Hsa-miR-1246, hsa-miR-320a and hsa-miR-196b-5p inhibitors can reduce the cytotoxicity of Ebola virus glycoprotein in vitro显示文摘Ebola virus(EBOV)causes a highly lethal hemorrhagic fever syndrome in humans and has been associated with mortality rates of up to 91%in Zaire, the most lethal strain.Though the viral envelope glycoprotein(GP)mediates widespread inflammation and cellular damage, these changes have mainly focused on alterations at the protein level, the role of microRNAs(miRNAs)in the molecular pathogenesis underlying this lethal disease is not fully understood. Here,we report that the miRNAs hsa-miR-1246,hsa-miR-320a and hsa-miR-196b-5p were induced in human umbilical vein endothelial cells(HUVECs)following expression of EBOV GP.Among the proteins encoded by predicted targets of these miRNAs,the adhesion-related molecules tissue factor pathway inhibitor(TFPI),dystroglycan1(DAG1)and the caspase 8 and FADD-like apoptosis regulator(CFLAR)were significantly downregulated in EBOV GP-expressing HUVECs.Moreover, inhibition of hsa-miR-1246,hsa-miR-320a and hsa-miR-196b-5p,or overexpression of TFPI,DAG1 and CFLAR rescued the cell viability that was induced by EBOV GP.Our results provide a novel molecular basis for EBOV pathogenesis and may contribute to the development of strategies to protect against future EBOV pandemics. | SHENG MiaoMiao ZHONG Ying CHEN Yang DU JianChao JU XiangWu ZHAO Chen ZHANG GuiGen ZHANG LiFang LIU KangTai YANG Ning XIE Peng LI DangSheng ZHANG Michael Q. | 3 | Strengthening effect of Cu-rich phase precipitation in 18Cr9Ni3CuNbN austenitic heat-resisting steel显示文摘The Cu-containing austenitic heat-resistant steel 18Cr9Ni3CuNbN, which is being used as superheater and reheater tube material for modern ultra-super-critical (USC) power plants all over the world, has been investigated at 650 ℃ long time aging till 10 000 h. SEM, TEM and 3DAP (three dimensional atom probe) have been used to follow microstructural changes with mechanical property variations. Experimental results show that Cu-rich phase and MX precipitate in the grains as well as M 23 C 6 precipitates at grain boundaries are the main precipitation strengthening phases in this steel. Among them Cu-rich phase is the most important strengthening phase. Homogeneous distribution of very fine nano-size Cu-rich phase has been formed at very early stage of 650 ℃ aging (less than 1 h). Cu atoms gradually concentrate to Cu-rich particles and the other elements (such as Fe, Cr, Ni etc) diffuse away from Curich particles to γ-matrix with the increasing of aging time at 650 ? C. The growth rate of Cu-rich phase at 650 ℃ long time aging is very slow and the average diameters of Cu-rich phase have been determined by TEM method. Cu-rich phase keeps in about 30 nm till 650 ℃ aging for 10 000 h. It shows that nano-size Cu-rich phase precipitation strengthening can be kept for long time aging at 650 ℃ because of its excellent stability at high temperatures. According to structure stability study and mechanical properties determination results the Cu-rich phase precipitation sequence and its strengthening mechanism model have been suggested and discussed. | Chengyu CHI Hongyao YU Jianxin DONG Xishan XIE Zhengqiang CUI Xiaofang CHEN Fusheng LIN | 2011 | Acta Metallurgica Sinica(English Letters)2011,24,2: | 6
Towards a semi-empirical model of the sea ice thickness based on hyperspectral remote sensing in the Bohai Sea显示文摘Sea ice thickness is one of the most important input parameters for the prevention and mitigation of sea ice disasters and the prediction of local sea environments and climates. Estimating the sea ice thickness is currently the most important issue in the study of sea ice remote sensing. With the Bohai Sea as the study area, a semiempirical model of the sea ice thickness(SEMSIT) that can be used to estimate the thickness of first-year ice based on existing water depth estimation models and hyperspectral remote sensing data according to an optical radiative transfer process in sea ice is proposed. In the model, the absorption and scattering properties of sea ice in different bands(spectral dimension information) are utilized. An integrated attenuation coefficient at the pixel level is estimated using the height of the reflectance peak at 1 088 nm. In addition, the surface reflectance of sea ice at the pixel level is estimated using the 1 550–1 750 nm band reflectance. The model is used to estimate the sea ice thickness with Hyperion images. The first validation results suggest that the proposed model and parameterization scheme can effectively reduce the estimation error associated with the sea ice thickness that is caused by temporal and spatial heterogeneities in the integrated attenuation coefficient and sea ice surface. A practical semi-empirical model and parameterization scheme that may be feasible for the sea ice thickness estimation using hyperspectral remote sensing data are potentially provided. | YUAN Shuai GU Wei LIU Chengyu XIE Feng | 2017 | Acta Oceanologica Sinica2017,36,1: | 4
