| 3 | Deficiency of two-pore segment channel 2 contributes to systemic lupus erythematosus via regulation of apoptosis and cell cycle显示文摘Background:Systemic lupus erythematosus(SLE)is a complex autoimmune disease,and the mechanism of SLE is yet to be fully elucidated.The aim of this study was to explore the role of two-pore segment channel 2(TPCN2)in SLE pathogenesis.Methods:Quantitative reverse transcription polymerase chain reaction(qRT-PCR)was used to detect the expression ofTPCN2 in SLE.We performed a loss-of-function assay by lentiviral construct in Jurkat and THP-1 cell.Knockdown ofTPCN2 were confirmed at the RNA level by qRT-PCR and protein level by Western blotting.Cell Count Kit-8 and flow cytometry were used to analyze the cell proliferation,apoptosis,and cell cycle ofTPCN2-deficient cells.In addition,gene expression profile ofTPCN2-deficient cells was analyzed by RNA sequencing(RNA-seq).Results:TPCN2 knockdown with short hairpin RNA(shRNA)-mediated lentiviruses inhibited cell proliferation,and induced apoptosis and cell-cycle arrest of G2/M phase in both Jurkat and THP-1 cells.We analyzed the transcriptome of knockdown-TPCN2-Jurkat cells,and screened the differential genes,which were enriched for the G2/M checkpoint,complement,and interleukin-6-Janus kinase-signal transducer and activator of transcription pathways,as well as changes in levels of forkhead box O,phosphatidylinositol 3-kinase/protein kinase B/mechanistic target of rapamycin,and T cell receptor pathways;moreover,TPCN2 significantly influenced cellular processes and biological regulation.Conclusion:TPCN2 might be a potential protective factor against SLE. | Keke Li Jingkai Xu Ke Xue Ruixing Yu Chengxu Li Wenmin Fei Xiaoli Ning Yang Han Ziyi Wang Jun Shu Yong Cui | 2022 | Chinese Medical Journal2022,,4: | 2 |

| 6 | Recrystallization nucleation under close-setδphase in a nickel-based superalloy during annealing显示文摘Annealing is one of the most efficient heat treatment to refine the grain microstructure in metal alloys.Prior to the annealing,an aging treatment can be implemented to precipitate the second phases for more nucleation sites in deformed mixed grains during subsequent recrystallization.However,the underlying mechanism behind the second phases and recrystallization has not yet been fully understood.Taking a typical deformed nickel(Ni)-based(GH4169)superalloy as an exapmle,this research aims to clairfy the connections between the recrystallization and delta(δ)phase(Ni_(3)Nb) during annealing.All deformed samples were also treated by aging and then annealing.The results show that the microstructure and component ofδphase dominated the recrystallization nucleation in the Ni-based alloy.Moreover,there was the presence of theδphase interval limiting nucleation(PILN)andδphase protecting nucleation(PPN),when a high content of close-set phase was precipitated from the matrix.The PILN determined the primary condition for the recrystallization nucleation that happended at the interval ofδphase larger than the critical nucleation size.The peak nucleation occurred in the PPN,where the recrystallization nucleus were free of the penetration by the adjacent grains,before the nucleation incubation time was reached.Thus,a new feasible JNtheory was proposed to determine the maximum domain of recrystallization nucleation under close-set precipitated phases.All the possible nucleation sites were assumed under PPN in the JNtheory.As such,the original microstructure can be expected to design for the annealing via the aging process.A quantitative evaluation of recrystallization nucleation can also achieved in the Ni-based alloys during aging. | Guanqiang Wang Mingsong Chen Yongcheng Lin Hongbin Li Yuqiang Jiang Yanyong Ma Chengxu Peng Jinliang Cai Quan Chen | 2022 | Journal of Materials Science & Technology2022,,20: | 1 |
