| 1 | Combination of iTRAQ proteomics and RNA-seq transcriptomics reveals multiple levels of regulation in phytoplasma-infected Ziziphus jujuba Mill显示文摘Jujube witches’broom(JWB)is caused by infection with a phytoplasma.A multi-omics approach was taken during graft infection of jujube by JWB-infected scion through the analysis of the plant transcriptome,proteome and phytohormone levels.A high number of differentially expressed genes(DEGs)were identified 37 weeks after grafting(WAG),followed by observation of typical symptoms of JWB at 48 WAG.At 37 WAG,the majority of the upregulated DEGs and differentially expressed proteins(DEPs)were related to flavonoid biosynthesis,phenylalanine metabolism and phenylpropanoid biosynthesis.Two of the four upregulated proteins were similar to jasmonate-induced protein-like.Among the downregulated genes,the two most populated GO terms were plant–pathogen interaction and plant hormone signal transduction(mainly for tryptophan metabolism).Moreover,phytoplasma infection resulted in reduced auxin content and increased jasmonate content,indicating that auxin and jasmonic acid have important roles in regulating jujube responses during the first and second stages of phytoplasma infection.At 48 WAG,the two largest groups of upregulated genes were involved in phenylpropanoid biosynthesis and flavonoid biosynthesis.Both genes and proteins involved in carbon metabolism and carbon fixation in photosynthetic organisms were downregulated,indicating that photosynthesis was affected by the third stage of phytoplasma infection. | Xia Ye Huiyu Wang Peng Chen Bing Fu Mengyang Zhang Jidong Li Xianbo Zheng Bin Tan Jiancan Feng | 2017 | Horticulture Research2017,4,1: | 9 |
| 2 | 3-year Treatment of Tenofovir Alafenamide vs.Tenofovir Disoproxil Fumarate for Chronic HBV Infection in China显示文摘Background and Aims:Tenofovir alafenamide(TAF)has similar efficacy to tenofovir disoproxil fumarate(TDF)but with improved renal and bone safety in chronic hepatitis B patients studied outside of China.We report 3-year results from two phase 3 studies with TAF in China(Clinicaltrials.gov:NCT02836249 and NCT02836236).Methods:Chinese hepatitis B e antigen(HBeAg)-positive and-negative chronic hepatitis B patients with viremia and elevated alanine aminotransferase were randomized 2:1 to TAF or TDF treatment groups and treated in a double-blind fashion for 144 weeks(3 years).Efficacy responses were assessed by individual study while safety was assessed by a pooled analysis.Results:Of the 334 patients(180 HBeAg-positive and 154 HBeAg-negative)randomized and treated,baseline characteristics were similar between groups.The overall mean age was 38 years and 73%were male.The mean HBV DNA was 6.4 log10 IU/mL.The median alanine aminotransferase was 88 U/L,and 37%had a history of antiviral use.At week 144,the proportion with HBV DNA<29 IU/mL was similar among the two groups,with TAF at 83%vs.TDF at 79%,and TAF at 93%vs.TDF at 92%for the HBeAg-positive and-negative patients,respectively.In each study,higher proportions of TAF than TDF patients showed normalized alanine aminotransferase(via the American Association for the Study of Liver Diseases and the China criteria)and showed loss of HBsAg;meanwhile,the HBeAg seroconversion rates were similar.Treatment was well-tolerated among the TAF patients,who showed a smaller median decline in creatinine clearance(−0.4 vs.−3.2 mL/min;p=0.014)and less percentage change in bone mineral density vs.TDF at hip(−0.95%vs.−1.93%)and spine(+0.35%vs.−1.40%).Conclusions:In chronic hepatitis B patients from China,TAF treatment provided efficacy similar to TDF but with better renal and bone safety at 3 years. | Jinlin Hou Qin Ning Zhongping Duan You Chen Qing Xie Fu-Sheng Wang Lunli Zhang Shanming Wu Hong Tang Jun Li Feng Lin Yongfeng Yang Guozhong Gong John FFlaherty Anuj Gaggar Shuyuan Mo Cong Cheng Gregory Camus Chengwei Chen Yan Huang Jidong Jia Mingxiang Zhang GS-US-320-0110 and GS-US-320-0108 China Investigators | 2021 | Journal of Clinical and Translational Hepatology2021,9,3: | 7 |

| 5 | Evaluation of menopausal status among breast cancer patients with chemotherapy-induced amenorrhea显示文摘Objective: In patients with chemotherapy-induced amenorrhea(CIA), the menopausal status is ambiguous and difficult to evaluate. This study aimed to establish a discriminative model to predict and classify the menopausal status of breast cancer patients with CIA.Methods: This is a single center hospital-based study from 2013 to 2016. The menopausal age distribution and accumulated incidence rate of CIA are described. Multivariate models were adjusted for established and potential confounding factors including age, serum concentration of estradiol(E2) and follicle-stimulating hormone(FSH),feeding, pregnancy, parity, abortions, and body mass index(BMI). The odds ratio(OR) and 95% confidence interval(95% CI) of different risk factors were estimated.Results: A total of 1,796 breast cancer patients were included in this study, among whom, 1,175(65.42%) were premenopausal patients and 621(34.58%) were post-menopause patients. Five hundred and fifty patients were included in CIA analysis, and a cumulative CIA rate of 81.64% was found in them. Age(OR: 1.856, 95% CI:1.732-1.990), serum concentration of E2(OR: 0.976, 95% CI: 0.972-0.980) and FSH(OR: 1.060, 95% CI:1.053-1.066), and menarche age(OR: 1.074, 95% CI: 1.009-1.144) were found to be associated with