1Metabolic dysregulation and emerging therapeutical targets for hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is an aggressive human cancer with increasing incidence worldwide.Multiple efforts have been made to explore pharmaceutical therapies to treat HCC,such as targeted tyrosine kinase inhibitors,immune based therapies and combination of chemotherapy.However,limitations exist in current strategies including chemoresistance for instance.Tumor initiation and progression is driven by reprogramming of metabolism,in particular during HCC development.Recently,metabolic associated fatty liver disease(MAFLD),a reappraisal of new nomenclature for nonalcoholic fatty liver disease(NAFLD),indicates growing appreciation of metabolism in the pathogenesis of liver disease,including HCC,thereby suggesting new strategies by targeting abnormal metabolism for HCC treatment.In this review,we introduce directions by highlighting the metabolic targets in glucose,fatty acid,amino acid and glutamine metabolism,which are suitable for HCC pharmaceutical intervention.We also summarize and discuss current pharmaceutical agents and studies targeting deregulated metabolism during HCC treatment.Furthermore,opportunities and challenges in the discovery and development of HCC therapy targeting metabolism are discussed.Danyu Du Chan Liu Mengyao Qin Xiao Zhang Tao Xi Shengtao Yuan Haiping Hao Jing Xiong 2022Acta Pharmaceutica Sinica B2022,12,2:27

3Ten cities cross-sectional questionnaire survey of children asthma and other allergies in China显示文摘Asthma,rhinitis and eczema(allergic or non-allergic)have increased throughout the world during the last decades,especially among children.Changes in the indoor environment are suspected to be important causes.China has experienced a dramatic change in indoor environmental exposures during the past two decades.However,such changes and their associations with children’s asthma and other health aspects have not been thoroughly studied.China,Children,Homes,Health(CCHH),Phase I,was a cross-sectional questionnaire survey of 48219 children 1–8 years old in 10 Chinese cities during 2010–2012.The questionnaire includes the International Study of Asthma and Allergies in Childhood(ISAAC)core health questions and additional questions regarding housing,life habits and outdoor environment.In health analyses,children aged 3–6 years old were included.The prevalences of doctor diagnosed asthma varied from 1.7%to 9.8%(mean 6.8%),a large increase from 0.91%in 1999 and 1.50%in2000.The prevalence of wheeze,rhinitis and atopic eczema(last 12 months)varied from 13.9%to 23.7%,24.0%to 50.8%and4.8%to 15.8%,respectively.Taiyuan had the lowest prevalences of all illnesses and Shanghai the highest,except for wheezewhere the highest value was for Urumqi.We found(1)no obvious association between disease prevalences and ambient PM10concentrations and(2)higher prevalences of disease in humid climates with hot summers and cold winters,but with no centrally heated buildings.Associations between the diseases and economic status as indexed by Gross Domestic Product(GDP)requires further study.ZHANG YinPing LI BaiZhan HUANG Chen YANG Xu QIAN Hua DENG QiHong ZHAO ZhuoHui LI AnGui ZHAO JiaNing ZHANG Xin QU Fang HU Yu YANG Qin WANG Juan ZHANG Ming WANG Fang ZHENG XiaoHong LU Chan LIU ZhiJian SUN YueXia MO JinHan ZHAO YiLi LIU Wei WANG TingTing NORBCK Dan BORNEHAG Carl-Gustaf SUNDELL Jan 2013Chinese Science Bulletin2013,58,34:22
4The recommendations of Chinese Parkinson’s disease and movement disorder society consensus on therapeutic management of Parkinson’s disease显示文摘Background:Parkinson’s disease(PD)is a chronic,progressive and debilitating disease,which affects over 2.5 million people in China.PD is characterized clinically by resting tremor,muscular rigidity,bradykinesia and postural instability.As the disease progresses,additional complications can arise such as non-motor and neurobehavioral symptoms.Pharmacological treatment and surgical intervention for PD have been implemented in China.Until 10 years ago,there was lack of standardization for the management of PD in different regions and among different physicians,leading to different treatment levels in different regions and different physicians.Since then,the Chinese Parkinson’s Disease and Movement Disorder Society have published three versions of guidelines for the management of PD in China,in 2006,2009 and 2014,respectively.Correspondingly,the overall level of treatment for PD in China improved.Objectives:To update the treatment guidelines based on current foreign and domestic practice guidelines and clinical evidence,and to improve the treatment options available to physicians