibility,the bone marrow-derived macrophages(BMMs)from mice were incubated on the hydrogels.With simulated by the low substrate stiffness,BMMs displayed an enhanced expression of CD86 on the cell surface and production of reactive oxygen species(ROS)in cells,and secreted more IL-1βand TNF-αin the supernatant.On the contrary,stressed by the medium stiffness,BMMs expressed more CD206,produced less ROS,and secreted more IL-4 and TGF-β.In vivo study by delivered the hydrogels subcutaneously in mice,more CD68+CD86+cells around the hydrogels with the low substrate stiffness were observed while more CD68+CD206+cells near by the middle stiffness hydrogels.In addition,the expressions of NIK,phosphorylated p65(pi-p65)and phosphorylated IκB(pi-IκB)were significantly increased after stimulation with low stiffness in BMMs.Taken together,these findings demonstrated that substrate stiffness could affect macrophages polarization.Low substrate stiffness promoted BMMs to shift to classically activated macrophages(M1)and the middle one to alternatively activated macrophages(M2),through modulating ROS-initiated NF-κB pathway.Therefore,we anticipated ECM-based substrate stiffness with immune modulation would be under consideration in the clinical applications if necessary.Mimi Chen Yu Zhang Pinghui Zhou Xingzhi Liu Huan Zhao Xichao Zhou Qiaoli Gu Bin Li Xuesong Zhu Qin Shi 2020Bioactive Materials2020,5,4:7

14The treatment of black-odorous water using tower bipolar electro-flocculation including the removal of phosphorus,turbidity,sulfion,and oxygen enrichment显示文摘The large amount of municipal wastewater discharged into urban rivers sometimes exceeds the rivers’self-purification capacity leading to black-odorous polluted water.Electro-flocculation has emerged as a powerful remediation technology.Electro-flocculation in a bubble column tower with a bipolar electrode(BPE)was tested in an attempt to overcome the high resistance and weak gas-floatation observed with a monopolar electrode(MPE)in treating such water.The BPE reactor tested had a Ti/Ta2O_(5)-IrO_(2)anode and a graphite cathode with an iron or aluminum bipolar electrode suspended between them.It was tested for its ability to reduce turbidity,phosphate and sulphion and to increase the concentration of dissolved oxygen.The inclusion of the bipolar electrode was found to distinctly improved the system’s conductivity.The system’s electro-flocculation and electrical floatation removed turbidity,phosphate and sulphion completely,and the dissolved oxygen level improved from 0.29 to 6.28 mg/L.An aluminum bipolar electrode performed better than an iron one.Changes in the structure of the microbial community confirmed a significant improvement in water quality.Huan He Qinjin Yu Chaochao Lai Chen Zhang Muhan Liu Bin Huang Hongping Pu Xuejun Pan 2021Frontiers of Environmental Science & Engineering2021,15,2:5
15Dose optimization of intrathecal administration of human umbilical cord mesenchymal stem cells for the treatment of subacute incomplete spinal cord injury显示文摘Human umbilical cord mesenchymal stem cells(hUC-MSCs)are a promising candidate for spinal cord injury(SCI)repair owing to their advantages of low immunogenicity and easy accessibility over other MSC sources.However,modest clinical efficacy hampered the progression of these cells to clinical translation.This discrepancy may be due to many variables,such as cell source,timing of implantation,route of administration,and relevant efficacious cell dose,which are critical factors that affect the efficacy of treatment of patients with SCI.Previously,we have evaluated the safety and efficacy of 4×10^(6) hUC-MSCs/kg in the treatment of subacute SCI by intrathecal implantation in rat models.To search for a more accurate dose range for clinical translation,we compared the effects of three different doses of hUC-MSCs-low(0.25×10^(6) cells/kg),medium(1×10^(6) cells/kg)and high(4×10^(6) cells/kg)-on subacute SCI repair through an elaborate combination of behavioral analyses,anatomical analyses,magnetic resonance imaging-diffusion tensor imaging(MRI-DTI),biotinylated dextran amine(BDA)tracing,electrophysiology,and quantification of mRNA levels of ion channels and neurotransmitter receptors.Our study demonstrated that the medium dose,but not the low dose,is as efficient as the high dose in producing the desired therapeutic outcomes.Furthermore,partial restoration of theγ-aminobutyric acid type A(GABAA)receptor expression by the effective doses indicates that GABAA receptors are possible candidates for therapeutic targeting of dormant relay pathways in injured spinal cord.Overall,this study revealed that intrathecal implantation of 1×10^(6) hUC-MSCs/kg is an alternative approach for treating subacute SCI.Ting-Ting Cao Huan Chen Mao Pang Si-Si Xu Hui-Quan Wen Bin Liu Li-Min Rong Mang-Mang Li 2022Neural Regeneration Research2022,17,8:5
