2Whole-genome sequencing provides insights into the genetic diversity and domestication of bitter gourd(Momordica spp.)显示文摘Bitter gourd(Momordica charantia)is a popular cultivated vegetable in Asian and African countries.To reveal the characteristics of the genomic structure,evolutionary trajectory,and genetic basis underlying the domestication of bitter gourd,we performed whole-genome sequencing of the cultivar Dali-11 and the wild small-fruited line TR and resequencing of 187 bitter gourd germplasms from 16 countries.The major gene clusters(Bi clusters)for the biosynthesis of cucurbitane triterpenoids,which confer a bitter taste,are highly conserved in cucumber,melon,and watermelon.Comparative analysis among cucurbit genomes revealed that the Bi cluster involved in cucurbitane triterpenoid biosynthesis is absent in bitter gourd.Phylogenetic analysis revealed that the TR group,including 21 bitter gourd germplasms,may belong to a new species or subspecies independent from M.charantia.Furthermore,we found that the remaining 166 M.charantia germplasms are geographically differentiated,and we identified 710,412,and 290 candidate domestication genes in the South Asia,Southeast Asia,and China populations,respectively.This study provides new insights into bitter gourd genetic diversity and domestication and will facilitate the future genomics-enabled improvement of bitter gourd.Junjie Cui Yan Yang Shaobo Luo Le Wang Rukui Huang Qingfang Wen Xiaoxia Han Nansheng Miao Jiaowen Cheng Ziji Liu Changyuan Zhang Chengcheng Feng Haisheng Zhu Jianwen Su Xinjian Wan Fang Hu Yu Niu Xiaoming Zheng Yulan Yang Dai Shan Zhensheng Dong Weiming He Narinder P.S.Dhillon Kailin Hu 2020Horticulture Research2020,7,1:7
3Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. 4Effect of cold rolling process on microstructure and mechanical properties of high strength β titanium alloy thin sheets显示文摘The microstructure and mechanical properties of Ti-3.5Al-5Mo-6V-3Cr-2Sn-0.5Fe high strength titanium alloy sheets prepared by unidirectional cold rolling and two-step cross cold rolling were investigated. Results showed that the β phase grains were refined significantly by cold rolling followed by solution treatment for a short time.Compared to unidirectional cold rolling, the short time solution treatment after two-step cross rolling could significantly reduce the non-uniformity of the microstructure of the alloy sheets. After aging treatment at 550 ℃,the anisotropy of the mechanical properties still existed in the unidirectional rolled sheets, and the tensile strength was highest along the rolling direction. 5Effect of phase separation structure on cementitious reactivity of blast furnace slag显示文摘Blast furnace slag samples with phase separation structure were prepared by re-melting and then water quenching process. By use of XRD,DTA and SEM technologies in combination with mechanical property experiment,the structure characteristics of samples were determined and their effects on cementitious reactivity were investigated. The results show that the samples with phase separation have better cementitious reactivity than sample with homogenous glass and sample with crystalline phases,which mainly contributes to its glass structure with coexistence of Ca-O rich phase and Si-O rich phase. 6Changes in soil microbial community response to precipitation events in a semi-arid steppe of the Xilin River Basin, China显示文摘In the context of climate change, precipitation is predicted to become more intense at the global scale. Such change may alter soil microbial communities and the microbially mediated carbon and nitrogen dynamics. In this study, we experimentally repackaged precipitation patterns during the growing season(from June to September) of 2012 in a semi-arid temperate steppe of the Xilin River Basin in Inner Mongolia of China, based on the 60-year growing season precipitation data. Specifically, a total amount of 240 mm simulated precipitation was assigned to experimental plots by taking the following treatments:(1) P6(6 extreme precipitation events, near the 1^(st) percentile);(2) P10(10 extreme precipitation events, near the 5^(th) percentile);(3) P16(16 moderate precipitation events, near the 50^(th) percentile); and(4) P24(24 events, 60-year average precipitation, near the 50^(th) percentile). At the end of the growing season, we analyzed soil microbial community structure and biomass, bacterial abundance, fungal abundance and bacterial composition, by using phospholipid fatty acid(PLFA), real-time quantitative polymerase chain reaction(RT-qPCR) and 16S rRNA gene clone library methods. The extreme precipitation events did not change soil microbial community structure(represented by the ratio of PLFA concentration in fungi to PLFA concentration in bacteria, and the ratio of PLFA concentration in gram-positive bacterial biomass to PLFA concentration in gram-negative bacterial biomass). However, the extreme precipitation events significantly increased soil microbial activity(represented by soil microbial biomass nitrogen and soil bacterial 16S rRNA gene copy numbers). Soil fungal community showed no significant response to precipitation events. According to the redundancy analysis, both soil microbial biomass nitrogen and soil ammonium nitrogen(NH_4-N) were found to be significant in shaping soil microbial community. Acidobacteria, Actinobacteria and Proteobacteria were the dominant phyla in soil bacterial composition, and responded differently to the extreme precipitation events. 