1Parkin deficiency accentuates chronic alcohol intake-induced tissue injury and autophagy defects in brain,liver and skeletal muscle显示文摘Alcoholism leads to organ injury including mitochondrial defect and apoptosis with evidence favoring a role for autophagy dysregulation in alcoholic damage.Parkin represents an autosomal recessive in herited gene for Parkins on's disease and an important member of selective autophagy for mitoch on dria.The associati on between Parkins on's disease and alcoholic injury remains elusive.This study aimed to examine the effect of parki n deficiency on chr onic alcohol in takeinduced organ injury in brain,liver and skeletal muscle(rectus femoris muscle).Adult parkinknockout(PRK-/-)and wild-type mice were placed on Liber-De Carli alcohol liquid diet(4%)for 12 weeks prior to assessment of liver enzymes,intraperitoneal glucose toleranee,protein carbonyl content apoptosis,hematoxylin and eosin morphological staining,and mitochondrial respiration(cytochrome c oxidase,NADH:cytochrome c reductase and succinate:cytochrome c reductase).Autophagy protein markers were monitored by western blot analysis.Our data revealed that chronic alcohol intake imposed liver injury as evidenced by elevated aspartate aminotransferase and alanine transaminase,glucose intoleranee,elevated protein carbonyl formation,apoptosis,focal inflammation,necrosis,microvesiculation,autophagy/mitophagy failure and dampened mitochondrial respiration(complexⅣ,complexesⅠandⅢ,and complexesⅡandⅢ)in the brain,liver and rectus femoris skeletal muscle.Although parkin ablation itself did not generate any notable effects on liver enzymes,insulin sensitivity,tissue carbonyl damage,apoptosis,tissue morphology,autophagy or mitochondrial respiration,it accentuated alcohol intake-induced tissue damage,apoptosis,morphological change,autophagy/mitophagy failure and mitochondrial injury without affecting insulin sensitivity.These data suggest that parkin plays an integral role in the preservation against alcohol-induced organ injury,apoptosis and mitochondrial damage.Hu Peng Xing Qin Sainan Chen Asli E Ceylan Maolong Dong Zhaofen Lin Jun Ren 2020Acta Biochimica et Biophysica Sinica2020,52,6:3
2Adaptive accurate tracking control of HFVs in the presence of dead-zone and hysteresis input nonlinearities显示文摘A novel accurate tracking controller is developed for the longitudinal dynamics of Hypersonic Flight Vehicles(HFVs)in the presence of large model uncertainties,external disturbances and actuator nonlinearities.Distinct from the state-of-the-art,besides being continuity,no restrictive conditions have been imposed on the HFVs dynamics.The system uncertainties are skillfully handled by being seen as bounded'disturbance terms'.In addition,by means of backstepping adaptive technique,the accurate tracking(i.e.tracking errors converge to zero as time approaches infinity)rather than bounded tracking(i.e.tracking errors converge to residual sets)has been achieved.What’s more,the accurate tracking problems for HFVs subject to actuator dead-zone and hysteresis are discussed,respectively.Then,all signals of closed-loop system are verified to be Semi-Global Uniformly Ultimate Boundness(SGUUB).Finally,the efficacy and superiority of the developed control strategy are confirmed by simulation results.Zehong DONG Yinghui LI Maolong LV Renwei ZUO 2021Chinese Journal of Aeronautics2021,34,5:3
3Development and molecular analysis of a novel acetohydroxyacid synthase rapeseed mutant with high resistance to sulfonylurea herbicides显示文摘With the increasing promotion of simplified rapeseed cultivation in recent years,the development of cultivars with high resistance to herbicides is urgently needed.We previously developed M342,which shows sulfonylurea herbicide resistance,by targeting acetohydroxyacid synthase(AHAS),a key enzyme in branched-chain amino acid synthesis.In the present study,we used a progeny line derived from M342 for an additional round of ethyl methane sulfonate mutagenesis,yielding the novel mutant DS3,which harbored two mutations in AHAS genes and showed high sulfonylurea resistance.One mutation was the substitution Trp574 Leu,as in M342,according to Arabidopsis protein sequencing.The other site was a newly recognized substitution,Pro197 Leu.A KASP marker targeting Pro197 Leu was developed and reliably predicted the response to sulfonylurea herbicides in the F2 population.The combination of Trp574 Leu and Pro197 Leu in DS3 produced a synergistic effect that greatly increased herbicide resistance.Analysis of the protein structures of AHAS1 and AHAS3 in wild-type and single-gene mutant plants revealed three-dimensional protein conformational changes that could account for differences in herbicide resistance characteristics including toxicity tolerance,AHAS enzyme activity,and AHAS gene expression.Yue Guo Changle Liu Weihua Long Jianqin Gao Jiefu Zhang Song Chen Huiming Pu Maolong Hu 2022The Crop Journal2022,10,1:3
