A single-cell transcriptome of mesenchymal stromal cells to fabricate bioactive hydroxyapatite materials for bone regeneration显示文摘The osteogenic microenvironment of bone-repairing materials plays a key role in accelerating bone regeneration but remains incompletely defined, which significantly limits the application of such bioactive materials. Here, the transcriptional landscapes of different osteogenic microenvironments, including three-dimensional(3D)hydroxyapatite(HA)scaffolds and osteogenic medium(OM),for mesenchymal stromal cells(MSCs)in vitro were mapped at single-cell resolution. Our findings suggested that an osteogenic process reminiscent of endochondral ossification occurred in HA scaffolds through sequential activation of osteogenic-related signaling pathways, along with inflammation and angiogenesis, but inhibition of adipogenesis and fibrosis. Moreover,we revealed the mechanism during OM-mediated osteogenesis involves the ZBTB16 and WNT signaling pathways.Heterogeneity of MSCs was also demonstrated. In vitro ossification of LRRC75A+MSCs was shown to have better utilization of WNT-related ossification process, and PCDH10+MSCs with superiority in hydroxyapatite-related osteogenic process. These findings provided further understanding of the cellular activity modulated by OM conditions and HA scaffolds, providing new insights for the improvement of osteogenic biomaterials. This atlas provides a blueprint for research on MSC heterogeneity and the osteogenic microenvironment of HA scaffolds and a database reference for the application of bioactive materials for bone regeneration. | Peng Guo Xizhe Liu Penghui Zhang Zhongyuan He Zhen Li Mauro Alini RGeoff Richards Sibylle Grad Martin J.Stoddart Guangqian Zhou Xuenong Zou Danny Chan Wei Tian Dafu Chen Manman Gao Zhiyu Zhou Shaoyu Liu | 2022 | Bioactive Materials2022,7,3: | 3
Predicting the degradation behavior of magnesium alloys with a diffusion-based theoretical model and in vitro corrosion testing显示文摘Magnesium alloys have shown great potential for their use in the medical device field, due to the promising biodegradability. However, it remains a challenge to characterize the degradation behavior of the Mg alloys in a quantitative manner. As such, controlling the degradation rate of the Mg alloys as per our needs is still hard, which greatly limits the practical application of the Mg alloys as a degradable biomaterial. This paper discussed a numerical model developed based on the diffusion theory, which can capture the experimental degradation behavior of the Mg alloys precisely. The numerical model is then implemented into a finite element scheme, where the model is calibrated with the data from our previous studies on the corrosion of the as-cast Mg-1 Ca and the as-rolled Mg-3 Ge binary alloys. The degradation behavior of a pin implant is predicted using the calibrated model to demonstrate the model’s capability. A standard flow is provided in a practical framework for obtaining the degradation behavior of any biomedical Mg alloys. This methodology was further verified via the comparison with enormous available experimental results. Lastly, the material parameters defined in this model were provided as a new kind of material property. | Zhenquan Shen Ming Zhao Dong Bian Danni Shen Xiaochen Zhou Jianing Liu Yang Liu Hui Guo Yufeng Zheng | 2019 | Journal of Materials Science & Technology2019,35,7: | 2

Crystallinity engineering of Au nanoparticles on graphene for in situ SERS monitoring of Fenton-like reaction显示文摘Fabrication of multifunctional nanoplatform to in situ monitor Fenton reaction is of vital importance to probe the underlying reaction process and design high-performance catalyst. Herein, a hybrid catalyst comprising of single-crystalline Au nanoparticles (SC Au NPs) on reduced graphene oxide (RGO) sheet was prepared, which not only exhibited an excellent ^(1)O_(2) mediated Fenton-like catalytic activity in promoting rhodamine 6G (R6G) degradation by activating H_(2)O_(2), but also displayed a sensitive surface-enhanced Raman spectroscopy (SERS) detection performance to R6G with a linear response range from 1.0×10^(-8) mol/L to 1.0×10^(-5) mol/L thus providing a powerful and versatile nanoplatform for in situ SERS monitoring Fenton-like catalytic reaction. The integration of catalytic and SERS activities into a single nanostructure are expected to provide great potentials for practical applications in environmental catalysis. | Danni Guo Lixia Zhao Hui Zhang | 2022 | Chinese Chemical Letters2022,33,3: | 1
