7Immobilization of acetylcholinesterase on one-dimensional gold nanoparticles for detection of organophosphorous insecticides显示文摘This paper reports a simple method for immobilization of acetylcholinesterase (AChE) on one-dimensional (1D) gold (Au) nanoparticles for detection of organophosphorous (OP) insecticides. 1D Au nanoparticles were prepared by electrodeposition in the pores of an alumina template which was subsequently removed by 2.0 M NaOH solution. They were characterized by XRD and FESEM. The immobilized AChE retained its biological activity and catalyzed the hydrolysis of acetylthiocholine to form thiocholine, which was subsequently oxidized to produce detectable signals. Based on the inhibition toward the enzymatic activity of AChE by OP insecticides, sensitive detection of methamidophos (an OP insecticide) was performed. Under optimal conditions, the sensors could be used for the determination of methamidophos ranging from 0.004 to 24 μg/mL with the detection limit of 0.001 μg/mL. The developed OP insecticide biosensors exhibited satisfactory stability and reproducibility. 8Contents of soil organic carbon and nitrogen in water-stable aggregates in abandoned agricultural lands in an arid ecosystem of Northwest China显示文摘Soil organic matter content in water-stable aggregates(WSA) in the arid ecosystems(abandoned agricultural lands especially) of China is poorly understood. In this study, we examined the WSA sizes and stability, and soil organic carbon(OC) and nitrogen(N) contents in agricultural lands with abandonment ages of 0, 3, 12, 20, 30 and 40 years, respectively, in the Minqin Oasis of Northwest China. The total soil OC and N contents at depths of 0–20, 20–40 and 40–60 cm in abandoned agricultural lands were compared to those in cultivated land(the control). Agricultural land abandonment significantly(P<0.05) influenced the distribution of MWD(mean weight diameter), and OC and N contents. There were significant increases in MWD and the proportion of macroaggregates(sizes >0.25 mm) as the age of agricultural land abandonment increased. The effect of abandonment ages of agricultural lands on MWD was determined by the changes of OC and N accumulation in WSA sizes >2 mm. The total OC and N contents presented a stratification phenomenon across soil depths in this arid ecosystem. That is, both of them decreased significantly at depths of 0–20 and 40–60 cm while increased at the depth of 20–40 cm. The WSA sizes <0.053 mm had the highest soil OC and N contents(accounting for 51.41%–55.59% and 42.61%–48.94% of their total, respectively). Soil OC and N contents in microaggregates(sizes 0.053–0.25 mm) were the dominant factors that influenced the variations of total OC and N contents in abandoned agricultural lands. The results of this study suggested that agricultural land abandonment may result in the recovery of WSA stability and the shifting of soil organic matter from the silt+clay(<0.053 mm) and microaggregate fractions to the macroaggregate fractions. 9Apelin activates the expression of inflammatory cytokines in microglial BV2 cells via PI-3K/Akt and MEK/Erk pathways显示文摘This paper aims to observe the changes of the inflammatory cytokines in microglial BV2 cells stimulated by apelin, and investigate the mechanism of inflammatory cytokines secretion after apelin stimulation. Immunofluorescence and quantitative real-time PCR were performed to observe expression of TNF-α, IL-1β, IL-10, MIP-1α, and MCP-1 in BV2 cells. Western blot was used to investigate the expression of phosphorylation PI-3K/Akt and phosphorylation Erk signaling pathways in BV2 cells after stimulation by apelin. Furthermore, PI-3K/Akt inhibitor(LY294402) and Erk inhibitor(U0126) were used as antagonists to detect the secretion mechanisms of cytokines in BV2 cells stimulated by apelin. Exogenous recombinant apelin activated the expression of TNF-α, IL-1β, MCP-1 and MIP-1α in BV2 cells by the detection of fluorescence expression and m RNA. Apelin also unregulated the protein expression of p-PI-3K/Akt and p-Erk in BV2 cells induced by apelin. LY294002 and U0126 inhibited activation of p-PI-3K/Akt and p-Erk expression by Western blot and attenuated the expression of inflammation factors in BV2 cells by fluorescence staining. 