in the management of PD.Summary:A variety of treatment recommendations in the treatment guidelines have been proposed,including physical activity and disease-modifying medication,which should be initiated at the early-stage of the disease.The principles of dosage titration should be followed to avoid acute adverse reactions to the drugs,to achieve a satisfactory clinical effect with a low dose and to reduce the incidence of long-term motor complications.Moreover,different treatment strategies should be considered at different stages of the disease.Importantly,treatment guidelines and personalized treatments should be valued equally.A set of treatment recommendations has been developed to assist physicians to improve and optimize clinical outcomes for patients with PD in China.Shengdi Chen Piu Chan Shenggang Sun Haibo Chen Baorong Zhang Weidong Le Chunfeng Liu Guoguang Peng Beisha Tang Lijuan Wang Yan Cheng Ming Shao Zhenguo Liu Zhenfu Wang Xiaochun Chen Mingwei Wang Xinhua Wan Huifang Shang Yiming Liu Pingyi Xu Jian Wang Tao Feng Xianwen Chen Xingyue Hu Anmu Xie Qin Xiao 2016Translational Neurodegeneration2016,5,1:23
5Benefits of Conservation Agriculture on Soil and Water Conservation and Its Progress in China显示文摘Conservation agriculture has been practised for three decades and has been spread widely. There are many nomenclatures surrounding conservation agriculture and differ to each other lightly. Conservation agriculture (CA) is a system approach to soil and water conservation, high crop productivity and profitability, in one word, it is a system approach to sustainable agriculture. Yet, because conservation agriculture is a knowledge-intensive and a complex system to learn and implement, and also because of traditions of intensive cultivation, adoption rates have been low, since to date, only about seven percent of the world's arable and permanent cropland area is farmed under conservation agriculture. The practice and wider extention of conservation agriculture thus requires a deeper understanding of its ecological underpinnings in order to manage its various elements for sustainable intensification, where the aim is to conserve soil and water and improve sustainability over the long term. 7Integrative single-cell analysis of transcriptome,DNA methylome and chromatin accessibility in mouse oocytes显示文摘Oocyte growth is a key step in forming mature eggs that are ready to be fertilized.The states and modifications of chromatin represent critical sources of information for this process.However,the dynamics and interrelations of these chromatin characteristics remain elusive.In this study,we developed an improved scCOOL-seq technique (iscCOOL-seq),which is a multiomics,single-cell and single-base resolution method with high mapping rates,and explored the chromatin accessibility landscape and its relationship to DNA methylation in growing mouse oocytes.The most dramatic change in chromatin accessibility occurs during oocyte growth initiation,accompanied with prominent transcriptome alterations and an elevated variation in DNA methylation levels among individual oocytes.Unlike CpG islands (CGIs),partially methylated domains (PMDs)are associated with a low density of nucleosome-depleted regions (NDRs)during the whole maturation period.Surprisingly,highly expressed genes are usually associated with NDRs at their transcriptional end sites (TESs).In addition,genes with de novo methylated gene bodies during oocyte maturation are already open at their promoters before oocyte growth initiation.Furthermore,epigenetic and transcription factors that might be involved in oocyte maturation are identified.Our work paves the way for dissecting the complex, yet highly coordinated,epigenetic alterations during mouse oocyte growth and the establishment of totipotency.Chan Gu Shanling Liu Qihong Wu Lin Zhang Fan Guo 2019Cell Research2019,29,2:14
8Transactivation of the TIEG1 confers growth inhibition of transforming growth factor-β-susceptible hepatocellular carcinoma cells显示文摘AIM:To investigate the role of transforming growth factor(TGF)-β-inducible early gene 1(TIEG1) in TGF-β-induced growth inhibition in hepatocellular carcinoma(HCC) cells.METHODS:Human hepatocyte and HCC cell lines with varied susceptibilities to TGF-β1 were tested by methylthiazoletetrazolium(MTT) assay.The expression changes of Smad2,Smad3,Smad4,Smad7,TIEG1 and TIEG2 gene following treatment with TGF-β1 in a TGF-β-sensitive hepatocyte cell line(MIHA),a TGF-β-sensitive hepatoma cell line(Hep3B) and two TGF-β-insensitive hepatoma cell lines(HepG2 and Bel7404) were examined.SiRNA targeting TIEG1 was transfected into Hep3B cells and the sensitivity of cells to TGF-β1 was