7Analysis of grinding mechanics and improved grinding force model based on randomized grain geometric characteristics显示文摘Too high grinding force will lead to a large increase in specific grinding energy, resulting in high temperature in grinding zone, especially for the aerospace difficult cutting metal materials,seriously affecting the surface quality and accuracy. At present, the theoretical models of grinding force are mostly based on the assumption of uniform or simplified morphological characteristics of grains, which is inconsistent with the actual grains. Especially for non-engineering grinding wheel,most geometric characteristics of grains are ignored, resulting in the calculation accuracy that cannot guide practical production. Based on this, an improved grinding force model based on random grain geometric characteristics is proposed in this paper. Firstly, the surface topography model of CBN grinding wheel is established, and the effective grain determination mechanism in grinding zone is revealed. Based on the known grinding force model and mechanical behavior of interaction between grains and workpiece in different stages, the concept of grain effective action area is proposed. The variation mechanism of effective action area under the influence of grain geometric and spatial characteristics is deeply analyzed, and the calculation method under random combination of five influencing parameters is obtained. The numerical simulation is carried out to reveal the dynamic variation process of grinding force in grinding zone. In order to verify the theoretical model, the experiments of dry grinding Ti-6Al-4 V are designed. The experimental results show that under different machining parameters, the results of numerical calculation and experimental measurement are in good agreement, and the minimum error value is only 2.1 %, which indicates that the calculation accuracy of grinding force model meets the requirements and is feasible. 8Nasal delivery of broadly neutralizing antibodies protects mice from lethal challenge with SARS-CoV-2 delta and omicron variants显示文摘Multiple new variants of severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)have constantly emerged,as the delta and omicron variants,which have developed resistance to currently gained neutralizing antibodies.This highlights a critical need to discover new therapeutic agents to overcome the variants mutations.Despite the availability of vaccines against coronavirus disease 2019(COVID-19),the use of broadly neutralizing antibodies has been considered as an alternative way for the prevention or treatment of SARS-Co V-2 variants infection.Here,we show that the nasal delivery of two previously characterized broadly neutralizing antibodies(F61 and H121)protected K18-h ACE2 mice against lethal challenge with SARS-Co V-2 variants.The broadly protective efficacy of the F61 or F61/F121 cocktail antibodies was evaluated by lethal challenge with the wild strain(WIV04)and multiple variants,including beta(B.1.351),delta(B.1.617.2),and omicron(B.1.1.529)at 200or 1000 TCID_(50),and the minimum antibody administration doses(5-1.25 mg/kg body weight)were also evaluated with delta and omicron challenge.Fully prophylactic protections were found in all challenged groups with both F61 and F61/H121 combination at the administration dose of 20 mg/kg body weight,and corresponding mice lung viral RNA showed negative,with almost all alveolar septa and cavities remaining normal.Furthermore,low-dose antibody treatment induced significant prophylactic protection against lethal challenge with delta and omicron variants,whereas the F61/H121 combination showed excellent results against omicron infection.Our findings indicated the potential use of broadly neutralizing monoclonal antibodies as prophylactic and therapeutic agent for protection of current emerged SARS-Co V-2 variants infection.Jia Lu Qiangling Yin Rongjuan Pei Qiu Zhang Yuanyuan Qu Yongbing Pan Lina Sun Ding Gao Cuiqin Liang Jingwen Yang Wei Wu Jiandong Li Zongqiang Cui Zejun Wang Xinguo Li Dexin Li Shiwen Wang Kai Duan Wuxiang Guan Mifang Liang Xiaoming Yang 2022Virologica Sinica2022,37,2:4
