1SARS-CoV-2 promotes RIPK1 activation to facilitate viral propagation显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is the ongoing global pandemic that poses substantial challenges to public health worldwide.A subset of COVID-19 patients experience systemic inflammatory response,known as cytokine storm,which may lead to death.Receptor-interacting serine/threonine-protein kinase 1(RIPK1)is an important mediator of inflammation and cell death.Here,we examined the interaction of RIPK1-mediated innate immunity with SARS-CoV-2 infection.We found evidence of RIPK1 activation in human COVID-19 lung pathological samples,and cultured human lung organoids and ACE2 transgenic mice infected by SARS-CoV-2.Inhibition of RIPK1 using multiple smallmolecule inhibitors reduced the viral load of SARS-CoV-2 in human lung organoids.Furthermore,therapeutic dosing of the RIPK1 inhibitor Nec-1s reduced mortality and lung viral load,and blocked the CNS manifestation of SARS-CoV-2 in ACE2 transgenic mice.Mechanistically,we found that the RNA-dependent RNA polymerase of SARS-CoV-2,NSP12,a highly conserved central component of coronaviral replication and transcription machinery,promoted the activation of RIPK1.Furthermore,NSP12323L variant,encoded by the SARS-CoV-2 C14408T variant first detected in Lombardy,Italy,that carries a Pro323Leu amino acid substitution in NSP12,showed increased ability to activate RIPK1.Inhibition of RIPK1 downregulated the transcriptional induction of proinflammatory cytokines and host factors including ACE2 and EGFR that promote viral entry into cells.Our results suggest that SARS-CoV-2 may have an unexpected and unusual ability to hijack the RIPK1-mediated host defense response to promote its own propagation and that inhibition of RIPK1 may provide a therapeutic option for the treatment of COVID-19.Gang Xu Ying Li Shengyuan Zhang Haoran Peng Yunyun Wang Dekang Li Taijie Jin Zhuohao He Yilun Tong Chunting Qi Guowei Wu Kangyun Dong Jizhou Gou Yang Liu Tongyang Xiao Jing Qu Liang Li Liang Liu Ping Zhao Zheng Zhang Junying Yuan 2021Cell Research2021,31,12:6

4Spatial Patterns of LULC and Driving Forces in the Transnational Area of Tumen River:A Comparative Analysis of the Sub-regions of China,the DPRK,and Russia显示文摘Understanding the spatial patterns of land-use and land-cover(LULC)and their driving forces in transnational areas is important for the sustainable development of these regions.However,the spatial patterns of LULC and their driving forces across multiple scales are poorly understood in transnational areas.In this study,we analyzed the spatial patterns of LULC and driving forces in the transnational area of Tumen River(TATR)in 2016 across two scales:the entire region and the sub-regions of China,the Democratic People’s Republic of Korea(DPRK),and Russia.Results showed that the LULC was dominated by broadleaf forest and dry farmland in the TATR in 2016,which accounted for 66.86%and 13.60%of the entire region,respectively.Meanwhile,the LULC in the three sub-regions exhibited noticeable differences.In the Chinese and the DPRK’s sub-regions,the area of broadleaf forest was greater than those for the other LULC types,while the Russian sub-region was dominated by broadleaf forest and grassland.The spatial patterns of LULC were mainly influenced by topography,climate,soil properties,and human activities.In addition,the driving forces of the spatial patterns of LULC in the TATR had an obvious scaling effect.Therefore,we suggest that effective policies and regulations with cooperation among China,the DPRK,and Russia are needed to plan the spatial patterns of LULC and improve the sustainable development of the TATR.NAN Ying WANG Bingbing ZHANG Da LIU Zhifeng QI Dekang ZHOU Haohao 2020Chinese Geographical Science2020,30,4:2
5Spatial patterns of net primary productivity and its driving forces:a multi-scale analysis in the transnational area of the Tumen River显示文摘Analyzing the spatial patterns of net primary productivity(NPP)and its driving forces in transnational areas provides a solid basis for understanding regional ecological processes and ecosystem services.However,the spatial patterns of NPP and its driving forces have been poorly understood on multiple scales in transnational areas.In this study,the spatial patterns of NPP in the transnational area of the Tumen River(TATR)in 2016 were simulated using the Carnegie Ames Stanford Approach(CASA)model,and its driving forces were analyzed using a stepwise multiple linear regression