1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29

3Clinical features of seizures after cerebral venous sinus thrombosis and its effect on outcome among Chinese Han population显示文摘background and purpose Clinical features of epileptic seizures after cerebral venous sinus thrombosis(CVST)among Chinese patients are not known,and it is still controversial whether seizures would affect the outcome of CVST.Methods In a Chinese hospital-based study of consecutive patients with CVST between 2003 and 2015,we described the clinical features of seizures and determined the predictors of seizure onset using multivariable logistic regression analysis.We also compared the in-hospital case-fatality and short-term functional outcome(modified Rankin Scale(mRS)at discharge)in patients with versus without seizures using ordinal regression analysis.results Among 151 patients with CVST,52(34.4%)presented seizures,of which 42(80.8%)were generalised seizures.Male gender(OR 6.32,95% CI 2.06 to 19.35,p=0.001),motor deficits(OR 4.89,95% CI 1.52 to 15.68,p=0.008),intracerebral haemorrhage(OR 3.93,95% CI 1.16 to 13.26,p=0.027),cerebral infarction(OR 3.78,95% CI 1.15 to 12.36,p=0.029)and superior sagittal sinus thrombosis(OR 3.38,95% CI 91.16 to 9.86,p=0.026)were independent predictors for seizures.The overall in-hospital case-fatality rate was 2.0%(3/151),and 21(13.9%)had mRS>2 at discharge.Compared with patients without seizures,patients with seizures were more likely to have a worse outcome(p=0.02)at discharge,independent of age,gender,clinical presentation,clot burden and presence of parenchymal lesions.conclusions In Chinese Han patients,compared with patients without seizures,patients with seizures after CVST had a worse outcome.Risk factors such as male gender,paresis,parenchymal lesion and superior sagittal sinus thrombosis were independently associated with seizure onset after CVST.Generalised seizure was the main form of seizures after CVST,which was obviously different to seizures after strokes of arterial origin.Hongyan Ding Yanan Xie Linxin Li Heling Chu Yuping Tang Qiang Dong Mei Cui 2017Stroke & Vascular Neurology2017,2,4:8

5An experiment-library least-squares method on on-line coal element contents analysis显示文摘A new experiment-library least-squares(experiment-LLs) method used in the neutron inelastic-scattering and thermal-capture analysis(NITA) technique for on-line coal analysis was developed,which has significantly decreased the non-linear radiation effects.In this method,sixty samples with preset elemental contents were made,from whose experimental spectra twenty single-element spectrum libraries corresponding to twenty kinds of coal were built using the least-squares method.The spectrum of the unknown sample was analyzed based on these twenty libraries to estimate its element contents.With the initial estimated result,the procedure of developing library and analysis was iterated to build a new library with the closest element contents to the unknown sample.Hence the experiment-LLs method can reduce non-linear radiation effects.The experiment-LLs method was performed on an improved coal analysis system which was equipped with a long-life 14 MeV pulsed-neutron generator,a bulk BGO detector with a temperature controller,a moisture meter and coal-smoothing device.The precisions of this system for ash content,water content,volatile content and calorific value have reached 1.0wt%,0.5wt%,1.0wt%,350 kJ/kg,respectively.WANG HuiDong LU JingBin LU YuPing YANG Dong MA KeYan YANG Kang LIU YuMin CHENG DaoWen 2012Science China(Physics,Mechanics & Astronomy)2012,55,11:6
6Relationship Between Hematoma Expansion Induced by Hypertension and Hyperglycemia and Blood-brain Barrier Disruption in Mice and Its Possible Mechanism:Role of Aquaporin-4 and Connexin43显示文摘We aimed to select an optimized hematoma expansion(HE)model and investigate the possible mechanism of blood-brain barrier(BBB)damage in mice.The results showed that HE occurred in the group with hypertension combined with hyperglycemia(HH-HE)from3 to 72 h after intracerebral hemorrhage;this was accompanied by neurological deficits and hardly influenced the survival rate.The receiver operating characteristic curve suggested the criterion for this model was hematoma volume expansion≥45.0%.Meanwhile,HH-HE aggravated BBB disruption.A protector of the BBB reduced HH-HE,while a BBB disruptor induced a further HH-HE.Aquaporin-4(AQP4)knock-out led to larger hematoma volume and more severe BBB disruption.Furthermore,hematoma volume and BBB disruption were reduced by multiple connexin43(Cx43)inhibitors in the wild-type group but not in the AQP4 knock-out group.In conclusion,the optimized HE model is induced by hypertension and hyperglycemia with the criterion of hematoma volume expanding≥45.0%.HH-HE leads to BBB disruption,which is dependent on AQP4 and Cx43.Heling Chu Zidan Gao Chuyi Huang Jing Dong Yuping Tang Qiang Dong 2020Neuroscience Bulletin2020,36,11:4