9Genetic analysis of fertility restoring genes for AL-type male sterility in wheat显示文摘In order to screen molecular markers linked to fertility restoring genes and further improve the breeding efficiency of restorer lines,in this study,wheat varieties 18A,18B and 99AR144-1 were used as experimental materials to establish F2 fertility-segregating population. Plant quantitative trait'major gene + polygene mixed model'separation analysis method and simple sequence repeat(SSR) molecular markers were adopted for genetic analysis of four generations,including the parents(P1 and P2),and hybrid(Fl and F2) populations. The results show that AL-type fertility restoring gene is controlled by two pairs of additive-dominant-epistatic genes and additive-dominant polygene;two primers linked to fertility restoring genes were selected by SSR molecular markers, including Xgwm95 on chromosome 2A and Barc61 on chromosome 1B,with the linkage distance of 15.0 cM and 18.0 cM,respectively. 10Antagonism of toll-like receptor 2 attenuates the formation and progression of abdominal aortic aneurysm显示文摘Abdominal aortic aneurysm(AAA) is an inflammatory vascular disorder with high mortality.Accumulating evidence shows that toll-like receptor 2(TLR2) plays a critical role in the regulation of wound-repairing process after tissue injury.We wondered if TLR2 signaling contributed to the pathogenesis of AAA and that targeting TLR2 would attenuate AAA development and progression.In this study,enhanced expression of TLR2 and its ligands were observed in human AAA tissue.Neutralization of TLR2 protected against AAA development and caused established AAA to regress in mouse models of AAA.In addition,TLR2-deficient mice also failed to develop AAA.The prophylactic and therapeutic effects of blocking TLR2 were accompanied by a significant resolution of inflammation and vascular remodeling,as indicated by the decreased expression or activity of MMP-2/9,α-SMA,inflammatory cytokines,and transcription factors NF-κB,AP-1 and STAT1/3 in AAA tissue.Mechanistically,blocking TLR2 decreased the expression and interaction of TLR2 and several endogenousligands,which diminished chronic inflammation and vascular remodeling in the vascular tissue of AAA.Our studies indicate that the interactions between TLR2 and its endogenous ligands contribute to the pathogenesis of AAA and that targeting TLR2 offers great potential toward the development of therapeutic agents against AAA.Huimin Yan Bing Cui Xiaowei Zhang Xiaoming Fu Jun Yan Xiaoxing Wang Xiaoxi Lv Zhong Chen Zhuowei Hu 2015Acta Pharmaceutica Sinica B2015,5,3:3

12Comprehensive identification and analyses of the Hsf gene family in the whole-genome of three Apiaceae species显示文摘Apiaceae is a major family from Apiales and includes many important vegetable and medicinal crops.Heat shock transcription factors(Hsf)play important roles in heat tolerance during plant development.Here,we conducted systematic analyses of the Hsf gene family in three Apiaceae species,including 17 Apium graveolens(celery),32 Coriandrum sativum(coriander),and 14 Daucus carota(carrot).A total of 73 Hsf genes were identified in three representative species,including Arabidopsis thaliana,Vitis vinifera,and Lactuca sativa.Whole-genome duplication played important roles in the Hsf gene family’s expansion within Apiaceae.Interestingly,we found that coriander had more Hsf genes than celery and carrot due to greater expansion and fewer losses.Twenty-seven branches of the phylogenetic tree underwent considerable positive selection in these Apiaceae species.We also explored the expression patterns of Hsf genes in three plant organs.Collectively,this study will serve as a rich gene resource for exploring the molecular mechanisms of heat tolerance.Additionally,this is the first study to report on the Hsf gene family in Apiaceae;thus,our research will provide guidance for future comparative and functional genomic studies on the Hsf gene family and others in Apiaceae.Qiaoying Pei Tong Yu TongWu Qihang Yang Ke Gong Rong Zhou Chunlin Cui Ying Yu Wei Zhao Xi Kang Rui Cao Xiaoming Song 2021Horticultural Plant Journal2021,7,5:3