10Protective effect of oxysophoridine on cerebral ischemia/reperfusion injury in mice显示文摘Oxysophoridine, a new alkaloid extracted from Sophora alopecuroides L., has been shown to have a protective effect against ischemic brain damage. In this study, a focal cerebral ischemia/reperfusion injury model was established using middle cerebral artery occlusion in mice. Both 62.5, 125, and 250 mg/kg oxysophoridine, via intraperitoneal injection, and 6 mg/kg nimodipine, via intragastric administration, were administered daily for 7 days before modeling. After 24 hours of reperfusion, mice were tested for neurological deficit, cerebral infarct size was assessed and brain tissue was collected. Results showed that oxysophoridine at 125, 250 mg/kg and 6 mg/kg nimodipine could reduce neurological deficit scores, cerebral infarct size and brain water content in mice. These results provided evidence that oxysophoridine plays a protective role in cerebral ischemia/reperfusion injury. In addition, oxysophoridine at 62.5, 125, and 250 mg/kg and 6 mg/kg nimodipine increased adenosine-triphosphate content, and decreased malondialdehyde and nitric oxide content. These compounds enhanced the activities of glutathione-peroxidase, superoxide dismutase, catalase, and lactate dehydrogenase, and decreased the activity of nitric oxide synthase. Protein and mRNA expression levels of N-methyl-D-aspartate receptor subunit NR1 were markedly inhibited in the presence of 250 mg/kg oxysophoridine and 6 mg/kg nimodipine. 11Gene characterization and molecular pathway analysis of reverse thermosensitive genic male sterility in eggplant(Solanum melongena L.)显示文摘The naturally occurring mutant eggplant line 05ms was identified with reverse thermosensitive genic male sterility(rTGMS),but its temperature-responsive fertility mechanisms remain largely unknown.Here,we studied the flower morphology,anther cellular structure,and genome-wide gene expression of this rTGMS line.Candidate genes for thermosensitive male sterility during the microspore development of 05ms and the temperature-insensitive line S63 under low-temperature(LT)and high-temperature(HT)conditions were identified.Under LT,tapetum cells were vacuolated and had delayed disintegration in 05ms.RNA-seq analysis indicated that DEGs were enriched in the KEGG pathways‘plant hormone signal transduction’,‘starch and sucrose metabolism’,and‘phenylpropanoid biosynthesis’.We identified two genes,4CLL1(Sme2.5_00368.1_g00010.1)and CKI1(Sme2.5_10056.1_g00002.1),which could potentially regulate eggplant anther development and may be candidate genes for rTGMS.Finally,we propose a working model of anther abortion for rTGMS in eggplant.CKI1 responds to LT stress and causes expression changes in genes related to anther development,such as 4CLL1,and the cellular structure of the tapetum becomes abnormal,causing male sterility.The findings of this study explain the underlying molecular mechanisms of male sterility in eggplant rTGMS lines.Bing Li Xueping Chen Yanrong Wu Aixia Gu Jingjing Zhang Shuangxia Luo Xiurui Gao Jianjun Zhao Xiuqing Pan Shuxing Shen 2019Horticulture Research2019,6,1:5
12Sand-fixing characteristics of Carex brunnescens and its application with straw checkerboard technique in restoration of degraded alpine meadows显示文摘Carex brunnescens(Pers.), a typical clonal species, is considered to be the only herb found to date that can develop on sand dunes in Maqu alpine region of northwestern China. However, the characteristics that C. brunnescens resists to harsh alpine environment have not been documented. In this study, we conducted a field investigation to determine the morphological, reproductive, and sand-fixing characteristics of C. brunnescens. Concomitantly, we transplanted the belowground rhizomes of C. brunnescens to sand dunes and compared the abilities to restore degraded alpine meadows among sand dunes that had no further treatment(SD+N), sand dunes that had straw checkerboard technique but no transplanted rhizomes of C. brunnescens(SD+SCT), and sand dunes that had both SCT