| 7 | The effect of soil moisture on the response by fungi and bacteria to nitrogen additions for N_(2)O production显示文摘In addition to bacteria,the contribution of fungi to nitrous oxide(N_(2)O)production has been recognized but the responses of these two broad and unrelated groups of microorganisms to global environmental changes,atmospheric nitrogen(N)deposition,and precipitation in terms of N_(2)O production are unclear.We studied how these two microbial-mediated N_(2)O production pathways responded to soil moisture conditions and to N addition in an N-limited temperate forest.Soils from a long-term N addition experiment in Changbai Mountain,northeastern China were incubated.Varied concentrations of cycloheximide and streptomycin,both inhibitors of fungal and bacterial activity,were used to determine the contributions of both to N_(2)O production in 66%,98%and 130%water-filled pore spaces(WFPS).The results showed that N_(2)O production decreased significantly with increasing cycloheximide concentration whereas streptomycin was only inhibiting N_(2)O emissions at 98%and 130%WFPS.The bacterial pathway of N_(2)O production in N-addition(Nadd)soil was significantly more dominant than that in untreated(Namb)soil.The difference in the fungal pathway of N_(2)O production between the soil with nitrogen addition and the untreated soil was not significant.Net N_(2)O emissions increased with increasing soil moisture,especially at 130%WFPS,a completely flooded condition.Bacteria dominated carbon dioxide(CO_(2))and N_(2)O emissions in Nadd soil and at 130%WFPS regardless of N status,while fungi dominated CO_(2)and N_(2)O emissions in soil without N addition at 66%and 98%WFPS.The results suggest that flooded soil is an important source of N_(2)O emissions and that bacteria might be better adapted to compete in fertile soils under anoxic conditions. | Lei Zhang Junqiang Zheng Xu Han Junhui Zhang Chengxu Li Shicong Geng Shijie Han | 2021 | Journal of Forestry Research2021,32,5: | 1 |

| 12 | Isolation of Chloroplasts from Marine Microalga Isochrysis galbana Parke for Their Lipid Composition Analysis显示文摘Marine microalga Isochrysis galbana is an important feed species with a high nutritional value.Different from other uni-cellular algae,its cell contains two chloroplasts which are the major sites for lipid synthesis.Here,we optimized a chloroplast isola-tion approach suitable for the isolation of I.galbana chloroplasts and determined the purity and integrity of the isolated chloroplasts through microscopic observations and enzyme activity assay.The chloroplast lipids were analyzed with a ultrahigh-performance li-quid chromatography-Q Exactive Orbitrap-mass spectrometry.This newly developed isolation approach is simple and reliable to isolate chloroplasts with high integrity and purity.The average yield of intact chloroplasts was 15.3%±0.1%.Glycolipids and acyl-glycerols were the main chloroplast lipids.Glycolipids accounted for 56.6%of chloroplast lipid.Digalactosyldiacylglycerol(DGDG),monogalactosyldiacylglycerol(MGDG)and sulfoquinovosyldiacylglycerol(SQDG)were the main glyceroglycolipids.The fatty acyl R1/R2 were mostly 18:4/16:1,18:3/16:1 and 18:4/18:5 in DGDGs,14:0/18:4,18:4/18:5,18:4/18:4 and 18:3/18:4 in MGDGs and 16:0/14:0,16:0/18:3,and 18:4/18:3 in SQDGs.In addition,diacylglycerol(DAG)was the most abundant acylglycerols;the content of 22:6/18:4-DAG was the highest.There was a little amount of glycosphingolipid(GSL)in chloroplast.Digalactosylmonoglyceride(DGMG),monogalactosylmonoglyceride(MGMG),sulfoquinovosylmonoacylglycerol(SQMG),monoglyceride(MAG),phospholi-pids(PLs),ceramide(Cer)and betaine lipids were nearly undetectable in chloroplast.The fatty acid proportions of DGDGs,MGDGs,SQDGs,DAGs,triglycerides(TAGs)and GSLs were either higher or lower than or similar to those of whole-cell.Collectively,our isolation approach is applicable to many aspects of chloroplast biology,and may offer a reference for the isolation of chloroplasts from other marine microalgae. | ZHONG Ye LI Yanrong XU Jilin CAO Jiayi ZHOU Chengxu YAN Xiaojun | 2022 | Journal of Ocean University of China2022,21,1: | 1 |