Predicting roof displacement of roadways in underground coal mines using adaptive neuro-fuzzy inference system optimized by various physics-based optimization algorithms显示文摘Due to the rapid industrialization and the development of the economy in each country, the demand for energy is increasing rapidly. The coal mines have to pace up the mining operations with large production to meet the energy demand. This requirement has led underground coal mines to go deeper with more difficult conditions, especially the mining hazards, such as large deformations, rockburst, coal burst, roof collapse,to name a few. Therefore, this study aims at investigating and predicting the stability of the roadways in underground coal mines exploited by longwall mining method, using various novel intelligent techniques based on physics-based optimization algorithms(i.e.multi-verse optimizer(MVO),equilibrium optimizer(EO),simulated annealing(SA),and Henry gas solubility optimization(HGSO)) and adaptive neuro-fuzzy inference system(ANFIS),named as MVO-ANFIS,EO-ANFIS,SA-ANFIS and HGSOANFIS models.Accordingly,162 roof displacement events were investigated based on the characteristics of surrounding rocks, such as cohesion,Young’s modulus, density, shear strength, angle of internal friction, uniaxial compressive strength, quench durability index, rock mass rating, and tensile strength. The MVO-ANFIS,EO-ANFIS,SA-ANFIS and HGSO-ANFIS models were then developed and evaluated based on this dataset for predicting roof displacements in roadways of underground mines. The results indicated that the proposed intelligent techniques could accurately predict the roof displacements in roadways of underground mines with an accuracy in the range of 83%-92%.Remarkably,the SA-ANFIS model yielded the most dominant accuracy(i.e.92%).Based on the accurate predictions from the proposed techniques, the reinforced solutions can be timely suggested to ensure the stability of roadways during exploiting coal, especially in the underground coal mines exploited by the longwall mining. | Chengyu Xie Hoang Nguyen Xuan-Nam Bui Van-Thieu Nguyen Jian Zhou | 2021 | Journal of Rock Mechanics and Geotechnical Engineering2021,13,6: | 4
Optimized functional linked neural network for predicting diaphragm wall deflection induced by braced excavations in clays显示文摘Deep excavation during the construction of underground systems can cause movement on the ground, especially in soft clay layers. At high levels, excessive ground movements can lead to severe damage to adjacent structures. In this study, finite element analyses(FEM)and the hardening small strain(HSS)model were performed to investigate the deflection of the diaphragm wall in the soft clay layer induced by braced excavations.Different geometric and mechanical properties of the wall were investigated to study the deflection behavior of the wall in soft clays.Accordingly,1090 hypothetical cases were surveyed and simulated based on the HSS model and FEM to evaluate the wall deflection behavior. The results were then used to develop an intelligent model for predicting wall deflection using the functional linked neural network(FLNN)with different functional expansions and activation functions.Although the FLNN is a novel approach to predict wall deflection;however,in order to improve the accuracy of the FLNN model in predicting wall deflection, three swarm-based optimization algorithms, such as artificial bee colony(ABC),Harris’s hawk’s optimization(HHO),and hunger games search(HGS),were hybridized to the FLNN model to generate three novel intelligent models, namely ABC-FLNN,HHO-FLNN,HGS-FLNN.The results of the hybrid models were then compared with the basic FLNN and MLP models. They revealed that FLNN is a good solution for predicting wall deflection, and the application of different functional expansions and activation functions has a significant effect on the outcome predictions of the wall deflection. It is remarkably interesting that the performance of the FLNN model was better than the MLP model with a mean absolute error(MAE)of 19.971,root-mean-squared error(RMSE)of 24.574,and determination coefficient(R^(2))of 0.878.Meanwhile,the performance of the MLP model only obtained an MAE of 20.321,RMSE of 27.091,and R^(2)of 0.851.Furthermore,the results also indicated that the proposed hybrid models,i.e.,ABC-FLNN,HHO-FLNN,HGS-FLNN,yielded more superior performances than those of the FLNN and MLP models in terms of the prediction of deflection behavior of diaphragm walls with an MAE in the range of 11.877 to 12.239,RMSE in the range of 15.821 to 16.045,and R^(2)in the range of 0.949 to 0.951.They can be used as an alternative tool to simulate diaphragm wall deflections under different conditions with a high degree of accuracy. | Chengyu Xie Hoang Nguyen Yosoon Choi Danial Jahed Armaghani | 2022 | Geoscience Frontiers2022,13,2: | 3
