| 2 | Pollution of organic compounds and heavy metals in a coal gangue dump of the Gequan Coal Mine, China显示文摘Samples around a coal gangue dump of the Gequan Coal Mine were collected in April 2009. GC (gas chromatography) and GC/MS (gas chromatography/mass spectrometry) were employed to analyze the composition of organic matter in the samples. ICP-MS (inductively coupled plasma mass spectrometry) was used to determine the concentrations of heavy metals. The contents of organic extracts are within the range of 140-750 mg/kg. Alkand aro-ratios are relatively high. Compared to those of the background sample (GQ13 ), the contents of saturated hydrocarbon compounds in all the samples are relatively high. The contents of polycyclic aromatic hydrocarbons (PAHs) are relatively high with the distance getting closer to the coal gangue dump. These indicate that organic matter in the samples is from coal particles of the coal gangue dump. | 3 | Nonhuman primate models of focal cerebral ischemia显示文摘Rodents have been widely used in the production of cerebral ischemia models. However, successful therapies have been proven on experimental rodent stroke model, and they have often failed to be effective when tested clinically. Therefore, nonhuman primates were recommended as the ideal alternatives, owing to their similarities with the human cerebrovascular system, brain metabolism, grey to white matter ratio and even their rich behavioral repertoire. The present review is a thorough summary of ten methods that establish nonhuman primate models of focal cerebral ischemia; electrocoagulation, endothelin-1-induced occlusion, microvascular clip occlusion, autologous blood clot embolization, balloon inflation, microcatheter embolization, coil embolization, surgical suture embolization, suture, and photochemical induction methods. This review addresses the advantages and disadvantages of each method, as well as precautions for each model, compared nonhuman primates with rodents, different species of nonhuman primates and different modeling methods. Finally it discusses various factors that need to be considered when modelling and the method of evaluation after modelling. | 4 | MVS-Pheno:A Portable and Low-Cost Phenotyping Platform for Maize Shoots Using Multiview Stereo 3D Reconstruction显示文摘Plant phenotyping technologies play important roles in plant research and agriculture.Detailed phenotypes of individual plants can guide the optimization of shoot architecture for plant breeding and are useful to analyze the morphological differences in response to environments for crop cultivation.Accordingly,high-throughput phenotyping technologies for individual plants grown in field conditions are urgently needed,and MVS-Pheno,a portable and low-cost phenotyping platform for individual plants,was developed.The platform is composed of four major components:a semiautomatic multiview stereo(MVS)image acquisition device,a data acquisition console,data processing and phenotype extraction software for maize shoots,and a data management system.The platform’s device is detachable and adjustable according to the size of the target shoot.Image sequences for each maize shoot can be captured within 60-120 seconds,yielding 3D point clouds of shoots are reconstructed using MVS-based commercial software,and the phenotypic traits at the organ and individual plant levels are then extracted by the software.The correlation coefficient(R^(2))between the extracted and manually measured plant height,leaf width,and leaf area values are 0.99,0.87,and 0.93,respectively.A data management system has also been developed to store and manage the acquired raw data,reconstructed point clouds,agronomic information,and resulting phenotypic traits.The platform offers an optional solution for high-throughput phenotyping of field-grown plants,which is especially useful for large populations or experiments across many different ecological regions. | Sheng Wu Weiliang Wen Yongjian Wang Jiangchuan Fan Chuanyu Wang Wenbo Gou Xinyu Guo | 2020 | Plant Phenomics2020,2,1: | 11 |
| 5 | Microbial aerosol characteristics in highly polluted and nearpristine environments featuring different climatic conditions显示文摘There is an increasing interest in understanding ambient bioaerosols due to their roles both in health and in climate. Here, we deployed an Ultraviolet Aerodynamic Particle Sizer to monitor viable(fluorescent) bioaerosol concentration levels at city centers(highly polluted) and their corresponding suburbs(near pristine)(total 40 locations) in 11 provinces featuring different climate zones in China between July 16 and 28, 2013. The concentration levels of viable bioaerosol particles(Bio PM) of [0.5 lm were measured, and corresponding percentages of Bio PM%(biological fraction of total PM) and Bio PM2.5%(biological fraction of PM2.5) in particulate matter(PM)and Bio PM, respectively, were determined. For some key cities, indoor viable bioaerosol levels were also obtained.In addition, bacterial structures of the air samples collected across these monitoring locations were studied using pyrosequencing. Bio PM concentration levels ranged from2.1 9 104 to 2.4 9 105/m3 for city centers [Bio PM% =6.4 %(±6.3 %)] and 0.5 9 104 to 4.7 9 105/m3 for suburbs [Bio PM% = 10 %(±8.7 %)]. Distinctive bioaerosol size distribution patterns were observed for different climate zones, e.g., some had fluorescence peaks at 3 lm, while the majority had peaks at 1 lm. Ambient bacterial aerosol community structures were also found different for different geophysical locations. Results suggest that there was a poor overall relationship between PM and Bio PM across 40 monitoring locations(R2= 0.081, two-tailed P value =0.07435). Generally, city centers had higher PM concentrations than suburbs, but not Bio PM and Bio PM%. Indoor bioaerosol levels were found at least tenfold higher than those corresponding outdoors. | 9 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 10 | Skeletal loading regulates breast cancer-associated osteolysis in a loading intensity-dependent fashion显示文摘Osteocytes are mechanosensitive bone cells, but little is known about their effects on tumor cells in response to mechanical stimulation. We treated breast cancer cells with osteocyte-derived conditioned medium(CM) and fluid flow-treated conditioned medium(FFCM) with 0.25 Pa and 1 Pa shear stress. Notably, CM and FFCM at 0.25 Pa induced the mesenchymal-to-epithelial transition(MET), but FFCM at 1 Pa induced the epithelial-to-mesenchymal transition(EMT). This suggested that the effects of fluid flow on conditioned media depend on flow intensity. Fluorescence resonance energy transfer(FRET)-based evaluation of Src activity and vinculin molecular force showed that osteopontin was involved in EMT and MET switching. A mouse model of tumorinduced osteolysis was tested using dynamic tibia loadings of 1, 2, and 5 N. The low 1 N loading suppressed tumor-induced osteolysis, but this beneficial effect was lost and reversed with loads at 2 and 5 N, respectively. | 17 | A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. Some results are as follows: 1) Rayleigh quotient accelerate technique is extended to nonselfadjoint problems. Generalized Rayleigh quotients of operator form and weak form are defined and the basic relationship between approximate eigenfunction and its generalized Rayleigh quotient is established. 2) New error estimates are obtained by replacing the ascent of exact eigenvalue with the ascent of finite element approximate eigenvalue. 3) Based on the work of Xu Jinchao and Zhou Aihui, finite element two-grid discretization schemes are established to solve nonselfadjoint elliptic differential operator eigenvalue problems and these schemes are used in both conforming finite element and non-conforming finite element.
