1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2EXPERIMENTAL STUDY OF FLOWFIELD STRUCTURE AROUND AN OGIVE-CYLINDER显示文摘The flowfield structure and their aerodynamic characteristics over an ogive cylinder were studied by means of flow visualization and surface pressure measurement in a water tunnel and a wind tunnel. The existence of multi asymmetric vortices over long slender bodies was experimentally confirmed at large angles of attack and in the subcritical Reynolds number range. The spatial 3 D characteristics of the multi vortices system were analyzed and a physical model was developed. The topological structure of different patterns in cross flow plane was studied and the mechanism governing the formation of asymmetric vortices and multi vortices was discussed from the viewpoint of stability of the topological structure. 3MALE GAMETOPHYTE DEFECTIVE 2,Encoding a Sialyltransferase-like Protein,is Required for Normal Pollen Germination and Pollen Tube Growth in Arabidopsis显示文摘Sialyltransferases(SiaTs) exist widely in vertebrates and play important roles in a variety of biological processes.In plants,several genes have also been identified to encode the proteins that share homology with the vertebrate SiaTs.However,very little is known about their functions in plants.Here we report the identification and characterization of a novel Arabidopsis gene,MALE GAMETOPHYTE DEFECTIVE 2(MGP2) that encodes a sialyltransferase-like protein.MGP2 was expressed in all tissues including pollen grains and pollen tubes.The MGP2 protein was targeted to Golgi apparatus.Knockout of MGP2 significantly inhibited the pollen germination and retarded pollen tube growth in vitro and in vivo,but did not affect female gametophytic functions.These results suggest that the sialyltransferase-like protein MGP2 is important for normal pollen germination and pollen tube growth,giving a novel insight into the biological roles of the sialyltransferase-like proteins in plants.Yi Deng Wei Wang Wen-Qing Li Chuan Xia Hong-Ze Liao Xue-Qin Zhang De Ye 2010Journal of Integrative Plant Biology2010,52,9:7
4Experimental study on the mechanical properties and consolidation mechanism of microbial grouted backfill显示文摘Backfill mining technology is the practice of returning waste materials underground for both disposal and geotechnical stability,however,a challenge with current technologies is that they commonly require cement-based binders which have a relatively high environmental impact.Finding alternatives to cement-based binders can improve environmental performance and this paper proposes microbial grouted backfill(MGB)as a potential solution.In this paper,the effects of the cementation solution concentration(CSC),volume ratio of bacterial solution to cementation solution(VRBC),particle sizes of the aggregates,and the number of grouting batches on the mechanical properties of MGB are studied.The experimental results show that MGB strength increased,up to a peak value,as CSC was increased,before decreasing as CSC was increased further.The results also show that MGB strength increased,up to a peak value,as VRBC decreased,before decreasing as the VRBC was decreased further.The peak strength was achieved at a CSC of 2 mol/L and a VRBC of 1:9.The strength of the MGB also increased as the number of grouting batches increased.Graded MGB samples showed the highest UCS,25.12 MPa,at particle sizes of 0.2 to 0.8 mm,while full(non-graded)MGB samples displayed mean UCS values ranging from1.56 MPa when the maximum particle size was 0.2 mm,up to 13 MPa when the maximum particle size was 1.2 mm.MGB samples are consolidated by the calcium carbonate that is precipitated during microbial metabolism,and the strength of MGB increases linearly as calcium carbonate content increases.The calcium carbonate minerals produced in MGB materials are primarily calcite,with secondary amounts of vaterite.Xuejie Deng Yu Li Fei Wang Xiaoming Shi Yinchao Yang Xichen Xu Yanli Huang Benjamin de Wit 2022International Journal of Mining Science and Technology2022,32,2:7