Comprehensive characterization of CRC with germline mutations reveals a distinct somatic mutational landscape and elevated cancer risk in the Chinese population显示文摘Objective:Hereditary colorectal cancer(CRC)accounts for approximately 5%–10%of all CRC cases. The full profile of CRC-related germline mutations and the corresponding somatic mutational profile have not been fully determined in the Chinese population. Methods:We performed the first population study investigating the germline mutation status in more than 1,000(n=1,923)Chinese patients with CRC and examined their relationship with the somatic mutational landscape.Germline alterations were examined with a 58-gene next-generation sequencing panel, and somatic alterations were examined with a 605-gene panel. Results:A total of 92 pathogenic(P)mutations were identified in 85 patients, and 81 likely pathogenic(LP)germline mutations were identified in 62 patients, accounting for 7.6%(147/1,923)of all patients.MSH2 and APC was the most mutated gene in the Lynch syndrome and non-Lynch syndrome groups, respectively.Patients with P/LP mutations had a significantly higher ratio of microsatellite instability, highly deficient mismatch repair, family history of CRC,and lower age.The somatic mutational landscape revealed a significantly higher mutational frequency in the P group and a trend toward higher copy number variations in the non-P group. The Lynch syndrome group had a significantly higher mutational frequency and tumor mutational burden than the nonLynch syndrome group.Clustering analysis revealed that the Notch signaling pathway was uniquely clustered in the Lynch syndrome group, and the MAPK and cAMP signaling pathways were uniquely clustered in the non-Lynch syndrome group.Population risk analysis indicated that the overall odds ratio was 11.13(95%CI:8.289–15.44)for the P group and 20.68(95%CI:12.89–33.18)for the LP group. Conclusions:Distinct features were revealed in Chinese patients with CRC with germline mutations. The Notch signaling pathway was uniquely clustered in the Lynch syndrome group, and the MAPK and cAMP signaling pathways were uniquely clustered in the non-Lynch syndrome group.Patients with P/LP germline mutations exhibited higher CRC risk. | Jianfei Yao Yunhuan Zhen Jing Fan Yuan Gong Yumeng Ye Shaohua Guo Hongyi Liu Xiaoyun Li Guosheng Li Pan Yang Xiaohui Wang Danni Liu Tanxiao Huang Huiya Cao Peisu Suo Yuemin Li Jingbo Yu Lele Song | 2022 | Cancer Biology & Medicine2022,19,5: | 1
Bioengineering extracellular vesicles as novel nanocarriers towards brain disorders显示文摘Despite noteworthy technological progress and promising preclinical trials, brain disorders are still the leading causes of death globally.Extracellular vesicles(EVs),nano-/micro-sized membrane vesicles carrying bioactive molecules, are involved in cellular communication.Based on their unique properties, including superior biocompatibility,non-immunogenicity,and blood-brain barrier(BBB)penetration,EVs can shield their cargos from immune clearance and transport them to specific site, which have attracted increasing interests as novel nanocarriers for brain disorders. However, considering the limitations of native EVs,such as poor encapsulation efficiency, inadequate targeting capability, uncontrolled drug release, and limited production, researchers bioengineer EVs to fully exploit the clinical potential.Herein, this review initially describes the basic properties, biogenesis, and uptake process of EVs from different subtypes. Then,we highlight the application of EVs derived from different sources for personalized therapy and novel strategies to construct bioengineered EVs for enhanced diagnosis and treatment of brain disorders.Besides,it also presents a systematic comparison between EVs and other brain-targeted nanocarriers. Finally, existing challenges and future perspectives of EVs have been discussed, hoping to bolster the research from benchtop to bedside. | Jie Wu Lei Ma Danni Sun Xinru Zhang Jiwei Cui Yingjiang Du Yumiao Guo Xue Wang Liuqing Di Ruoning Wang | 2023 | Nano Research2023,16,2: | 1