Predicting rock size distribution in mine blasting using various novel soft computing models based on meta-heuristics and machine learning algorithms显示文摘Blasting is well-known as an effective method for fragmenting or moving rock in open-pit mines. To evaluate the quality of blasting, the size of rock distribution is used as a critical criterion in blasting operations.A high percentage of oversized rocks generated by blasting operations can lead to economic and environmental damage. Therefore, this study proposed four novel intelligent models to predict the size of rock distribution in mine blasting in order to optimize blasting parameters,as well as the efficiency of blasting operation in open mines.Accordingly,a nature-inspired algorithm(i.e.,firefly algorithm-FFA)and different machine learning algorithms(i.e.,gradient boosting machine(GBM),support vector machine(SVM),Gaussian process(GP),and artificial neural network(ANN))were combined for this aim, abbreviated as FFA-GBM,FFA-SVM,FFA-GP,and FFA-ANN,respectively.Subsequently,predicted results from the abovementioned models were compared with each other using three statistical indicators(e.g.,mean absolute error,root-mean-squared error, and correlation coefficient)and color intensity method. For developing and simulating the size of rock in blasting operations,136 blasting events with their images were collected and analyzed by the Split-Desktop software. In which,111 events were randomly selected for the development and optimization of the models.Subsequently, the remaining 25 blasting events were applied to confirm the accuracy of the proposed models.Herein, blast design parameters were regarded as input variables to predict the size of rock in blasting operations. Finally, the obtained results revealed that the FFA is a robust optimization algorithm for estimating rock fragmentation in bench blasting.Among the models developed in this study,FFA-GBM provided the highest accuracy in predicting the size of fragmented rocks. The other techniques(i.e.,FFA-SVM,FFA-GP,and FFA-ANN)yielded lower computational stability and efficiency.Hence, the FFA-GBM model can be used as a powerful and precise soft computing tool that can be applied to practical engineering cases aiming to improve the quality of blasting and rock fragmentation. | Chengyu Xie Hoang Nguyen Xuan-Nam Bui Yosoon Choi Jian Zhou Thao Nguyen-Trang | 2021 | Geoscience Frontiers2021,12,3: | 3
Hydrologic implications of the isotopic kinetic fractionation of open-water evaporation显示文摘The kinetic fractionation of open-water evaporation against the stable water isotope H_2 ^(18)O is an important mechanism underlying many hydrologic studies that use ^(18)O as an isotopic tracer. A recent in-situ measurement of the isotopic water vapor flux over a lake indicates that the kinetic effect is much weaker(kinetic factor 6.2‰) than assumed previously(kinetic factor14.2‰) by lake isotopic budget studies. This study investigates the implications of the weak kinetic effect for studies of deuterium excess-humidity relationships, regional moisture recycling, and global evapotranspiration partitioning. The results indicate that the low kinetic factor is consistent with the deuterium excess-humidity relationships observed over open oceans. The moisture recycling rate in the Great Lakes region derived from the isotopic tracer method with the low kinetic factor is a much better agreement with those from atmospheric modeling studies than if the default kinetic factor of 14.2‰ is used. The ratio of transpiration to evapotranspiration at global scale decreases from 84±9%(with the default kinetic factor) to 76±19%(with the low kinetic factor), the latter of which is in slightly better agreement with other non-isotopic partitioning results. | Wei XIAO Yufei QIAN Xuhui LEE Wei WANG Mi ZHANG Xuefa WEN Shoudong LIU Yongbo HU Chengyu XIE Zhen ZHANG Xuesong ZHANG Xiaoyan ZHAO Fucun ZHANG | 2018 | Science China Earth Sciences2018,61,10: | 1

