1Blocking meningeal lymphatic drainage aggravates Parkinson’s disease-like pathology in mice overexpressing mutated α-synuclein显示文摘Background:Abnormal aggregation of brainα-synuclein is a central step in the pathogenesis of Parkinson’s disease(PD),thus,it is reliable to promote the clearance ofα-synuclein to prevent and treat PD.Recent studies have revealed an essential role of glymphatic system and meningeal lymphatic vessels in the clearance of brain macromolecules,however,their pathophysiological aspects remain elusive.Method:Meningeal lymphatic drainage of 18-week-old A53T mice was blocked via ligating the deep cervical lymph nodes.Six weeks later,glymphatic functions and PD-like phenotypes were systemically analyzed.Results:Glymphatic influx of cerebrospinal fluid tracer was reduced in A53T mice,accompanied with perivascular aggregation ofα-synuclein and impaired polarization of aquaporin 4 expression in substantia nigra.Cervical lymphatic ligation aggravated glymphatic dysfunction of A53T mice,causing more severe accumulation ofα-synuclein,glial activation,inflammation,dopaminergic neuronal loss and motor deficits.Conclusion:The results suggest that brain lymphatic clearance dysfunction may be an aggravating factor in PD pathology.Wenyan Zou Tinglin Pu Weixi Feng Ming Lu Ying Zheng Renhong Du Ming Xiao Gang Hu 2019Translational Neurodegeneration2019,8,1:17
2Regulatory Effect of E2,IL-6 and IL-8 on the Growth of Epithelial Ovarian Cancer Cells显示文摘To determine the regulatory effects of estrogen and cytokine IL-6 and IL-8 on the growth of epithelial ovarian cancer (OVCA), we first examined the status of estrogen receptors (ERα and ERβ), IL-6 receptor (IL-6Rα and gp130), and IL-8 receptor (IL-8RA and IL-8RB) on five epithelial OVCA cell lines by semiquantitative RT-PCR and Western blot analysis. Results showed that the expressions of these receptors were variable on the five cells.Those OVCA cells expressing the receptors were selected to study related molecular mechanism. MTT assay was performed to observe the effects of 17β-estradiol (E2), IL-6 and IL-8 on cell proliferation. We discovered that E2 markedly promoted the proliferation of CAOV-3 and OVCAR-3 cell in a time- and dose-dependent manner.Tamoxifen (Txf), an ER inhibitor, completely blocked the proliferation of the E2-induced cells, and IL-6- or/and IL-8-neutralizing antibody only showed partially blocking activity. IL-6 and IL-8 were able to significantly stimulate CAOV-3 and OVCAR-3 cell proliferation in a time- and dose-dependent manner, which had a potential synergistic effect on CAOV-3 cells but not on OVCAR-3 cells. The cell proliferation induced by these two cytokines was abolished completely by their specific neutralizing antibodies, partially by Txf, but not by unrelated goat IgG.Taken together, our results suggested that estrogen, IL-6 and IL-8 could modulate OVCA growth by forming a reciprocal cascade with amplifying effect. 3Characterization of uncultivated magnetotactic bacteria from the sediments of Yuehu Lake, China显示文摘Marine magnetotactic bacteria were collected from the intertidal sediments of Yuehu Lake(China), where their abundance reached 103–104 ind./cm3. Diverse morphotypes of magnetotactic bacteria were observed, including cocci and oval, vibrio-, spirillum-, rod-, elliptical-, handle- and bar-shaped forms. The magnetococci were the most abundant, and had flagella arranged in parallel within a bundle. The majority of magnetosomes were arranged in one, two or multiple chains, although irregular arrangements were also evident. All the results of high-resolution transmission electron microscopy(HRTEM) analysis show that magnetosome crystals were composed of Fe3O4, and their morphology was specific to particular cell morphotypes. By the 16 S r RNA gene sequence analysis, we found fourteen operational taxonomic units(OTUs) which were related to magnetotactic bacteria. Among these, thirteen belonged to the Alphaproteobacteria and one to the Gammaproteobacteria.Compared with known axenic and uncultured marine magnetotactic bacteria, the 16 S r RNA gene sequences of most magnetotactic bacteria collected from the Yuehu Lake exhibited sequence identities ranging from 90.1% to96.2%(<97%). 