1Pediatric reference intervals in China(PRINCE):design and rationale for a large,multicenter collaborative cross-sectional study显示文摘There is a lack of accurate pediatric reference intervals(RIs) in China, with most commonly used RIs established without consideration of the effect of age and gender. The Pediatric Reference Intervals in China(PRINCE) project aims to establish and verify pediatric RIs for 31 common laboratory measurands.The project will be a large, multicenter cross-sectional study:14,490 healthy children and adolescents aged up to 19 years will be surveyed by 10 children's hospitals and one pediatric department of a university hospital. To evaluate the feasibility and efficiency of the study methods, 602 children were surveyed in the pilot phase of the PRINCE study in April 2017: it found that some measurands were distinctly age dependent and that there were differences between values for males and females. The results of the pilot study affirmed the necessity of the PRINCE project for Chinese pediatrics. The pilot also indicated potential difficulties in the full survey, e.g., difficulties in recruiting children aged under 3 years and insufficient collection of blood samples from infants. 2DICER1 mutations in twelve Chinese patients with pleuropulmonary blastoma显示文摘Our aim is to examine the impact of DICER1 mutations on the pathogenesis of pleuropulmonary blastoma(PPB) by evaluating the mutation frequency and investigating the family history of Chinese patients with PPB. The family histories of 12 children with PPB recruited consecutively were surveyed. Blood samples from patients and their first-degree relatives were tested for DICER1 mutations. Whole-genome sequencing of blood samples and formalin-fixed and paraffin-embedded(FFPE) tumor tissue was performed in one family with twins. Twelve patients with PPB included six type II and six type III cases. Seven of the12 patients harbored DICER1 mutations, six of which were frameshift or nonsense mutations. Another case carried a germline DICER1 mutation affecting the splice site. FFPE sample had a nonsense mutation in TDG and missense mutations in DICER1.In addition, two cases with DICER1 mutations were found to have lung cysts preceding the diagnosis of PPB. Furthermore, one patient had a family history remarkable for thyroid diseases. Our results indicate that the germline mutation frequency in Chinese patients with PPB is similar to the ones reported for patients from USA, UK, and Japan. 4Optimization of Ultrasonic-enzymatic Extraction of Total Flavonoids from Cardamine tangutorum O.E.Schulz by Response Surface Methodology and Content Determination显示文摘[Objectives]This study aimed to optimize the extraction process of total flavonoids in Cardamine tangutorum O.E.Schulz.[Methods]On the basis of single-factor experiments,the ultrasonic-enzymatic extraction process of total flavonoids in C.tangutorum O.E.Schulz was optimized by Box-Behnken design from aspects of ethanol volume fraction,extraction time,extraction temperature and enzyme addition,with yield of total flavonoids as the evaluation index.[Results]The optimal extraction process of total flavonoids in C.tangutorum O.E.Schulz was as follows:ethanol volume fraction of 70%,ultrasonic extraction time of 43 min,ultrasonic extraction temperature of 37℃and cellulose addition of 0.07 mg.Under the optimal conditions,the yield of total flavonoids from C.tangutorum O.E.Schulz reached 75.90 mg/g.[Conclusions]The optimized extraction process was reasonable and feasible,and it could provide a reference for the extraction of total flavonoids in C.tangutorum O.E.Schulz.This study provided a foundation for the quality control of food/medicinal materials and the development and research of big health products of C.tangutorum O.E.Schulz.Xiaoxia CAI Bochao LI Xuexue LI Hongna SU Wenbing LI Yuan LIU 2020Medicinal Plant2020,11,6:2
5Effect of TiO_2 nanoparticle aggregation on marine microalgae Isochrysis galbana显示文摘TiO_2 nanoparticles(NPs) could adversely impact aquatic ecosystems. However, the aggregation of these NPs could attenuate this effect. In this work, the biological effects of TiO_2 NPs on a marine microalgae Isochrysis galbana were investigated. The aggregation kinetics of TiO_2 NPs under different conditions was also investigated to determine and understand these effects. Results showed that, though TiO_2 NPs had no obvious impact on the size and reproducibility of algal cells under testing conditions, they caused a negative effect on algal chlorophyll, which led to a reduction in photosynthesis. Furthermore, fast aggregation of TiO_2 NPs occurred under all conditions, especially at the pH close to the p Hzpc. 6Steroid receptor coactivator-1:The central intermediator linking multiple signals and functions in the brain and spinal cord显示文摘The effects