model.We found that the total amount of NPP in the TATR in 2016 was approximately 14.53 TgC.The amount of NPP on the Chinese side(6.23 TgC)was larger than those on the other two sides,accounting for 42.88%of the total volume of the entire region.Among different land-use and land-cover(LULC)types,the amount of NPP of the broadleaf forest was the largest(11.22 TgC),while the amount of NPP of the bare land was the smallest.The NPP per unit area was about 603.21 gC/(m2 yr)across the entire region,while the NPP per unit area on the Chinese side was the largest,followed by the Russian side and the DPRK's side.The spatial patterns of NPP were influenced by climate,topography,soil texture,and human activities.In addition,the driving forces of the spatial patterns of NPP in the TATR had an obvious scaling effect,which was mainly caused by the spatial heterogeneity of climate,topography,soil texture,and human activities.We suggest that effective land management policies with cooperation among China,the DPRK,and Russia are needed to maintain NPP and improve environmental sustainability in the TATR.Jianwen WANG Da ZHANG Ying NAN Zhifeng LIU Dekang QI 2020Frontiers of Earth Science2020,14,1:1

9Transmembrane transport of polycyclic aromatic hydrocarbons by bacteria and functional regulation of membrane proteins显示文摘In recent years,increasing research has been conducted on transmembrane transport processes and the mechanisms behind the microbial breakdown of polycyclic aromatic hydrocarbons(PAHs),including the role of membrane proteins in transmembrane transport and the mode of transmission.This article explains the adsorption,uptake and transmembrane transport of PAHs by bacteria,the regulation of membrane protein function during the transmembrane transport.There are three different regulation mechanisms for uptake,depending on the state and size of the oil droplets relative to the size of the microbial cells,which are(i)direct adhesion,(ii)emulsification and pseudosolubilization,and(iii)interfacial uptake.Furthermore,two main transmembrane transport modes are introduced,which are(i)active transport and(ii)passive uptake and active efflux mechanism.Meanwhile,introduce the proteomics and single cell analysis technology used to address these areas of research,such as Isobaric tags for relative and absolute quantitation(iTRAQ)technology and Nano Secondary ion mass spectrometry(Nano-SIMS).Additionally,analyze the changes in morphology and structure and the characteristics of microbial cell membranes in the process of transmembrane transport.Finally,recognize the microscopic mechanism of PAHs biodegradation in terms of cell and membrane proteins are of great theoretical and practical significance for understanding the factors that influence the efficient degradation of PAHs contaminants in soil and for remediating the PAHs contamination in this area with biotechnology.Hongqi Wang Ruhan Jiang Dekang Kong Zili Liu Xiaoxiong Wu Jie Xu Yi Li 2020Frontiers of Environmental Science & Engineering2020,14,1:0
10A Novel Synchronization Method in Terahertz Large-Scale Antenna Array System显示文摘We focus on the problems of the accurate time delay estimation,the design of training pilots,and hybrid matrix optimization within the large-scale antenna array Terahertz(THz)broadband communication system.In contrast to the existing researches based on narrow-band arrays,we hereby shed light on the time delay estimation of broadband arrays.In THz broadband communication systems,the data symbol duration is relatively short when comparing with the dimension of the antenna array.In large-scale antenna systems,signals received in each antenna are no longer different phase-shifted copies of the same symbol,but completely different symbols in which occasion traditional narrowband structure is no longer suitable.Based on the above conclusion,firstly,we put forward a system model based on large-scale antenna arrays and Time delay line(TDL)structure.Secondly,we deduce the Cramer-Rao lower bound(CRLB)of the time delay estimation,and present a time delay estimation algorithm that could reach the CRLB.Thirdly,by minimizing the CRLB,we address the design of the training pilot and optimized TDL structure under the condition of constant envelope training pilot and modulus TDL structure.Finally,we disclose the numerical simulation results.According to the simulation results,the aforementioned method is workable in reaching the CRLB,the TDL structure can significantly surpass that of the traditional model,and the optimal pilot design method outperforms the pseudo-random pilot structure.ZHENG Chen DING Xuhui LIU Dekang BU Xiangyuan AN Sining 2021Chinese Journal of Electronics2021,30,5:0

