1Application of Principal Component Analysis(PCA)to the Evaluation and Screening of Multiactivity Fungi显示文摘Continued innovation in screening methodologies remains important for the discovery of high-quality multiactive fungi,which have been of great significance to the development of new drugs.Mangrove-derived fungi,which are well recognized as prolific sources of natural products,are worth sustained attention and further study.In this study,118 fungi,which mainly included Aspergillus spp.(34.62%)and Penicillium spp.(15.38%),were isolated from the mangrove ecosystem of the Maowei Sea,and 83.1%of the cultured fungi showed at least one bioactivity in four antibacterial and three antioxidant assays.To accurately evaluate the fungal bioactivities,the fungi with multiple bioactivities were successfully evaluated and screened by principal component analysis(PCA),and this analysis provided a dataset for comparing and selecting multibioactive fungi.Among the 118 mangrove-derived fungi tested in this study,Aspergillus spp.showed the best comprehensive activity.Fungi such as A.clavatonanicus,A.flavipes and A.citrinoterreus,which exhibited high comprehensive bioactivity as determined by the PCA,have great potential in the exploitation of natural products and the development of new drugs.This study demonstrated the first use of PCA as a time-saving,scientific method with a strong ability to evaluate and screen multiactive fungi,which indicated that this method can affect the discovery and development of new drugs.YANG Zonglin SHI Yaqi LI Pinglin PAN Kanghong LI Guoqiang LI Xianguo YAO Shuo ZHANG Dahai 2022Journal of Ocean University of China2022,21,3:4
2EFFECTS OF INJECTION STRATEGY ON D.I. DIESEL ENGINE COMBUSTION显示文摘Experiments are conducted to develop an understanding of how split injections can affect the combustion and emission characteristics of a D.I. diesel engine with a common-rail injection system. The ratio of the amount of fuel injected between two injection pulses and the injection interval is varied keeping the injected fuel quantity constant. Results show that under the 70D90-10 injection pattern, the engine achieves the lower NOx-smoke emissions and BSFC compared with the single injection pattern. The heat release rate and the temperature show that the split injections increase the initial premixed burn and retards the diffusion burn. With the balance of these two effects, the maximum in-cylinder temperature decreases while the 50% heat release point is held at almost the same crank angle. 3Polarization-sensitive and wide-spectrum photovoltaic detector based on quasi-1D ZrGeTe_(4)nanoribbon显示文摘Low-dimensional semiconductors with in-plane anisotropy and narrow bandgap have been extensively applied to polarized detection in the near-infrared(NIR)region.However,the narrow bandgap can cause noise owing to the high dark current in photodetectors.This article reports quasi-1D ZrGeTe_(4)nanoribbonbased photodetectors with low dark current and broadband polarization detection.The photodetector was fabricated by evaporating 50-nm-thick Au electrodes on a ZrGeTe_(4)nanoribbon.Benefiting from the photovoltaic characteristics in the ZrGeTe_(4)nanoribbon and Au electrodes,these photodetectors can operate without bias voltage,with decreased dark current,and improved device performance.Furthermore,the quasi-1D ZrGeTe_(4)nanoribbon-based photodetectors demonstrate a polarization sensitivity in a broadband from visible(VIS)to the NIR region,such as a high photoresponsivity of 625.65 mA W1,large external quantum efficiency of 145.9%at 532 nm,and photocurrent anisotropy ratio of 2.04 at 1064 nm.They exhibit a novel perpendicular optical reversal of 90in polarization-sensitive photodetection,angle-resolved absorption spectra,and azimuth-dependent reflectance difference microscopy(ADRDM)from VIS to the NIR region,as opposed to other nanoribbon-based polarization-sensitive photodetectors.This work paves the way for utilizing photovoltaic photodetectors based on low-dimensional materials for broad-spectrum polarized photodetection.Ruixue Bai Tao Xiong Jinshu Zhou Yue-Yang Liu Wanfu Shen Chunguang Hu Faguang Yan Kaiyou Wang Dahai Wei Jingbo Li Juehan Yang Zhongming Wei 2022InfoMat2022,4,3:2