the patients' menopausal status. According to multivariate analysis, the discriminative model to predict the menopausal status is Logit(P)=-28.396+0.536 Age-0.014 E2+0.031 FSH. The sensitivities for this model were higher than 85%, and its specificities were higher than 89%.Conclusions: The discriminative model obtained from this study for predicting menstrual state is important for premenopausal patients with CIA. | 10 | Characterizing and optimizing TPC-C workloads on large-scale systems using SSD arrays显示文摘Transaction processing performance council benchmark C(TPC-C) is the de facto standard for evaluating the performance of high-end computers running on-line transaction processing applications. Differing from other standard benchmarks, the transaction processing performance council only defines specifications for the TPC-C benchmark, but does not provide any standard implementation for end-users. Due to the complexity of the TPC-C workload, it is a challenging task to obtain optimal performance for TPC-C evaluation on a large-scale high-end computer. In this paper, we designed and implemented a large-scale TPC-C evaluation system based on the latest TPC-C specification using solid-state drive(SSD) storage devices. By analyzing the characteristics of the TPC-C workload, we propose a series of system-level optimization methods to improve the TPC-C performance. First, we propose an approach based on Small File table space to organize the test data in a round-robin method on all of the disk array partitions; this can make full use of the underlying disk arrays.Second, we propose using a NOOP-based disk scheduling algorithm to reduce the utilization rate of processors and improve the average input/output service time. Third, to improve the system translation lookaside buffer hit rate and reduce the processor overhead, we take advantage of the huge page technique to manage a large amount of memory resources. Lastly, we propose a locality-aware interrupt mapping strategy based on the asymmetry characteristic of non-uniform memory access systems to improve the system performance. Using these optimization methods, we performed the TPC-C test on two large-scale high-end computers using SSD arrays. The experimental results show that our methods can effectively improve the TPC-C performance. In this situation, some ideas about Chinese courts can reach automation or get better result through the research of machine learning, such as similar documents recommendation, workload evaluation based on similarity of judgement documents and prediction of possible relevant statutes. In trying to achieve all above mentioned, and also in face of the characteristics of Chinese judgement document, we propose a topic model based approach to measure the text similarity of Chinese judgement document, which is based on TF-IDF, Latent Dirichlet Allocation (LDA), Labeled Latent Dirichlet Allocation (LLDA) and other treatments. Combining with the characteristics of Chinese judgment document,we focus on the specific steps of approach, the preprocessing of corpus, the parameters choices of training and the evaluation of similarity measure result. Besides, implementing the approach for prediction of possible statutes and regarding the prediction accuracy as the evaluation metric, we designed experiments to demonstrate the reasonability of decisions in the process of design and the high performance of our approach on text similarity measure. The puncture direction of optimizing prostate biopsy was a 45° angle to the sagittal plane from front, middle, and back. The two cores from each lateral lobe were horizontally inwardly inclined 45°. Results: A total of 45 patients with initial negative biopsy for cancer were received the optimizing prostate biopsy scheme. The cancer detection rate was 17.8%(8/45), and prostate intraepithelial neoplasm(PIN) was 6.7%(3/45). The patients receiving repeat transrectal prostate biopsies were pathologically diagnosed as lower Gleason grade prostate cancers. Conclusion: The cancer detection rate of repeat biopsy prostate cancer is lower than that of initial biopsy. | 20 | 扩大穿刺对PSA≥20ng/mL的患者合适吗?(英文)显示文摘Objective: The aim of this study was to investigate whether it was necessary to increase the number of cores at initial prostate biopsy with patients of prostate-specific antigen(PSA) ≥ 20 ng/mL and to explore an appropriate individualized transrectal ultrasonograhpy(TRUS)-guided prostate biopsy for the detection of prostate cancer in men suspicious of prostate cancer. Methods: A total of 115 patients with PSA ≥ 20 ng/mL and suspicious of prostate cancer were prospectively randomized to perform TRUS-guided biopsy. Patients were randomized to a '6 + X' cores or a '10 + X' cores protocol. The primary end point was cancer detection rate. Secondary end points were cancer characteristics, rate of complications and the level of pain experienced by patients during TRUS-guided prostate biopsy. Results: Preoperative variables were similar in both groups. The overall prostate cancer detection rate was 73.9%. The '10 + X' cores strategy increased cancer detection rate only 9.7% in patients with PSA ≥ 20 ng/mL but < 50 ng/mL, while there was no difference between the two strategies for cancer detection in patients with PSA ≥ 50.1 ng/mL. The number of extended biopsy cores and pain score of extended biopsy in prostate cancer patients increased significantly(P < 0.001).
