1Special Section:SARS-CoV-2 Preliminary evidence from a multicenter prospective observational study of the safety and efficacy of chloroquine for the treatment of COVID-19显示文摘Effective therapies are urgently needed for the SARS-CoV-2 pandemic.Chloroquine has been proved to have antiviral effect against coronavirus in vitro.In this study,we aimed to assess the efficacy and safety of chloroquine with different doses in COVID-19.In this multicenter prospective observational study,we enrolled patients older than 18 years old with confirmed SARS-CoV-2 infection excluding critical cases from 12 hospitals in Guangdong and Hubei Provinces.Eligible patients received chloroquinephosphate s00 mg,orally,once(half dose)or twice(full dose)daily.Patients treated with non-chloroquine therapy were included as historical controls.The primary endpoint is the time to undetectable viral RNA.Secondary outcomes include the proportion of patients with undetectable viral RNA by day 10 and 14,hospitalization time,duration of fever,and adverse events.A total of 197 patients completed chloroquine treatment,and 176patients were included as historical controls.The median time to achieve an undetectable viral RNA was shorter in chloroquine than in non-chloroquine(absolute difference in medians-6.0 days;95%CI-6.0 to—4.0).The duration of fever is shorter in chloroquine(geometric mean ratio 0.6;95%CIO.5 to 0.8).No serious adverse events were observed in the chloroquine group.Patients treated with half dose experienced lower rate of adverse events than with full dose.Although randomized trials are needed for further evaluation,this study provides evidence for safety and efficacy of chloroquine in COVID-19 and suggests that chloroquine can be a cost-effective therapy for combating the COVID-19 pandemic.Mingxing Huang Man Li Fei Xiao Pengfei Pang Jiabi Liang Tiantian Tang Shaoxuan Liu Binghui Chen Jingxian Shu Yingying You Yang Li Meiwen Tang Jianhui Zhou Guanmin Jiang Jingfen Xiang Wenxin Hong Songmei He Zhaoqin Wang Jianhua Feng Changqing Lin Yinong Ye Zhilong Wu Yaocai Li Bei Zhong Ruilin Sun Zhongsi Hong Jing Liu Huili Chen Xiaohua Wang Zhonghe Li Duanqing Pei Lin Tian Jinyu Xia Shanping Jiang Nanshan Zhong Hong Shan 2020National Science Review2020,7,9:20
2Cropland redistribution to marginal lands undermines environmental sustainability显示文摘Cropland redistribution to marginal land has been reported worldwide;however,the resulting impacts on environmental sustainability have not been investigated sufficiently.Here we investigated the environmental impacts of cropland redistribution in China.As a result of urbanization-induced loss of high-quality croplands in south China(~8.5 t ha^(-1)),croplands expanded to marginal lands in northeast(~4.5 t ha^(-1)) and northwest China(~2.9 t ha^(-1)) during 1990-2015 to pursue food security.However,the reclamation in these low-yield and ecologically vulnerable zones considerably undermined local environmental sustainability,for example increasing wind erosion(+3.47%),irrigation water consumption(+34.42%),fertilizer use(+20.02%) and decreasing natural habitats(-3.11%).Forecasts show that further reclamation in marginal lands per current policies would exacerbate environmental costs by 2050.The future cropland security risk will be remarkably intensified because of the conflict between food production and environmental sustainability.Our research suggests that globally emerging reclamation of marginal lands should be restricted and crop yield boost should be encouraged for both food security and environmental benefits.Wenhui Kuang Jiyuan Liu Hanqin Tian Hao Shi Jinwei Dong Changqing Song Xiaoyong Li Guoming Du Yali Hou Dengsheng Lu Wenfeng Chi Tao Pan Shuwen Zhang Rafiq Hamdi Zherui Yin Huimin Yan Changzhen Yan Shixin Wu Rendong Li Jiuchun Yang Yinyin Dou Wenbin Wu Liqiao Liang Bao Xiang Shiqi Yang 2022National Science Review2022,9,1:13
3Gossypol: phytoalexin of cotton显示文摘Sesquiterpenoids are a class of 15-carbon secondary metabolites that play diverse roles in plant adaptation to environment.Cotton plants accumulate a large amount of sesquiterpene aldehydes(including gossypol)as phytoalexins against pathogens and herbivores.They are stored in pigment glands of aerial organs and in epidermal layers of roots.Several enzymes of gossypol biosynthesis pathway have been characterized,including 3-hydroxy-3-methylglutaryl coenzyme A reductase(HMGR)and farnesyl diphosphate synthase(FPS)that catalyze the formation of the precursor farnesyl diphosphate(FPP),(+)-δ-cadinene synthase(CDN)which is the first enzyme committed to gossypol biosynthesis,and the downstream enzymes of CYP706B1 and methyltransferase.Expressions of these genes are tightly regulated during cotton plants development and induced by jasmonate and fungi elicitors.The transcription factor Ga WRKY1 has been shown to be involved in gossypol pathway regulation.Recent development of new genomic platforms and methods and releases of diploid and tetraploid cotton genome sequences will greatly facilitate the elucidation of gossypol biosynthetic pathway and its regulation.Xiu Tian Juxin Ruan Jinquan Huang Xin Fang Yingbo Mao Lingjian Wang Xiaoya Chen Changqing Yang 2016Science China(Life Sciences)2016,59,2:10