and transplanted rhizomes of C. brunnescens(SD+SCT+P). We found that belowground vertical rhizomes and horizontal rhizomes(including branching rhizomes and main rhizomes) of C. brunnescens were highly developed and that population reproduction was dominated by horizontal rhizomes. C. brunnescens exhibited a significant sand-fixation effect under following conditions: population density was 145–156 ramets/m^2, vegetation cover was 31.2%–39.3%, total length of belowground rhizomes was 11,223 cm/m^2, total length of belowground first-order roots was 9161–10,524 cm/m^2, fresh weight of aboveground part was 198.5–212.6 g/m^2, and fresh weight of belowground part was 578.8–612.4 g/m^2. It should be particularly noted that SD+SCT+P treatment(sand dunes that had both straw checkerboard technique and transplanted rhizomes of C. brunnescens) was the best and SD+N(sand dunes that had no further treatment) was the worst in terms of following biotic indicators: total number of reproductive ramets, total number of belowground rhizomes, and fresh weight of aboveground and belowground parts of C. brunnescens, contents of soil organic carbon, available nitrogen, microbial biomass carbon, and microbial biomass nitrogen. It implies that applying SCT in sand dunes and transplanting belowground rhizomes to sand dunes with SCT could improve both soil fertility and growth of C. brunnescens. 15An animal model of cerebral palsy induced by prenatal exposure to lipopolysaccharide and hypoxia显示文摘BACKGROUND:Neonatal cerebral palsy is mainly caused by prenatal factors.At present, an animal model of prenatal infection and early postnatal hypoxia does not exist.OBJECTIVE:To observe morphology and motor performance following prenatal infection and hypoxic insult-induced brain damage of neonatal rats to verify the feasibility to establish a model of cerebral palsy.DESIGN, TIME AND SETTING:A randomized, controlled, animal experiment was performed at the Laboratories of Xinjiang Center for Disease Control and Prevention from September 2007 to June 2008.MATERIALS:The hypoxic incubator was purchased from Shanghai Pediatric Medical Institute, China.Lipopolysaccharide (LPS, Escherichia coli, O55:B5) was purchased from Sigma-Aldrich (St.Louis, MO, USA).METHODS:A total of 27 Wistar rats, aged 7 days, were randomly assigned to sham-surgery group (n=15) with no carotid artery incision or hypoxia treatment, hypoxia/ischemia (H/I) group (n=12) undergoing ligature of the right common carotid artery followed by exposure to hypoxia at postnatal day 7 (P7), and LPS/H group (n=19), in which pregnant rats were exposed in utero to LPS followed by prenatal hypoxia at embryonic day 16.MAIN OUTCOME MEASURES:Behavior, compound muscle action potential, and pathological changes were observed in 28-day-old rats.RESULTS:The footprint repeat space showed that left limb footprint repeatability in the H/I and LPS/H groups was lower than in the sham-surgery group (P<0.05).The space between the footprints was larger and unstable.Hind limb quadricep compound muscle action potential in the H/I and LPS/H groups showed lower wave amplitude compared with the sham-surgery group (P<0.05).Hematoxylin and eosin staining showed irregular cells around the ventricle, as well as periventricular leukomalacia.CONCLUSION:An animal model of cerebral palsy was established, which simulated the human condition most likely associated with occurrence of this disease.This model could be used for experimental studies related to cerebral palsy.Gang Chen Yanrong HU Wei Liu Jiang Li Linbao Wen Jianxin Li Lihui Zhao Xiaopeng Yang Yi Zhu Zhenzhu Sun Guangming Chi 2010Neural Regeneration Research2010,5,14:4

