| 1 | Thin films as an emerging platform for drug delivery显示文摘Pharmaceutical scientists throughout the world are trying to explore thin films as a novel drug delivery tool. Thin films have been identified as an alternative approach to conventional dosage forms. The thin films are considered to be convenient to swallow, selfadministrable, and fast dissolving dosage form, all of which make it as a versatile platform for drug delivery. This delivery system has been used for both systemic and local action via several routes such as oral, buccal, sublingual, ocular, and transdermal routes. The design of efficient thin films requires a comprehensive knowledge of the pharmacological and pharmaceutical properties of drugs and polymers along with an appropriate selection of manufacturing processes. Therefore, the aim of this review is to provide an overview of the critical factors affecting the formulation of thin films, including the physico-chemical properties of polymers and drugs, anatomical and physiological constraints, as well as the characterization methods and quality specifications to circumvent the difficulties associated with formulation design. | 9 | Multivalent network modifier upregulates bioactivity of multispecies biofilm-resistant polyalkenoate cement显示文摘Polyalkenoate cement(PAC)is a promising material for regenerative hard tissue therapy.The ionically rich glass component of PAC encourages bioactive interaction via.the release of essential ions.However,PAC bioactivity is restricted owing to(i)structurally inherent cationic network formers and(ii)surface bacterial biofilm formation.These two factors cause a deficiency in ion release,further complicated by secondary infections and premature therapeutic failure.Here,a multivalent zwitterionic network modifier(mZM)is presented for upregulation of ionic exchange and bioactivity enhancement.By introducing a non-zero charged mZM into PACs,an increase in the proportion of non-bridging oxygen occurs.The network modification promotes ion channel formation,causing a multiple-fold increase in ion release and surface deposition of hydroxy-carbonate apatite(ca.74%).Experiments ex vivo and animal models also demonstrate the efficient remineralization ability of the mZM.Furthermore,divalent cationic interaction results in bacterial biofilm reduction(ca.68%)while also influencing a shift in the biofilm species composition,which favors commensal growth.Therefore,PAC modification with mZM offers a promising solution for upregulation of bioactivity,even aiding in customization by targeting site-specific regenerative therapy in future applications. | Ji-Yeong Kim Woojin Choi Utkarsh Mangal Ji-Young Seo Tae-Yun Kang Joohee Lee Taeho Kim Jung-Yul Cha Kee-Joon Lee Kwang-Mahn Kim Jin-Man Kim Dohyun Kim Jae-Sung Kwon Jinkee Hong Sung-Hwan Choi | 2022 | Bioactive Materials2022,7,8: | 0 |
| 10 | U-IMPACT:a universal 3D microfluidic cell culture platform显示文摘The development of organs-on-a-chip has resulted in advances in the reconstruction of 3D cellular microenvironments.However,there remain limitations regarding applicability and manufacturability.Here,we present an injection-molded plastic array 3D universal culture platform(U-IMPACT)for various biological applications in a single platform,such as cocultures of various cell types,and spheroids(e.g.,tumor spheroids,neurospheres)and tissues(e.g.,microvessels).The U-IMPACT consists of three channels and a spheroid zone with a 96-well plate form factor.Specifically,organoids or spheroids(~500μm)can be located in designated areas,while cell suspensions or cellladen hydrogels can be selectively placed in three channels.For stable multichannel patterning,we developed a new patterning method based on capillary action,utilizing capillary channels and the native contact angle of the materials without any modification.We derived the optimal material hydrophilicity(contact angle of the body,45–90°;substrate,<30°)for robust patterning through experiments and theoretical calculations.We demonstrated that the U-IMPACT can implement 3D tumor microenvironments for angiogenesis,vascularization,and tumor cell migration.Furthermore,we cultured neurospheres from induced neural stem cells.The U-IMPACT can serve as a multifunctional organ-on-achip platform for high-content and high-throughput screening. | Seung-Ryeol Lee Youngtaek Kim Suryong Kim Jiho Kim Seonghyuk Park Stephen Rhee Dohyun Park Byungjun Lee Kyusuk Baek Ho-Young Kim Noo Li Jeon | 2022 | Microsystems & Nanoengineering2022,8,6: | 0 |