5Compression-related EMIC waves drive relativistic electron precipitation显示文摘With coordinated observations of the NOAA 15 satellite and OUL magnetometer station in Finland, we report that the electromagnetic ion cyclotron(EMIC) waves which were stimulated by the compression of the magnetosphere drive relativistic electron precipitation in geoquiescence on 1 Jan 2007. After an enhancement of solar wind dynamic pressure(SWDP), a dayside Pc1 pulsation was observed by the OUL station. Such a Pc1 pulsation is caused by an EMIC wave which propagates from the generation source to lower altitudes. Simultaneously, the NOAA 15 satellite registered an enhancement of precipitating electron count rates with energies >3 Me V within the anisotropic zone of protons. This phenomenon is coincident with the quasi-linear theoretical calculation presented in this paper. 8Protective Effect of Procyanidin B2 on Acute Liver Injury Induced by Aflatoxin B1 in Rats显示文摘Objective This study aimed to explore the protective effect of procyanidin B2(PCB2)on acute liver injury induced by aflatoxin B1(AFB1)in rats.Methods Forty Sprague Dawley rats were randomly divided into control,AFB1,AFB1+PCB2,and PCB2 groups.The latter two groups were administrated PCB2 intragastrically(30 mg/kg body weight)for 7 d,whereas the control and AFB1 groups were given the same dose of double distilled water intragastrically.On the sixth day of treatment,the AFB1 and AFB1+PCB2 groups were intraperitoneally injected with AFB1(2 mg/kg).The control and PCB2 groups were intraperitoneally administered the same dose of dimethyl sulfoxide(DMSO).On the eighth day,all rats were euthanized:serum and liver tissue were isolated for further examination.Hepatic histological features were assessed by hematoxylin and eosin-stained sections.Weight,organ coefficient(liver,spleen,and kidney),liver function(serum alanine aminotransferase,aspartate aminotransferase,alkaline phosphatase,total bilirubin,and direct bilirubin),oxidative index(catalase,glutathione,superoxide dismutase,malondialdehyde,and 8-hydroxy-2′-deoxyguanosine),inflammation factor[hepatic interleukin-6(IL-6)m RNA expression and serum IL-6],and bcl-2/bax ratio were measured.Results AFB1 significantly caused hepatic histopathological damage,abnormal liver function,oxidative stress,inflammation,and bcl-2/bax ratio reduction compared with DMSO-treated controls.Our results indicate that PCB2 treatment can partially reverse the adverse liver conditions induced by AFB1.Conclusion Our findings indicate that PCB2 exhibits a protective effect on acute liver injury induced by AFB1.DENG Zhi Jie ZHAO Jing Fang HUANG Feng SUN Gui Li GAO Wei LU Li XIAO De Qiang 2020Biomedical and Environmental Sciences2020,33,4:4
9AEG1通过激活NF-κB和AP-1诱导肝癌HepG2细胞COX-2表达(英文)显示文摘Objective:The aim of this study was to investigate whether astrocyte elevated gene 1 (AEG1) regulates COX-2 expression in human hepatoma HepG2 cells and related pathways involved in this process. Methods:Human hepatoma HepG2 cells were transfected with pcDNA3.1(-)-AEG1 plasmid or psilencer2.0-AEG1-shRNA1 plasmid to up/down-regulate AEG1 expression, pcDNA3.1(-) and psilencer 2.0 empty vector plasmids were transfected respectively as control. Real-time RT-PCR was carried out to measure the expression levels of AEG1 and COX-2 mRNA. The expression levels of AEG1 and COX-2 protein were detected by Western blot. NF-κB signaling was blocked by PDTC, and AP-1 signaling was blocked by curcumin. Results:AEG1 mRNA and protein levels were increased after pcDNA3.1(-)-AEG1 transfection, and decreased after psilencer2.0-AEG1-shRNAs transfection. COX-2 mRNA and protein levels were increased in AEG1-overexpressing cells and decreased in AEG1-knockdown cells. Phosphorylations of p65 and c-jun were up-regulated in AEG1-overexpressing cells. Both PDTC and curcumin reduced COX-2 expression in HepG2 cells with AEG1 overexpression. Conclusion:AEG1-overexpressing and-knockdown HepG2 cells are established successfully. 12Study of the retina algorithm on FPGA for fast tracking显示文摘Real-time track reconstruction in high-energy physics experiments at colliders running at high luminosity is very challenging for trigger systems. To perform pattern recognition and track fitting, artificial retina or Hough transformation algorithms have been introduced to the field typically implemented on state-of-the-art field programmable gate array(FPGA) devices. In this paper, we report on two FPGA implementations of the retina algorithm: one using a mixed Floating-Point core and the other using Fixed-Point and Look-Up Table, and detailed measurements of the retina performance are investigated and compared.