Notes for genera:Ascomycota显示文摘Knowledge of the relationships and thus the classification of fungi, has developed rapidly with increasingly widespread use of molecular techniques, over the past 10–15 years, and continues to accelerate.Several genera have been found to be polyphyletic, and their generic concepts have subsequently been emended.New names have thus been introduced for species which are phylogenetically distinct from the type species of particular genera. The ending of the separate naming of morphs of the same species in 2011,has also caused changes in fungal generic names. In order to facilitate access to all important changes,it was desirable to compile these in a single document. The present article provides a list of generic names of Ascomycota(approximately 6500 accepted names published to the end of 2016),including those which are lichen-forming.Notes and summaries of the changes since the last edition of‘Ainsworth&Bisby’s Dictionary of the Fungi’in 2008 are provided. The notes include the number of accepted species, classification, type species(with location of the type material),culture availability,life-styles,distribution,and selected publications that have appeared since 2008.This work is intended to provide the foundation for updating the ascomycete component of the'Without prejudice list of generic names of Fungi'published in 2013,which will be developed into a list of protected generic names. This will be subjected to the XIXth International Botanical Congress in Shenzhen in July 2017 agreeing to a modification in the rules relating to protected lists, and scrutiny by procedures determined by the Nomenclature Committee for Fungi(NCF).The previously invalidly published generic names Barriopsis,Collophora(as Collophorina),Cryomyces,Dematiopleospora,Heterospora(as Heterosporicola),Lithophila,Palmomyces(as Palmaria)and Saxomyces are validated,as are two previously invalid family names,Bartaliniaceae and Wiesneriomycetaceae.Four species of Lalaria, which were invalidly published are transferred to Taphrina and validated as new combinations.Catenomycopsis Tibell&Constant.is reduced under Chaenothecopsis Vain.,while Dichomera Cooke is reduced under Botryosphaeria Ces.&De Not.(Art.59). | Nalin N.Wijayawardene Kevin D.Hyde Kunhiraman C.Rajeshkumar David L.Hawksworth Hugo Madrid Paul M.Kirk Uwe Braun Rajshree V.Singh Pedro W.Crous Martin Kukwa Robert Lücking Cletus P.Kurtzman Andrey Yurkov Danny Haelewaters Andre´Aptroot H.Thorsten Lumbsch Einar Timdal Damien Ertz Javier Etayo Alan J.L.Phillips Johannes Z.Groenewald Moslem Papizadeh Laura Selbmann Monika C.Dayarathne Gothamie Weerakoon E.B.Gareth Jones Satinee Suetrong Qing Tian Rafael F.Castanda-Ruiz Ali H.Bahkali Ka-Lai Pang Kazuaki Tanaka Dong Qin Dai Jariya Sakayaroj Martina Hujslová Lorenzo Lombard Belle D.Shenoy Ave Suija Sajeewa S.N.Maharachchikumbura Kasun M.Thambugala Dhanushka N.Wanasinghe Bharati O.Sharma Subhash Gaikwad Gargee Pandit Laura Zucconi Silvano Onofri Eleonora Egidi Huzefa A.Raja Rampai Kodsueb Marcela E.S.Caceres Sergio Perez-Ortega Patrícia O.Fiuza Josiane Santana Monteiro Larissa N.Vasilyeva Roger G.Shivas Maria Prieto Mats Wedin Ibai Olariaga Adebola Azeez Lateef Yamini Agrawal Seyed Abolhassan Shahzadeh Fazeli Mohammad Ali Amoozegar Guo Zhu Zhao Walter P.Pfliegler Gunjan Sharma Magdalena Oset Mohamed A.Abdel-Wahab Susumu Takamatsu Konstanze Bensch Nimali Indeewari de Silva AndréDe Kesel Anuruddha Karunarathna Saranyaphat Boonmee Donald H.Pfister Yong-Zhong Lu Zong-Long Luo Nattawut Boonyuen Dinushani A.Daranagama Indunil C.Senanayake Subashini C.Jayasiri Milan C.Samarakoon Xiang-Yu Zeng Mingkwan Doilom Luis Quijada Sillma Rampadarath Gabriela Heredia Asha J.Dissanayake Ruvishika S.Jayawardana Rekhani H.Perera Li Zhou Tang Chayanard Phukhamsakda Margarita Hernańdez-Restrepo Xiaoya Ma Saowaluck Tibpromma Luis F.P.Gusmao Darshani Weerahewa Samantha C.Karunarathna | 2017 | Fungal Diversity2017,,5: | 0