examined.Overexpression of TIEG1 was induced by lentiviral-mediated transduction in TGF-β1-resistant hepatoma cell lines(Bel7404 and HepG2).MTT assay and 4',6-Diamidino-2-phenylindole staining were used to identify cell viability and apoptosis,respectively.The expression level of stathmin was measured by reverse transcriptase polymerase chain reaction and Western-blotting analysis,and stathmin promoter activity by TIEG1 was monitored by a luciferase reporter gene system.RESULTS:TIEG1 was significantly upregulated by TGF-β1 in the TGF-β1-sensitive HCC cell line,Hep3B,but not in the resistant cell lines.The suppression of TIEG1 by siRNAs decreased the sensitivity of Hep3B cells to TGF-β1,whereas the overexpression of TIEG1 mediated growth inhibition and apoptosis in TGF-β1-resistant HCC cell lines,which resembled those of TGF-β1-sensitive HCC cells treated with TGF-β1.Our data further suggested that stathmin was a direct target of TIEG1,as stathmin was signif icantly downregulated by TIEG1 overexpression,and stathmin promoter activity was inhibited by TIEG1 in a dose-dependent manner.CONCLUSION:Our data suggest that transactivation of TIEG1 conferred growth inhibition of TGF-β-susceptible human HCC cells.Lei Jiang Yiu-Kay Lai Jin-Fang Zhang Chu-Yan Chan Gang Lu Marie CM Lin Ming-Liang He Ji-Cheng Li Hsiang-Fu Kung 2012World Journal of Gastroenterology2012,18,17:13
9Chemical screen identifies a geroprotective role of quercetin in premature aging显示文摘Aging increases the risk of various diseases. The main goal of aging research is to find therapies that attenuate aging and alleviate aging-related diseases. In this study, we screened a natural product library for geroprotective compounds using Werner syndrome (WS) human mesenchymal stem cells (hMSCs), a premature aging model that we recently established. Ten candidate compounds were identified and quercetin was investigated in detail due to its leading effects. Mechanistic studies revealed that quercetin alleviated senescence via the enhancement of cell proliferation and restoration of heterochromatin architecture in WS hMSCs. RNA-sequencing analysis revealed the transcriptional commonalities and differences in the geroprotective effects by quercetin and Vitamin C. Besides WS hMSCs, quercetin also attenuated cellular senescence in Hutchinson-Gilford progeria syndrome (HGPS) and physiological-aging hMSCs. 13First observation of tropospheric nitrogen dioxide from the Environmental Trace Gases Monitoring Instrument onboard the GaoFen-5 satellite显示文摘The Environmental Trace Gases Monitoring Instrument(EMI)is the first Chinese satellite-borne UV–Vis spectrometer aiming to measure the distribution of atmospheric trace gases on a global scale.The EMI instrument onboard the GaoFen-5 satellite was launched on 9 May 2018.In this paper,we present the tropospheric nitrogen dioxide(NO2)vertical column density(VCD)retrieval algorithm dedicated to EMI measurement.We report the first successful retrieval of tropospheric NO_(2) VCD from the EMI instrument.Our retrieval improved the original EMI NO_(2) prototype algorithm by modifying the settings of the spectral fit and air mass factor calculations to account for the on-orbit instrumental performance changes.The retrieved EMI NO_(2) VCDs generally show good spatiotemporal agreement with the satellite-borne Ozone Monitoring Instrument and TROPOspheric Monitoring Instrument(correlation coefficient R of ~0.9,bias<50%).A comparison with ground-based MAX-DOAS(Multi-Axis Differential Optical Absorption Spectroscopy)observations also shows good correlation with an R of 0.82.The results indicate that the EMI NO_(2) retrieval algorithm derives reliable and precise results,and this algorithm can feasibly produce stable operational products that can contribute to global air pollution monitoring.Chengxin Zhang Cheng Liu Ka Lok Chan Qihou Hu Haoran Liu Bo Li Chengzhi Xing Wei Tan Haijin Zhou Fuqi Si Jianguo Liu 2020Light(Science & Applications)2020,9,1:11
14Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases,especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed.Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung,heart and artery,the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging.With age,expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier,cardiomyocytes,and vascular endothelial cells.We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore,treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether,our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly.Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu 2021Cell Research2021,31,4:10
