6Clinical significance of connective tissue growth factor in hepatitis B virus-induced hepatic fibrosis显示文摘AIM:To determine the utility of connective tissue growth factor(CCN2/CTGF) for assessing hepatic fibrosis in hepatitis B virus(HBV)-induced chronic liver diseases(CLD-B).METHODS:Enzyme-linked immunosorbent assay was used to measure CCN2 in sera from 107 patients with chronic hepatitis B(CHB) and 39 patients with HBVinduced active liver cirrhosis and 30 healthy individuals.Liver samples from 31 patients with CHB,8 patients with HBV-induced liver cirrhosis and 8 HBV carriers with normal liver histology were examined for transforming growth factor β-1(TGF-β1) or CCN2 mRNA levels by in situ hybridization,and computer image analysis was performed to measure integrated optimal density(IOD) of CCN2 mRNA-positive cells in liver tissues.Histological inflammation grading and fibrosis staging were evaluated by H and E staining and Van Gieson's method.RESULTS:Serum CCN2 concentrations were,respectively,4.0-or 4.9-fold higher in patients with CHB or active liver cirrhosis as compared to healthy individuals(P < 0.01).There was good consistency between the levels of CCN2 in sera and CCN2 mRNA expression in liver tissues(r = 0.87,P < 0.01).The levels of CCN2 in sera were increased with the enhancement of histological fibrosis staging in patients with CLD-B(r = 0.85,P < 0.01).Serum CCN2 was a reliable marker for the assessment of liver fibrosis,with areas under the receiver operating characteristic(ROC) curves(AUC) of 0.94 or 0.85 for,respectively,distinguishing normal liver controls from patients with F1 stage liver fibrosis or discriminating between mild and significant fibrosis.CONCLUSION:Detection of serum CCN2 in patients with CLD-B may have clinical significance for assessment of severity of hepatic fibrosis.Rong-Li Piao David R Brigstock Jie Zhu Man-Li Zhang Run-Ping Gao 2012World Journal of Gastroenterology2012,18,18:12
7Sedimentary cycles of trace elements in Salawusu River Valley since 150 ka BP显示文摘The paper makes some analyses on 11 trace elements in the Milanggouwan stratigraphical section in the Salawusu River valley, which is regarded as a prototype geology-palaeoclimate record since 150 ka BP. The results show that the content and variation of trace elements has experienced remarkably regular changes in the pace with coarse and fine sedimentary cycles of palaeo-aeolian sands to its overlying fluvio-lacustrine facies or/and palaeosols. The trace elements with chemical properties of relatively active (V, Sr, Cu, Ni, As) and relatively stable (P, Pb, Rb, Mn, Nb, Zr) are a manifestation of the corresponding 27 changeable cycles between peak and valley values, appearing a multi-fluctuational process line of relative gathering and migration since then. The low numerical value distribution of these two types of trace elements in the aeolian sand facies represents erosion and accumulation under wind force during the cold-dry climate. Whereas their enrichments in both fluvio-lacustrine facies and palaeosols are related to the valley抯 special low-lying physiognomic position between the Ordos Plateau and the Loess Plateau under the warm and humid climate conditions. 9Electro-Pulse-Boring(EPB): Novel Super-Deep Drilling Technology for Low Cost Electricity显示文摘The inexhaustible heat deposit in great depths(5–10km) is a scientific fact. Such deposit occurs around the globe. Thereby,everybody is enabled to generate autonomously clean and renewable energy,ample electricity and heat. The economical exploration and exploitation of this superdeep geothermal heat deposit requires a novel drilling technique,because the currently only deep drilling method(Rotary) is limited to about 5km,due to the rising costs,depending exponentially on depth. Electro-pulse-boring(EPB) is a valuable option to Rotary drilling. EPB,originally investigated in Russia,is ready to be developed for industrialization. The feasibility of EPB is proven by many boreholes drilled up to 200 m in granite(crystalline). Estimates show outstanding low costs for drilling by EPB: 100 €/m for a borehole with a large diameter such as 20'(50 cm),independent on depth and applicable likewise for sediments and crystalline rocks,such as granite. The current rate of penetration(ROP) of 3 m per hour is planned to be augmented up to 35m per hour,and