A uranyl based coordination polymer showing response to low-dosage ionizing radiations down to 10^(−5) Gy显示文摘Radiation detection material is a central component of nuclear technology finding applications in many critical fields.Developing a highly radiation-sensitive material that shows a facilely detectable response to ultra-low dosage of radiation is a long-term research target and remains to be a challenge.Previously reported most optimal chemical radiation dosimeter can detect low-dosage X-andγ-ray radiations down to 10^(−4) Gy.We document here a new photoresponsive coordination polymer showing upgraded radiation detection capabilities with the detection limit on the radiation dose one order of magnitude lower than the previous record. The radiation induced photoluminescence quenching process was elucidated by multiple spectroscopic characterizations. | Jian Xie Yaxing Wang Wei Liu Chengyu Liang Yugang Zhang Lanhua Chen Daopeng Sheng Zhifang Chai Shuao Wang | 2020 | Science China Chemistry2020,63,11: | 0
Selective Hydrogenation of Phenylacetylene by Carbon Monoxide and Water显示文摘Catalytic selective hydrogenation of alkynes to the corresponding alkenes is an important process in industrial production.Modulating the selective hydrogenation of alkynes to the alkenes requires ingenuity since alkenes can easily be converted into the corresponding alkanes under reductive conditions.Applying different reductive reagents to prevent the direct usage of H_(2)can avoid difficulties in hydrogen storage and transportation.Herein,we demonstrate a tandem process to hydrogenate phenylacetylene by CO and H_(2)Oviathecouplingof thelow-temperaturewater-gas shift reaction and selective hydrogenation of phenylacetylene utilizing theα-MoC catalyst. The reductive reagent,CO,not only produces H_(2)from H_(2)O to drive the reaction forward, but it also regulates the selectivity of styrene by preventing further hydrogenation. | Xuetao Qin Ruiqi Zhang Ming Xu Yao Xu Lirong Zheng Chengyu Li Shixiang Yu Jie Yan Jinglin Xie Genghuang Wu Junfeng Rong Meng Wang Ding Ma | 2023 | CCS Chemistry2023,5,10: | 0
Avian influenza viruses suppress innate immunity by inducingtrans-transcriptional readthrough via SSU72显示文摘Innate immunity plays critical antiviral roles. The highly virulent avian influenza viruses (AIVs) H5N1, H7N9, and H5N6 can betterescape host innate immune responses than the less virulent seasonal H1N1 virus. Here, we report a mechanism by whichtranscriptional readthrough (TRT)-mediated suppression of innate immunity occurs post AIV infection. By using cell lines, mouselungs, and patient PBMCs, we showed that genes on the complementary strand (“trans” genes) influenced by TRT were involved inthe disruption of host antiviral responses during AIV infection. The trans-TRT enhanced viral lethality, and TRT abolishmentincreased cell viability and STAT1/2 expression. The viral NS1 protein directly bound to SSU72, and degradation of SSU72 inducedTRT. SSU72 overexpression reduced TRT and alleviated mouse lung injury. Our results suggest that AIVs infection induce TRT byreducing SSU72 expression, thereby impairing host immune responses, a molecular mechanism acting through the NS1-SSU72-trans-TRT-STAT1/2 axis. Thus, restoration of SSU72 expression might be a potential strategy for preventing AIV pandemics. | Yan Zhao Fengming Huang Zhen Zou Yuhai Bi Yang Yang Cong Zhang Qiang Liu Daozhen Shang Yiwu Yan Xiangwu Ju Song Mei Peng Xie Xiao Li Mingyao Tian Shuguang Tan Huijun Lu Zongsheng Han Kangtai Liu Yuqing Zhang Junbo Liang Zhu Liang Qingchao Zhang Jiahui Chang William JLiu Cong Feng Tanshi Li Michael Q.Zhang Xiaoyue Wang George FGao Yingxia Liu Ningyi Jin Chengyu Jiang | 2022 | Cellular & Molecular Immunology2022,19,6: | 0
OpNAC1 transcription factor regulates the biosynthesis of the anticancer drug camptothecin by targeting loganic acid O-methyltransferase in Ophiorrhiza pumila显示文摘Camptothecin(CPT) is an anticancer pentacyclic quinoline alkaloid widely used to treat cancer patients worldwide. However, the biosynthetic pathway and transcriptional regulation of camptothecin are largely unknown. Ophiorrhiza pumila, the herbaceous plant from the Rubiaceae family, has emerged as a model plant for studying camptothecin biosynthesis and regulation. In this study, a high-quality reference genome of O. pumila with estimated size of ~456.90Mb was reported, and the accumulation level of camptothecin in roots was higher than that in stems and leaves. Based on its spatial distribution in the plant, we examined gene functions and expression by combining genomics with transcriptomic analysis. Two loganic acid