4The Notch pathway attenuates burn-induced acute lung injury in rats by repressing reactive oxygen species显示文摘Background:Acute lung injury(ALI)is a common complication following severe burns.The underlying mechanisms of ALI are incompletely understood;thus,available treatments are not sufficient to repair the lung tissue after ALI.Methods:To investigate the relationship between the Notch pathway and burn-induced lung injury,we established a rat burn injury model by scalding and verified lung injury via lung injury evaluations,including hematoxylin and eosin(H&E)staining,lung injury scoring,bronchoalveolar lavage fluid and wet/dry ratio analyses,myeloperoxidase immunohistochemical staining and reac-tive oxygen species(ROS)accumulation analysis.To explore whether burn injury affects Notch1 expression,we detected the expression of Notch1 and Hes1 after burn injury.Then,we extracted pulmonary microvascular endothelial cells(PMVECs)and conducted Notch pathway inhibition and activation experiments,via aγ-secretase inhibitor(GSI)and OP9-DLL1 coculture,respectively,to verify the regulatory effect of the Notch pathway on ROS accumulation and apoptosis in burn-serum-stimulated PMVECs.To investigate the regulatory effect of the Notch pathway on ROS accumulation,we detected the expression of oxidative-stress-related molecules such as superoxide dismutase,nicotinamide adenine dinucleotide phosphate(NADPH)oxidase(NOX)2,NOX4 and cleaved caspase-3.NOX4-specific small interfering RNA(siRNA)and the inhibitor GKT137831 were used to verify the regulatory effect of the Notch pathway on ROS via NOX4.Results:We successfully established a burn model and revealed that lung injury,excessive ROS accumulation and an inflammatory response occurred.Notch1 detection showed that the expression of Notch1 was significantly increased after burn injury.In PMVECs challenged with burn serum,ROS and cell death were elevated.Moreover,when the Notch pathway was suppressed by GSI,ROS and cell apoptosis levels were significantly increased.Conversely,these parameters were reduced when the Notch pathway was activated by OP9-DLL1.Mechanistically,the inhibition of NOX4 by siRNA and GKT137831 showed that the Notch pathway reduced ROS production and cell apoptosis by downregulating the expression of NOX4 in PMVECs.Conclusions:The Notch pathway reduced ROS production and apoptosis by downregulating the expression of NOX4 in burn-stimulated PMVECs.The Notch-NOX4 pathway may be a novel therapeutic target to treat burn-induced ALI.Weixia Cai Kuo Shen Peng Ji Yanhui Jia Shichao Han Wanfu Zhang Xiaolong Hu Xuekang Yang Juntao Han Dahai Hu 2022Burns & Trauma2022,10,1:2

11Silencing JA hydroxylases in Nicotiana attenuata enhances jasmonic acid-isoleucine-mediated defenses against Spodoptera litura显示文摘Jasmonic acid(JA)plays important roles in plant resistance to insect herbivores.One important derivative of JA is 12-OH-JA,which is produced by two independent pathways:direct hydroxylation of JA by jasmonate-induced oxygenases(JOXs)or hydrolyzation of 12-OH-JA-Ile.Yet the function of 12-OH-JA in plant-herbivore interactions remains largely unknown.In this study,we silenced four JOX homologs independently in the wild tobacco Nicotiana attenuata by virus-induced gene silencing(VIGS),and found that all four JOX homologs are involved in JA hydroxylation.Simultaneously silencing the four JA hydroxylases in VIGS-NaJOXs plants decreased herbivory-induced 12-OH-JA by 33%,but JA and JA-Ile levels increased by 45%and 30%,respectively,compared to those in control plants.Compared to direct hydroxylation from JA,hydrolyzation from 12-OH-JA-Ile is equally important for herbivory-induced 12-OHJA accumulation:in the 12-OH-JA-Ile deficient irJAR4/6 plants,12-OH-JA decreased 34%.Moreover,VIGSNaJOXs plants exhibited enhanced resistance to the generalist herbivore Spodoptera litura.The poor larval performance was strongly correlated with high levels of several JA-Ile-dependent direct defense metabolites in the VIGS-NaJOXs plants.When we simultaneously silenced all four JA hydroxylases in the JAIle-deficient irJAR4/6 background,the enhanced herbivore resistance diminished,demonstrating that enhanced herbivore resistance resulted from elevated JA-Ile levels.Given that silencing these NaJOX-like genes did not detectably alter plant growth but highly increased plant defense levels,we propose that JOX genes are potential targets for genetic improvement of herbivore-resistant crops.Jinxiang Tang Dahai Yang Jianqiang Wu Suiyun Chen Lei Wang 2020Plant Diversity2020,42,2:1