| 13 | Few-layered MoS_(2)anchored on 2D porous C_(3)N_(4)nanosheets for Ptfree photocatalytic hydrogen evolution显示文摘The Pt-free photocatalytic hydrogen evolution(PHE)has been the focus in the photocatalytic field.The catalytic system with the large accessible surface and good mass-transfer ability,as well as the intimate combination of co-catalyst with semiconductor is promising for the promotion of the application.Here,we have reported the design of the two-dimensional(2D)porous C_(3)N_(4)nanosheets(PCN NS)intimately combined with few-layered MoS_(2)for the high-effective Pt-free PHE.The PCN NS were synthesized based on peeling the melamine–cyanuric acid precursor(MC precursor)by the triphenylphosphine(TP)molecular followed by the calcination,mainly due to the matched size of the(100)plane distance of the precursor(0.8 nm)and the height of TP molecular.The porous structure is favorable for the mass-transfer and the 2D structure having large accessible surface,both of which are positive to promote the photocatalytic ability.The few-layered MoS_(2)are grown on PCN to give 2D MoS_(2)/PCN composites based on anchoring phosphomolybdic acid(PMo_(12))cluster on polyetherimide(PEI)-modified PCN followed by the vulcanization.The few-layered MoS_(2)have abundant edge active sites,and its intimate combination with porous PCN NS is favorable for the faster transfer and separation of the electrons.The characterization together with the advantage of 2D porous structure can largely promote the photocatalytic ability.The MoS_(2)/PCN showed good PHE activity with the high hydrogen production activity of 4,270.8μmol·h^(−1)·g^(−1)under the simulated sunlight condition(AM1.5),which was 7.9 times of the corresponding MoS_(2)/bulk C_(3)N_(4)and 12.7 times of the 1 wt.%Pt/bulk C_(3)N_(4).The study is potentially meaningful for the synthesis of PCN-based catalytic systems. | Nan Wang Dongxu Wang Aiping Wu Siyu Wang Zhihui Li Chengxu Jin Youming Dong Fanyi Kong Chungui Tian Honggang Fu | 2023 | Nano Research2023,16,2: | 1 |
| 14 | Analysis of the track system in bumpy unstructured hard road environment by vibration test显示文摘The complex terrain environment in the hilly land directly affects the operational reliability of agricultural robots.In order to study the impact of road irregularity on walking chassis vibration,the 3CYLZ-750 remote-controlled weeding machine which is applied to orchards was taken as the object of study,and the rear roller was selected as the object of observation to reveal the rules under which the vibration of the track chassis changes as there is a sudden change in road surface elevation.A column-type test-to-pass method based on unit excitation was proposed in this study.The excitation behavior and action process were analyzed by category.A critical acceleration prediction model was built and verified by virtual simulation and hard road surface excitation testing.The results showed that at the forward velocity of 0-2.5 km/h and exciter height of 20-100 mm,the vertical vibration acceleration of the target roller was significantly affected by Track Contact Point Centrifugal Acceleration(TCPCA).As TCPCA increased,the change rate of vertical vibration acceleration decreased,reaching a minimum of[−13.8,28.8];as TCPCA decreased,the vertical vibration acceleration tended to increase positively at a maximum variation range of[−13.3,42.2].The measured and simulated macroscopic change rules were consistent with the theoretical analysis,further verifying the correctness of variable extraction,and providing a research basis for the accurate modification and improvement of the model.The research conclusions can lay a theoretical foundation for analyzing the walking reliability of the track chassis,and provide a design basis and technical support for the development of a tracked agricultural robot chassis for the hilly land in the future. | Mutong Li Junlue Li Lin He Jie He Lian Hu Chengxu Lyu | 2022 | International Journal of Agricultural and Biological Engineering2022,15,4: | 1 |