16MOF-Derived ZnS Nanodots/Ti_(3)C_(2)T_(x) MXene Hybrids Boosting Superior Lithium Storage Performance显示文摘ZnS has great potentials as an anode for lithium storage because of its high theoretical capacity and resource abundance;however,the large volume expansion accompanied with structural collapse and low conductivity of ZnS cause severe capacity fading and inferior rate capability during lithium storage. Herein,0D-2 D ZnS nanodots/Ti_(3)C_(2)T_x MXene hybrids are prepared by anchoring ZnS nanodots on Ti_(3)C_(2)T_(x) MXene nanosheets through coordination modulation between MXene and MOF precursor(ZIF-8) followed with sulfidation. The MXene substratecoupled with the ZnS nanodots can synergistically accommodate volume variation of ZnS over charge–discharge to realize stable cyclability. As revealed by XPS characterizations and DFT calculations,the strong interfacial interaction between ZnS nanodots and MXene nanosheets can boost fast electron/lithium-ion transfer to achieve excellent electrochemical activity and kinetics for lithium storage. Thereby,the as-prepared ZnS nanodots/MXene hybrid exhibits a high capacity of 726.8 mAh g^(-1) at 30 mA g^(-1),superior cyclic stability(462.8 mAh g^(-1) after 1000 cycles at 0.5 A g^(-1)),and excellent rate performance. 17Increased early activation of CD56^(dim)CD16^(dim)/-natural killer cells in immunological non-responders correlates with CD4^(+)T-cell recovery显示文摘Background:Natural killer(NK)cells play a critical role in suppressing human immunodeficiency virus-1(HIV-1)infection,but knowledge on whether and how NK cells affect immune reconstitution in HIV-1-infected individuals who receive antiretroviral therapy(ART)is limited.Methods:We performed a case-control study with 35 healthy individuals and 66 HIV-1-infected patients including 32 immunological non-responders(INRs)with poor CD4+T-cell recovery(<500 cells/μL after 4 years of ART)and 34 immunological responders(IRs)with improved CD4+T-cell recovery(>500 cells/μL after 4 years of ART).NK cell phenotype,receptor repertoire,and early activation in INRs and IRs were investigated by flow cytometry.Results:A significantly higher proportion of CD56dimCD16dim/-NK cells was observed in INRs than IRs before ART and after 4 years of ART.The number of CD56dimCD16dim/-NK cells was inversely correlated with CD4+T-cell counts in INRs before ART(r=-0.344,P=0.050).The more CD69-expressing NK cells there were,the lower the CD4+T-cell counts andΔCD4,and these correlations were observed in INRs after ART(r=-0.416,P=0.019;r=-0.509,P=0.003,respectively).Additionally,CD69-expressing CD56dimCD16dim/-NK cells were more abundant in INRs than those in IRs(P=0.018)after ART,both of which had an inverse association trend towards significance with CD4+T-cell counts.The expression of the activating receptors NKG2C,NKG2D,and NKp46 on CD56dimCD16dim/-NK cell subsets were higher in IRs than that in INRs after 4 years of ART(all P<0.01).Strong inverse correlations were observed between CD69 expression and NKG2C,NKG2A-NKG2C+,NKG2D,and NKp46 expression on CD56dimCD16dim/-NK cells in INRs after ART(NKG2C:r=-0.491,P=0.004;NKG2A-NKG2C+:r=-0.434,P=0.013;NKG2D:r=-0.405,P=0.021;NKp46:r=-0.457,P=0.008,respectively).Conclusions:INRs had a larger number of CD56dimCD16dim/-NK cells characterized by higher activation levels than did IRs after ART.The increase in the CD56dimCD16dim/-NK cell subset may play an adverse role in immune reconstitution.Further functional studies of CD56dimCD16dim/-NK cells in INRs are urgently needed to inform targeted interventions to optimize immune recovery.Qiu-Yue Zhang Xin Zhang Bin Su Li-Feng Liu Xiao-Dong Yang Bin Tang Huan Xia Ping Ma Tong Zhang Hao Wu 2020Chinese Medical Journal2020,,24:3
18Identification and functional analysis of heterogeneous FOXP3^+ Treg cell subpopulations in human pancreatic ductal adenocarcinoma显示文摘CD4^+CD25^+regulatory T(Treg) cells express the transcription factor FOXP3 and play an essential role in preventing autoimmunity. Abundant Treg cell accumulation in tumors and tumor draining lymph nodes(TDLNs) has been reported to correlate with both poor and favorable prognosis in various cancers, which suggests that Tregs may have multiple effects on antitumor immunity. However, the heterogeneity of tumor^-and TDLN^-infiltrating Treg cells remains unclear. Here we provide heterogeneity analysis of tumor infiltrating human CD4^+Treg cells and their matched adjacent tissues and TDLNs. We defined three different subpopulations of tumor^-and TDLN^-infiltrating Treg cells by Helios and CCR8 expression in pancreatic ductal adenocarcinoma(PDAC) and confirmed their functional heterogeneity. Helios^+CCR8^+Treg cells with potent suppressor function and limited IL^-2 and IFN^-c secretion were identified in tumors and TDLNs. On the contrary, Helios^-CCR8^-Treg cells have impaired suppressive activity, and elevated expression of pro^-inflammatory cytokines. More advanced grades of PDAC have predominantly Helios^+CCR8^+Treg cells and few Helios^-CCR8^-Treg cells both in tumors and TDLNs that suggests poor prognosis. 19Study on the Influence of Airgun Excitation Conditions on Airgun Signals and Travel Time Variation Measurements显示文摘Because of the different excitation conditions of the airgun source,there will be subtle differences in airgun signals.Travel time variation of airgun signals often mix into source information which can’t fully reflect the evolution of the medium.This article uses the airgun signals from the Binchuan Transmitting Seismic Station to analyze the airgun signal′s characteristics of phase and correlation.We conducted a comparative analysis of the effects of the pair difference method and the deconvolution method on eliminating the influence of the excitation conditions in travel time variation.The results show that:(1)The pressure pulse and its subsequent wave of airgun source wavelet are less affected by excitation conditions that we can use it to obtain high-precision excitation moments.(2)Deconvolution can improve the correlation of the airgun signal.(3)The pair difference method can’t eliminate the influence of excitation conditions in travel time variation.Deconvolution can reduce excitation condition interference and the influence of the excitation condition in the travel time variation after deconvolution of the vertical signal is significantly reduced.XIANG Ya YANG Runhai WANG Bin ZHOU Yunyao LU Yongqing WU Huan QI Junwei PENG Jing 2019Earthquake Research in China2019,33,2:3