13Towards high-power mid-IR light source tunable from 3.8 to 4.5 µm by HBr-filled hollow-core silica fibres显示文摘Fibre lasers operating at the mid-IR have attracted enormous interest due to the plethora of applications in defence,security,medicine,and so on.However,no continuous-wave(CW)fibre lasers beyond 4μm based on rare-earth-doped fibres have been demonstrated thus far.Here,we report efficient mid-IR laser emission from HBr-filled silica hollow-core fibres(HCFs)for the first time.By pumping with a self-developed thulium-doped fibre amplifier seeded by several diode lasers over the range of 1940–1983 nm,narrow linewidth mid-IR emission from 3810 to 4496 nm has been achieved with a maximum laser power of about 500 mW and a slope efficiency of approximately 18%.To the best of our knowledge,the wavelength of 4496 nm with strong absorption in silica-based fibres is the longest emission wavelength from a CW fibre laser,and the span of 686 nm is also the largest tuning range achieved to date for any CW fibre laser.By further reducing the HCF transmission loss,increasing the pump power,improving the coupling efficiency,and optimizing the fibre length together with the pressure,the laser efficiency and output power are expected to increase significantly.This work opens new opportunities for broadly tunable high-power mid-IR fibre lasers,especially beyond 4μm.Zhiyue Zhou Zefeng Wang Wei Huang Yulong Cui Hao Li Meng Wang Xiaoming Xi Shoufei Gao Yingying Wang 2022Light(Science & Applications)2022,11,1:3
14Observation and research of deep underground multi-physical fields—Huainan–848 m deep experiment显示文摘Compared with the surface,the deep environment has the advantages of allowing“super-quiet and ultra-clean”-geophysical field observation with low vibration noise and little electromagnetic interference,which are conducive to the realization of long-term and high-precision observation of multi-physical fields,thus enabling the solution of a series of geoscience problems.In the Panyidong Coal Mine,where there are extensive underground tunnels at the depth of 848 m below sea level,we carried out the first deep-underground geophysical observations,including radioactivity,gravity,magnetic,magnetotelluric,background vibration and six-component seismic observations.We concluded from these measurements that(1)the background of deep subsurface gravity noise in the long-period frequency band less than 2 Hz is nearly two orders of magnitude weaker than that in the surface observation environment;(2)the underground electric field is obviously weaker than the surface electric field,and the relatively high frequency of the underground field,greater than 1 Hz,is more than two orders of magnitude weaker than that of the surface electric field;the east-west magnetic field underground is approximately the same as that at the surface;the relatively high-frequency north-south magnetic field underground,below 10 Hz,is at least one order of magnitude lower than that at the surface,showing that the underground has a clean electromagnetic environment;(3)in addition to the highfrequency and single-frequency noises introduced by underground human activities,the deep underground space has a significantly lower background vibration noise than the surface,which is very beneficial to the detection of weak earthquake and gravity signals;and(4)the underground roadway support system built with ferromagnetic material interferes the geomagnetic field.We also found that for deep observation in the“ultra-quiet and ultra-clean”environment,the existing geophysical equipment and observation technology have problems of poor adaptability and insufficient precision as well as data cleaning problems,such as the effective separation of the signal and noise of deep observation data.It is also urgent to interpret and comprehensively utilize these high-precision multi-physics observation data.Yun WANG Yaxin YANG Heping SUN Chengliang XIE Qisheng ZHANG Xiaoming CUI Chang CHEN Yongsheng HE Qiangqiang MIAO Chaomin MU Lianghui GUO Jiwen TENG 2023Science China Earth Sciences2023,66,1:3
15Evolution of kaolinite subgroup minerals and mixed-layer illite/smectite in the Paleogene Damintun Depression in Liaohe Basin of China and its implication for paleotemperature显示文摘The oil-rich Damintun Depression is located in the Liaohe Basin, Northeast China, and was formed during the Paleogene. The major oil-producing strata in the depression are mudstone and shale. To explore the burial diagenetic history of the basin and the formation thresholds of hydrocarbons, the characters of the kaolinite subgroup minerals and mixed-layer illite/smectite in the mudstone and the shale are studied by using X-ray diffraction, electron probe, scanning electron microscope, and Fourier infrared spectrum. The kaolinite subgroup consists of kaolinite and halloysite. The kaolinite is flake-like or vermiform-like. The halloysite is in long tubular shape and its length is related to its iron content. A longer tube has lower iron content. The crystallinity of kaolinite is 0.40 °2θ, and its degree of order increases from 0.03 to 1.17 with the burial depth. Kaolinite is in disorder when the buried depth is less than or equal to 2479 m , and it is partially ordered when the buried depth is greater than 2479 m. Kaolinite is supposed to turn into dickite when the depth is greater than 2550 m, but low penetrability and low porosity of the shale and mudstone prevent such a change. The mixed-layer illite/smectite changes from disorder to order continually as