| 2 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 3 | Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. | 4 | Numerical Analysis of Nd:YAG Pulsed Laser Polishing CVD Self-standing Diamond Film显示文摘Chemical vapor deposited(CVD) diamond film has broad application foreground in high-tech fields.But polycrystalline CVD self-standing diamond thick film has rough surface and non-uniform thickness that adversely affect its extensive applications.Laser polishing is a useful method to smooth self-standing diamond film.At present,attentions have been focused on experimental research on laser polishing,but the revealing of theoretical model and the forecast of polishing process are vacant.The paper presents a finite element model to simulate and analyze the mechanism of laser polishing diamond based on laser thermal conduction theory.The experimental investigation is also carried out on Nd:YAG pulsed laser smoothing diamond thick film.The simulation results have good accordance with the results of experimental results.The temperature and thermal stress fields are investigated at different incidence angles and parameters of Nd:YAG pulsed laser.The pyramidal-like roughness of diamond thick film leads to the non-homogeneous temperature fields.The temperature at the peak of diamond film is much higher than that in the valley,which leads to the smoothing of diamond thick film.The effect of laser parameters on the surface roughness and thickness of graphite transition layer is also carried out.The results show that high power density laser makes the diamond surface rapid heating,evaporation and sublimation after its graphitization.It is also found that the good polish quality of diamond thick film can be obtained by a combination of large incident angle,moderate laser pulsed energy,large repetition rate and moderate laser pulse width.The results obtained here provide the theoretical basis for laser polishing diamond film with high efficiency and high quality. | XU Feng HU Haifeng ZUO Dunwen XU Chun QING Zhenghua WANG Min | 2013 | Chinese Journal of Mechanical Engineering2013,26,1: | 6 |

While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. | 6 | High Physical Activity is Associated with an Improved Lipid Profile and Resting Heart Rate among Healthy Middle-aged Chinese People显示文摘Objective To investigate the effects of physical activity(PA) on dyslipidemia and elevated resting heart rate(RHR) in a large-scale cross-sectional study in China. Methods We recruited community-based individuals who were 40-60 years old using a cluster sampling method. The PA levels of the participants were classified as low, moderate, or high, using the International Physical Activity Questionnaire. Dyslipidemia was defined as the detection of abnormalities in lipid indicators, and 4 lipid parameters were evaluated using fasting blood samples. Multivariate logistic regression analyses were used to evaluate the associations of PA with dyslipidemia and RHR. Results A total of 10,321 participants(38.88% men) were included in this study. The percentages of individuals with high, moderate, and low PA levels were 46.5%, 43.9%, and 9.6%, respectively. In both men and women, high PA provided odds ratios of 0.88 [95% confidence interval(CI): 0.83, 0.94] for dyslipidemia and 0.82(95% CI: 0.73, 0.92) for elevated RHR, compared to participants with low PA. | 9 | Anti-free radical,anti-oxidative ability and anti-fatigue effects of Huanshaodan An experiment of aging mice显示文摘BACKGROUND: In the theory of traditional Chinese medicine,aging is mainly thought renal deficiency caused renal failure,mainly involving decline of kidney-Yang and deficiency of kidney-essence. Huanshaodan,a Chinese traditional preparation for kidney-replenishing essence,was used to be the preparation for reinforcing renal deficiency and preventing aging for aged people. OBJECTIVE: To observe the effects of Huanshaodan on swimming durance and the abilities of catalase (CAT) in serum and monoamine oxidase-B (MAO-B) in brain tissue as well as in vitro anti-oxidative ability of aging mouse. DESIGN: A controlled animal experiment. SETTING: College of Basic Medicine,Hunan University of Traditional Chinese Medicine. MATERIALS: Fifty-four healthy NIH mice,aged 18 months old,of either gender,weighing (48.9±5.4)g,and one SD male rat,aged 16 months old,weighing 51.7 g,were provided by Animal Experimental Center,Hunan University of Traditional Chinese Medicine. Thirty NIH mice were randomly chosen for swimming test,and divided into experimental group and control group,with 15 in each; The other 24 NIH mice were