| 2 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 7 | Use of Support Vector Regression Based on Mean Impact Value Model to Identify Active Compounds in a Combination of Curcuma longa L.and Glycyrrhiza extracts显示文摘A support vector regression based on the mean impact value (MIV) model was constructed to identify the bioactive compounds inhibiting proliferation of He La cells in a combination of turmeric (Curcuma longa L.)and liquorice (Glycyrrhiza) extracts.The quantitative chemical fingerprint from 50 batches of turmeric and liquorice extracts was established using high performance liquid chromatography hyphenated to an ultraviolet visible detector.Qualitative results were obtained using ultra performance liquid chromatography coupled with electrospray ionization quadrupole time-of-flight tandem mass spectrometry.A total of 46 peaks (peaks 1–15 from turmeric and 16–46 from liquorice) were selected as 'common peaks' for analysis.The inhibitory effect of the combined extracts on He La cells was measured by MTT (3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay.It was found that 15 compounds (peaks:8,12,30,24,46,11,14,9,3,1,44,18,7,45 and 43)possessing high absolute MIV exhibited a significant correlation with the cytotoxicity against He La cells; most of these have already been confirmed with potential cytotoxicity in previous research.The important potential application of the present model can be extended to help discover active compounds from complex herbal medicine prior to traditional bioassay-guided separation.It is considered that this could be a useful tool for redeveloping herbal medicine based on the use of these active compounds. | Jianlan Jiang Qingjie Tan Weifeng Li Xinyun Du Ningzhi Liu | 2017 | Transactions of Tianjin University2017,23,3: | 2 |

| 16 | Characteristics and Mechanism of Upper Permian Reef Reservoirs in the Eastern Longgang Area, Northeastern Sichuan Basin, China显示文摘As a typical kind of lithologic reservoirs,reef reservoirs are generally featured by their large single-well reservoir thickness,good reservoir physical properties,and high gas well productivity.The Upper Permian Changxing Formation is an important natural gas exploration target in the Sichuan Basin,which hosts a large reef gas reservoir and is mainly distributed along the Kaijiang-Liangping Trough.Comprehensive analyses are implemented to investigate reservoir characteristics and identify controlling factors of reef reservoirs in Changxing Formation in the Eastern Longgang Area,Northeastern Sichuan Basin,including core,logging,and seismic data analyses.Changxing Formation reservoirs in the study area mainly occur in the reef-shoal complex,which are featured by wide distribution,large thickness and generally good physical properties.Reservoir rocks are dominantly composed of bioclastic dolomite and silty-fine dolomite(with grain phantom structure),while the main reservoir space consists of residual intergranular pores,intergranular dissolution pores,and karst vugs.In the seismic profiles,typical mound-shaped chaotic reflections can be clearly seen.It is suggested by the main reservoir development controlling factor analysis that the distributions of reef reservoirs are typically controlled by sedimentary facies belts,while the scale of the reef-shoal complex is determined by the pene-sedimentary micro-paleo-geomorphology.Dolomitization can not only significantly preserve the primary pores but also enhance the permeability of rocks.Moreover,karstification is the key to high-quality reef-shoal reservoirs. | Lei Tan Hong Liu Yuzhe Tang Bing Luo Ya Zhang Yuran Yang Yisha Liao Weifeng Du Xiran Yang | 2020 | Petroleum2020,6,2: | 1 |

| 20 | Incorporation of intra-city human mobility into urban growth simulation:A case study in Beijing显示文摘The effective modeling of urban growth is crucial for urban planning and analyzing the causes of land-use dynamics.As urbanization has slowed down in most megacities,improved urban growth modeling with minor changes has become a crucial open issue for these cities.Most existing models are based on stationary factors and spatial proximity,which are unlikely to depict spatial connectivity between regions.This research attempts to leverage the power of real-world human mobility and consider intra-city spatial interaction as an imperative driver in the context of urban growth simulation.Specifically,the gravity model,which considers both the scale and distance effects of geographical locations within cities,is employed to characterize the connection between land areas using individual trajectory data from a macro perspective.It then becomes possible to integrate human mobility factors into a neural-network-based cellular automata(ANN-CA)for urban growth modeling in Beijing from 2013 to 2016.The results indicate that the proposed model outperforms traditional models in terms of the overall accuracy with a 0.60%improvement in Cohen’s Kappa coefficient and a 0.41%improvement in the figure of merit.In addition,the improvements are even more significant in districts with strong relationships with the central area of Beijing.For example,we find that the Kappa coefficients in three districts(Chaoyang,Daxing,and Shunyi)are considerably higher by more than 2.00%,suggesting the possible existence of a positive link between intense human interaction and urban growth.This paper provides valuable insights into how fine-grained human mobility data can be integrated into urban growth simulation,helping us to better understand the human-land relationship. | WANG Siying FEI Teng LI Weifeng ZHANG Anqi GUO Huagui DU Yunyan | 2022 | Journal of Geographical Sciences2022,32,5: | 0 |
