| 1 | Genome-and transcriptome-wide association studies provide insights into the genetic basis of natural variation of seed oil content in Brassica napus显示文摘Seed oil content(SOC)is a highly important and complex trait in oil crops.Here,we decipher the genetic basis of natural variation in SOC of Brassica napus by genome-and transcriptome-wide association studies using 505 inbred lines.We mapped reliable quantitative trait loci(QTLs)that control SOC in eight environments,evaluated the effect of each QTL on SOC,and analyzed selection in QTL regions during breeding.Six-hundred and ninety-two genes and four gene modules significantly associated with SOC were identified by analyzing population transcriptomes from seeds.A gene prioritization framework,POCKET(prioritizing the candidate genes by incorporating information on knowledge-based gene sets,effects of variants,genome-wide association studies,and transcriptome-wide association studies),was implemented to determine the causal genes in the QTL regions based on multi-omic datasets.A pair of homologous genes,BnPMT6s,in two QTLs were identified and experimentally demonstrated to negatively regulate SOC.This study provides rich genetic resources for improving SOC and valuable insights toward understanding the complex machinery that directs oil accumulation in the seeds of B.napus and other oil crops. | Shan Tang Hu Zhao Shaoping Lu Liangqian Yu Guofang Zhang Yuting Zhang Qing-Yong Yang Yongming Zhou Xuemin Wang Wei Ma Weibo Xie Liang Guo | 2021 | Molecular Plant2021,14,3: | 6 |

| 4 | Association of plasminogen activator inhibitor-1 4G/5G promoter polymorphism with recurrent cerebral infarction in China’s North Jiangsu Province显示文摘BACKGROUND: Many international studies have shown that plasminogen activator inhibitor-1 (PAI-1) 4G/5G promoter polymorphism does not increase the risk for cerebral infarction. OBJECTIVE: Using PCR methodology and agarose electrophoresis to detect PAI-1 4G/5G promoter polymorphism in patients with recurrent cerebral infarction in the North Jiangsu Province of China, and to compare results with healthy subjects and patients with first-occurrence cerebral infarction in the same region. DESIGN, TIME AND SETTING: Non-randomized, concurrent, control trial. A total of 122 cerebral infarction patients were admitted to Xuzhou Medical College Hospital's Department of Neurology and Xuzhou Central Hospital’s Department of Neurology between July 2003 and August 2006. PARTICIPANTS: The patients consisted of 63 males and 59 females, aged (62 ± 10) years. They were divided into first-occurrence (n = 58) and recurrence (n = 64) groups. In addition, 50 healthy subjects that underwent physical examination in the outpatient department, including 26 males and 24 females, aged (60 ±12) years, were selected as controls. METHODS AND MAIN OUTCOME MEASURES: PAI-1 4G/5G promoter polymorphism was detected and analyzed using PCR methodology and agarose electrophoresis. RESULTS: Significant differences were determined in terms of genotypic frequency and allele frequency of PAI-1 4G/5G promoter polymorphism, in patients with first-occurrence or recurrent cerebral infarction, when compared with healthy subjects (P < 0.05). There was, however, no significant difference between the first-occurrence and recurrence groups (P > 0.05). CONCLUSION: PAI-1 4G/5G promoter polymorphism is genetic risk factor for cerebral infarction in China. | 5 | Vascular Endothelial Growth Factor‑Recruiting Nanofiber Bandages Promote Multifunctional Skin Regeneration via Improved Angiogenesis and Immunomodulation显示文摘Tissue injury leads to gradients of chemoattractants,which drive multiple processes for tissue repair,including the inflam-matory response as well as endogenous cell recruitment.However,a limited time window for the gradients of chemoattract-ants as well as their poor stability at the injury site may not translate into healthy tissue repair.Consequently,intelligent multifunctional scaffolds with the capability to stabilize injury-induced cytokines and chemokines hold great promise for tissue repair.Vascular endothelial growth factor(VEGF)plays a significant role in wound healing by promoting angiogen-esis.The overarching objective of this research was to develop intelligent multifunctional scaffolds with the capability to endogenously recruit VEGF and promote wound healing via angiogenic and immunomodulatory dual functions.Prominin-1-derived peptide(PR1P)was encapsulated into electrospun poly(L-lactide-coglycolide)/gelatin(P/G)-based bandages.The sustained release of PR1P recruited VEGF in situ,thereby stabilizing the protein concentration peak in vivo and affording a reparative microenvironment with an adequate angiogenic ability at the wound site.Meanwhile,PR1P-recruited VEGF-induced macrophage reprogramming towards M2-like phenotypes further conferred immunomodulatory functions to the bandages.These dual functions of proangiogenesis and immunomodulation formed a cascade amplification,which regulated matrix metalloproteinases(MMP-9)as well as inflammatory factors(nuclear factor(NF)-κb,tumor necrosis factor(TNF)-α)in the wound microenvironment via the VEGF/macrophages/microenvironment axis.Consequently,the bandages realized multifunctional regeneration in splinted excisional wounds in rats,with or without diabetes,affording a higher skin append-age neogenesis,sensory function,and collagen remodeling.Conclusively,our approach encompassing in situ recruitment of VEGF at the injury site with the capability to promote immunomodulation-mediated tissue repair affords a promising avenue for scarless wound regeneration,which may also have implications for other tissue engineering disciplines. | Yi Chen Zhengchao Yuan Weiyan Sun Muhammad Shafiq Jun Zhu Jiafei Chen Hai Tang Ling Hu Weikang Lin Yanxi Zeng Long Wang Lei Zhang Yunlang She Hui Zheng Guofang Zhao Dong Xie Xiumei Mo Chang Chen | 2023 | Advanced Fiber Materials2023,5,1: | 1 |