used for enzyme activity assay,and also divided into experimental group and control group,with 12 in each. SD rat was used for in vitro anti-oxidative ability test. Huanshaodan water decoction was composed of Cheqianzi,Wuweizi,Huaishan,Danggui,Huangbai,Shudi,Baizhi,Niuxi,Baishen,Tusizi,Buguzhi,Roucongrong and Heshouwu 13 Chinese herbs. METHODS: This study was carried out in the Second Laboratory,Department of Biochemistry,Hunan University of Traditional Chinese Medicine in June 2006. Swimming and enzyme activity assay: Mice in the two experimental groups were intragastrically administrated with 10 μL/ g Huanshaodan water decoction. Mice in the two control groups were intragastrically administrated with the same amount of normal saline. All the mice were intragastrically administrated for 5 days,and they were free to access to medicine in the other 2 days in a week. Each mouse was administrated for 8 weeks. MAIN OUTCOME MEASURES: ① Forty days after administration,mice in the experimental group and control group for swimming test were loaded at tails and allowed to swim in the water-tank. Swimming durance was recorded. ② Following the method of Chen Qi,the activities of CAT in serum and MAO-B in brain tissue as well as the inhibitory rate of each medicine on malonaldehyde (MDA) content in the in vitro rat hepatic tissue were determined; Meanwhile,the inhibitory rate of different doses of Tusizi liquid to MDA content in the rat hepatic tissue in vitro was also assayed. RESULTS: Fifty-four NIH mice and one SD rat were recruited in this experiment. Three mice died in the swimming test,and all the other animals were involved in the final analysis. ① Swimming durance of mice in the experimental group was significantly longer than that in the control group (t =7.502,P < 0.01). The activity of CAT in serum of mice in the experimental group was significantly higher than that in the control group (t =13.307,P < 0.01). ② The activity of MAO-B in brain tissue of mice in the experimental group was significantly lower than that in the control group (t =13.27,P < 0.01). ③ The inhibitory rate of Cheqianzi,Wuweizi,Huaishan,Danggui,Huangbai,Shudi,Baizhi,Niuxi,Baishen,Tusizi,Buguzhi,Roucongrong and Heshouwu 13 Chinese herbs of Huanshaodan to MDA in the rat hepatic tissue in vitro was –62.9,–95.1,–34.9,–65.1,–99.1,–87.2,–94.1,–20.0,–67.0,–83.7,–91.0,–98.4,–93.0,respectively. ④ The inhibitory rate of low to high dose of Tusizi liquid to MDA content in the rat hepatic tissue in vitro was –3.41,–18.1,–26.6,–83.7,respectively. Vaccine development requires an understanding of the kinetics of neutralizing antibody(NAb) responses to severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).Methods In total, 605 serum samples from 125 COVID-19 patients(from January 1 to March 14, 2020)varying in age, sex, severity of symptoms, and presence of underlying diseases were collected, and antibody titers were measured using a micro-neutralization assay with wild-type SARS-CoV-2.Results NAbs were detectable approximately 10 days post-onset(dpo) of symptoms and peaked at approximately 20 dpo. The NAb levels were slightly higher in young males and severe cases, while no significant difference was observed for the other classifications. In follow-up cases, the NAb titer had increased or stabilized in 18 cases, whereas it had decreased in 26 cases, and in one case NAbs were undetectable at the end of our observation. Although a decreasing trend in NAb titer was observed in many cases, the NAb level was generally still protective.Conclusion We demonstrated that NAb levels vary among all categories of COVID-19 patients. China.However,current studies focus on antimicrobial and anticoagulation agents rather than tumors.The molecular identities of antihepatoma bioactive components in centipedes have not yet been extensively investigated.It is a challenge to isolate and characterize the effective components of centipedes due to limited peptide purification technologies for animal-derived medicines.AIM To purify,characterize,and synthesize the bioactive components with the strongest antihepatoma activity from centipedes and determine the antihepatoma mechanism.METHODS An antihepatoma peptide(scolopentide)was isolated and identified from the centipede scolopendra subspinipes mutilans using a combination of enzymatic hydrolysis,a Sephadex G-25 column,and two steps of high-performance liquid chromatography(HPLC).Additionally,the CCK8 assay was used to select the extracted fraction with the strongest antihepatoma activity.The molecular weight of the extracted scolopentide was