of steroid hormones are believed to be mediated by their nuclear receptors(NRs).The p160 coactivator family,including steroid receptor coactivator-1(SRC-1),2 and 3,has been shown to physically interact with NRs to enhance their transactivational activities.Among which SRC-1 has been predominantly localized in the central nervous system including brain and spinal cord.It is not only localized in neurons but also detectable in neuroglial cells(mainly localized in the nuclei but also detectable in the extra-nuclear components).Although the expression of SRC-1 is regulated by many steroids,it is also regulated by some non-steroidal factors such as injury,sound and light.Functionally,SRC-1 has been implied in normal function such as development and ageing,learning and memory,central regulation on reproductive behaviors,motor and food intake.Pathologically,SRC-1 may play a role in the regulation of neuropsychiatric disorders(including stress,depression,anxiety,and autism spectrum disorder),metabolite homeostasis and obesity as well as tumorigenesis.Under most conditions,the related mechanisms are far from elucidation;although it may regulate spatial memory through Rictor/mTORC2-actin polymerization related synaptic plasticity.Several inhibitors and stimulator of SRC-1 have shown anti-cancer potentials,but whether these small molecules could be used to modulate ageing and central disorder related neuropathology remain unclear.Therefore,to elucidate when and how SRC-1 is turned on and off under different stimuli is very interesting and great challenge for neuroscientists.Zhaoyou Meng Xiaoya Wang Dongmei Zhang Zhen Lan Xiaoxia Cai Chen Bian Jiqiang Zhang 2022Genes & Diseases2022,9,5:1

8Identification of Growth-promoting Bacteria from Rhizosphere of Pastures and Their Effects on Growth of Lotus corniculatus L.显示文摘[Objectives] This study was conducted to explore the interaction between nitrogen-fixing and phosphate-solubilizing strains and the optimal combination of different functional strains,in order to provide a theoretical basis for the development of PGPR compound fertilizers suitable for local environment.[Methods] In this study,16S rDNA gene sequence analysis was used to identify fast-growing and competitive strains from pasture nodules and rhizosphere soils in Guizhou Province,and three representative Rhizobia and phosphorus-solubilizing bacteria were chosen for the test of bacterial combination when reducing 50% of nitrogen and 30% of phosphorus.The effects of different strain combinations on the plant height,root length,aboveground and underground biomass of Lotus corniculatus L.were investigated,and the total nitrogen and total phosphorus contents of the plants were determined.[Results] The mixed bacterial agents could promote the increase of root biomass,and the effects of A1,A3,B3 and C3 were the most obvious.The fresh weight and dry weight of the roots of L.corniculatus increased by 30.35%-168.45% and 26.43%-180.00%,respectively,and A3,B3,B2 and C3 had the best effects.The total phosphorus content of the plants increased by 12.79%-55.25% compared with the CK2;and most of the bacterial agents with significant growth-promoting effects showed decreased total nitrogen contents,while those with non-significant growth-promoting effects showed significantly-increased total nitrogen contents,which were not as much as the CK1.Comprehensively,the most productive combination was C3,namely R27-2 Rhinohizobium fredii and P33-3 Stenotrophomonas rhizophila.[Conclusions] This study can provide a theoretical basis for the production and promotion of bacterial fertilizers.Lu CAI Yajiao LI Xiaoxia LIU Xin WEI Jianhong SHU 2019Agricultural Biotechnology2019,8,5:1

18A HAase/NIR responsive surface on titanium implants for treating bacterial infection and improving osseointegration显示文摘Bacterial infection and insufficient osseointegration are critical factors affecting the long-term success of titanium-based implants.Unfortunately,the direct application of antibiotic on Ti implants easily leads to poor cytocompatibility,as well as the production of drug-resistant bacteria.So,in this work,we designed a prospective antibacterial strategy by combining photothermal and ciprofloxacin(CIP).The synergistic effect of photothermal and antibiotic may provide an effective bacteriostatic efficacy without sacrificing osteogenesis at a mild condition of moderate temperature and less antibiotic.Herein,CIP was loaded into mesoporous polydopamine(MPDA)nanoparticles(MPDA@CIP),which were anchored on the surface of titanium and finally covered with sodium hyaluronate-catechol(HAc)coating.The hydrophilic HAc layer could inhibit the early adhesion of bacteria,and some bacteria could secrete bacterial hyaluronidase to accelerate the degradation of HAc.This enabled smart enzyme-triggered release