again,irrespective whether in sedimentary or crystalline formations. Consequently,a 10km deep borehole with 50 cm will ultimately be drilled within 12 days. EPB will create new markets,such as:(i) EPB shallow drilling for geotechnics,energy piles,measures in order to mitigate natural hazards,etc.,(ii) EPB deep drilling(3–5km) for hydro-geothermics,exploration campaigns etc. and(iii) EPB super-deep drilling(5–10km) for petro-geothermics,enabling the economic generation of electricity. The autonomous and unlimited supply with cost efficient electricity,besides ample heat,ensures reliably clean and renewable energy,thus,high supply security. Such development will provide a substantial relief to cope with the global challenge to limit the climate change below 2 oC. 10Inactivation of SACE_3446, a TetR family transcriptional regulator, stimulates erythromycin production in Saccharopolyspora erythraea显示文摘Erythromycin A is a widely used antibiotic produced by Saccharopolyspora erythraea;however,its biosynthetic cluster lacks a regulatory gene,limiting the yield enhancement via regulation engineering of S.erythraea.Herein,six TetR family transcriptional regulators(TFRs)belonging to three genomic context types were individually inactivated in S.erythraea A226,and one of them,SACE_3446,was proved to play a negative role in regulating erythromycin biosynthesis.EMSA and qRT-PCR analysis revealed that SACE_3446 covering intact N-terminal DNA binding domain specifically bound to the promoter regions of erythromycin biosynthetic gene eryAI,the resistant gene ermE and the adjacent gene SACE_3447(encoding a longchain fatty-acid CoA ligase),and repressed their transcription.Furthermore,we explored the interaction relationships of SACE_3446 and previously identified TFRs(SACE_3986 and SACE_7301)associated with erythromycin production.Given demonstrated relatively independent regulation mode of SACE_3446 and SACE_3986 in erythromycin biosynthesis,we individually and concomitantly inactivated them in an industrial S.erythraea WB.Compared with WB,the WBΔ3446 and WBΔ3446Δ3986 mutants respectively displayed 36%and 65%yield enhancement of erythromycin A,following significantly elevated transcription of eryAI and ermE.When cultured in a 5 L fermentor,erythromycin A ofWBΔ3446 and WBΔ3446Δ3986 successively reached 4095 mg/L and 4670 mg/L with 23%and 41%production improvement relative to WB.The strategy reported here will be useful to improve antibiotics production in other industrial actinomycete.Hang Wu Yansheng Wang Li Yuan Yongrong Mao Weiwei Wang Lin Zhu Panpan Wu Chengzhang Fu Rolf Muller David T.Weaver Lixin Zhang Buchang Zhang 2016Synthetic and Systems Biotechnology2016,1,1:7

13Hierarchical highly ordered SnO_(2) nanobowl branched ZnO nanowires for ultrasensitive and selective hydrogen sulfide gas sensing显示文摘Highly sensitive and selective hydrogen sulfide(H_(2)S)sensors based on hierarchical highly ordered SnO_(2) nanobowl branched ZnO nanowires(NWs)were synthesized via a sequential process combining hard template processing,atomic-layer deposition,and hydrothermal processing.The hierarchical sensing materials were prepared in situ on microelectromechanical systems,which are expected to achieve high-performance gas sensors with superior sensitivity,long-term stability and repeatability,as well as low power consumption.Specifically,the hierarchical nanobowl SnO_(2)@ZnO NW sensor displayed a high sensitivity of 6.24,a fast response and recovery speed(i.e.,14 s and 39 s,respectively),and an excellent selectivity when detecting 1 ppm H_(2)S at 250°C,whose rate of resistance change(i.e.,5.24)is 2.6 times higher than that of the pristine SnO_(2) nanobowl sensor.The improved sensing performance could be attributed to the increased specific surface area,the formation of heterojunctions and homojunctions,as well as the additional reaction between ZnO and H_(2)S,which were confirmed by electrochemical characterization and band alignment analysis.Moreover,the well-structured hierarchical sensors maintained stable performance after a month,suggesting excellent stability and repeatability.In summary,such well-designed hierarchical highly ordered nanobowl SnO_(2)@ZnO NW gas sensors demonstrate favorable potential for enhanced sensitive and selective H_(2)S detection with long-term stability and repeatability.Li-Yuan Zhu Kai-Ping Yuan Jia-He Yang Cheng-Zhou Hang Hong-Ping Ma Xin-Ming Ji Anjana Devi Hong-Liang Lu David Wei Zhang 2020Microsystems & Nanoengineering2020,6,1:7

