2酵母培养物对斑点叉尾鮰幼鱼生长性能的影响显示文摘以斑点叉尾鮰为对象,研究饲料中添加不同比例的酵母培养物[达农威水产益康(DVAQTM)]对其幼鱼成活、生长和饲料系数的影响。基础饲料蛋白含量为32%,赖氨酸含量为需要量的85%(斑点叉尾鮰赖氨酸在饲料蛋白质中的适宜含量5.1%),其余必需氨基酸均满足其需要量,分别向基础饲料中添加酵母培养物0.03%0、.06%和0.12%,且基础饲料中减少一定量的玉米,制成试验饲料,进行为期12周的生长试验。试验在水族箱(75 L)中进行,每个处理设4个重复,每个重复放养10尾鮰幼鱼(平均体重为4.5 g)。研究结果表明0,.06%和0.12%添加组鮰成活率均为100%,略高于对照组(97%)。0.06%添加组鮰幼鱼增重率较对照组平均提高19%,饲料系数降低13%。0.12%添加组较对照组增重率平均提高22%,饲料系数降低15%。因此,饲料中添加酵母培养物可提高斑点叉尾鮰幼鱼的生长性能。H.Randall Robinette Constance Young 孟繁伊 2011饲料工业2011,32,6:10

4UFMylation of RPL26 links translocation-associated quality control to endoplasmic reticulum protein homeostasis显示文摘Protein biogenesis at the endoplasmic reticulum(ER)in eukaryotic cells is monitored by a protein quality control system named ER-associated protein degradation(ERAD).While there has been substantial progress in understanding how ERAD eliminates defective polypeptides generated from erroneous folding,how cells remove nascent chains stalled in the translocon during co-translational protein insertion into the ER is unclear.Here we show that ribosome stalling during protein translocation induces the attachment of UFM1,a ubiquitin-like modifier,to two conserved lysine residues near the COOH-terminus of the 60S ribosomal subunit RPL26(uL24)at the ER.Strikingly,RPL26 UFMylation enables the degradation of stalled nascent chains,but unlike ERAD or previously established cytosolic ribosome-associated quality control(RQC),which uses proteasome to degrade their client proteins,ribosome UFMylation promotes the targeting of a translocation-arrested ER protein to lysosomes for degradation.RPL26 UFMylation is upregulated during erythroid differentiation to cope with increased secretory flow,and compromising UFMylation impairs protein secretion,and ultimately hemoglobin production.We propose that in metazoan,co-translational protein translocation into the ER is safeguarded by a UFMylation-dependent protein quality control mechanism,which when impaired causes anemia in mice and abnormal neuronal development in humans.Lihui Wang Yue Xu Heather Rogers Layla Saidi Constance Tom Noguchi Honglin Li Jonathan Wilson Yewdell Nicholas Raymond Guydosh Yihong Ye 2020Cell Research2020,30,1:5
5MELD score,insulin-like growth factor 1 and cytokines on bone density in end-stage liver disease显示文摘AIM:To determine the contributions of insulin-like growth factor 1 (IGF-1),cytokines and liver disease severity to bone mineral density in patients pre-transplantation.METHODS:Serum IGF-1,tumor necrosis factor-α (TNFα) and interleukin 6 (IL-6) were measured and the Model for End-Stage Liver Disease (MELD) score calculated in 121 adult patients referred to a single centre for liver transplantation.Bone mineral density (BMD) of the lumbar spine and femoral neck were assessed via dual energy X-ray absorptiometry.Demographics,liver disease etiology,medication use and relevant biochemistry were recorded.RESULTS:A total of 117 subjects were included,with low BMD seen in 68.6%,irrespective of disease etiol-ogy.In multivariable analysis,low body mass index (BMI),increased bone turnover and low IGF-1 were independent predictors of low spinal bone density.At the hip,BMI,IGF-1 and vitamin D status were predictive.Despite prevalent elevations of TNFα and IL-6,levels did not correlate with degree of bone loss.The MELD score failed to predict low BMD in this pre-transplant population.CONCLUSION:Osteopenia/osteoporosis is common in advanced liver disease.Low serum IGF-1 is weakly predictive but serum cytokine and MELD score fail to predict the severity of bone disease.Rebecca Mitchell Jill McDermid Mang M Ma Constance L Chik 2011World Journal of Hepatology2011,3,6:5
6Erythropoietin modulates bone marrow stromal cell differentiation显示文摘Erythropoietin is essential for bone marrow erythropoiesis and erythropoietin receptor on non-erythroid cells including bone marrow stromal cells suggests systemic effects of erythropoietin. Tg6 mice with chronic erythropoietin overexpression have a high hematocrit, reduced trabecular and cortical bone and bone marrow adipocytes, and decreased bone morphogenic protein 2 driven ectopic bone and adipocyte formation. Erythropoietin treatment(1 200 IU·kg–1) for 10 days similarly exhibit increased hematocrit,reduced bone and bone marrow adipocytes without increased osteoclasts, and reduced bone morphogenic protein signaling in the bone marrow. Interestingly, endogenous erythropoietin is required for normal differentiation of bone marrow stromal cells to osteoblasts and bone marrow adipocytes.ΔEpoR_E mice with erythroid restricted erythropoietin receptor exhibit reduced trabecular bone, increased bone marrow adipocytes, and decreased bone morphogenic protein 2 ectopic bone formation. Erythropoietin treated ΔEpoR_E mice achieved hematocrit similar to wild-type mice without reduced bone, suggesting that bone reduction with erythropoietin treatment is associated with non-erythropoietic erythropoietin response. Bone marrow stromal cells from wild-type,Tg6, and ΔEpoR_E-mice were transplanted into immunodeficient mice to assess development into a bone/marrow organ. Like endogenous bone formation, Tg6 bone marrow cells exhibited reduced differentiation to bone and adipocytes indicating that high erythropoietin inhibits osteogenesis and adipogenesis, while ΔEpoR_E bone marrow cells formed ectopic bones with reduced trabecular regions and increased adipocytes, indicating that loss of erythropoietin signaling favors adipogenesis at the expense of osteogenesis. 20Pilot testing of CO_(2) capture from a coal-fired power plant-Part 2:Results from 1-MWe pilot tests显示文摘Using a 1-MWe slipstream pilot plant,solid-sorbent-based post-combustion CO_(2) capture was tested at a coal-fired power plant.Results from pilot testing were used to develop a preliminary full-scale commercial design.The sorbent selected for pilot-scale evaluation during this project consisted of an ion-exchange resin that incorporated amines covalently bonded to the substrate.A unique temperature-swing-absorption(TSA)process was developed that incorporated a three-stage fluidized-bed adsorber integrated with a single-stage fluidized-bed regenerator.Overall,following start-up and commissioning challenges that are often associated with first-of-a-kind pilots,the pilot plant operated as designed and expected,with a few key exceptions.The two primary exceptions were associated with:(i)handling characteristics of the sorbent,which were sufficiently different at operating temperature than at ambient temperature when design specifications were established with lab-scale testing;and(ii)CO_(2) adsorption in the transport line between the regenerator and adsorber that preloaded the sorbent with CO_(2) prior to entering the adsorber.Results from the pilot programme demonstrate that solid-sorbent-based post-combustion capture can be utilized to achieve 90%CO_(2) capture from coal-fired power plants.Sharon Sjostrom Constance Senior 2020Clean Energy2020,4,1:2
