| 1 | Regulatory effects of antitumor agent matrine on FOXO and PI3K-AKT pathway in castration-resistant prostate cancer cells显示文摘We previously demonstrated that matrine could inhibit the proliferating, migrating, as well as invading processes of both PC-3 and DU145 cells. However, the underlying molecular mechanisms have not yet been clearly defined. In this study, using various techniques such as high throughput sequencing technology, bioinformatics, quantitative real-time PCR, and immunoblot analysis,we aimed to understand whether matrine serves as a novel regulator of FOXO and PI3K-AKT signaling pathway. DU145 and PC-3 cell lines were cultured for 24 h in vitro. Cells were treated with either matrine or control serum for 48 h, followed by extraction of total RNA. The RNA was sequenced using HiSeq 2500 high-throughput sequencing platform (Illumina). A gene library was established and quality analysis of read data carried out. Integrated database from the website DAVID was used to analyze Gene Ontology (GO), and Kyoto encyclopedia of genes and genomes (KEGG) pathway of differential genes was used for pathway analysis, screening for fold differences of more than two times. The FOXO and PI3K-AKT signaling pathways were screened, and expression levels of mRNA and core protein detected by real-time PCR and immunoblotting, respectively. High throughput sequencing and GO analysis revealed that differentially expressed genes before and after treatment played an important role in cell metabolic process, growth process, anatomical structure formation, cellular component organization, and biological regulation. KEGG signal pathway analysis revealed that FOXO and PI3K-AKT signal pathways had a significant difference between before and after matrine-treated androgen-independent prostate cancer cells PC-3 and DU145. Real-time PCR showed that matrine treatment led to a significant increase in the expression levels of FOXO1A, FOXO3A, FOXO4, and FOXO6 in DU145 and PC-3 cells (P<0.01 or P<0.05), whereas the PI3K expression levels decreased (P<0.01). Similarly, immunoblotting revealed a significant increase (P<0.05) in the expression levels of FOXO1A FOXO3A, FOXO4, and FOXO6 in both PC-3 and DU145 cells, whereas PI3K expression levels decreased (P<0.05). Matrine had a broad regulating effect on the mRNA expression profiles of both PC-3 and DU145 cells. Matrine may inhibit cell proliferation, migration, as well as invasion, and induce apoptosis in both PC-3 and DU145 cells through FOXO and PI3K-AKT signaling pathways. | 2 | Targeted Mutagenesis of NAC Transcription Factor Gene,OsN AC041,Leading to Salt Sensitivity in Rice显示文摘Salinity is a major abiotic stress factor that seriously affects plant growth. Many genes are involved in the response to salt stress with various metabolism pathways. A number of plant transcription factor family genes have been found to be involved in the salt stress response, and NAM, ATAF and CUC(NAC) transcription factors are thought to act as active regulators during abiotic stress, especially salt stress. In this study, we detected a rice NAC transcription factor coding gene, OsNAC041, and confirmed that it influenced the germination of seeds under salt stress and salt tolerance of plants. OsNAC041 was primarily expressed in the leaves and located in the nucleus. Furthermore, the CRISPR/Cas9 method was used to obtain a targeted osnac041 mutant, of which the plant height was higher than that of the wild-type, showing increased salt sensitivity. Moreover, RNA-seq analysis revealed a number of differentially expressed genes(DEGs) involved in several important signaling pathways in the osnac041 mutant. Subsequently, Kyoto Encyclopedia of Genes and Genomes annotation also revealed differential expression of DEGs associated with mitogen-activated protein kinase signaling, peroxisome, eukaryotictype ABC transporters, photosynthesis and plant hormones, which are involved in stress-related signaling pathways. Overall, our study suggested that OsNAC041 was involved in the salt stress response in rice. | 3 | One-step synthesis of monodispersed Pt nanoparticles anchored on 3D graphene foams and its application for electrocatalytic hydrogen evolution显示文摘Although platinum-based materials are regarded as the state-of-the-art electro-catalysts for hydrogen evolution reaction(HER),high cost and quantity scarcity hamper their scale-up utilization in industrial deployment.Herein,a one-step strategy was developed to synthesize multi-walled carbon nanotubes and reduced graphene oxide supported Pt nanoparticle hydrogel(PtNP/rGO-MWCNT),in which only ascorbic acid was used as the reductant for one-pot reduction of both GO and chloroplatinic acid.The hydrogel can be directly used as a flexible binder-free catalytic electrode to achieve high performance of HER.Compared to conventional strategies,the current strategy not only significantly reduces the Pt loading to 3.48 wt%,simplifies the synthesis process,but also eliminates the use of any polymer binders,thus decreasing the series resistance and improving catalytic activity.An overpotential of only 11 mV was achieved on as-prepared PtNP/rGO-MWCNT to drive a geometrical current density of 10 mA/cm2 in0.5 mol/L H2 SO4,with its catalytic activity being kept over 15 h.In acidic medium,the HER activity of the PtNP/rGO-MWCNT catalyst exceeds most of the reported Pt-based electro-catalysts and is 3-fold higher than that obtained on commercial Pt/C electrode. | Dan Zhou Bei Jiang Rui Yang Xiandeng Hou Chengbin Zheng | 2020 | Chinese Chemical Letters2020,31,6: | 3 |