15Neuroprotectants attenuate hypobaric hypoxia-induced brain injuries in cynomolgus monkeys显示文摘Hypobaric hypoxia (HH) exposure can cause serious brain injury as well as life-threatening cerebral edema in severe cases. Previous studies on the mechanisms of HH-induced brain injury have been conducted primarily using non-primate animal models that are genetically distant to humans, thus hindering the development of disease treatment. Here, we report that cynomolgus monkeys (Macaca fascicularis) exposed to acute HH developed human-like HH syndrome involving severe brain injury and abnormal behavior. Transcriptome profiling of white blood cells and brain tissue from monkeys exposed to increasing altitude revealed the central role of the HIF-1 and other novel signaling pathways, such as the vitamin D receptor (VDR) signaling pathway, in co-regulating HH-induced inflammation processes. We also observed profound transcriptomic alterations in brains after exposure to acute HH, including the activation of angiogenesis and impairment of aerobic respiration and protein folding processes, which likely underlie the pathological effects of HH-induced brain injury. Administration of progesterone (PROG) and steroid neuroprotectant 5α-androst-3β,5,6β-triol (TRIOL) significantly attenuated brain injuries and rescued the transcriptomic changes induced by acute HH. Functional investigation of the affected genes suggested that these two neuroprotectants protect the brain by targeting different pathways, with PROG enhancing erythropoiesis and TRIOL suppressing glutamate-induced excitotoxicity. 16Analysis on the influence factors of Bitcoin’s price based on VEC model显示文摘Background:Bitcoin,the most innovate digital currency as of now,created since 2008,even through experienced its ups and downs,still keeps drawing attentions to all parts of society.It relies on peer-to-peer network,achieved decentralization,anonymous and transparent.As the most representative digital currency,people curious to study how Bitcoin’price changes in the past.Methods:In this paper,we use monthly data from 2011 to 2016 to build a VEC model to exam how economic factors such as Custom price index,US dollar index,Dow jones industry average,Federal Funds Rate and gold price influence Bitcoin price.Result:From empirical analysis we find that all these variables do have a long-term influence.US dollar index is the biggest influence on Bitcoin price while gold price influence the least.Conclusion:From our result,we conclude that for now Bitcoin can be treated as a speculative asset,however,it is far from being a proper credit currency.Yechen Zhu David Dickinson Jianjun Li 2017Financial Innovation2017,3,1:5
17Artemisinin Biosynthesis in Non-glandular Trichome Cells of Artemisia annua显示文摘Artemisinin-based combination therapy(ACT)forms the first line of malaria treatment.However,the yield fluctuation of artemisinin has remained an unsolved problem in meeting the global demand for ACT.This problem is mainly caused by the glandular trichome(GT)-specific biosynthesis of artemisinin in all currently used Artemisia annua cultivars.Here,we report that non-GT cells of self-pollinated inbred A.annua plants can express the artemisinin biosynthetic pathway.Gene expression analysis demonstrated the transcription of six known pathway genes in GT-free leaves and calli of inbred A.annua plants.LC-qTOF-MS/MS analysis showed that these two types of GT-free materials produce artemisinin,artemisinic acid,and arteannuin B.Detailed IR-MALDESI image profiling revealed that these three metabolites and dihydroartemisinin are localized in non-GT cells of leaves of inbred A.annua plants.Moreover,we employed all the above approaches to examine artemisinin biosynthesis in the reported XL annua glandless(gl)mutant.The resulting data demonstrated that leaves of regenerated gl plantlets biosynthesize artemisinin.Codectively,these findings not only add new knowledge leading to a revision of the current dogma of artemisinin biosynthesis inannua but also may expedite innovation of novel metabolic engineering approaches for high and stable production of artemisinin in the future.Rika Judd M.Caleb Bagley Mingzhuo Li Yue Zhu Caiyan Lei Seyit Yuzuak Mans Ekelof Gaobin Pu Xiting Zhao David C.Muddiman De-Yu Xie 2019Molecular Plant2019,12,5:5