| 17 | Summarization and comparison of dermoscopic features on different subtypes of rosacea显示文摘Background:The dermoscopic features of rosacea have already been reported.However,the current findings are incomplete,and little is known about phymatous rosacea.Hence,this study aimed to summarize and compare the dermoscopic features and patterns of three rosacea subtypes(erythematotelangiectatic[ETR],papulopustular[PPR],and phymatous[PHR])in the Chinese Han population and to evaluate whether these features differ with patients’genders,ages,and durations.Methods:Dermoscopic images of 87 rosacea patients were collected in non-polarized and polarized dermoscopy contact modes at 20-fold magnification.Dermoscopic features,including vessels,scales,follicular findings,and other structures,were summarized and evaluated.Results::The reticular linear vessels and red diffuse structureless areas of ETR were distinctive.For PPR,red diffuse structureless areas,reticular linear vessels,yellow scales,follicular plugs,and follicular pustules were typical dermoscopic criteria.The common dermoscopic features of PHR were:orange diffuse structureless areas,linear vessels with branches,perifollicular white color,orange focal structureless areas,and white lines.The following features statistically differed among the three rosacea subtypes:reticular linear vessels(P<0.001),unspecific linear vessels(P=0.005),linear vessels with branches(P<0.001),yellow scales(P=0.001),follicular plugs(P<0.001),perifollicular white color(P<0.001),red diffuse structureless areas(P=0.022),orange diffuse structureless areas(P<0.001),red focal structureless areas(P=0.002),orange focal structureless areas(P=0.003),white lines(P<0.001),follicular pustules(P<0.001),and black vellus hairs(P<0.001).Conclusions:The dermoscopic patterns of ETR are red diffuse structureless areas and reticular linear vessels.For PPR,the pattern comprehends combinations of red diffuse structureless areas,reticular linear vessels,yellow scales,follicular plugs,and follicular pustules.Meanwhile,PHR is characterized by remarkable orange diffuse structureless areas,linear vessels with branches,perifollicular white color,orange focal structureless areas,and white lines. | Wenmin Fei Yang Han Ang Li Keke Li Xiaoli Ning Chengxu Li Wenju Wang Rusong Meng Yong Cui | 2022 | Chinese Medical Journal2022,,12: | 1 |

| 20 | ERCC1对胃癌患者手术及化疗后生存预测的意义(英文)显示文摘Objective:The aim of this study was to evaluate the effect of the excision repair cross-complementing(ERCC1) expression on survival in advanced gastric cancer patients who underwent surgical resection and treated with oxaliplatin-based adjuvant chemotherapy.Methods:Sixty-three patients who underwent surgical resection for cure and treated with oxaliplatin-based adjuvant chemotherapy were included in this study.The expressions of ERCC1 of gastric cancer were examined by immunohistochemistry and the patients were categorized into ERCC1-(+) and ERCC1-(-) groups.The relation between ERCC1 expression and survival of patients was examined.Results:Of the 63 eligible patients,36 patients(57.1%) had tumor with a positive expression of ERCC1 and the remaining 27 patients had tumor with a negative ERCC1 expression.Expression differences of ERCC1 didn't correlated with age(P = 0.827),gender(P = 0.12),differentiation(P = 0.113),histological type(P = 0.942),site of tumor(P = 0.221),size of tumor(P = 0.608),stage(P = 0.815) and lymphatic invasion(P = 0.165).Overall survival(OS) was significantly longer in patients without ERCC1 expression,when compared to patients with ERCC1 expression(P = 0.023).Multivariate analysis revealed that ERCC1 expression significantly impacted on OS(HR:4.049;P = 0.000).Conclusion:We concluded that resected and treated with oxaliplatin-based adjuvant chemotherapy gastric cancer patients without ERCC1 expression have a better survival when compared to patients with ERCC1 expression.ERCC1 expression will hopefully provide a rational basis for improving adjuvant chemotherapeutic strategies for gastric cancer patients.ERCC1,itself,may be a prognostic factor for gastric cancer. | Qiang Li Chengxue Dang Zhigang Liu Ruifang Sun | 2011 | The Chinese-German Journal of Clinical Oncology2011,10,2: | 0 |