13Spin-decoupled metalens with intensity-tunable multiple focal points显示文摘The control of spin electromagnetic(EM)waves is of great significance in optical communications.Although geometric metasurfaces have shown unprecedented capability to manipulate the wavefronts of spin EM waves,it is still challenging to independently manipulate each spin state and intensity distribution,which inevitably degrades metasurface-based devices for further applications.Here we propose and experimentally demonstrate an approach to designing spin-decoupled metalenses based on pure geometric phase,i.e.,geometric metasurfaces with predesigned phase modulation possessing functionalities of both convex lenses and concave lenses.Under the illumination of left-/right-handed circularly polarized(LCP or RCP)terahertz(THz)waves,these metalenses can generate transversely/longitudinally distributed RCP/LCP multiple focal points.Since the helicity-dependent multiple focal points are locked to the polarization state of incident THz waves,the relative intensity between two orthogonal components can be controlled with different weights of LCP and RCP THz waves,leading to the intensity-tunable functionality.This robust approach for simultaneously manipulating orthogonal spin states and energy distributions of spin EM waves will open a new avenue for designing multifunctional devices and integrated communication systems.BINGSHUANG YAO XIAOFEI ZANG YANG ZHU DAHAI YU JINGYA XIE LIN CHEN SEN HAN YIMING ZHU SONGLIN ZHUANG 2021Photonics Research2021,9,6:1
14An exploratory study on road tunnel with semi-transparent photovoltaic canopy-From energy saving and fire safety perspectives显示文摘Road tunnels consume a large amount of energy,especially in the Canadian cold climate,where the roads are heated electrically or deicing during the winter.For a more sustainable and resilient road tunnel energy system,we conducted an exploratory study on installing a semi-transparent photovoltaic(STPV)canopy at the entrances and exits of a tunnel under a river.The proposed system generates solar-powered electricity,improves thermal and visual conditions,and reduces energy loads.In this study,field measurements of road surface temperature and air temperature were conducted,and numerical simulations with and without STPV were performed to study air and road surface temperatures under different traffic speeds.The field measurements show the road surface temperatures are higher than the air temperature on average.The interior air and road surface temperature were measured to be above 0°C,even though the outdoor temperature is far below 0°C,thus significantly reducing the need for deicing in winter using salts.The simulations show that the air and surface temperatures elevate due to the solar transmission heat through the STPV canopy,thus reducing deicing energy consumption significantly.The fire safety analysis also showed that the proposed system's top opening should be located near the tunnel entrance instead of the canopy entrance for better smoke exhaust during a fire.Dahai Qi Senwen Yang Chang Shu Xin Zhang Liangzhu(Leon)Wang Andreas Athienitis 2022Building Simulation2022,15,4:1
15Multi-tissue scRNA-seq reveals immune cell landscape of turbot (Scophthalmus maximus)显示文摘In vertebrates,bony fishes possess not only innate immune cells but also T and B cells that are equivalent to those in mammals.However,the precise sub-cluster of immune cells in teleost fish remains largely unknown.Herein,we developed a dynamic bacterial infection model in turbot(Scophthalmus maximus)and created a fish immune cell landscape(FICL)for a primary lymphoid organ(head kidney),a secondary lymphoid organ(spleen),and barrier tissues(gill and posterior intestine).Moreover,through comprehensive characterization of the expression profiles of 16 clusters,including dendritic cells-like(DCs-like),macrophages(Mos),neutrophils,NK cells,as well as 12 sub-clusters of T and B cells,we found that CD8+CTLs,CD4-CD8-T,Th17 and ILC3-2 like cells posssa bifunctional role associated with cytotoxicity and immunoregulation during bacterial infection.To our knowledge,these results could provide a useful resource for a better understanding of immune cells in teleost fish and could act as a comprehensive knowledge base for assessing the evolutionary mechanism of adaptive immunity in vertebrates.Weijie Chen Jianchang Huang Wei Wang Ying Wang Hao Chen Qiyao Wang Yuanxing Zhang Qin Liu Dahai Yang 2022Fundamental Research2022,2,4:0
