| 1 | Effect of Chaiqinchengqi decoction on inositol requiring enzyme 1α in alveolar macrophages of dogs with acute necrotising pancreatitis induced by sodium taurocholate显示文摘OBJECTIVE: To investigate the effect of Chaiqinchengqi decoction(CQCQD) on inositol requiring enzyme 1α(IRE1α) in alveolar macrophages(AMs)of the dog model of acute necrotising pancreatitis(ANP) induced by sodium taurocholate.METHODS: Fifteen beagle dogs were randomised into a control group,ANP group and CQCQD group(n = 5 per group). ANP was induced by a retrograde duct injection of 50 mg/kg of 5% sodium taurocholate. The dogs in the control group received injections of the same volume of saline as the sodium taurocholate. After the models were induced,the dogs in the CQCQD group were administered 10 m L/kg CQCQD every 2 h for 6 h. Two hours after the last administration of either CQCQD or saline,they were sacrificed by anaesthesia. AMs were collected to determine the IRE1α and Interleukin-1β(IL-1β)m RNA and protein expression,and pancreatic tissues were collected for histopathology analysis.RESULTS: Compared with the ANP group,the m RNA and protein expression of IRE1α and the protein expression of IL-1β of AMs in the CQCQD group were significantly down-regulated,and the pancreatic histopathology score of the CQCQD group also was lower. | 3 | Changes in Soil Enzyme Activity and Nutrient Content in Different Years of Continuous Cropping Tobacco Fields显示文摘In order to find out changes in the main fertility indicators of continuous cropping tobacco fields in Henan Province with years of continuous cropping,5 typical tobacco fields( Jia County,Shaoling District,Fangcheng County,Xiangcheng County,and Song County) were studied. Indicators in question included activity of main enzymes( sucrase,urease,catalase,and acid phosphatase),p H value,organic matters,total nitrogen,available phosphorus and available potassium. The results indicate that in most tobacco fields,the activity of sucrase and urease increased in two years of continuous cropping,later,it showed a declining trend with increase in years of continuous cropping; in some fields,it directly declined with increase in years of continuous cropping; the activity of catalase increased in two years of continuous cropping,later,it showed a declining trend with increase in years of continuous cropping,and it became stable after three years of continuous cropping;generally,the activity of acid phosphatase declined with increase in the years of continuous cropping within 3 years continuous cropping,later,it became stable. Except Fangcheng County,the soil p H value of other districts( counties) generally declined with increase in years of continuous cropping,and it was in the range of 5. 6-6. 8. With the increase in continuous cropping,the soil organic matters( SOM) in tobacco fields of Jia County and Song County declined,changes of SOM in tobacco fields of Fangcheng County and Xiangcheng County were not obvious,and SOM showed the trend of first decline then rise in Shaoling District. In most tobacco fields( Jia County,Xiangcheng County,and Song County),the total nitrogen showed a declining trend; in Shaoling District,it showed a trend of first decline then rise; in Fangcheng County,it showed a rising trend. The available phosphorus firstly slightly declined then rose in tobacco fields of Shaoling District and Song County; it showed a trend of first rise then decline in Jia County; there were no obvious changes in Fangcheng County and Xiangcheng County. Changes in the available potassium were different in tobacco fields. In sum,there are certain rules for changes in soil enzyme activity and nutrient content in tobacco fields in Henan Province,but changes are not completely the same. It is concluded that the site conditions and farming activities exert a certain influence on soil enzyme activity,p H value,organic matters,and nitrogen,phosphorus and potassium content. | 8 | Interaction between neutrophil extracellular traps and cardiomyocytes contributes to atrial fibrillation progression显示文摘Atrial fibrillation(AF)is a frequent arrhythmia associated with cardiovascular morbidity and mortality.Neutrophil extracellular traps(NETs)are DNA fragments with cytoplasm proteins released from neutrophils,which are involved in various cardiovascular diseases.To elucidate the role of NETs in AF,we investigated the effect of NETs on AF progression and the secretion of NETs in AF.Results showed that:NETs induced the autophagic apoptosis of cardiomyocytes,and NETs also led to mitochondrial injury by promoting mitochondrial depolarization and ROS production.Ongoing tachy-pacing led to the structural loss of cardiomyocytes and provided potent stimuli to induce NETs secretion from neutrophils.In the meanwhile,increased Ang II in AF facilitated NETs formation through the upregulation of AKT phosphorylation,while it could not directly initiate NETosis as the autophagy was not induced.In vivo,DNase I was administrated to abrogate NETs formation,and AF-related fibrosis was ameliorated as expected.Correspondingly,the duration of the induced AF was reduced.Our study addresses the formation mechanism of NETs in AF and demonstrates the lethal effects of NETs on cardiomyocytes through the induction of mitochondrial injury and autophagic cell death,which comprehensively describes the positive feedback comprised of NETs and stimuli secreted by cardiomyocytes that sustains the progression of AF and AF related fibrosis. | Li He Ruiqi Liu Honghua Yue Xiaoxin Zhang Xiaohui Pan Yutao Sun Jun Shi Guonian Zhu Chaoyi Qin Yingqiang Guo | 2023 | Signal Transduction and Targeted Therapy2023,8,8: | 1 |