| 11 | Method for process-based modeling of combat scenarios using interaction analysis weapon systems显示文摘With technological advancements,weapon system development has become increasingly complex and costly.Using modeling and simulation(M&S)technology in the conceptual design stage is effective in reducing the development time and cost of weapons.One way to reduce the complexity and trial-and-error associated with weapon development using M&S technology is to develop combat scenarios to review the functions assigned to new weapons.Although the M&S technology is applicable,it is difficult to analyze how effectively the weapons are functioning,because of the dynamic features inherent in combat scenario modeling,which considers interrelations among different weapon entities.To support review of weapon functions including these characteristics,this study develops a process-based modeling(PBM)method to model the interactions between weapons in the combat scenario.This method includes the following three steps:(1)construct virtual models by converting the weapons and the weapon functions into their corresponding components;(2)generate the combat process from the combat scenario,which is derived from the interrelations among weapons under consideration using reasoning rules;(3)develop a process-based model that describes weapon functions by combining the combat process with virtual models.Then,a PBM system based on this method is implemented.The case study executed on this system shows that it is useful in deriving process-based models from various combat scenarios,analyzing weapon functions using the derived models,and reducing weapon development issues in the conceptual design stage. | Dongsu JEONG Dohyun KIM Yoonho SEO | 2021 | Frontiers of Information Technology & Electronic Engineering2021,22,3: | 0 |

| 13 | Vaccinia-related kinase 2 variants differentially affect breast cancer growth by regulating kinase activity显示文摘Genetic information is transcribed from genomic DNA to mRNA,which is then translated into threedimensional proteins.mRNAs can undergo various post-transcriptional modifications,including RNA editing that alters mRNA sequences,ultimately affecting protein function.In this study,RNA editing was identified at the 499th base(c.499)of human vaccinia-related kinase 2(VRK2).This RNA editing changes the amino acid in the catalytic domain of VRK2 from isoleucine(with adenine base)to valine(with guanine base).Isoleucine-containing VRK2 has higher kinase activity than the valine-containing VRK2,which leads to an increase in tumor cell proliferation.Earlier we reported that VRK2 directly interacts with dystrobrevin-binding protein(dysbindin)and results in reducing its stability.Herein,we demonstrate that isoleucine-containing VRK2 decreases the level of dysbindin than valinecontaining VRK2.Dysbindin interacts with cyclin D and thereby regulates its expression and function.The reduction in the level of dysbindin by isoleucine-containing VRK2 further enhances the cyclin D expression,resulting in increased tumor growth and reduction in survival rates.It has also been observed that in patient samples,VRK2 level was elevated in breast cancer tissue compared to normal breast tissue.Additionally,the isoleucine form of VRK2 exhibited a greater increase in breast cancer tissue.Therefore,it is concluded that VRK2,especially dependent on the 167th variant amino acid,can be one of the indexes of tumor progression and proliferation. | SEUNG-HEE GWAK JUHYUN LEE EUNJI OH DOHYUN LEE WONSHIK HAN JONGMIN KIM KYONG-TAI KIM | 2024 | Oncology Research2024,32,2: | 0 |

| 15 | Graphene-based saturable absorber and mode-locked laser behaviors under gamma-ray radiation显示文摘We investigate optical and electrical behaviors of a graphene saturable absorber (SA) and mode-locking performance of a graphene-SA-based mode-locked Er fiber laser in gamma-ray radiation. When irradiated up to 4.8 kGy at ~ 100 Gy/hr dose rate, the overall nonlinear transmittance in transverse electric mode was increased, while maintaining modulation depth to >10%. The corresponding polarization.dependent loss was reduced at a 1.2-dB/kGy rate. In the electrical properties, the charge carrier mobility was reduced, and the Dirac voltage shift was increased to positive under gamma-ray radiation. The radiation-induced optical and electrical changes turned out to be almost recovered after a few days. In addition, we confirmed that the graphene-SA-based laser showed stable CW mode-locking operation while the inserted graphene SA was irradiated for 2-kGy at a 45-Gy/hr dose rate, which corresponds to >40 years of operation in low Earth orbit satellites.To the best of our knowledge, this is the first evaluation of graphene SAs and graphene-SA-based mode-locked lasers in gamma-ray radiation, and the measured results confirm the high potential of graphene SAs and graphene-SA-based lasers in various outer-space environments as well as other radiation environments, including particle accelerators and radiationbased medical instruments. | Dohyun Kim Nam Hun Park Hyunju Lee Jaegoan Lee Dong-Il Yeom Jungwon Kim | 2019 | Photonics Research2019,7,7: | 0 |