| 9 | Ordered Mesoporous Intermetallic PtP_(2) Nanoparticles with Enhanced Electrocatalytic Activity and Stability for Hydrogen Evolution显示文摘Ordered mesoporous noble metals(NMs)have displayed unique catalytic and electrocatalytic performance distinctive from traditional nanoparticles.Despite great efforts,the range of mesoporous NMs is mainly limited to single metals and their metal alloys with random atomic arrangements.Herein,we report a simple solid-phase synthesis of novel mesoporous intermetallic noble metal-nonmetal(MI-PtX_(2))nanoparticles with hierarchical orderliness as highly efficient electrocatalysts for hydrogen evolution reaction(HER).The synthesis relies on a Pt-to-PtX_(2) evolution with mesoporous Pt confined in thermally stable KIT-6(Korea Advanced Institute of Science and Technology-6)as the concurrent template.Meanwhile,the method could be extended readily to control structures and compositions of mesoporous intermetallic nanoparticles such as hollow structures and ternary MI-PtMP_(2).Samples featured rhombic dodecahedral morphology,ordered gyroid mesostructure,and cubic/hexagonal intermetallic phase,producing abundant undercoordinated sites and optimized surface electronic structures.These features kinetically accelerate H_(2)O dissociation to remarkably enhance electrocatalytic HER performance.The optimum MI-PtP_(2) disclosed ultrahigh mass/specific activity(3.31 A mg_(pt)^(−1)/7.75 mA cm_(pt)^(−2))and superior stability(only 15.2%of mass activity loss after an accelerated durability test for 30,000 cycles),suppressing the reported electrocatalysts.Our work opens up new opportunities for designing and synthesizing novel hierarchically ordered mesoporous electrocatalysts with targeted functions for a variety of applications. | Yanzhi Wang Lizhi Sun Hao Lv Chengbin Zheng Ben Liu | 2023 | CCS Chemistry2023,5,8: | 0 |
| 10 | Reduction of mercury(Ⅱ)by electrons contained in carbon dots:An environmentally friendly cold vapor generation for mercury analysis显示文摘In this work,the reduction of mercury ions(Hg2+)to elemental mercury(Hg0)was easily achieved using highly reductive carbon dots(r-CDs),which synthesized from sucrose by a simple and cost-effective method.After a careful mechanistic study,the reduction was probably accomplished with the large numbers of electrons contained in r-CDs rather than the oxidation of its functional groups.Additionally,a3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay showed that the r-CDs were nontoxic to wildlife and human beings.Consequently,the r-CDs were used as an alternative to toxic reductants(SnCl2 or NaBH4)for the sensitive and in situ determination of mercury by cold vapor gene ration(CVG)coupled to a miniature point discharge optical emission spectrometer(μPD-OES).Limit of detection of 0.05μg/L was obtained for Hg2+,with relative standard deviation(RSD)less than 5.4%at a concentration of 5μg/L.The accuracy of r-CDs induced CVG-μPD-OES was validated by the determination of mercury in a certified reference material(DOLT-5,dogfish liver)and five natural water samples collected from different rivers and lakes in Chengdu City,Since r-CDs are nontoxic and prepared from abundant and inexpensive sucrose,the r-CDs induced CVG-μPD-OES retains the great potential for the inexpensive and enviro nmentally friendly field analysis of mercury in natural water.The accuracy of the proposed method was validated by the analysis of a certified reference material and several water samples with satisfactory results. | Tao Chen Yao Lin Haochen Li Rui Yang Xiandeng Hou Baozhan Zheng Chengbin Zheng | 2020 | Chinese Chemical Letters2020,31,10: | 0 |
| 11 | Inhibition of MALT1 paracaspase activity improves lesion recovery following spinal cord injury显示文摘Spinal cord injury(SCI) is a devastating traumatic injury that causes persistent, severe motor and sensory dysfunction. Immune responses are involved in functional recovery after SCI. Mucosa-associated lymphoid tissue lymphoma translocation 1(MALT1) has been shown to regulate the survival and differentiation of immune cells and to play a critical role in many diseases, but its function in lesion recovery after SCI remains unclear. In this paper, we generated KI(knock in) mice with a point mutation(C472 G) in the active center of MALT1 and found that the KI mice exhibited improved functional recovery after SCI.Fewer macrophages were recruited to the injury site in KI mice and these macrophages differentiated into anti-inflammatory macrophages. Moreover, macrophages from KI mice exhibited reduced phosphorylation of p65, which in turn resulted in decreased SOCS3 expression and increased pSTAT6 levels.Similar results were obtained upon inhibition of MALT1 paracaspase with the small molecule inhibitor‘‘MI-2' or the more specific inhibitor ‘‘MLT-827'. In patients with SCI, peripheral blood mononuclear cells(PBMC) displayed increased MALT1 paracaspase. Human macrophages showed reduced proinflammatory and increased anti-inflammatory characteristics following the inhibition of MALT1 paracaspase.