the buried depth increases. Its disorder (R0I/S), as defined by illite layer content (I%), is smaller than 50% at depths less than 2550.25 m. 16Low-frequency repetitive transcranial magnetic simulation prevents chronic epileptic seizure显示文摘Although low-frequency repetitive transcranial magnetic simulation can potentially treat epilepsy,its underlying mechanism remains unclear.This study investigated the influence of low-frequency repetitive transcranial magnetic simulation on changes in several nonlinear dynamic electroencephalographic parameters in rats with chronic epilepsy and explored the mechanism underlying repetitive transcranial magnetic simulation-induced antiepileptic effects.An epilepsy model was established using lithium-pilocarpine intraperitoneal injection into adult Sprague-Dawley rats,which were then treated with repetitive transcranial magnetic simulation for 7 consecutive days.Nonlinear electroencephalographic parameters were obtained from the rats at 7,14,and 28 days post-stimulation.Results showed significantly lower mean correlation-dimension and Kolmogorov-entropy values for stimulated rats than for non-stimulated rats.At 28 days,the complexity and point-wise correlation dimensional values were lower in stimulated rats.Low-frequency repetitive transcranial magnetic simulation has suppressive effects on electrical activity in epileptic rats,thus explaining its effectiveness in treating epilepsy.Yinxu Wang Xiaoming Wang Sha Ke Juan Tan Litian Hu Yaodan Zhang Wenjuan Cui 2013Neural Regeneration Research2013,8,27:2
17Application of superconductive gravity technique on the constraints of core-mantle coupling parameters显示文摘The parameters, i.e. the Period and the Quality factor, of the Earth's free core nutation (FCN) are closely related to the dissipative coupling between the core and the mantle. Based on the FCN parameters obtained from the actual observations and theoretical simulation, significantly constrained in this study were several key parameters near the core-mantle boundary (CMB), related to the core and mantle coupling, including viscosity at the top of liquid core, conductivity at the bottom of the mantle, and dynamic ellipticity of the CMB. In order to choose high quality observations from global stations of the superconducting gravimeters (SG) on the Global Geodynamics Project (GGP) network, we adopted two criteria, the standard deviations of harmonic analysis on tidal observations and the quality of the FCN parameters calculated with the observations from single station. After the mean ocean tidal effects of the recent ocean tidal models were removed, the FCN parameters were retrieved by stacking the tidal gravity observations from the GGP network. The results were in a good agreement with those in the recent research by using the SG and/or the VLBI observations. Combined with an FCN theoretical model deduced by angular momentum method, the viscous and electromagnetic coupling parameters near the CMB were evaluated. Numerical results indicated that the viscosity at the top of the liquid core was in the range from 6.6×102 to 2.6×103 Pa·s, which was in good agreement with those obtained from the Earth's nutation, the FCN and variations in the length of day (LOD). The conductivity at the bottom of the mantle should be as large as 2.6×106-1.0×107 S m-1 to match the FCN quality factors from the actual observations. 18Conditioned medium-electrospun fiber biomaterials for skin regeneration显示文摘Conditioned medium(CM)contains variety of factors secreted by cells,which directly regulate cellular processes,showing tremendous potential in regenerative medicine.Here,for the first time,we proposed a novel regenerative therapy mediated by biodegradable micro-nano electrospun fibers loaded with highly active conditioned medium of adipose-derived stem cells(ADSC-CM).ADSC-CM was successfully loaded into the nanofibers with biological protection and controllable sustained-release properties by emulsion electrospinning and protein freeze-drying technologies.In vitro,ADSC-CM released by the fibers accelerated the migration rate of fibroblasts;inhibited the over proliferation of fibroblasts by inducing apoptosis and damaging cell membrane;in addition,ADSC-CM inhibited the transformation of fibroblasts into myofibroblasts and suppressed excessive production of extracellular matrix(ECM).In vivo,the application of CM-biomaterials significantly accelerated wound closure and improved regeneration outcome,showing superior pro-regenerative performance.This study pioneered the application of CM-biomaterials in regenerative medicine,and confirmed the practicability and significant biological effects of this innovative biomaterials.Lu Chen Liying Cheng Zhen Wang Jianming Zhang Xiyuan Mao Zhimo Liu Yuguang Zhang Wenguo Cui Xiaoming Sun 2021Bioactive Materials2021,6,2:2