7Three-dimensional properties of mesoscale cyclonic warm-core and anticyclonic cold-core eddies in the South China Sea显示文摘In general,a mesoscale cyclonic(anticyclonic)eddy has a colder(warmer)core,and it is considered as a cold(warm)eddy.However,recently research found that there are a number of'abnormal'mesoscale cyclonic(anticyclonic)eddies associated with warm(cold)cores in the South China Sea(SCS).These'abnormal'eddies pose a challenge to previous works on eddy detection,characteristic analysis,eddy-induced heat and salt transports,and even on mesoscale eddy dynamics.Based on a 9-year(2000–2008)numerical modelling data,the cyclonic warm-core eddies(CWEs)and anticyclonic cold-core eddies(ACEs)in the SCS are analyzed.This study found that the highest incidence area of the'abnormal'eddies is the northwest of Luzon Strait.In terms of the eddy snapshot counting method,8620 CWEs and 9879 ACEs are detected,accounting for 14.6%and 15.8%of the total eddy number,respectively.The size of the'abnormal'eddies is usually smaller than that of the'normal'eddies,with the radius only around 50 km.In the generation time aspect,they usually appear within the 0.1–0.3 interval in the normalized eddy lifespan.The survival time of CWEs(ACEs)occupies 16.3%(17.1%)of the total eddy lifespan.Based on two case studies,the intrusion of Kuroshio warm water is considered as a key mechanism for the generation of these'abnormal'eddies near the northeastern SCS.Wenjin Sun Yu Liu Gengxin Chen Wei Tan Xiayan Lin Yuping Guan Changming Dong 2021Acta Oceanologica Sinica2021,40,10:3

9VPS4B mutation impairs the osteogenic differentiation of dental follicle cells derived from a patient with dentin dysplasia type I显示文摘A splicing mutation in VPS4B can cause dentin dysplasia type I(DD-I),a hereditary autosomal-dominant disorder characterized by rootless teeth,the etiology of which is genetically heterogeneous.In our study,dental follicle cells(DFCs)were isolated and cultured from a patient with DD-I and compared with those from an age-matched,healthy control.In a previous study,this DD-I patient was confirmed to have a loss-of-function splicing mutation in VPS4B(IVS7+46C>G).The results from this study showed that the isolated DFCs were vimentin-positive and CK14-negative,indicating that the isolated cells were derived from the mesenchyme.DFCs harboring the VPS4B mutation had a significantly higher proliferation rate from day 3 to day 8 than control DFCs,indicating that VPS4B is involved in cell proliferation.The cells were then replenished with osteogenic medium to investigate how the VPS4B mutation affected osteogenic differentiation.Induction of osteogenesis,detected by alizarin red and alkaline phosphatase staining in vitro,was decreased in the DFCs from the DD-I patient compared to the control DFCs.Furthermore,we also found that the VPS4B mutation in the DD-I patient downregulated the expression of osteoblast-related genes,such as ALP,BSP,OCN,RUNX2,and their encoded proteins.These outcomes confirmed that the DD-I-associated VPS4B mutation could decrease the capacity of DFCs to differentiate during the mineralization process and may also impair physiological root formation and bone remodeling.This might provide valuable insights and implications for exploring the pathological mechanisms underlying DD-I root development.Qiang Li Fangli Lu Tianxuan Chen Ke Zhang Yuping Lu Xiaocong Li Yingying Wang Ling Liu Qing Tian Fu Xiong Dong Chen 2020International Journal of Oral Science2020,12,3:2