5A fundamental theorem for eco-environmental surface modelling and its applications显示文摘We propose a fundamental theorem for eco-environmental surface modelling(FTEEM) in order to apply it into the fields of ecology and environmental science more easily after the fundamental theorem for Earth’s surface system modeling(FTESM). The Beijing-Tianjin-Hebei(BTH) region is taken as a case area to conduct empirical studies of algorithms for spatial upscaling, spatial downscaling, spatial interpolation, data fusion and model-data assimilation, which are based on high accuracy surface modelling(HASM), corresponding with corollaries of FTEEM. The case studies demonstrate how eco-environmental surface modelling is substantially improved when both extrinsic and intrinsic information are used along with an appropriate method of HASM. Compared with classic algorithms, the HASM-based algorithm for spatial upscaling reduced the root-meansquare error of the BTH elevation surface by 9 m. The HASM-based algorithm for spatial downscaling reduced the relative error of future scenarios of annual mean temperature by 16%. The HASM-based algorithm for spatial interpolation reduced the relative error of change trend of annual mean precipitation by 0.2%. The HASM-based algorithm for data fusion reduced the relative error of change trend of annual mean temperature by 70%. The HASM-based algorithm for model-data assimilation reduced the relative error of carbon stocks by 40%. 8Autologous fibroblasts induce fibrosis of the nucleus pulposus to maintain the stability of degenerative intervertebral discs显示文摘Lumbar degenerative disc diseases cause low back pain(LBP). The maintenance of the height and stability of the intervertebral disc(IVD) space is an effective treatment for LBP. The following study evaluated the effects of fibroblast injection on intervertebral disc degeneration(IDD) in a preclinical setting. Compared with the IDD group, the fibroblast treatment group demonstrated effective maintenance of IVD height, reduced endplate degeneration, and improved nuclear magnetic resonance signals and overall histological structure. In doing so, fibrotic IVDs maintained the stability and biomechanics of the vertebra. This finding is in agreement with clinical findings that human nucleus pulposus(NP) fibrosis is essential for the maintenance of IVD height and mechanical properties in patients following percutaneous endoscopic lumbar discectomy(PELD). Mechanistically, we demonstrated that injected fibroblasts not only proliferated but also induced NP cells to adopt a fibrotic phenotype via the secretion of TGF-β.Finally, to better mimic human conditions, the efficacy of autologous fibroblast injection in the treatment of IDD was further examined in a nonhuman primate cynomolgus monkey model due to their capacity for upright posture. We showed that the injection of fibroblasts could maintain the IVD height and rescue IVD signals in cynomolgus monkeys. Taken together, the results of our study reveal that autologous fibroblast injection can enhance the natural process of fibrosis during acute and subacute stages of stress-induced IDD. 9Disruption of the bHLH transcription factor Abnormal Tapetum 1 causes male sterility in watermelon显示文摘Although male sterility has been identified as a useful trait for hybrid vigor utilization and hybrid seed production,its underlying molecular mechanisms in Cucurbitaceae species are still largely unclear.Here,a spontaneous male-sterile watermelon mutant,Se18,was reported to have abnormal tapetum development,which resulted in completely aborted pollen grains.Map-based cloning demonstrated that the causal gene Citrullus lanatus Abnormal Tapetum 1(ClATM1)encodes a basic helix-loop-helix(bHLH)transcription factor with a 10-bp deletion and produces a truncated protein without the bHLH interaction and functional(BIF)domain in Se18 plants.qRT–PCR and RNA in situ hybridization showed that ClATM1 is specifically expressed in the tapetum layer and in microsporocytes during stages 6–8a of anther development.The genetic function of ClATM1 in regulating anther development was verified by CRISPR/Cas9-mediated mutagenesis.Moreover,ClATM1 was significantly downregulated in the Se18 mutant,displaying a clear dose effect at the transcriptional level.Subsequent dual-luciferase reporter,β-glucuronidase(GUS)activity,and yeast one-hybrid assays indicated that ClATM1 could activate its own transcriptional expression through promoter binding.Collectively,ClATM1 is the first male sterility gene cloned from watermelon,and its self-regulatory activity provides new insights into the molecular mechanism underlying anther development in plants.Ruimin Zhang Jingjing Chang Jiayue Li Guangpu Lan Changqing Xuan Hao Li Jianxiang Ma Yong Zhang Jianqiang Yang Shujuan Tian Li Yuan Xian Zhang Chunhua Wei 2021Horticulture Research2021,8,1:3
10Start-up Characteristics of Linear Compressors in a Refrigeration System显示文摘This work investigates the start-up characteristics of linear compressors in a refrigeration system through experiment and simulation.Experiments are carried out by a refrigeration test system with a linear compressor controlled by a LabVIEW platform.A simulation model that considers the nonlinear process of gas force is set up on the basis of Runge-Kutta method for linear compressors.Compared with the experimental results,the simulation errors are within 15%,including the unstable state.The influences of ambient temperature and power frequency on linear compressors are studied through experiments.Unstable phenomena exist at 25℃ ambient temperature compared with the designed ambient temperature of 35℃.The unsteadiness mechanism is analyzed by simulation.Simulation analysis indicated that two sensitive stages of linear compressors,namely,starting to pump and touching top dead center,are unstable.Furthermore,properly increasing equivalent mass (approximately 3%) or spring stiffness doring the design stage can be a practical method to improve the stability of linear compressors.ZOU Huiming LI Xuan TANG Mingsheng TIAN Changqing 2021Journal of Thermal Science2021,30,2:1