4Cardamonin protects against lipopolysaccharide-induced myocardial contractile dysfunction in mice through Nrf2-regulated mechanism显示文摘In patients with sepsis,lipopolysaccharide(LPS)from the outer membrane of gram-negative bacteria triggers cardiac dysfunction and heart failure,but target therapy for septic cardiomyopathy remains unavailable.In this study we evaluated the beneficial effects of cardamonin(CAR),a flavone existing in Alpinia plant,on endotoxemia-induced cardiac dysfunction and the underlying mechanisms with focus on oxidative stress and apoptosis.Adult mice were exposed to LPS(4 mg/kg,i.p.for 6 h)prior to functional or biochemical assessments.CAR(20 mg/kg,p.o.)was administered to mice immediately prior to LPS challenge.We found that LPS challenge compromised cardiac contractile function,evidenced by compromised fractional shortening,peak shortening,maximal velocity of shortening/relengthening,enlarged LV end systolic diameter and prolonged relengthening in echocardiography,and induced apoptosis,overt oxidative stress(O_(2)^(−)production and reduced antioxidant defense)associated with inflammation,phosphorylation of NF-κB and cytosolic translocation of transcriptional factor Nrf2.These deteriorative effects were greatly attenuated or mitigated by CAR administration.However,H&E and Masson’s trichrome staining analysis revealed that neither LPS challenge nor CAR administration significantly affected cardiomyocyte cross-sectional area and interstitial fibrosis.Mouse cardiomyocytes were treated with LPS(4µg/mL)for 6 h in the absence or presence of CAR(10μM)in vitro.We found that addition of CAR suppressed LPS-induced defect in cardiomyocyte shortening,which was nullified by the Nrf2 inhibitor ML-385 or the NF-κB activator prostratin.Taken together,our results suggest that CAR administration protects against LPS-induced cardiac contractile abnormality,oxidative stress,apoptosis,and inflammation through Nrf2-and NF-κB-dependent mechanism.Ying Tan Hong-hong Wan Ming-ming Sun Wen-jing Zhang Maolong Dong Wei Ge Jun Ren Hu Peng 2021Acta Pharmacologica Sinica2021,42,3:2
5Numerical study on transverse deformation characteristics of shield tunnel subject to local soil loosening显示文摘In this study,a refined numerical model for segmental lining of a shield tunnel,which contains detailed models of reinforcement and connecting bolts,is established using finite element software.The model is first validated by the results from a full-scale model test.Then,based on the load-structure method,this numerical model is adopted to investigate the internal force distribution and the transverse deformation characteristics of the shield tunnel when it is subject to local soil loosening.The influence of loosening position,loosening range,and loosening extent on the mechanical response is extensively studied through comprehensive numerical analyses.The results show that the main influence of local soil loosening on the ring is to disturb the force balance and change the constraint conditions,thus changing the deformation pattern and force state.After the loosening occurs,the bending moment of the ring in the loosening range increases and the axial force decreases.The vertical convergence of the ring is the largest and the equivalent stiffness of the ring is the smallest when the local soil loosened at the haunch and the loosening range a is 90°.The vertical convergence of the ring increases with increasing of the loosening extent,and the equivalent stiffness decreases linearly with increasing of the loosening extent.The results can enhance our understanding of mechanical behaviors of segmental lining associated soil loosening,and will show a possible way for detecting soil loosening based on the measured deformation and internal forces.Dong Su Weijie Chen Xuetao Wang Maolong Huang Xiaochao Pang Xiangsheng Chen 2022Underground Space2022,7,1:2