characterized by quadrupole time of flight mass spectrometry(QTOF MS),and the sequence was matched by using the Mascot search engine.Based on the sequence and molecular weight,scolopentide was synthesized using solid-phase peptide synthesis methods.The synthetic scolopentide was confirmed by MS and HPLC.The antineoplastic effect of extracted scolopentide was confirmed by CCK8 assay and morphological changes again in vitro.The antihepatoma effect of synthetic scolopentide was assessed by the CCK8 assay and Hoechst staining in vitro and tumor volume and tumor weight in vivo.In the tumor xenograft experiments,qualified model mice(male 5-week-old BALB/c nude mice)were randomly divided into 2 groups(n=6):The scolopentide group(0.15 mL/d,via intraperitoneal injection of synthetic scolopentide,500 mg/kg/d)and the vehicle group(0.15 mL/d,via intraperitoneal injection of normal saline).The mice were euthanized by cervical dislocation after 14 d of continuous treatment.Mechanistically,flow cytometry was conducted to evaluate the apoptosis rate of HepG2 cells after treatment with extracted scolopentide in vitro.A Hoechst staining assay was also used to observe apoptosis in HepG2 cells after treatment with synthetic scolopentide in vitro.CCK8 assays and morphological changes were used to compare the cytotoxicity of synthetic scolopentide to liver cancer cells and normal liver cells in vitro.Molecular docking was performed to clarify whether scolopentide tightly bound to death receptor 4(DR4)and DR5.qRT-PCR was used to measure the mRNA expression of DR4,DR5,fas-associated death domain protein(FADD),Caspase-8,Caspase-3,cytochrome c(Cyto-C),B-cell lymphoma-2(Bcl-2),Bcl-2-associated X protein(Bax),x-chromosome linked inhibitor-of-apoptosis protein and Cellular fas-associated death domain-like interleukin-1βconverting enzyme inhibitory protein in hepatocarcinoma subcutaneous xenograft tumors from mice.Western blot assays were used to measure the protein expression of DR4,DR5,FADD,Caspase-8,Caspase-3,and Cyto-C in the tumor tissues.The reactive oxygen species(ROS)of tumor tissues were tested.RESULTS In the process of purification,characterization and synthesis of scolopentide,the optimal enzymatic hydrolysis conditions(extract ratio:5.86%,IC_(50):0.310 mg/mL)were as follows:Trypsin at 0.1 g(300 U/g,centipede-trypsin ratio of 20:1),enzymolysis temperature of 46°C,and enzymolysis time of 4 h,which was superior to freeze-thawing with liquid nitrogen(IC_(50):3.07 mg/mL).A peptide with the strongest antihepatoma activity(scolopentide)was further purified through a Sephadex G-25 column(obtained A2)and two steps of HPLC(obtained B5 and C3).The molecular weight of the extracted scolopentide was 1018.997 Da,and the peptide sequence was RAQNHYCK,as characterized by QTOF MS and Mascot.Scolopentide was synthesized in vitro with a qualified molecular weight(1018.8 Da)and purity(98.014%),which was characterized by MS and HPLC.Extracted scolopentide still had an antineoplastic effect in vitro,which inhibited the proliferation of Eca-109(IC_(50):76.27μg/mL),HepG2(IC_(50):22.06μg/mL),and A549(IC_(50):35.13μg/mL)cells,especially HepG2 cells.Synthetic scolopentide inhibited the proliferation of HepG2 cells(treated 6,12,and 24 h)in a concentration-dependent manner in vitro,and the inhibitory effects were the strongest at 12 h(IC_(50):208.11μg/mL).Synthetic scolopentide also inhibited the tumor volume(Vehicle vs Scolopentide,P=0.0003)and weight(Vehicle vs Scolopentide,P=0.0022)in the tumor xenograft experiment.Mechanistically,flow cytometry suggested that the apoptosis ratios of HepG2 cells after treatment with extracted scolopentide were 5.01%(0μg/mL),12.13%(10μg/mL),16.52%(20μg/mL),and 23.20%(40μg/mL).Hoechst staining revealed apoptosis in HepG2 cells after treatment with synthetic scolopentide in vitro.The CCK8 assay and morphological changes indicated that synthetic scolopentide was cytotoxic and was significantly stronger in HepG2 cells than in L02 cells.Molecular docking suggested that scolopentide tightly bound to DR4 and DR5,and the binding free energies were-10.4 kcal/mol and-7.1 kcal/mol,respectively.In subcutaneous xenograft tumors from mice,quantitative real-time polymerase chain reaction and western blotting suggested that scolopentide activated DR4 and DR5 and induced apoptosis in SMMC-7721 Liver cancer cells by promoting the expression of FADD,caspase-8 and caspase-3 through a mitochondria-independent pathway.CONCLUSION Scolopentide,an antihepatoma peptide purified from centipedes,may inspire new antihepatoma agents.Scolopentide activates DR4 and DR5 and induces