17Influences of Film Thickness on the Electrical Properties of TaN_x Thin Films Deposited by Reactive DC Magnetron Sputtering显示文摘TaNx thin films were deposited on commercial polished Al2O3 ceramic substrates by reactive dc magnetron sputtering. The influences of the film thickness on the electrical properties of the samples were examined in detail. It is found that the film thickness does not influence the phase structures of the TaNx thin films. The sheet resistances of the samples shift from 173 Ω/sq. to 7.5 Ω/sq. with the film thickness shifting from 30 nm to 280 nm. With the increase of the film thickness from 30 nm to 280 nm, the temperature coefficient of resistance (TCR) of the samples shifts from negative value to positive value. When the film thickness is about 100 nm, TaNx thin films exhibits a near-zero TCR value (approximately -15×10-6/℃). This fact implies that TaNx thin films with a null TCR can be obtained by adjusting the film thickness. 18Recoverable Mn~Ⅲ (salen) supported on diamine modified zirconium poly(styrene-isopropenyl phosphonate)-phosphate as an efficient catalyst for epoxidation of unfunctionalized olefins显示文摘Chiral Mn Ⅲ (salen) (Jacobsen's catalyst) was axially immobilized onto a new type of organic polymer-inorganic hybrid materialzirconium poly(styrene-isopropenyl phosphonate)-phosphate(ZPS-IPPA) with different linkage lengths and evaluated as catalysts for the epoxidation of unfunctionalized olefins. The results demonstrated that the prepared catalysts exhibited moderate to good activity and enantioselectivity in the asymmetric epoxidation of unfunctionalized olefins. Furthermore, the immobilized catalysts were relatively stable and could be conveniently separated from the reaction system by simple precipitation in hexane. Moreover, higher enantioselectivity was obtained with catalyst 2c than that of homogeneous counterpart catalyzed even after eight times. 19Genetic diversity of the wild Asian lotus (Nelumbo nucifera) from Northern China显示文摘Asian lotus(Nelumbo nucifera Gaertn.)is an aquatic plant with ornamental,cultural,economic,and ecological values.China has abundant germplasm resources of Asian lotus.However,in many areas,the wild Asian lotuses have been destroyed and the germplasms is now facing extinction.In addition,the knowledge of the genetic diversity of the wild Asian lotus in China is poor.To identify and protect the germplasms of Chinese wild Asian lotus,eleven genomic-SSR primers,three EST-SSR primers,and three chloroplast DNA primers were used to investigate the genetic diversity among 69 samples of wild Asian lotus from 25 locations in northern China.The genetic diversity of 27 samples of wild Asian lotus from southern China and other countries,the ancient Asian lotus,Asian lotus cultivars from China,and Asian-American hybrids was also compared.The genetic diversity of the wild Asian lotus from northern China was characterized as medium,and themean values of observed heterozygosity(Ho)and expected heterozygosity(He)were 0.087 and 0.552,respectively.Based on a UPGMA dendrogram and STRUCTURE analysis,the wild Asian lotus samples in northern China were divided into three groups.The wild Asian lotus samples from northern China contained 16 haplotypes.The Nei’s genetic distance between the wild Asian lotus samples from the Songhua River basin and the Liao River basin in northeastern China was relatively small,and these germplasms might be relatively primitive compared to those from other regions.This study provides essential information regarding the genetic diversity of the wild Asian lotus resources in northern China,and provides a basis for further analysis of population-level genetic evolution through high-throughput sequencing.Yanrong Fu Fengluan Liu Shuo Li Daike Tian Li Dong Yuchu Chen Yan Su 2021Horticultural Plant Journal2021,7,5:3
20Preparation and characterization of microcrack-free thick YBa_2Cu_3O_(7-δ) films显示文摘High quality epitaxial YBa2Cu3O7-δ (YBCO) superconducting films were fabricated on (00l) LaAlO3 substrates using the direct-current sputtering method. The attainment of an unusually high film thickness (up to 2.0 μm) without mi-crocracking was attributed in part to the presence of pores correlated with yttrium-rich composition in the films. The influ-ence of the film thickness on the microstructure was investigated by X-ray diffraction conventional scan (θ-2θ,ω-scan,pole figure) and high-resolution reciprocal space mapping. The films were c-axis oriented with no a-axis-oriented grains up to the thickness of 2 μm. The surface morphology and the critical current density (Jc) strongly depended on the film thickness.