13Transcriptomic but not genomic variability confers phenotype of breast cancer stem cells显示文摘Background:Breast cancer stem cells(BCSCs)are considered responsible for cancer relapse and drug resistance.Understanding the identity of BCSCs may open new avenues in breast cancer therapy.Although several discoveries have been made on BCSC characterization,the factors critical to the origination of BCSCs are largely unclear.This study aimed to determine whether genomic mutations contribute to the acquisition of cancer stem-like phenotype and to investigate the genetic and transcriptional features of BCSCs.Methods:We detected potential BCSC phenotype-associated mutation hotspot regions by using whole-genome sequencing on parental cancer cells and derived serial-generation spheres in increasing order of BCSC frequency,and then performed target deep DNA sequencing at bulk-cell and single-cell levels.To identify the transcriptional program associated with BCSCs,bulk-cell and single-cell RNA sequencing was performed.Results:By using whole-genome sequencing of bulk cells,potential BCSC phenotype-associated mutation hotspot regions were detected.Validation by target deep DNA sequencing,at both bulk-cell and single-cell levels,revealed no genetic changes specifically associated with BCSC phenotype.Moreover,single-cell RNA sequencing showed profound transcriptomic variability in cancer cells at the single-cell level that predicted BCSC features.Notably,this transcriptomic variability was enriched during the transcription of 74 genes,revealed as BCSC markers.Breast cancer patients with a high risk of relapse exhibited higher expression levels of these BCSC markers than those with a low risk of relapse,thereby highlighting the clinical significance of predicting breast cancer prognosis with these BCSC markers.Conclusions:Transcriptomic variability,not genetic mutations,distinguishes BCSCs from non-BCSCs.The identified 74 BCSC markers have the potential of becoming novel targets for breast cancer therapy.Mengying Tong Ziqian Deng Mengying Yang Chang Xu Xiaolong Zhang Qingzheng Zhang Yuwei Liao Xiaodi Deng Dekang Lv Xuehong Zhang Yu Zhang Peiying Li Luyao Song Bicheng Wang Aisha Al-Dherasi Zhiguang Li Quentin Liu 2018Cancer Communications2018,38,1:0

15A Novel Angle of Arrival Tracking Method in Large-Scale-Array Systems显示文摘In this paper,we investigate the problem of angle of arrival(AOA)tracking for the largescale array in terahertz(THz)communication,which has a large size and a narrow beam,highly demanding an accurate angle estimation.On the one hand,the system usually adopts a hybrid structure with limited radio-frequency(RF)chains,which increases the difficulty of angle estimation;on the other hand,the rapid mobility of users also brings new challenges to angle estimation.To address the above challenges,a two-stage tracking framework is proposed in this paper,which employs the random phase matrix and orthogonal long pilots in the first stage to reduce the complicated multi-user estimation to multiple single-user estimations,followed by using both wide and narrow beams in the second stage to serve high-speed and low-speed users.Furthermore,a generalized-approximated-message-passing(GAMP)method is proposed for facilitating a low-accuracy estimation of the angles,followed by adopting a modified expectation-maximization(EM)algorithm based phase estimation to unbiased estimate the instantaneous angle with the help of high-gain characteristics of the beams.The proposed structure can not only simplify the estimation complexity,but also improve the estimation accuracy due to its capability of transferring the non-linear problem of angle observation into a linear gaussian model.In addition,the Kalman tracking framework is employed for performing a continuous angle tracking.Numerical results show that the angle estimation based on the random phase matrix in the initial stage can obtain a high enough estimation accuracy,while the GAMP algorithm implemented in the second stage can quickly capture the angle range under the Rayleigh limit.The performance of the proposed EM-based tracking method is shown to outperform the traditional extended Kalman filter(EKF)method.Chen Zheng Dekang Liu Xuhui Ding Xiangyuan Bu Zhongshan Zhang 2022China Communications2022,19,9:0