Hallmark guided identification and characterization of a novel immune-relevant signature for prognostication of recurrence in stageⅠ-Ⅲlung adenocarcinoma显示文摘The high risk of postoperative mortality in lung adenocarcinoma(LUAD)patients is principally driven by cancer recurrence and low response rates to adjuvant treatment. Here,A combined cohort containing 1,026 stageⅠ-Ⅲpatients was divided into the learning(n Z 678)and validation datasets(n Z 348).The former was used to establish a 16-mRNA risk signature for recurrence prediction with multiple statistical algorithms, which was verified in the valida-tion set.Univariate and multivariate analyses confirmed it as an independent indicator for both recurrence-free survival(RFS)and overall survival(OS).Distinct molecular characteristics between the two groups including genomic alterations, and hallmark pathways were compre-hensively analyzed.Remarkably, the classifier was tightly linked to immune infiltrations,high-lighting the critical role of immune surveillance in prolonging survival for LUAD.Moreover, the classifier was a valuable predictor for therapeutic responses in patients, and the low-risk group was more likely to yield clinical benefits from immunotherapy.A transcription factor regulato-ry proteineprotein interaction network(TF-PPI-network)was constructed via weighted gene co-expression network analysis(WGCNA)concerning the hub genes of the signature. The con-structed multidimensional nomogram dramatically increased the predictive accuracy.There-fore,our signature provides a forceful basis for individualized LUAD management with promising potential implications. | Yongqiang Zhang Zhao Yang Yuqin Tang Chengbin Guo Danni Lin d Linling Cheng Xun Hu Kang Zhang Gen Li | 2023 | Genes & Diseases2023,10,4: | 0
Rare peptide anchors of HLA class I alleles contribute to the COVID-19 disease severity and T cell memory显示文摘Understanding how human leukocyte antigen(HLA)polymorphism affects both the susceptibility and severity of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection will help to identify individuals at higher risk to better manage and prioritize vaccination at the clinical level and explain the differences in epidemic trends in different regions at the epidemiological level. This study compared the frequencies of HLA class I alleles(HLA-A,B)in 214 coronavirus disease 2019(COVID-19)patients with different disease severity and 35 healthy controls and analyzed the correlations between specific HLA alleles and disease severity and T cell memory. The results showed no significant difference in HLA allele frequencies between COVID-19 patients and healthy controls(P>0.05).The allele HLA-B*13:02 was significantly correlated with the disease severity of COVID-19 patients(P=0.006).After adjustment for age and disease severity, the T cell responses of COVID-19 convalescents with the allele HLA-B*40:01 may be lower at six months(P=0.044)and 12 months(P=0.069).Moreover,these results may be due to their rare peptide anchors by analyzing the binding peptide motifs of these HLA alleles. The study may be valuable for investigating the potential association of specific HLA alleles with SARS-CoV-2 infection. | Xin Wang Jie Zhang Peipei Guo Yuanyuan Guo Xiaonan Yang Maoshun Liu Danni Zhang Yaxin Guo Jianbo Zhan Kun Cai Jikin Zhou Shaobo Dong Jun Liu | 2023 | Biosafety and Health2023,5,6: | 0