4Basic helix-loop-helix(bHLH)transcription factor MdbHLH3 negatively affects the storage performance of postharvest apple fruit显示文摘The storage period of fleshy fruits greatly affects their quality and selection,and is largely controlled by genetic factors.Therefore,it is imperative to elucidate how genetic factors affect fruit ripening and its storage.Here,we evaluated the postharvest storage properties of the basic helix-loop-helix(bHLH)transcription factor MdbHLH3-overexpressing transgenic Royal gala apple fruits.During storage,the contents of starch,malic acid,fructose,glucose,and sucrose in fruits of three MdbHLH3 transgenic lines were always higher than those of the wild-type(WT)control.Interestingly,the sugar-acid ratio also showed the same trend during fruit storage.Additionally,the fruit firmness decreased with increasing storage time,and the contents of cell wall components such as water-soluble pectin and cellulose in transgenic fruits were higher than those in control fruits,while the firmness of transgenic fruits was lower than that in WT control fruits.Though the ethylene release rate in both showed the same trend(firstly increasing,then decreasing,and finally peaking)in 90-day stored fruits,transgenic apples had higher ethylene levels than the WT control throughout storage.Furthermore,the activities of membrane peroxidase,antioxidant enzymes,and fruit ripening enzymes in all transgenic fruits were significantly higher than those in the WT control.Thus,our findings show how MdbHLH3 negatively regulates and reduces apple storage time.This may prove useful for not only developing biotechnological strategies,but also support traditional breeding programs,to help improve the storage time of fleshy fruits.Wenyan Wang Jianqiang Yu Mengchi Du Jiahui Wang Dagang Hu 2022Horticultural Plant Journal2022,8,6:2

6The Investigation on Single Event Function Failure for DC/DC Converters with Three Single Terminal Topological Structures显示文摘Heavy ion radiation experiments have been done to DC/DC converters with different topological structures for space applications. The test results were analyzed about the function failure of three topological structures caused by single event effects. The relationship between the function failure and the input supply voltage, the output load current and the topological structure of the module were discussed. Based on the analysis of the variation relationship among the source/drain terminal voltage of MOSFETs and the input voltage and the output load, the sensitivity factors associated with the function failure caused by single event effects were discussed. 8Efficient Catalytic Conversion of Polysulfides by Biomimetic Design of “Branch‑Leaf” Electrode for High‑Energy Sodium–Sulfur Batteries显示文摘Rechargeable room temperature sodium–sulfur(RT Na–S)batteries are seriously limited by low sulfur utilization and sluggish electrochemical reaction activity of polysulfide intermediates.Herein,a 3D“branch-leaf”biomimetic design proposed for high performance Na–S batteries,where the leaves constructed from Co nanoparticles on carbon nanofibers(CNF)are fully to expose the active sites of Co.The CNF network acts as conductive“branches”to ensure adequate electron and electrolyte supply for the Co leaves.As an effective electrocatalytic battery system,the 3D“branch-leaf”conductive network with abundant active sites and voids can effectively trap polysulfides and provide plentiful electron/ions pathways for electrochemical reaction.DFT calculation reveals that the Co nanoparticles can induce the formation of a unique Co–S–Na molecular layer on the Co surface,which can enable a fast reduction reaction of the polysulfides.Therefore,the prepared“branch-leaf”CNF-L@Co/S electrode exhibits a high initial specific capacity of 1201 mAh g^−1 at 0.1 C and superior rate performance.Wenyan Du Kangqi Shen Yuruo Qi Wei Gao Mengli Tao Guangyuan Du Shu‑juan Bao Mingyang Chen Yuming Chen Maowen Xu 2021Nano-Micro Letters2021,13,3:1
9Synthesis of chitosan-based grafting magnetic flocculants for flocculation of kaolin suspensions显示文摘A series of novel chitosan-based magnetic flocculants FS@CTS-P(AM-DMC)was prepared by molecular structure control.The characterization results showed that FS@CTS-P(AM-DMC)had a uniform size of about 21.46 nm,featuring a typical core-shell structure,and the average coating layer thickness of CTS-P(AM-DMC)was about 5.03 nm.FS@CTS-P(AM-DMC)exhibited excellent flocculation performance for kaolin suspension,achieved 92.54% turbidity removal efficiency under dosage of 150 mg/L,pH 7.0,even at high turbidity(2000 NTU)with a removal efficiency of 96.96%.The flocculation mechanism was revealed to be dominated by charge neutralization under acidic and neutral conditions,while adsorption and bridging effects play an important role in alkaline environments.The properties of magnetic aggregates during flocculation,breakage,and regeneration were studied at different pH levels and dosages.In the process of magnetophoretic,magnetic particles collide and adsorb with kaolin particles continuously due to magnetic and electrostatic attraction,transform into magnetic chain clusters,and then further form three-dimensional network magnetic aggregates that can capture free kaolin particles and other chain clusters.Particle image velocimetry confirmed the formation of eddy current of magnetic flocs and experienced three stages:acceleration,stabilization,and deceleration.Chuang Liu Xiaoyu Wang Sicong Du Wenyan Liang 2024Journal of Environmental Sciences2024,,5:0
