| 1 | SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. | 2 | Tolerance to low phosphorus in rice varieties is conferred by regulation of root growth显示文摘Phosphorus use efficiency(PUE)can be improved through cultivation techniques and breeding.However,little is known about rice(Oryza sativa L.)agronomic and physiological traits associated with high PUE.We characterized the agronomic and physiological traits of rice varieties with different tolerances to low phosphorus in nutrient solution.Two varieties with strong tolerance to low phosphorus(STVs)and two with weak tolerance(WTVs)were grown at normal(NP,control)and low phosphorus(LP,1/20 of NP)concentrations.Plants grown at LP produced significantly lower grain yield than those grown at NP.WTVs yields were lower than STVs yields.Compared to NP,LP significantly increased phosphorus translocation efficiency(PTE),internal phosphorus efficiency(IPE)and phosphorus harvest index(PHI).Under the LP condition,PTE and IPE were higher for STVs than for WTVs.LP also reduced tiller number,shoot biomass,leaf area index(LAI),leaf photosynthetic rate,and mean root diameter of both kinds of varieties at the main growth stages,but to a lower extent in STVs.LP significantly increased the number of productive tillers,root biomass,root-shoot ratio,root bleeding rate,and root acid phosphatase(RAP)activity.Total root length,root oxidation activity(ROA),and root total and active absorbing surface areas for STVs were significantly increased under LP,whereas the opposite responses were observed for WTVs.Total root length,ROA,root bleeding rate,root active absorbing surface area,and RAP activity were positively and significantly correlated with grain yield,PTE,and IPE.These results suggest that the tolerance of rice varieties to a low-phosphorus growth condition is closely associated with root growth with higher biomass and activity. | Yaping Deng Chuanbao Men Shengfeng Qiao Wenjie Wang Junfei Gu Lijun Liu Zujian Zhang Hao Zhang Zhiqin Wang Jianchang Yang | 2020 | The Crop Journal2020,8,4: | 8 |

| 4 | DNA damage repair promotion in colonic epithelial cells by andrographolide downregulated cGAS-STING pathway activation and contributed to the relief of CPT-11-induced intestinal mucositis显示文摘Gastrointestinal mucositis is one of the most debilitating side effects of the chemotherapeutic agent irinotecan(CPT-11). Andrographolide, a natural bicyclic diterpenoid lactone, has been reported to possess anti-colitis activity. In this study, andrographolide treatment was found to significantly relieve CPT-11-induced colitis in tumor-bearing mice without decreasing the tumor suppression effect of CPT-11. CPT-11 causes DNA damage and the release of double-stranded DNA(ds DNA) from the intestine, leading to cyclic-GMP-AMP synthase(c GAS)-stimulator of interferon genes(STING)-mediated colitis, which was significantly decreased by andrographolide both in vivo and in vitro. Mechanistic studies revealed that andrographolide could promote homologous recombination(HR) repair and downregulate ds DNA-c GAS-STING signaling and contribute to the improvement of CPT-11-induced gastrointestinal mucositis. | 7 | Memory access integrity:detecting fine-grained memory access errors in binary code显示文摘As one of the most notorious programming errors,memory access errors still hurt modern software security.Particularly,they are hidden deeply in important software systems written in memory unsafe languages like C/C++.Plenty of work have been proposed to detect bugs leading to memory access errors.However,all existing works lack the ability to handle two challenges.First,they are not able to tackle fine-grained memory access errors,e.g.,data overflow inside one data structure.These errors are usually overlooked for a long time since they happen inside one memory block and do not lead to program crash.Second,most existing works rely on source code or debugging information to recover memory boundary information,so they cannot be directly applied to detection of memory access errors in binary code.However,searching memory access errors in binary code is a very common scenario in software vulnerability detection and exploitation.In order to overcome these challenges,we propose Memory Access Integrity(MAI),a dynamic method to detect finegrained memory access errors in off-the-shelf binary executables.The core idea is to recover fine-grained accessing policy between memory access behaviors and memory ranges,and then detect memory access errors based on the policy.The key insight in our work is that memory accessing patterns reveal information for recovering the boundary of memory objects and the accessing policy.Based on these recovered information,our method maintains a new memory model to simulate the life cycle of memory objects and report errors when any accessing policy is violated.We evaluate our tool on popular CTF datasets and real world softwares.Compared with the state of the art detection tool,the evaluation result demonstrates that our tool can detect fine-grained memory access errors effectively and efficiently.As the practical impact,our tool has detected three 0-day memory access errors in an audio decoder. | Wenjie Li Dongpeng Xu WeiWu Xiaorui Gong Xiaobo Xiang Yan Wang Fangming gu Qianxiang Zeng | 2019 | Cybersecurity2019,2,1: | 1 |