16CTNNAL1 deficiency suppresses CFTR expression in HDM-induced asthma mouse model through ROCK1-CAL signaling pathway显示文摘The downregulation of adhesion molecule catenin alpha-like 1(CTNNAL1)in airway epithelial cells of asthma patients and house dust mite(HDM)-induced asthma animal models was illustrated in our previous study.It is assumed to contribute to airway inflammation and mucus hypersecretion.In this work,we further explore the underlying mechanism of CTNNAL1 in asthma.CTNNAL1-silenced female mice exhibit a decreased level of cystic fibrosis transmembrane conductance regulator(CFTR),a cAMP-activated and ATP-gated Cl–channel that correlates with mucus hypersecretion.Our previous study demonstrated that ROCK1 expression decreases but ROCK2 expression increases in the lungs of a CTNNAL1-silenced mouse model.Inhibition of ROCK1 leads to a reduction in CFTR expression in CTNNAL1-overexpressing and CTNNAL1-silenced human bronchial epithelial(HBE)cells.It has been reported that ROCK1 is a downstream target of RhoA and that activation of RhoA increases CFTR expression after CTNNAL1 deficiency in vitro and in vivo.The above results indicate that CTNNAL1 regulates CFTR expression through the ROCK1 pathway.In addition,the expression of CFTR-associated ligand(CAL)is increased after CTNNAL1 silencing,and immunoprecipitation results confirm the interaction between ROCK1 and CAL.Inhibition of CAL does not influence ROCK1 expression but increases CFTR expression in CTNNAL1-silenced HBE cells.These data suggest that CTNNAL1 deficiency decreases CFTR expression in the HDM-induced asthma mouse model through the ROCK1-CAL signaling pathway.Di Wu Jiahui Zhu Fang Yang Riwang Li Lexin Liu Dahai Liu Chi Liu Xiangping Qu Huijun Liu Ming Ji Xiaoqun Qin Lan Hua Yang Xiang 2023Acta Biochimica et Biophysica Sinica2023,55,10:0
17Assessment of popular techniques for co-processing municipal solid waste in Chinese cement kilns显示文摘Cement kiln co-processing techniques have been developed in the past 20 years in China,and more than 60 factories now use fermentation,screening,and gasification pre-treatment techniques to coprocess municipal solid waste(MSW).There three complete MSW pre-treatment techniques,coprocessing procedures,and environmental risk assessments have been described in few publications.In this study,we assessed the effectiveness of each technique.The results suggested that the pollutant content released by each pre-treatment technology was lower than the emission standard.To reveal the mechanisms of pollutant migration and enrichment,the substances in the kiln and kiln products are investigated.The input of co-processing materials(Co-M)produced by fermentation caused formation of polychlorinated dibenzo-p-dioxins and dibenzoftiran(PCDD/Fs)in the bypass flue gas(By-gas)in excess of the regulatory standard.The Co-M input produced by the screening and gasifier technologies caused the total organic carbon(TOC)concentration to exceed the standard.In addition,the NOx,TOC,and PCDD/Fs in the By-gas exceeded the regulatory standard.Raw meal was the primary chlorine and heavy metals input stream,and clinker(CK)and cement kiln dust(CKD)accounted for>90%of the total chlorine output stream.Flue gas and CKD were the primary volatile heavy metal(Hg)output streams.Greater than 70%of the semi-volatile heavy metals(Cd,Pb,T1 and Se)distributed in hot raw meal and bypass cement kiln dust.The low-volatility heavy metals were concentrated in the CK.These results indicated that co-processing techniques used in China still require improvement.Hua Long Yang Liao Changhao Cui Meijia Liu Zeiwei Liu Li Li Wenzheng Hu Dahai Yan 2022Frontiers of Environmental Science & Engineering2022,16,4:0

19miR-378 and its host gene Ppargc1β exhibit independent expression in mouse skeletal muscle显示文摘MicroRNAs (miRNAs) are implicated in multiple biological processes in physiological and pathological settings. Nearly half of the known miRNAs are classified as ‘intronic’ miRNAs because they are embedded within the introns of protein-coding or noncoding genes. Such miRNAs were thought to be processed from primary host gene transcripts and share the promoter of their host. Recent analyses predicted that some intronic miRNAs might be transcribed and regulated as independent units, but there is little direct evidence for this in a specific biological context. Here, we focused on miR-378, which is located within the first intron of the peroxisome proliferator-activated receptor γ coactivator 1-beta (Ppargc1β) gene and critically regulates skeletal muscle cell differentiation and muscle regeneration. We demonstrate that miR-378 and Ppargc1β exhibit distinct expression patterns during skeletal muscle cell differentiation. In terminally differentiated adult skeletal muscle tissues of mice, miR-378 is predominantly expressed in glycolytic muscle, whereas Ppargc1β is mainly expressed in oxidative soleus muscle. Mechanistically, miR-378, but not Ppargc1β, is regulated by the transcription factor, MyoD, in muscle cells.