of antimicrobials at the site of infection on-demand and avoided unwanted side effects on normal tissues.In addition,NIR light irradiation had a positive influence on both CIP release and MPDA nanoparticle’s photothermal effect.Moreover,before anchoring MPDA@CIP,by the construction of hydroxyapatite microstructure on Ti sur-face with micro-arc oxidation and alkali heat treatment,the ability of bone formation of Ti could be promoted also.Both in vitro as well as in vivo assays demonstrated that functional Ti has an excellent antibacterial effect and osteogenic ability.Dan Li Danyang Wang Ye He Bailong Tao Xiaoxia Liu Yulu Yang Lu Tan Yuchen Zhang Jingwei Hu Weihu Yang Yu Tang Kaiyong Cai 2023Journal of Materials Science & Technology2023,,12:0
19Determination and Multidimensional Analysis of 29 Inorganic Elements in Cardamine tangutorum O.E.Schulz of Different Origins显示文摘[Objectives]This study aimed to determine the contents of inorganic elements in Cardamine tangutorum O.E.Schulz samples from different origins to provide a scientific reference for the quality control,safety evaluation and clinical medicinal use of C.tangutorum O.E.Schulz.[Methods]The contents of Al,As,B,Ba,Ca,Cd,Co,Cr,Cu,Fe,Hg,K,Mg,Mn,Mo,Na,Ni,P,Pb,Pd,Rb,Se,Si,Sn,Sr,Ti,Tl,V and Zn in the C.tangutorum O.E.Schulz samples were determined simultaneously by wet digestion and inductively coupled plasma optical emission spectroscopy(ICP-OES),and the determination results were analyzed by principal component analysis.[Results]A total of 28 inorganic elements were detected in the samples of C.tangutorum O.E.Schulz from different origins.The contents of heavy metals did not exceed the limits.Among the elements,the determined values of Hg in all samples were below the detection limit,so it was not detected.The contents of Ca,Al,P and Fe were relatively high,and they were essential nutrients in the human body.There were significant differences among different elements.Samples from different origins show similar patterns in the distribution of main elements.Correlation analysis shows that 15 pairs of inorganic elements out of 24 elements had extremely significant positive correlations,and 26 pairs of elements had significant positive correlations.The principal component analysis determined Al,As,Ba,Ca,Cd,Fe,Mn,Ni,P,Pb,Pd,Se,V and Zn were the characteristic inorganic elements in the samples of C.tangutorum O.E.Schulz.[Conclusions]The contents of toxic heavy metals in C.tangutorum O.E.Schulz samples from different origins met the edible requirements.The method was simple,rapid and accurate,and could be used to analyze and determine the inorganic elements in the samples of C.tangutorum O.E.Schulz,providing a theoretical basis for the better development and utilization of C.tangutorum O.E.Schulz.Xiaoxia CAI Bochao LI Junzhang QUBIE Yuebu HAILAI Jianlong LAN Wenbing LI Yuan LIU 2020Medicinal Plant2020,11,6:0
20Rapid and sensitive detection of cucumber mosaic virus by reverse transcription loop-mediated isothermal amplification显示文摘Cucumber mosaic virus(CMV),as a member of Cucumovirus genus,is one of the most common lily viruses and has been reported in many countries [1,2].The CMV-infected plants may show developmental abnormalities of leaves,flowers,and bulbs [1].The symptoms will be extremely severe in plants infected by lily symptomless virus(LSV)[1].So far,CMV has posed a threat to cultivars of lily production.Consequently,it is important to develop an efficient approach to achieve rapid and sensitive detection of CMV.Many visual and serological methods have been reported to detect CMV,such as double antibody sandwich enzyme-linked immunosorbent assay [1],immunosorbent electron microscopy [1],enzyme-linked immunosorbent assay(ELISA)and reverse transcriptase-polymerase chain reaction(RT-PCR)[2].However,these methods are time-consuming and require skilled operator and specialized equipment.Loop-mediated isothermal amplification(LAMP)method provides a possible solution for these problems [3].LAMP was developed by Notomi et al.[4] with high specificity and sensitivity because of Bst DNA polymerase.LAMP method not only is suitable for field and large-scale detection of animal virus with more cost-effective but also could be applied to detect foodborne bacterial pathogens and other viruses with high performances [5].For a naked-eye inspection,SYBR Green I was added to the LAMP reaction products to visualize the results.In this study,we developed a rapid and sensitive LAMP method to detect CMV in lily plants and a simple reverse transcription loop-mediated isothermal amplification(RT-LAMP)to detect field CMV samples in lily bulbs.Xiaorui Fan Yanan Du Youming Cai Yongchun Zhang Xiao Zhao Jieling Liang Dan Yang Qi Zhang Xiaoxia Zhang Wanjing Zhang Yan Xu Kai Zhao 2019Acta Biochimica et Biophysica Sinica2019,51,2:0