8Detection of Prognostic Biomarkers for Hepatocellular Carcinoma through CircRNA-associated CeRNA Analysis显示文摘Background and Aims:The prognosis of hepatocellular carcinoma(HCC)is extremely poor;therefore,there is an urgent need for novel prognostic molecular biomarkers of HCC.The current investigation utilized circular(circ)RNA-associated competing endogenous(ce)RNAs analysis in order to identify significant prognostic biomarkers of HCC.Methods:CircRNAs and mRNAs that were differentially expressed between normal and HCC tissues were identi-fied.Their respective functions were predicted with Gene Ontology enrichment and Kyoto Encyclopedia of Genes and Genomes enrichment analyses.A nomogram was used for model verification.Results:A ceRNA network composed of differentially expressed circRNAs and mRNAs was con-structed.Significant hub nodes in the ceRNA network were hsa_circ_0004662,hsa_circ_0005735,hsa_circ_0006990,hsa_circ_0018403 and hsa_circ_0100609.By using this in-formation,a prognostic risk assessment tool was developed based on the expressions of seven genes(PLOD2,TARS,RNF19B,CCT2,RAN,C5orf30 and MCM10).Furthermore,multivariate Cox regression analysis revealed risk and T-stage parameters as independent prognostic factors.The nomograms that were constructed from risk and T-stage groups were used to further assess the prediction of HCC patient survival rates.The nomogram,which consisted of risk and T-stage scores assessment models,was found to be an independent factor for predicting prognosis of HCC.Conclusions:Five circRNAs,including hsa_circ_0004662,hsa_circ_0005735,hsa_circ_0006990,hsa_circ_0018403 and hsa_circ_0100609,that may play key roles in the pro-gression of HCC were identified.Seven gene signatures were identified,which were associated with the aforemen-tioned circRNAs,including PLOD2,TARS,RNF19B,CCT2,RAN,C5orf30 and MCM10,all of which were significant genes involved in the pathophysiology of HCC.These genes may be used as a prognosticating tool in HCC patients.Li Han Maolong Wang Yuling Yang Hanlin Xu Lili Wei Xia Huang 2022Journal of Clinical and Translational Hepatology2022,10,1:1

11Experimental investigation on the invert stability of operating railway tunnels with different drainage systems using 3D printing technology显示文摘In recent years, the invert anomalies of operating railway tunnels in water-rich areas occur frequently,which greatly affect the transportation capacity of the railway lines. Tunnel drainage system is a crucial factor to ensure the invert stability by regulating the external water pressure(EWP). By means of a threedimensional(3D) printing model, this paper experimentally investigates the deformation behavior of the invert for the tunnels with the traditional drainage system(TDS) widely used in China and its optimized drainage system(ODS) with bottom drainage function. Six test groups with a total of 110 test conditions were designed to consider the design factors and environmental factors in engineering practice,including layout of the drainage system, blockage of the drainage system and groundwater level fluctuation. It was found that there are significant differences in the water discharge, EWP and invert stability for the tunnels with the two drainage systems. Even with a dense arrangement of the external blind tubes, TDS was still difficult to eliminate the excessive EWP below the invert, which is the main cause for the invert instability. Blockage of drainage system further increased the invert uplift and aggravated the track irregularity, especially when the blockage degree is more than 50%. However, ODS can prevent these invert anomalies by reasonably controlling the EWP at tunnel bottom. Even when the groundwater level reached 60 m and the blind tubes were fully blocked, the invert stability can still be maintained and the railway track experienced a settlement of only 1.8 mm. 13Gas accumulation conditions and key technologies for exploration & development of Sinian and Cambrian gas reservoirs in Anyue gasfield显示文摘Exploration of the Sinian-Lower Paleozoic formations in the Sichuan Basin starts from the 1950s,and is tortuous and complex.To summarize the exploration experiences is of great significance of reference for exploration of oil and gas in deep carbonate rocks of the Sichuan Basin.Gas accumulation in the Anyue oversize gasfield is chiefly controlled by the Deyang-Anyue intra-platform rift and central Sichuan paleouplift.The intra-platform rift controls the scale hydrocarbon generation center and effective hydrocarbon plays.The Anyue gasfield is characterized by near-source hydrocarbon accumulation.The marginal zone of intra-platform rift and high part of the paleouplift control the scale distribution of favorable facies belts of reservoir development,and overlapped by