1Ulva prolifera green-tide outbreaks and their environmental impact in the Yellow Sea, China显示文摘The Ulva prolifera green tides in the Yellow Sea, China, which have been occurring since 2007, are a serious environmental problem attracting worldwide attention. Despite extensive research, the outbreak mechanisms have not been fully understood. Comprehensive analysis of anthropogenic and natural biotic and abiotic factors reveals that human activities, regional physicochemical conditions and algal physiological characteristics as well as ocean warming and biological interactions(with microorganism or other macroalgae) are closely related to the occurrence of green tides. Dynamics of these factors and their interactions could explain why green tides suddenly occurred in 2007 and decreased abruptly in 2017.Moreover, the consequence of green tides is serious. The decay of macroalgal biomass could result in hypoxia and acidification, possibly induce red tide and even have a long-lasting impact on coastal carbon cycles and the ecosystem. 3Investigation and experimental validation of curcumin-related mechanisms against hepatocellular carcinoma based on network pharmacology显示文摘Objective:To determine the potential molecular mechanisms underlying the therapeutic effect of curcumin on hepatocellular carcinoma(HCC)by network pharmacology and experimental in vitro validation.Methods:The predictive targets of curcumin or HCC were collected from several databases.the identified overlapping targets were crossed with Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses using the Database for Annotation,Visualization,and Integrated Discovery(DAVID)platform.Two of the candidate pathways were selected to conduct an experimental verification.The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide tetrazolium(MTT)assay was used to determine the effect of curcumin on the viability of Hep G2 and LO2 cells.The apoptosis and autophagy of Hep G2 cells were respectively detected by flow cytometry and transmission electron microscopy.Besides,western blot and real-time polymerase chain reaction(PCR)were employed to verify the p53 apoptotic pathway and adenosine 5’-monophosphate(AMP)-activated protein kinase(AMPK)autophagy pathway.Hep G2 cells were pretreated with pifithrin-α(PFT-α)and GSK690693 for further investigation.Results:The 167 pathways analyzed by KEGG included apoptosis,autophagy,p53,and AMPK pathways.The GO enrichment analysis demonstrated that curcumin was involved in cellular response to drug,regulation of apoptotic pathway,and so on.The in vitro experiments also confirmed that curcumin can inhibit the growth of Hep G2 cells by promoting the apoptosis of p53 pathway and autophagy through the AMPK pathway.Furthermore,the protein and messenger RNA(m RNA)of the two pathways were downregulated in the inhibitor-pretreated group compared with the experimental group.The damage-regulated autophagy modulator(DRAM)in the PFT-α-pretreated group was downregulated,and p62 in the GSK690693-pretreated group was upregulated.Conclusions:Curcumin can treat HCC through the p53 apoptotic pathway and the AMPK/Unc-51-like kinase 1(ULK1)autophagy pathway,in which the mutual transformation of autophagy and apoptosis may occur through DRAM and p62.Yang CHEN Qian LI Sisi REN Ting CHEN Bingtao ZHAI Jiangxue CHENG Xiaoyan SHI Liang SONG Yu FAN Dongyan GUO 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,8:3
4Enriched Opportunistic Pathogens Revealed by Metagenomic Sequencing Hint Potential Linkages between Pharyngeal Microbiota and COVID-19显示文摘As a respiratory tract virus,SARS-CoV-2 infected people through contacting with the upper respiratory tract first.Previous studies indicated that microbiota could modulate immune response against pathogen infection.In the present study,we performed metagenomic sequencing of pharyngeal swabs from eleven patients with COVID-19 and eleven Non-COVID-19 patients who had similar symptoms such as fever and cough.Through metagenomic analysis of the above two groups and a healthy group from the public data,there are 6502 species identified in the samples.Specifically,the Pielou index indicated a lower evenness of the microbiota in the COVID-19 group than that in the Non-COVID-19 group.Combined with the linear discriminant analysis(LDA)and the generalized linear model,eighty-one bacterial species were found with increased abundance in the COVID-19 group,where 51 species were enriched more than 8 folds.The top three enriched genera were Streptococcus,Prevotella and Campylobacter containing some opportunistic pathogens.More interestingly,through experiments,we found that two Streptococcus strains,S.suis and S.agalactiae,could stimulate the expression of ACE2 of Vero cells in vitro,which may promote SARS-CoV-2 infection.Therefore,these enriched pathogens in the pharynxes of COVID-19 patients may involve in the virus-host interactions to affect SARS-CoV-2 infection and cause potential secondary bacterial infections through changing the expression of the viral receptor ACE2 and/or modulate the host’s immune system.Dongyan Xiong Caroline Muema Xiaoxu Zhang Xinming Pan Jin Xiong Hang Yang Junping Yu Hongping Wei 2021Virologica Sinica2021,36,5:3