10Gradient carbonyl-iron/carbon-fiber reinforced composite metamaterial for ultra-broadband electromagnetic wave absorption by multi-scale integrated design显示文摘The demand of high-end electromagnetic wave absorbing materials puts forward higher requirements on comprehensive performances of small thickness,lightweight,broadband,and strong absorption.Herein,a novel multi-layer stepped metamaterial absorber with gradient electromagnetic properties is proposed.The complex permittivity and permeability of each layer are tailored via the proportion of carbonyliron and carbon-fiber dispersing into the epoxy resin.The proposed metamaterial is further optimized via adjusting the electromagnetic parameters and geometric sizes of each layer.Comparing with the four-layer composite with gradient electromagnetic properties which could only realize reflection loss(RL)of less than−6 dB in 2.0-40 GHz,the optimized stepped metamaterial with the same thickness and electromagnetic properties realizes less than−10 dB in the relevant frequency range.Additionally,the RL of less than−15 dB is achieved in the frequency range of 11.2-21.4 GHz and 28.5-40 GHz.The multiple electromagnetic wave absorption mechanism is discussed based on the experimental and simulation results,which is believed to be attributed to the synergy effect induced by multi-scale structures of the metamaterial.Therefore,combining multi-layer structures and periodic stepped structures into a novel gradient absorbing metamaterial would give new insights into designing microwave absorption devices for broadband electromagnetic protections.Qian Zhou Tiantian Shi Bei Xue Shengyue Gu Wei Ren Fang Ye Xiaomeng Fan Wenyan Duan Zihan Zhang Lifei Du 2023International Journal of Minerals,Metallurgy and Materials2023,30,6:0
11Long-Term Extreme Wave Characteristics in the Water Adjacent to China Based on ERA5 Reanalysis Data显示文摘Extreme waves have a profound impact on coastal infrastructure;thus,understanding the variation law of risky analysis and disaster prevention in coastal zones is necessary.This paper analyzed the spatiotemporal characteristics of extreme wave heights adjacent to China from 1979 to 2018 based on the ERA5 datasets.Nonstationary extreme value analysis is undertaken in eight repre-sentative points to investigate the trends in the values of 50-and 100-year wave heights.Results show that the mean value of extreme waves is the largest in the eastern part of Taiwan Island and the smallest in the Bohai Sea from 1979 to 2018.Only the extreme wave height in the northeastern part of Taiwan Island shows a significant increase trend in the study area.Nonstationary analysis shows remarkable variations in the values of 50-and 100-year significant wave heights in eight points.Considering the annual mean change,E1,E2,S1,and S2 present an increasing trend,while S3 shows a decreasing trend.Most points for the seasonal mean change demon-strate an increasing trend in spring and winter,while other points show a decreasing trend in summer and autumn.Notably,the E1 point growth rate is large in autumn,which is related to the change in typhoon intensity and the northward movement of the typhoon path.DU Wenyan ZHANG Xuri SHI Hongyuan LI Guanyu ZHOU Zhengqiao YOU Zaijin ZHANG Kuncheng 2024Journal of Ocean University of China2024,23,1:0
12Hsa_Circ_0000826 inhibits the proliferation of colorectal cancer by targeting AUF1显示文摘Many circular RNAs(circRNAs)are reported to be abnormally expressed during the progression of various tumors,and these circRNAs can be used as anti-tumor targets.Therefore,it is important to identify circRNAs that can be used effectively for the clinical diagnosis and treatment of colorectal cancer(CRC).Here,we report that hsa_Circ_0000826(Circ_0000826),a circ RNA with significantly reduced expression level in CRC tissues,is associated with a poor prognosis in patients.The silencing of Circ_0000826 promotes the proliferation of CRC cells.Conversely,the overexpression of Circ_0000826 restricted CRC cell proliferation both in vitro and in vivo.Furthermore,Circ_0000826 could target AU-rich element RNA-binding protein 1(AUF1).AUF1,known as heterogeneous nuclear ribonucleoprotein D(hnRNP D),could bind to the c-MYC 3’-UTR and promote c-MYC expression.When Circ_0000826 binds to AUF1,it competitively inhibits the binding of AUF1 to the c-MYC 3’-UTR,which inhibits the c-MYC expression and cell proliferation.These results provide novel insights into the functional mechanism of Circ_0000826 action in CRC progression and indicate its potential use as a therapeutic target in CRC.Zheying Zhang Wenyan Fan Qingzu Gao Yifei Han Jingyu Ma Wuji Gao Yuhan Hu Huifang Zhu Rui Yang Haijun Wang Baoshun Du Zuoyang Zhang Jiateng Zhong 2023Journal of Genetics and Genomics2023,50,3:0