The wheat sucrose synthase gene TaSus1 is a determinant of grain number per spike显示文摘Some haplotypes of the sucrose synthase gene TaSus1 are associated with thousand-grain weight(TGW)in wheat(Triticum aestivum L.).However,no mutations have been identified within the gene to test this association. The effects of TaSus1 on grain number per spike(GNS)also are largely unknown. Our previous genome-wide association study identified TaSus-A1 as a candidate gene controlling fertile spikelet number per spike(FSN).In the present study,we generated two independent mutants for the three TaSus1 homoeologs by CRISPR/Cas9-mediated genome editing. The triple mutants displayed lower FSN,GNS,grain number per spikelet(GNST),and TGW than wild-type plants. In 306 hexaploid wheat accessions, two single-nucleotide polymorphisms in TaSus-A1 contributed differently to GNS.Introgression of the two alleles into a wheat genetic background confirmed their effects. The alleles differed in geographical distribution among the accessions. | Liping Shen Lili Zhang Changbin Yin Xiaowan Xu Yangyang Liu Kuocheng Shen He Wu Zhiwen Sun Ke Wang Zhonghu He Xueyong Zhang Chenyang Hao Jian Hou Aoyue Bi Xuebo Zhao Daxing Xu Botao Ye Xuchang Yu Ziying Wang Danni Liu Yuanfeng Hao Fei Lu Zifeng Guo | 2024 | The Crop Journal2024,12,1: | 0
Sustained release of magnesium and zinc ions synergistically accelerates wound healing显示文摘Skin wounds are a major medical challenge that threaten human health.Functional hydrogel dressings demonstrate great potential to promote wound healing. In this study, magnesium(Mg)and zinc(Zn)are introduced into methacrylate gelatin(GelMA)hydrogel via low-temperature magnetic stirring and photocuring, and their effects on skin wounds and the underlying mechanisms are investigated.Degradation testing confirmed that the GelMA/Mg/Zn hydrogel released magnesium ions(Mg^(2+))and zinc ions(Zn^(2+))in a sustained manner. The Mg^(2+) and Zn^(2+)not only enhanced the migration of human skin fibroblasts(HSFs)and human immortalized keratinocytes(HaCats),but also promoted the transformation of HSFs into myofibroblasts and accelerated the production and remodeling of extracellular matrix. Moreover, the GelMA/Mg/Zn hydrogel enhanced the healing of full-thickness skin defects in rats via accelerated collagen deposition, angiogenesis and skin wound re-epithelialization.We also identified the mechanisms through which GelMA/Mg/Zn hydrogel promoted wound healing:the Mg^(2+) promoted Zn^(2+)entry into HSFs and increased the concentration of Zn^(2+)in HSFs,which effectively induced HSFs to differentiate into myofibroblasts by activating the STAT3 signaling pathway. The synergistic effect of Mg^(2+) and Zn^(2+)promoted wound healing. In conclusion, our study provides a promising strategy for skin wounds regeneration. | Fan Yang Yijia Xue Feilong Wang Danni Guo Yunjiao He Xiao Zhao Fanyu Yan Yuqian Xu Dandan Xia Yunsong Liu | 2023 | Bioactive Materials2023,,8: | 0
Identification of the lysine and histidine transporter family in Camellia sinensis and the characterizations in nitrogen utilization显示文摘In plants, the lysine and histidine transporter(LHT)family represent a class of proteins that mediate the uptake, translocation, and utilization of amino acids. The tea plant(Camellia sinensis)is a perennial evergreen with a relatively high level of amino acids. However, systematic identification and molecular characterization of the LHT gene family has rarely been reported in tea plants. In this study,22 CsLHTs were identified from the‘Shuchazao’genome and classified into two groups. The modeled three-dimensional structure and the conserved domains presented a high similarity among the LHTs proteins. Moreover,it was predicted that a few genes were conserved through the analysis of the physiochemical characters, structures and cis-elements in promoters. The expression patterns in tea plants revealed that CsLHT7 was mainly expressed in the roots, and CsLHT4 and CsLHT11 exhibited relatively high expression in both the roots and leaves. Moreover, the expression of all three genes could be induced by organic nitrogen.Additionally, heterogeneous expression of CsLHT4,CsLHT7 and CsLHT11 in Arabidopsis thaliana decreased the aerial parts biomass compared with that in WT plants while significantly increased the rosette biomass only for CsLHT11transgenic plants versus WT plants.Overall, our results provide fundamental information about CsLHTs and potential genes in N utilization for further analysis in tea plants. | Wei Huang Danni Ma Fawad Zaman Xulei Hao Li Xia E Zhang Pu Wang Mingle Wang Fei Guo Yu Wang Dejiang Ni Hua Zhao | 2024 | Horticultural Plant Journal2024,10,1: | 0