| 9 | SZ-5 Cabin’s Height Changes during Three Super-storms in 2003显示文摘In this work,the daily height variations of SZ-5(Shenzhou-5) cabin from 22 October to 28 November in 2003 are analyzed,which includes the period of the Halloween Storm and the Great November Storm.The significant orbital decays have been observed at the end of October and in late November due to the great solar flares and the severe geomagnetic storms.According to the equation of the air-drag-force on a spacecraft and the SZ-5 orbital decay information,the relative daily average thermospheric density changes during the three 2003 super-storms are derived and the results are compared with the Naval Research Laboratory Mass Spectrometer Incoherent Scatter Radar Extended Model(NRLMSISE-00).The results show that the daily average thermospheric density(at the altitude of SZ-5,about 350 km) in storm time enhances to approximately 200% as much as that in the quiet time but the empirical model may somewhat underestimate the average thermospheric density changes and the daily contributions of geomagnetic storms to the density enhancements during these severe space weather events. | HUANG Cong LIU Dandan GUO Jing ZHANG Xiaoxin | 2019 | 空间科学学报2019,39,6: | 0 |

| 13 | Screening of Fermentation Starters for Organic Fertilizer Composted from Chicken Manure显示文摘In order to find fermentation starters that can promote rapid and effective composting of chicken manure,shorten the fermentation period,and improve the compost quality,fermentation starters with different microbial combination ratios(C1,C2,C3,C4 and C5) were designed and fermentation experiment was carried out on these five fermentation starters,commercially available organic fertilizer starter(SS),and chicken manure without inoculating fermentation starter. In the process of the fermentation,the changes in the fermentation temperature,water content and pH value were monitored; the effects of organic manure on the germination rate and growth of rapeseed seeds were measured;viable count,water content,and pH value of prepared fermentation starters were measured; main indicators( organic matter,nitrogen,phosphorus,and potassium) of organic fertilizer generated from fermentation were measured. From the indicators,the formula C3 and C4 were better,in the beginning of fermentation,the temperature rose rapidly,and it reached 40 ℃ in the second day; the water content dropped to below 35% in the 18 th day; the pH value underwent the process of declining-rising-stable,and the overall value showed slight rise,but the change was not great( 0-0. 5). C3 and C4 fermentation products had better effects on the germination and growth of rapeseeds. The germination rate of the rapeseed seeds was 100% and 97. 5% and the bud length was 15. 94 mm and 14. 57 mm,respectively,and the root length was45. 97 mm and 39. 44 mm,respectively. The content of organic matter in fermented organic manure was 86. 62% and 85. 17% respectively,and the total content of nitrogen,phosphorus and potassium was 8. 85% and 8. 22% respectively,and the pH was 7. 5 and 8. 0 respectively.All of these complied with the industry standard NY525-2012( organic matter ≥ 45%,and nitrogen,phosphorus,and potassium ≥ 5%,pH 5. 5-8. 5). | 14 | Characterizing and engineering promoters for metabolic engineering of Ogataea polymorpha显示文摘Bio-manufacturing via microbial cell factory requires large promoter library for fine-tuned metabolic engi-neering.Ogataea polymorpha,one of the methylotrophic yeasts,possesses advantages in broad substrate spec-trum,thermal-tolerance,and capacity to achieve high-density fermentation.However,a limited number of available promoters hinders the engineering of O.polymorpha for bio-productions.Here,we systematically characterized native promoters in O.polymorpha by both GFP fluorescence and fatty alcohol biosynthesis.Ten constitutive promoters(P_(PDH),P_(PYK),P_(FBA),P_(PGM),P_(GLK),P_(TRI),P(GPI),P_(ADH1),P_(TEF1) and P_(GCW14))were obtained with the activity range of 13%–130% of the common promoter P_(GAP)(the promoter of glyceraldehyde-3-phosphate de-hydrogenase),among which P_(PDH) and P_(GCW14) were further verified by biosynthesis of fatty alcohol.Furthermore,the inducible promoters,including ethanol-induced P_(ICL1),rhamnose-induced P_(LRA3) and P_( LRA4),and a bidirectional promoter(P_(Mal)-P_(Per))that is strongly induced by sucrose,further expanded the promoter toolbox in O.polymorpha.Finally,a series of hybrid promoters were constructed via engineering upstream activation sequence(UAS),which increased the activity of native promoter P LRA3 by 4.7–10.4 times without obvious leakage expression.Therefore,this study provided a group of constitutive,inducible,and hybrid promoters for metabolic engineering of O.polymorpha,and also a feasible strategy for rationally regulating the promoter strength. | Chunxiao Yan Wei Yu Xiaoxin Zhai Lun Yao Xiaoyu Guo Jiaoqi Gao Yongjin J.Zhou | 2022 | Synthetic and Systems Biotechnology2022,7,1: | 0 |