O-methyltransferase(OpLAMTs)were identified in strictosidine-producing plant O.pumila, and enzyme catalysis assays showed that OpLAMT1 and not OpLAMT2 could convert loganic acid into loganin. Further knock-out of OpL AMT1expression led to the elimination of loganin and camptothecin accumulation in O. pumila hairy roots.Four key residues were identified in OpLAMT1 protein crucial for the catalytic activity of loganic acid to loganin. By co-expression network, we identified a NAC transcription factor, OpNAC1, as a candidate gene for regulating camptothecin biosynthesis.Transgenic hairy roots and biochemical assays demonstrated that OpNAC1 suppressed OpLAMT1 expression. Here, we reported on two camptothecin metabolic engineering strategies paving the road for industrial-scale production of camptothecin in CPT-producing plants. | Xiaolong Hao Can Wang Wei Zhou Qingyan Ruan Chenhong Xie Yinkai Yang Chengyu Xiao Yan Cai Jingyi Wang Yao Wang Xuebin Zhang Itay Maoz Guoyin Kai | 2023 | Journal of Integrative Plant Biology2023,65,1: | 0
Sorption and desorption of sex hormones in soil-and sediment-water systems:A review显示文摘Sex hormones are a group of potent endocrine disruptors that can be released into agricultural soils and sediment via wastewater discharge and manure fertilization.Sorption represents a critical determinant for the transport potential and risks of sex hormones in the environment. Therefore, this study reviewed the sorption and desorption mechanisms of sex hormones in soil-and sedimentwater systems, and summarized the effects of various factors on sorption and desorption processes.A total of 359 set of sorption data were collected from the literature.Sex hormones were mostly described by the linear model. The sorption magnitudes(logKoc)of estrogens, androgens, and progestins were in the range of 2.77–3.90,2.55–4.18,and 2.61–4.39,respectively.The average logKoc values of the sex hormones were significantly correlated with their logKow values(R^(2)=0.13,p<0.05),while the R^(2) values were much lower than those when fewer sex hormones were included for analysis. In addition, the Kd values of most sex hormones were significantly correlated with the OC%of soils and sediment(R^(2)=0.16–0.99,p<0.05),but were insignificantly correlated with the particle size distribution and surface area. These results indicated that hydrophobic partitioning interaction and other specific interactions are responsible for sex hormone uptake in soil-and sediment-water systems. The sorption of sex hormones in soil-and sediment-water systems can also be affected by other environmental variables, including pH,temperature, and ionic strength.Future studies should focus on the coupled leaching-sorption processes in manure-water-soil systems under field-scale conditions. | Xiong Dai Xingjian Yang Bihai Xie Jiajia Jiao Xiuping Jiang Chengyu Chen Zhen Zhang Zhili He Hang Lin Weisong Chen Yongtao Li | 2022 | Soil Ecology Letters2022,4,1: | 0
Catch organism assemblages along artificial reefs area and adjacent waters in Haizhou Bay显示文摘To better understand the community patterns mediated by connectivity in artificial reefs of coastal areas, it is necessary to understand the distribution and coexistence of organisms with artificial reefs area and adjacent waters. This study was conducted to examine main catches assemblages collected by trawls in Haizhou Bay,which included five habitats: the artificial reef area(AR), aquaculture area(AA), natural area(NA), estuary area(EA) and comprehensive effect area(CEA). The result shows that the total abundances of species in the five habitats were highly different(univariate PERMANOVA: P = 0.001, n = 24), but some species were also unique in their habitat(e.g. Scapharca subcrenata and Glossaulax didyma in AA). The body size distribution of specific species between habitats are different. For Collichthys lucidus, their body size in AR(14.63 cm ± 1.64 cm) and EA(14.3 cm ± 0.85 cm) is higher than that in NA(10.65 cm ± 1.64 cm), CEA(11.28 cm ± 1.85 cm) and AA(12.1 cm ±0.43 cm), which indicates the potential connection from AR to EA mediated by their adult population. We concluded that artificial reefs in AR can be considered key components that have the ability to support species assemblages in adjacent habitats. This study has implications for the conservation and monitoring of species assemblages in coastal areas in terms of that artificial reefs can be applied in different stages of habitat protection implementation and in different combinations of scenarios. | Shike Gao Bin Xie Chengyu Huang Xiao Zhang Shuo Zhang Wenwen Yu | 2024 | Acta Oceanologica Sinica2024,43,2: | 0