paleokarstic reformation,the reservoirs are widely distributed.The intra-platform rift and paleouplift jointly control formation of the scale traps and hydrocarbon enrichment.Gas reservoir features and major controlling factors for hydrocarbon accumulation have some differences in the main play strata,such as Longwangmiao Formation,and Member 4 and Member 2 of Dengying Formation.The gas reservoir in the Longwangmiao Formation is a lithologic gas reservoir under the tectonic setting,and the inherited paleouplift is a key for reservoir formation and hydrocarbon accumulation.The gas reservoir in the Member 4 of Dengying Formation is mainly controlled by large structural-stratigraphic composite traps,and the gas reservoir in the Member 2 of Dengying Formation is the structural trap.Distribution of the large gasfields in the Dengying Formation is controlled by the intra-platform rift,near-source hydrocarbon-reservoir configuration controlled by the ancient erosion plane and stratigraphic sealing in the updip direction.For exploration and development of the Anyue gasfield,four series of technologies are developed,namely,reservoir seismic fine description technology,well logging fine evaluation technology of gas layers,fast drilling technology of complex strata,and stimulation technology for high temperature and high pressure gas layers.Technical progresses provide powerful guarantee for efficient exploration and development of the oversize gasfields.Yueming Yang Yu Yang Guang Yang Jiarong Song LongWen Chuanguang Deng Maolong Xia Qi Ran Guobin Duan Bing Luo Bing Xie 2018Petroleum Research2018,3,3:1
14Low-voltage and high-gain WSe_(2)avalanche phototransistor with an out-of-plane WSe_(2)/WS_(2)heterojunction显示文摘The properties of photodetectors based on two-dimensional materials can be significantly enhanced by avalanche effect.However,a high avalanche breakdown voltage is needed to reach impact ionization,which leads to high power consumption.Here,we report the unique features of a low-voltage avalanche phototransistor formed by an in-plane WSe_(2)field effect transistor(FET)with an out-of-plane WSe_(2)/WS_(2)P–N heterojunction(HJ FET).The avalanche breakdown voltage in the device can be decreased from−31 to−8.5 V when compared with that in WSe_(2)FET.The inherent mechanism is mainly related to the redistributed electric field in the WSe_(2)channel after the formation of the out-of-plane P–N heterojunction.When the bias voltage is−16.5 V,the photoresponsivity in the HJ FET is enhanced from 1.5 to 135 A/W,which is significantly higher than that in the WSe_(2)FET because of the obvious reduction of the avalanche breakdown voltage.Moreover,HJ FET shows a higher responsivity than WSe_(2)FET in the range of 400–1,100 nm under low bias voltage.This phenomenon is caused by accelerating electron–hole spatial separation in the heterojunction.These results indicate that the use of an WSe_(2)FET with an out-of-plane WSe_(2)/WS_(2)heterojunction is ideal for high-performance photodetectors with low power consumption.Lingyao Meng Ningning Zhang Maolong Yang Xixi Yuan Maliang Liu Huiyong Hu Liming Wang 2023Nano Research2023,16,2:1

19Approach for uncertain multi-objective programming problems with correlated objective functions under C_(EV) criterion显示文摘An uncertain multi-objective programming problem is a special type of mathematical multi-objective programming involving uncertain variables. This type of problem is important because there are several uncertain variables in real-world problems.Therefore, research on the uncertain multi-objective programming problem is highly relevant, particularly those problems whose objective functions are correlated. In this paper, an approach that solves an uncertain multi-objective programming problem under the expected-variance value criterion is proposed. First, we define the basic framework of the approach and review concepts such as a Pareto efficient solution and expected-variance value criterion using an order relation between various uncertain variables.Second, the uncertain multi-objective problem is converted into an uncertain single-objective programming problem via a linear weighted method or ideal point method. Then the problem is transformed into a deterministic single objective programming problem under the expected-variance value criterion. Third, four lemmas and two theorems are proved to illustrate that the optimal solution of the deterministic single-objective programming problem is an efficient solution to the original uncertainty problem.