18CSCO_3 CYCLES IN SALAWUSU RIVER BASINSINCE 150KA B.P.显示文摘ABSTRACT: This paper, with Milanggouwan stratigraphic section as a typical section of the Salawusu River Basin, explores the relation between CaCO3 content distribution and climate change since 150 ka B. P. and concludes that: 1) The low-high changes of CaCO3 content in the section has a remarkable corresponding relation with the sedimentary cycles of ancient aeolian sand and overlying fluviolacustrine facies or palaeosols. 2) CaCO3 distribution in aeolian sand is relatively meagre, ranging from 0. 8% - 7. 18%, or on an average 2. 50% but relatively enriches in the fluviolacustrine faceis and palaeosols, ranging from 2. 20% - 14. 9% , or on an average 5. 74%. This implies that they have different climatic backgrounds. The former was the product of erosion, transport and deposition by wind under arid and cold climatic conditions, whereas the latter was related to its special low-lying geomorphic position between the Ordos Plateau and Loess Plateau and warm-humid climatic environment. When the climatic became warm and humid, fluviolacustrine and swamp facies developed, soil-forming action strengthened, and low-lying catchment condition was favorable to CaCO3 accumulation. 3) The basic cause responsible for the multicycle of CaCO3 migration and accumulation in the Milanggouwan section may be the multiple alterations of winter and summer monsoons over the Mu Us Desert under the influences of climatic fluctuation of glacial and interglacial periods in the Northern Hemisphere since 150ka B. 19Road traffic injury mortality and morbidity by country development status, 2011- 2017显示文摘Purpose:This research examined road traffic injury mortality and morbidity disparities across of country development status,and discussed the possibility of reducing country disparities by various actions to accelerate the pace of achieving Sustainable Development Goals target 3.6 e to halve the number of global deaths and injuries from road traffic accidents by 2020.Methods:Data for road traffic mortality,morbidity,and socio-demographic index(SDI)were extracted by country from the estimates of the Global Burden of Disease study,and the implementation of the three types of national actions(legislation,prioritized vehicle safety standards,and trauma-related post-crash care service)were extracted from the Global Status Report on Road Safety byWorld Health Organization.We fitted joinpoint regression analysis to identify and quantify the significant rate changes from 2011 to 2017.Results:Age-adjusted road traffic mortality decreased substantially for all the five SDI categories from 2011 to 2017(by 7.52%e16.08%).Age-adjusted road traffic mortality decreased significantly as SDI increased in the study time period,while age-adjusted morbidity generally increased as SDI increased.Subgroup analysis by road user yielded similar results,but with two major differences during the study period of 2011 to 2017:(1)pedestrians in the high SDI countries experienced the lowest mortality(1.68e1.90 per 100,000 population)and morbidity(110.45e112.72 per 100,000 population for incidence and 487.48e491.24 per 100,000 population for prevalence),and(2)motor vehicle occupants in the high SDI countries had the lowest mortality(4.07e4.50 per 100,000 population)but the highest morbidity(428.74e467.78 per 100,000 population for incidence and 1025.70e1116.60 per 100,000 population for prevalence).Implementation of the three types of national actions remained nearly unchanged in all five SDI categories from 2011 to 2017 and was consistently stronger in the higher SDI countries than in the lower SDI countries.Lower income nations comprise the heaviest burden of global road traffic injuries and deaths.Conclusion:Global road traffic deaths would decrease substantially if the large mortality disparities across country development status were reduced through full implementation of proven national actions including legislation and law enforcement,prioritized vehicle safety standards and trauma-related postcrash care services.Jie-Yi He Wang-Xin Xiao David CSchwebel Mo-Tao Zhu Pei-Shan Ning Li Li Xun-Jie Cheng Jun-Jie Hua Guo-Qing Hu 2021Chinese Journal of Traumatology2021,24,2:4