10An AIEE polyelectrolyte as a light-up fluorescent probe for heparin sensing in full detection range显示文摘Polyelectrolyte PTPEPyH, containing tetraphenylethene (TPE) and pyridinium, was synthesized. Its optical properties were investigated with spectroscopies and the results showed that there were two emission-enhanced stages in the interaction between positive-charged PTPEPyH and negative-charged biomacromolecule heparin. The mechanism was very different due to the reduced non-radiative energy loss and the change of surroundings. 11Magnetic transformation of Mn from anti-ferromagnetism to ferromagnetism in FeCoNiZMn_(x)(Z=Si,Al,Sn,Ge)high entropy alloys显示文摘We design high entropy alloys(HEAs) with different induction elements(Si/Al/Sn).In order to keep the crystal structure invariant and to investigate how the increment in saturation magnetization(Ms)is caused only by the change of electron spin state,each set of HEAs contains a different amount of Mn.Synergistic effects among induction elements that induce the magnetic transformation of Mn from anti-ferromagnetism to ferromagnetism are found.Ms of added Mn reduces when a particular induction element(Si_(0.4)/Al_(0.4)/Sn_(0.4)) exists,while a larger increment of Ms appears when two induction elements coexist,Si_(0.4)Al_(0.4)(25.79 emu/g) and Sn_(0.4)Al_(0.4)(15.43 emu/g).This is reflected in the microcosmic magnetic structure for the emergence of closed domains due to large demagnetization energy,which is confirmed by the Lorentz transmission electron microscope(LTEM) data.The calculated magnetic moments and the exchange integral constants from density functional theory based on the Exact Muffin-Tin Orbits fo rmalism reveal that the magnetic state and the strength of fe rromagnetic and anti-ferromagnetic coupling determine the variation of Ms in different chemical environments.The difference in energy levels of coexisting multiple induction elements also leads to a larger increment of Ms,Si_(0.4)Al_(0.4)Sn_(0.4)(29.78 emu/g),and Si_(0.4)Al_(0.4)Ge_(0.4)Sn_(0.4)(31.00 emu/g).Bin Zhang Yuping Duan Haifeng Zhang Shuo Huang Guojia Ma Tongmin Wang Xinglong Dong Nan Jia 2021Journal of Materials Science & Technology2021,,9:2
12Switching emissions of two tetraphenylethene derivatives with solvent vapor, mechanical, and thermal stimuli显示文摘Two derivatives of tetraphenylethene (TPE) were synthesized through one step cross McMurry coupling reaction. Both luminogens exhibit aggregation-induced emission (AIE) and crystallization induced emission enhancement (CIEE). The emissions of both luminogens could be switched between blue and green through reversible modulation of morphology with thermal, organic solvent fuming and mechanical stimuli. 14Effect of Cu on Discontinuous Precipitation of a Symmetrical AlZn Alloy显示文摘The effect of Cu additions on discontinuous precipitation of AlZn alloy were studied with optical microscopy, scanning electron microscopy and X-ray diffraction. It is found that the effect of addition of 2 at. pct Cu on cellular nucleation site is not remarkable, while the effect of aging temperature on cellular nucleation site of AlZn and AlZn-2Cu alloys is fairly obvious. The cell growth rate of discontinuous precipitation of AlZn alloy is remarkably accelerated with the addition of 2 at. pct Cu. The discontinuous precipitation microstructure of AlZn alloy is not apparently affected with the addition of 2 at. pct Cu and the cellular front is still fine microstructure of spinodal decomposition. The phase constituents consist of a fcc Al-rich phase, a hcp Zn-rich phase and a CuZn4 phase. 18Organic Host-Guest Materials with Bright Red Room-Temperature Phosphorescence for Persistent Bioimaging显示文摘Organic room-temperature phosphorescence(RTP)materials have attracted immense attention in bioimaging due to their long emission lifetime and large Stokes shift.RTP materials with long emission wavelength can improve the penetration depth for bioimaging.However,the design of red persistent RTP materials is still challenging.In this study,a fused-ring structure has been proposed to effectively decrease the triplet energy level,thus extending the emission wavelength of phosphorescence.In addition,the fused-ring structure exhibits a high molar extinction coefficient(ɛ)and high luminescence efficiency due to the rigid structure.A new class of crystalline hosts(iminodibenzyl,IDB)are developed to stabilize the triplet excitons that are generated from the fused-ring molecules.The maximum RTP wavelength of doping materials can reach 635 nm with a lifetime of 9.35 ms.Water-disperse nanoparticles are successfully prepared for in vivo time-resolved bioimaging,which eliminates the background fluorescence interference from biological tissues.These reveal a delicate design strategy for the construction of long-wavelength emissive RTP materials for high-resolution bioimaging.Yueyue Si Yeyun Zhao Wenbo Dai Shisheng Cui Peng Sun Jianbing Shi Bin Tong Zhengxu Cai Yuping Dong 2023Chinese Journal of Chemistry2023,41,13:1