| 16 | Impact of adverse childhood experiences on the symptom severity of different mental disorders: a cross-diagnostic study显示文摘Background Adverse childhood experiences have a significant impact on different mental disorders.Objective To compare differences in adverse childhood experiences among those with different mental disorders and their relationships in a cross-disorder manner.Methods The study included 1513 individuals aged≥18 years:339 patients with substance use disorders,125 patients with schizophrenia,342 patients with depression,136 patients with bipolar disorder,431 patients with obsessive-compulsive disorder(OCD),and 140 healthy controls.The Early Trauma Inventory Self Report-Short Form was used to investigate childhood traumatic experiences,and the Addiction Severity Index,Positive and Negative Syndrome Scale,Hamilton Depression Scale,Young Mania Rating Scale,and Yale-Brown Obsessive-Compulsive Scale were used to assess mental disorder severity.Correlation and multivariate logistic regression were analysed between adverse childhood experiences and clinical features.Results Levels of adverse childhood experiences were significantly different among different mental disorders.Moreover,25.8%of patients with substance use disorders reported childhood trauma,which was significantly higher than found in the other four psychiatric disorder groups.Emotional abuse scores were positively correlated with disease severity:the higher the total trauma score,the more severe the mental disorder.Conclusions Adverse childhood experiences are a common phenomenon in those with mental disorders,and the level of trauma affects mental disorder severity.Emotional abuse is closely related to many mental disorders.The incidence or severity of mental disorders can be reduced in the future by reducing the incidence of adverse childhood experiences or by timely intervention in childhood trauma. | Wenjie Gu Qing Zhao Chengmei Yuan Zhenghui Yi Min Zhao Zhen Wang | 2022 | General Psychiatry2022,35,2: | 1 |

| 20 | Early Diagnosis of Bipolar Disorder Coming Soon:Application of an Oxidative Stress Injury Biomarker(BIOS)Model显示文摘Early distinction of bipolar disorder(BD)from major depressive disorder(MDD)is difficult since no tools are available to estimate the risk of BD.In this study,we aimed to develop and validate a model of oxidative stress injury for predicting BD.Data were collected from 1252 BD and 1359 MDD patients,including 64 MDD patients identified as converting to BD from 2009 through 2018.30 variables from a randomly-selected subsample of 1827(70%)patients were used to develop the model,including age,sex,oxidative stress markers(uric acid,bilirubin,albumin,and prealbumin),sex hormones,cytokines,thyroid and liver function,and glycolipid metabolism.Univariate analyses and the Least Absolute Shrinkage and Selection Operator were applied for data dimension reduction and variable selection.Multivariable logistic regression was used to construct a model for predicting bipolar disorder by oxidative stress biomarkers(BIOS)on a nomogram.Internal validation was assessed in the remaining 784 patients(30%),and independent external validation was done with data from 3797 matched patients from five other hospitals in China.10 predictors,mainly oxidative stress markers,were shown on the nomogram.The BIOS model showed good discrimination in the training sample,with an AUC of 75.1%(95%CI:72.9%–77.3%),sensitivity of 0.66,and specificity of 0.73.The discrimination was good both in internal validation(AUC 72.1%,68.6%–75.6%)and external validation(AUC 65.7%,63.9%–67.5%).In this study,we developed a nomogram centered on oxidative stress injury,which could help in the individualized prediction of BD.For better real-world practice,a set of measurements,especially on oxidative stress markers,should be emphasized using big data in psychiatry. | Zhiang Niu Xiaohui Wu Yuncheng Zhu Lu Yang Yifan Shi Yun Wang Hong Qiu Wenjie Gu Yina Wu Xiangyun Long Zheng Lu Shaohua Hu Zhijian Yao Haichen Yang Tiebang Liu Yong Xia Zhiyu Chen Jun Chen Yiru Fang | 2022 | Neuroscience Bulletin2022,38,9: | 1 |