| 9 | Place-pitch perception in electrical hearing with post-lingually deafened cochlear implant users显示文摘The main goal of this study was to systematically investigate place-pitch perception in electrical hearing and the relative relationship between place-pitch perception ability,speech understanding and musical pitch discrimination by cochlear implant(CI) users.Electrode pitch ranking test was carried out to evaluate the place-pitch perception ability of CI users. | 10 | A peptide chain release factor 2a gene regulates maize kernel development by modulating mitochondrial function显示文摘Mitochondrial protein translation that is essential for aerobic energy production includes four essential steps of the mitochondrial ribosome cycle,namely,initiation,elongation,termination of the polypeptide,and ribosome recycling.Translation termination initiates when a stop codon enters the A site of the mitochondrial ribosome where it is recognized by a dedicated peptide release factor(RF).However,RFs and mechanisms involved in translation in plant mitochondria,especially in monocotyledons,remain largely unknown.Here,we identified a crumpled kernel(crk5 allele)mutant,with significantly decreased kernel size,100-kernel weight,and an embryo-lethal phenotype.The Crk5 allele was isolated using map-based cloning and found to encode a mitochondrial localization RF2a.As it is an ortholog of Arabidopsis mitochondrial RF2a,we named the gene ZmmtRF2a.ZmmtRF2a is missing the 5th–7th exons in the crk5 resulting in deletion of domains containing motifs GGQ and SPF that are essential for release activity of RF,mitochondrial ribosome binding,and stop codon recognition.Western blot and qRT-PCR analyses indicate that the crk5 mutation results in abnormal mitochondrion structure and function.Intriguingly,we observed a feedback loop in the crk5 with up-regulated transcript levels detected for several mitochondrial ribosome and mitochondrial-related components,in particular mitochondrial complexes CI,CIV,and a ribosome assembly related PPR.Together,our data support a crucial role for ZmmtRF2a in regulation of mitochondrial structure and function in maize. | Hui Zhang Yijian Feng Kunyang Song Guofang Li Jiao Jin Jingjing Gao Yongtian Qin Hongqiu Wang Jinpeng Cheng Zonghua Liu Jihua Tang Zhiyuan Fu | 2023 | The Crop Journal2023,11,6: | 0 |

| 12 | A novel nomogram based on routine clinical indicators for screening for Wilson's disease显示文摘Background and aims:There is currently no single model for predicting Wilson's disease(WD).We aimed to create a nomogram using daily clinical parameters to improve the accuracy of WD diagnosis in patients with abnormal liver function.Methods:Between July 2016 and December 2020,we identified 90 WD patients with abnormal liver function who had homozygous or compound heterozygous mutations in the ATP7B gene.The control group included 128 patients with similar liver function but no WD during the same time period.To create a nomogram,we screened potential predictive variables using the least absolute shrinkage and selection operator model and multivariate logistic regression.Results:We developed a nomogram for screening for WD based on six predictive factors:serum copper,direct bilirubin,uric acid,cholinesterase,prealbumin,and reticulocyte percentage.In the training cohort,the area under curve(AUC)of the nomogram reached 0.967(95%confidence interval(CI)0.946e0.988),while the area under the precision-recall curve was 0.961.Based on the optimal cutpoint of 213.55,our nomogram performed well,with a sensitivity of 96%and a specificity of 87%.In the validation cohort,the AUC of the nomogram was as high as 0.991(95%CI 0.970e1.000).Conclusions:We developed a nomogram that can predict the risk of WD prior to the detection of serum ceruloplasmin or urinary copper,greatly increasing screening efficiency for patients with abnormal liver function. | Jiahui Pang Shuru Chen Weiqiang Gan Guofang Tang Yusheng Jie Zhanyi Li Yutian Chong Youming Chen Jiao Gong Xinhua Li Yongyu Mei | 2023 | Liver Research2023,7,1: | 0 |