7Impacts of El Nino on the somatic condition of Humboldt squid based on the beak morphology显示文摘The Humboldt squid Dosidicus gigas has a short life span, and environmental variability plays a significant role in regulating its population dynamics and distribution. An analysis of 1 096 samples of D. gigas collected by the Chinese commercial fishing vessels during 2013, 2014, and 2016 off the Peruvian Exclusive Economic Zone, was conducted to evaluate the impacts of El Nino events on the somatic condition of D. gigas. This study indicates that the slopes of all beak variables in relation to mantle length (ML) for females were greater than those of males during 2013, 2014, and 2016, and slopes of the upper crest length and the lower rostrum length significantly differed between females and males in 2013 (P<0.05). Variation in the slopes for beak variables among years was studied;no significant difference was observed (ANCOVA, P>0.05). The Fulton's condition coefficients (K) of females and males in 2013 and 2014 were significantly greater than those in 2016 (P<0.01). The K values of females were greater than those of males in 2013, 2014, and 2016, and K values significantly differed between females and males in 2013. In normal years, the chlorophyll a (Chl a) concentration showed an N-shaped variability from January to December. However, in the El Nino period, it tended to weaken the upwelling coupled with warm and low Chl a concentration waters. 13Advanced glycation end-products change placental barrier function and tight junction in rats with gestational diabetes mellitus via the receptor for advanced glycation end products/nuclear factor-κB pathway显示文摘The placenta plays an important role in nutrient transport to maintain the growth and development of the embryo.Gestational diabetes mellitus(GDM),the most common complication during pregnancy,highly affects placental function in late gestation.Advanced glycation end-products(AGEs),a complex and heterogeneous group of compounds engaged by the receptor for AGEs(RAGE),are closely associated with diabetes-related complications.In this study,AGEs induced a decrease in the expression of tight junction(TJ)proteins in BeWo cells and increased the paracellular permeability of trophoblast cells by regulating RAGE/NF-κB.Sprague-Dawley(SD)rats injected with 100 mg/kg AGEs-rat serum albumin(RSA)via the tail vein from embryo day 2 were set as the placental barrier dysfunction model group(n=10).The effect of AGEs on placental permeability was determined using the Evans-Blue dye extravasation method.The ultrastructure of the placenta samples was observed by transmission electron microscopy.The effects of AGEs on the placenta were confirmed by treating rats with RAGE antagonist FPS-ZM1 and soluble forms of RAGE(sRAGE).AGEs treatment increased placental permeability and disrupted the tight junctions in pregnant rat placenta,but has no effect on blood glucose.The expression of TJ-related proteins,including ZO-1,Occludin,and Claudin 5,were downregulated after AGEs treatment.Further,AGEs treatment increased the expression of RAGE and nuclear factor-κB in the placenta of rats and upregulated the levels of vascular endothelial growth factor.The effects of AGEs on the placenta were blocked by RAGE antagonist FPS-ZM1 and sRAGE.This study demonstrates the mechanism underlying AGEs-induced disturbance in placental function in pregnant rats and highlights the potential of AGEs in the treatment of GDM.YUEHUA SHI QIUYING YAN QIN LI WEI QIAN DONGYAN QIAO DONGDONG SUN HONG YU 2023BIOCELL2023,47,1:0

15Effects of Standardized Rehabilitation on Quality of Life of Stroke Patients at Convalescence and Sequelae Stages显示文摘Objective:To explore the effects of standardized rehabilitation on quality of life (QOL) of stroke patients at convalescence and sequelae stages. Method:A total of 251 stroke patients were randomly divided into a standardized rehabilitation group and a control group. The simplified Fugl-Meyer assessment(FMA) scale, the 36-item short-form health survey questionnaire (SF-36) and functional comprehensive assessment (FCA) were administered before as well as after 3, 6 and at 12 months a follow-up study respectively. Statistical analysis was conducted based on the evaluations at 4 testing time points. Result:There was no significant difference in FMA,SF-36 and FCA scores of the two groups before and after 3 months treatments, while FMA,SF-36 and FCA scores of the rehabilitation group were obviously higher than those of the control group either after 6 months treatments or of followup study. Moreover, after 6 months treatments FMA score was apparently higher than the score at the beginning and after 3 months treatments. The FMA, SF36 and FAC scores during the followup visit decreased when compared with scores after 6 months treatments, but increased significantly when compared with the scores at the beginning and after 3 months treatments. 