14A Multi-Scale Method for PM2.5 Forecasting with Multi-Source Big Data显示文摘In the age of big data,the Internet big data can finely reflect public attention to air pollution,which greatly impact ambient PM2.5 concentrations;however,it has not been applied to PM2.5 prediction yet.Therefore,this study introduces such informative Internet big data as an effective predictor for PM2.5,in addition to other big data.To capture the multi-scale relationship between PM2.5 concentrations and multi-source big data,a novel multi-source big data and multi-scale forecasting methodology is proposed for PM2.5.Three major steps are taken:1)Multi-source big data process,to collect big data from different sources(e.g.,devices and Internet)and extract the hidden predictive features;2)Multi-scale analysis,to address the non-uniformity and nonalignment of timescales by withdrawing the scale-aligned modes hidden in multi-source data;3)PM2.5 prediction,entailing individual prediction at each timescale and ensemble prediction for the final results.The empirical study focuses on the top highly-polluted cities and shows that the proposed multi-source big data and multi-scale forecasting method outperforms its original forms(with neither big data nor multi-scale analysis),semi-extended variants(with big data and without multi-scale analysis)and similar counterparts(with big data but from a single source and multi-scale analysis)in accuracy.YUAN Wenyan DU Hongchuan LI Jieyi LI Ling 2023Journal of Systems Science & Complexity2023,36,2:0
15Hydroxyapatite reinforced inorganic-organic hybrid nanocomposite as high-performance adsorbents for bilirubin removal in vitro and in pig models显示文摘Highly efficient removal of bilirubin from whole blood directly by hemoperfusion for liver failure therapy remains a challenge in the clinical field due to the low adsorption capacity,poor mechanical strength and low biocompatibility of adsorbents.In this work,a new class of nanocomposite adsorbents was constructed through an inorganic-organic co-crosslinked nanocomposite network between vinyltriethoxysilane(VTES)-functionalized hydroxyapatite nanoparticles(V-Hap)and non-ionic styrene-divinylbenzene(PS-DVB)resins(PS-DVB/V-Hap)using suspension polymerization.Notably,our adsorbent demonstrated substantially improved mechanical performance compared to the pure polymer,with the hardness and modulus increasing by nearly 3 and 2.5 times,respectively.Moreover,due to the development of a mesoporous structure,the prepared PS-DVB/V-Hap3 exhibited an ideal adsorption capacity of 40.27 mg·g^(-1).More importantly,the obtained adsorbent beads showed outstanding blood compatibility and biocompatibility.Furthermore,in vivo extracorporeal hemoperfusion verified the efficacy and biosafety of the adsorbent for directly removing bilirubin from whole blood in pig models,and this material could potentially prevent liver damage and improve clinical outcomes.Taken together,the results suggest that PS-DVB/V-Hap3 beads can be used in commercial adsorption columns to threat hyperbilirubinemia patients through hemoperfusion,thus replacing the existing techniques where plasma separation is initially required.Yamin Chai Zhuang Liu Yunzheng Du Lichun Wang Jinyan Lu Qian Zhang a Wenyan Han Tingting Wang Yameng Yu Lisha Sun Lailiang Ou 2021Bioactive Materials2021,6,12:0
