| 2 | An efficient papaya leaf distortion mosaic potyvirus vector for virus-induced gene silencing in papaya显示文摘Papaya(Carica papaya L.)is regarded as an excellent model for genomic studies of tropical trees because of its short generation time and its small genome that has been sequenced.However,functional genomic studies in papaya depend on laborious genetic transformations because no rapid tools exist for this species.Here,we developed a highly efficient virus-induced gene silencing(VIGS)vector for use in papaya by modifying an artificially attenuated infectious clone of papaya leaf distortion mosaic virus(PLDMV;genus:Potyvirus),PLDMV-E,into a stable Nimble Cloning(NC)-based PLDMV vector,pPLDMV-NC,in Escherichia coli.The target fragments for gene silencing can easily be cloned into pPLDMV-NC without multiple digestion and ligation steps.Using this PLDMV VIGS system,we silenced and characterized five endogenous genes in papaya,including two common VIGS marker genes,namely,phytoene desaturase,Mg-chelatase H subunit,putative GIBBERELLIN(GA)-INSENSITIVE DWARF1A and 1B encoding GA receptors;and the cytochrome P450 gene CYP83B1,which encodes a key enzyme involved in benzylglucosinolate biosynthesis.The results demonstrate that our newly developed PLDMV VIGS vector is a rapid and convenient tool for functional genomic studies in papaya. | Decai Tuo Pu Yan Guangyuan Zhao Hongguang Cui Guopeng Zhu Yang Liu Xiukun Yang He Wang Xiaoying Li Wentao Shen Peng Zhou | 2021 | Horticulture Research2021,8,1: | 2 |

| 11 | Engineering surface oxygen vacancy of mesoporous CeO_(2) nanosheets assembled microspheres for boosting solar-driven photocatalytic performance显示文摘Surface oxygen vacancy defects of mesoporous CeO_(2)nanosheets assembled microspheres(D-CeO_(2))are engineered by polymer precipitation,hydrothermal and surface hydrogenation strategies.The resultant D-CeO_(2)with a main pore diameter of 9.3 nm has a large specific surface area(~102.3 m^(2)/g)and high thermal stability.The mesoporous nanosheets assembled microsphere structure prevents the nanosheets from aggregation,which is beneficial to effective mass transfer and shortens the migration distance of charge carriers.After surface hydrogenation,the photoresponse extends to long wavelength region,combing with the band gap from 2.63 eV reduced to 2.39 eV.Under AM 1.5 G radiation,the photocatalytic degradation rate of tetracycline(TC)can be up to 99.99%,which is three times as high as that of pristine CeO_(2)microspheres.The excellent solar-driven photocatalytic performance can be attributed to the efficient surface oxygen vacancy engineering and the mesoporous nanosheets assembled microsphere structure,which narrows the band gap,shortens the migration distance of carriers,promotes the spatial separation of photogenerated electron-hole pairs and favors mass transfer.The strategy provides new insights for fabricating other high-efficient oxide photocatalysts. | Decai Yang Yachao Xu Kai Pan Chuanxin Yu Jiaxing Wu Mingxia Li Fan Yang Yang Qu Wei Zhou | 2022 | Chinese Chemical Letters2022,33,1: | 0 |

| 14 | Identification of novel loci influencing refractive error in East Asian populations using an extreme phenotype design显示文摘The global 'myopia boom' has raised significant international concerns. Despite a higher myopia prevalence in Asia, previous large-scale genome-wide association studies(GWASs) were mostly based on European descendants. Here, we report a GWAS of spherical equivalent(SE) in 1852 Chinese Han individuals with extreme SE from Guangzhou(631 <-6.00 D and 574 > 0.00 D) and Wenzhou(593 <-6.00 D and54 >-1.75 D), followed by a replication study in two independent cohorts with totaling 3538 East Asian individuals. The discovery GWAS and meta-analysis identify three novel loci, which show genome-wide significant associations with SE, including 1 q25.2 FAM163 A, 10 p11.22 NRP1/PRAD3, and 10 p11.21 ANKRD30 A/MTRNR2 L7, together explaining 3.34% of SE variance. 10 p11.21 is successfully replicated.The allele frequencies of all three loci show significant differences between major continental groups(P < 0.001). The SE reducing(more myopic) allele of rs10913877(1 q25.2 FAM163 A) demonstrates the highest frequency in East Asians and much lower frequencies in Europeans and Africans(EAS = 0.60,EUR = 0.20, and AFR = 0.18). The gene-based analysis additionally identifies three novel genes associated with SE, including EI24, LHX5, and ARPP19. | 15 | The joint driving effects of climate and weather changes caused the Chamoli glacier-rock avalanche in the high altitudes of the India Himalaya显示文摘Ice avalanches are one of the most devastating mountain hazards,and can pose a great risk to the security of the surrounding area.Although ice avalanches have been widely observed in mountainous regions around the world,only a few ice avalanche events have been studied comprehensively,due to the lack of available data.In this study,in response to the recent catastrophic rock-ice avalanche(7 February 2021)at Chamoli in the India Himalaya,we used high-resolution satellite images and found that this event was actually a glacier-rock landslide,where the collapse of the rock-ice body was caused by the sliding of the bedrock beneath the glacier,for which the source area and volume loss were about 2.89×10^(5) m^(2) and 2.46×10^(7) m^(3),respectively,corresponding to an average elevation change of about−85 m.Furthermore,visual analysis of the dense time-series satellite images shows that the overall downward sliding of the collapsed rock-ice body initiated around the summer of 2017,and thereafter exhibited clear seasonality(mainly in summer).Meteorological analysis reveals a strong rainfall anomaly in the initiation period of the sliding and a remarkable winter warming anomaly in the 40 days before the collapse.Comparisons of multi-temporal digital elevation models(DEMs)further suggest that the glacier geometry in the collapsed areas was likely changing(i.e.,accelerated surface thinning in the lower part of the glaciers and insignificant change in the upper part),which is consistent with the region-wide climate warming.Finally,by combining the above findings and a geomorphic analysis,we conclude that the rock-ice avalanche event was mainly caused by the joint effects of climate and weather changes acting on a steeply sloping and fracture-prone geological condition.The findings of this study provide new and valuable evidence for the study of slope/glacier instability at high altitudes.This study also highlights that,for the Himalaya and other high mountain ranges,there is an urgent need to identify the glaciers that have a high risk of ice avalanches. | Yushan ZHOU Xin LI Donghai ZHENG Zhiwei LI Baosheng AN Yingzheng WANG Decai JIANG Jianbin SU Bin CAO | 2021 | Science China Earth Sciences2021,64,11: | 0 |