16Theory of the Solution of Inventive Problems (TRIZ) and Computer Aided Design (CAD) for Micro-electro-mechanical Systems (MEMS)显示文摘Satellite remote sensing technology[1] is widely used in all walks of life ,which plays an increasingly remarkable results in natural disasters(sudden and major).With more and more launch and application of high resolution satellite, the texture information in remote sensing imagery becomes much more abundant. In the age of big data, for the infrared remote sensor has short life, which annoys many people. In the packaging process, due to a difference in thermal expansion coefficients [2] between the flip-chip bonded MEMS device and the substrate, cooling after bonding can cause the MEMS to buckle. Combine TAIZ theory with the flexure design in CAD to solve the problem. It can be obtained that increasing fold length can reduce warpage. 17Knowledge Acquisition from Forestry Machinery Patent Based on the Algorithm for Closed Weighted Pattern Mining显示文摘The application of big data mining can create over a trillion dollars value. Patents contain a great deal of new technologies and new methods which have unique value in the product innovation. In order to improve the effectiveness of big data mining and aid the innovation of products of forestry machinery, the algorithm for closed weighted pattern mining is applied to acquire the function knowledge in the patents of forestry machinery. Compared with the other algorithms for mining patterns, the algorithm is more suitable for the characteristics of patent data. It not only takes into account the importance of different items to reduce the search space effectively, but also avoids achieving excessive uninteresting patterns below the premise that assures quality. 19Noninvasive Low-Frequency Pulsed Focused Ultrasound Therapy for Rheumatoid Arthritis in Mice显示文摘Rheumatoid arthritis(RA)is a common autoimmune disease characterized by chronic and progressive inflammation of the synovium.Focused ultrasound therapy is an increasingly attractive alternative for treating RA owing to its noninvasiveness;however,it remains unclear which immune subsets respond to ultrasound stimulation.In this study,we showed that spleen-targeted low-frequency pulsed focused ultrasound(LFPFU)effectively improved the severity of arthritis in an arthritis mouse model established in DBA/1J mice.Additionally,we performed in-depth immune profiling of spleen samples from RA mice,RA mice that underwent ultrasound therapy,and healthy controls using mass cytometry along with extensive antibody panels and identified the immune composition of 14 cell populations,including CD4^(+)/CD8^(+)T cells,B cells,natural killer cells,and dendritic cells.Moreover,multidimensional analysis according to cell-surface markers and phenotypes helped in identifying 4 and 5 cell subpopulations among T and myeloid cells,respectively,with 6 T cell subsets and 3 myeloid cell subsets responsive to ultrasound therapy among the 3 groups.Of these cell subsets,CD8^(+)T cell subsets showed a unique response to ultrasound stimulation in RA mice.Specifically,CD8^(+)T cells show a noticeable correlation with the degree of arthritis progression and could serve as an indicator for spleen-focused ultrasound-based therapy.Furthermore,single-cell RNA sequencing of spleen cells revealed the importance of T,B,and myeloid cell populations in the anti-inflammatory pathway.These results elucidated the unique cell subsets and transcriptome of splenic cells responsive to LFPFU and demonstrated the potential of spleen-focused ultrasound stimulation in the treatment of inflammatory diseases.Xuqiao Hu Fei Li Jieying Zeng Zhenru Zhou Zhaoyang Wang Jing Chen Dongyan Cao Yifan Hong Laixin Huang Yongsheng Chen Jinfeng Xu Fajn Dong Rongmin Yu Harong Zhen 2023Research2023,,3:0
20Research on the influence of contact surface constraint on mechanical properties of rock-concrete composite specimens under compressive loads显示文摘The contact form of rock-concrete has a crucial influence on the failure characteristics of the stability of rock-concrete engineering.To study the influence of contact surface on the mechanical properties of rock-concrete composite specimens under compressive loads,the two different contact forms of rock-concrete composite specimens are designed,the mechanical properties of these two different specimens are analyzed under triaxial compressive condition,and analysis comparison on the stress-strain curves and failure forms of the two specimens is carried out.The influence of contact surface constraint on the mechanical properties of rock-concrete composite specimens is obtained.Results show that the stress and strain of rock-concrete composite specimens with contact surface constraint are obviously higher than those without.Averagely,compared with composite specimens without the contact surface,the existence of contact surface constraint can increase the axial peak stress of composite specimens by 24%and the axial peak strain by 16%.According to the characteristics of the fracture surface,the theory of microcrack development is used to explain the contact surface constraint of rock-concrete composite specimens,which explains the difference of mechanical properties between the two rock-concrete composite specimens in the experiment.Research results cannot only enrich the research content of the mechanics of rock contact,but also can serve as a valuable reference for the understanding of the corresponding mechanics mechanism of other similar composite specimens.Baoyun ZHAO Yang LIU Dongyan LIU Wei HUANG Xiaoping WANG Guibao YU Shu LIU 2020Frontiers of Structural and Civil Engineering2020,14,2:0