16Genomic analysis of a pure culture of magnetotactic bacterium Terasakiella sp.SH-1显示文摘Magnetotactic bacteria(MTB)display magnetotaxis ability because of biomineralization of intracellular nanometer-sized,membrane-bound organelles termed magnetosomes.Despite having been discovered more than half a century,only a few representatives of MTB have been isolated and cultured in the laboratory.In this study,we report the genomic characterization of a novel marine magnetotactic spirillum strain SH-1 belonging to the genus Terasakiella that was recently isolated.A gene encoding haloalkane dehalogenase,which is involved in the degradation of chlorocyclohexane,chlorobenzene,chloroalkane,and chloroalkene,was identified.SH-1 genome contained cysCHI and soxBAZYX genes,thus potentially capable of assimilatory sulfate reduction to H_(2)S and using thiosulfate as electron donors and oxidizing it to sulfate.Genome of SH-1 also contained genes encoding periplasmic dissimilatory nitrate reductases(napAB),assimilatory nitrate reductase(nasA)and assimilatory nitrite reductases(nasB),suggesting that it is capable of gaining energy by converting nitrate to ammonia.The pure culture of Terasakiella sp.SH-1 together with its genomic results off ers new opportunities to examine biology,physiology,and biomineralization mechanisms of MTB.Haijian DU Wenyan ZHANG Wei LIN Hongmiao PAN Tian XIAO Long-Fei WU 2021Journal of Oceanology and Limnology2021,39,6:0
173D printing-based drug-loaded implanted prosthesis to prevent breast cancer recurrence post-conserving surgery显示文摘Systemic chemotherapy of breast cancer is commonly delivered as a large dose and has toxic side effects.Local chemotherapy would overcome the shortcomings of systemic reconstruction and could play an important role in breast cancer surgery according to personalized demand.The application of three-dimensional(3D)printing technology makes personalized customization possible.We designed and prepared a prosthesis containing paclitaxel(PTX)and doxorubicin(DOX)microspheres(PPDM)based on 3D printing to prevent tumor recurrence and metastasis after breast conserving surgery.Polydimethysiloxane has good biocompatibility and was used as a drug carrier in this study.The average particle size of the PTX and DOX microspheres were approximately 3.1μm and 2.2μm,respectively.The drug loading of PTX and DOX microspheres was 4.2% and 2.1%,respectively.In vitro drug release studies demonstrated that the 3D-printed prosthesis loaded with PTX and DOXmicrospheres could release the drugs continuously for more than 3 weeks and thereby suppress cancer recurrence with reduced side effects.The PTX and DOX microspheres not only exerted a synergistic effect,but also achieved a good sustained release effect.In vivo evaluation showed that the PPDM could effectively inhibit breast cancer recurrence andmetastasis inmice with breast cancer.PPDM are expected to achieve postoperative chemotherapy for breast cancer and be highly efficient to prevent local breast cancer recurrence and metastasis.Wenyan Hao Zengjuan Zheng Lin Zhu Lulu Pang JinqiuMa Siqing Zhu Lina Du Yiguang Jin 2021Asian Journal of Pharmaceutical Sciences2021,16,1:0

20Efficacy and Safety of Sofosbuvir-based Regimens in Hepatitis C Patients With Decompensated Cirrhosis:A Systematic Review and Meta-analysis显示文摘Background and Aims:Decompensated cirrhotic patients with hepatitis C(HCV)are often under-represented in clinical trials.We aimed to evaluate pooled data on the efficacy and safety of sofosbuvir(SOF)-based regimens in these patients.Methods:We conducted a systemic review and meta-analysis by searching multiple databases for studies published from October 2010 to October 2020.Outcomes of interest were sustained virologic response(SVR)and safety of SOFbased regimens in decompensated HCV patients.Two reviewers independently performed the study selection and data extraction.Results:We included 33 studies that enrolled 5,302 HCV patients.The pooled SVR rate in decompensated patients with SOF-based regimens was 85.1%(95%CI:82.8–87.3).Patients on SOF/velpatasvir±ribavirin achieved a significantly higher SVR(91.0%,95%CI:87.7–93.9)than that of SOF/ledipasvir±ribavirin[(86.3%,95%CI:84.6–87.8);p=0.004],or on SOF/daclatasvir±ribavirin(82.4%,95%CI:78.2–86.2%;p<0.001).Adding ribavirin to SOFbased regimens(pooled SVR 84.9%,95%CI:81.7–87.9)did not significantly increase the SVR[83.8%(95%CI:76.8–89.8%;p=0.76)]in decompensated patients,which was also true in subgroup analyses for each regimen within the same treatment duration.However,adding ribavirin significantly increased the frequency of adverse events from 52.9%(95%CI:28.0–77.1)to 89.2%(95%CI:68.1–99.9)and frequency of severe events.The pooled incidence of hepatocellular carcinoma and case-fatality of decompensated patients were 3.1%(95%CI:1.5–5.0)and 4.6%(95%CI:3.1–6.3),respectively.The overall heterogeneity was high.There was no publication bias.Conclusions:The analysis found that 12 weeks of SOF/velpatasvir without ribavirin is the preferred therapy,with a significantly higher SVR compared with other SOF-based regimens in decompensated HCV patients.Wenyan Zhang Jing Zhang Shan Tang Yali Liu Xiaofei Du Lixia Qiu Menglu Liu Haibin Yu Calvin Q.Pan 2023Journal of Clinical and Translational Hepatology2023,11,1:0