15Evidence of compliance with and effectiveness of guidelines for noninvasive prenatal testing in China:a retrospective study of 189,809 cases显示文摘In China,the medical guidelines recommend performing noninvasive prenatal testing(NIPT)with caution for pregnant women aged 35 years or older.However,the Mother and Child Health Care Law suggests that all primiparous women whose age is older than 35 years undergo prenatal diagnosis.These two inconsistent suggestions/recommendations have made obstetricians confused about whether to offer NIPT to these older pregnant women.To face this issue and find out the solution we performed a retrospective study of 189,809 NIPT samples collected from 28 provincial-leveled administrative units in China.Of 1,564 women with high-risk pregnancies who underwent NIPT,459(29.3%)did not participate in follow-up.The compound sensitivity and specificity of NIPT for trisomies 21,18 and 13 detection was 99.1%(95%CI,98.0%–99.6%)and 99.9%(95%CI,98.8%–99.9%),respectively.In secundiparous women,NIPT showed high sensitivity and specificity similar to that in primiparous women.The observed risk for trisomies 21 and 18 significantly increased when the maternal age was 39 and older.After the publication of the current NIPT policy,the follow-up rate at our center was 97.9%;however,a large number of women are not in maternal and infant care networks nationwide,and that makes the follow-up rate outside our center relatively low.Our study shows that to balance the prevention of major aneuploidies and the limited resources for prenatal diagnosis,the cut-off age of 35 for invasive prenatal diagnosis might be unnecessary.Although the NIPT guidelines are well written,how to practice it effectively,especially in less industrialized areas,is worth discussing.Chan Tian Tao Deng Xiuhuang Zhu Chen Gong Yangyu Zhao Yuan Wei Rong Li Xiufeng Xu Miaonan He Zhiwei Zhang Jing Cheng Mol BenWillem Jie Qiao 2020Science China(Life Sciences)2020,63,3:10

17Enabling Industrial Internet of Things(IIoT) towards an emerging smart energy system显示文摘The increasing penetration of renewable energy on the transmission and distribution power network is driving the adoption of two-way power flow control, data and communications needed to meet the dependency of balancing generation and load. Thus, creating an environment where power and information flow seamlessly in real time to enable reliable and economically viable energy delivery, the advent of Internet of Energy(IoE) as well as the rising of Internet of Things(IoT) based smart systems.The evolution of IT to Io T has shown that an information network can be connected in an autonomous way via routers from operating system(OS) based computers and devices to build a highly intelligent eco-system. Conceptually, we are applying the same methodology to the Io E concept so that Energy Operating System(EOS) based assets and devices can be developed into a distributed energy network via energy gateway and self-organized into a smart energy eco-system.This paper introduces a laboratory based IIo T driven software and controls platform developed on the NICE Nano-grid as part of a NICE smart system Initiative for Shenhua group. 20Stabilization of heterochromatin by CLOCK promotes stem cell rejuvenation and cartilage regeneration显示文摘Accumulating evidence indicates an association between the circadian clock and the aging process.However,it remains elusive whether the deregulation of circadian clock proteins underlies stem cell aging and whether they are targetable for the alleviation of aging-associated syndromes.Here,we identified a transcription factor-independent role of CLOCK,a core component of the molecular circadian clock machinery,in counteracting human mesenchymal stem cell(hMSC)decay.CLOCK expression was decreased during hMSC aging.In addition,CLOCK deficiency accelerated hMSC senescence,whereas the overexpression of CLOCK,even as a transcriptionally inactive form,rejuvenated physiologically and pathologically aged hMSCs.Mechanistic studies revealed that CLOCK formed complexes with nuclear lamina proteins and KAP1,thus maintaining heterochromatin architecture and stabilizing repetitive genomic sequences.Finally,gene therapy with lentiviral vectors encoding CLOCK promoted cartilage regeneration and attenuated age-related articular degeneration in mice.These findings demonstrate a noncanonical role of CLOCK in stabilizing heterochromatin,promoting tissue regeneration,and mitigating aging-associated chronic diseases.Chuqian Liang Zunpeng Liu Moshi Song Wei Li Zeming Wu Zehua Wang Qiaoran Wang Si Wang Kaowen Yan Liang Sun Tomoaki Hishida Yanning Cai Juan Carlos lzpisua Belmonte Pedro Guillen Piu Chan Qi Zhou Weiqi Zhang Jing Qu Guang-Hui Liu 2021Cell Research2021,31,2:8
