1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2High-frequency MnZn soft magnetic ferrite by engineering grain boundaries with multiple-ion doping显示文摘MnZn soft magnetic ferrites have been widely utilized in power electronics,owing to the combined merits of high permeability and low energy loss.However,their deployment would result in a drastic increase in power dissipation at>3 MHz,thus limiting the scope extent of miniaturization,together with their efficiency.Here,we report a high-performance MnZn ferrite by doping multiple ions(La,Ti,Si,Ca)at grain boundaries,achieving the most optimized power loss of 267 kW/m^(3) at 5 MHz(10 m T,100℃)and initial permeability of 644,which is much better than the previously reported results and commercial products.Such an improvement is attributed to weakened magnetic exchange coupling at grain-boundary regions,associated with a significant transition from the multi-to mono-domain structures,originating physically from large crystallographic mis-orientations(>25°).The present study bears important significance in understanding the intrinsic correlation between the crystallographic mis-orientation and magnetic domain structure,and provides an alternative way for optimizing high-frequency soft magnetic ferrites.Mi Yan Shengbo Yi Xiuyuan Fan Zhenghua Zhang Jiaying Jin Guohua Bai 2021Journal of Materials Science & Technology2021,,20:2

4Paclitaxel liposome for injection (Lipusu) plus cisplatin versus gemcitabine plus cisplatin in the first-line treatment of locally advanced or metastatic lung squamous cell carcinoma: A multicenter, randomized, open-label, parallel controlled clinical study显示文摘Background:Lipusu is the first commercialized liposomal formulation of pacli-taxel and has demonstrated promising efficacy against locally advanced lung squamous cell carcinoma(LSCC)in a small-scale study.Here,we conducted a multicenter,randomized,phase 3 study to compare the efficacy and safety of cis-platin plus Lipusu(LP)versus cisplatin plus gemcitabine(GP)as first-line treat-ment in locally advanced or metastatic LSCC.Methods:Patients enrolled were aged between 18 to 75 years,had locally advanced(clinical stage IIIB,ineligible for concurrent chemoradiation or surgery)or metastatic(Stage IV)LSCC,had no previous systemic chemother-apy and at least one measurable lesion as per the Response Evaluation Criteria in Solid Tumors(version 1.1)before administration of the trial drug.The primary endpoint was progression-free survival(PFS).The secondary endpoints included objective response rate(ORR),disease control rate(DCR),overall survival(OS),and safety profiles.To explore the possible predictive value of plasma cytokines for LP treatment,plasma samples were collected from the LP group at baseline and first efficacy evaluation time and were then subjected to analysis by 45-Plex ProcartaPlex Panel 1 to detect the presence of 45 cytokines using the Luminex xMAP technology.The correlation between treatment outcomes and dynamic changes in the levels of cytokines were evaluated in preliminary analyses.Results:The median duration of follow-up was 15.4 months.237 patients in the LP group and 253 patients in the GP group were included in the per protocol set(PPS).In the PPS,the median PFS was 5.2 months versus 5.5 months in the LP and GP group(hazard rtio[HR]:1.03,P=0.742)respectively.The median OS was 14.6 months versus 12.5 months in the LP and GP group(HR:0.83,P=0.215).The ORR(41.8%versus 45.9%,P=0.412)and DCR(90.3%versus 88.1%,P=0.443)were also similar between the LP and GP group.A significantly lower proportion of patients in the LP group experienced adverse events(AEs)leading to treatment interruptions(10.9%versus 26.4%,P<0.001)or treatment termination(14.3%versus 23.1%,P=0.011).The analysis of cytokine levels in the LP group showed that low baseline levels of 27 cytokines were associated with an increased ORR,and 15 cytokines were associated with improved PFS,with 14 cytokines,including TNF-a,IFN-y,IL-6,and IL-8,demonstrating an overlapping trend.Conclusion:The LP regimen demonstrated similar PFS,OS,ORR and DCR as the GP regimen for patients with locally advanced or metastatic LSCC but had more favorable toxicity profiles.The study also identified a spectrum of different cytokines that could be potentially associated with the clinical benefit in patients who received the LP regimen.Jie Zhang Yueyin Pan Qin Shi Guojun Zhang Liyan Jiang Xiaorong Dong Kangsheng Gu Huijuan Wang Xiaochun Zhang Nong Yang Yuping Li Jianping Xiong Tienan Yi Min Peng Yong Song Yun Fan Jiuwei Cui Gongyan Chen Wei Tan Aimin Zang Qisen Guo Guangqiang Zhao Ziping Wang Jianxing He Wenxiu Yao Xiaohong Wu Kai Chen Xiaohua Hu Chunhong Hu Lu Yue Da Jiang Guangfa Wang Junfeng Liu Guohua Yu Junling Li Jianling Bai Wenmin Xie Weihong Zhao Lihong Wu Caicun Zhou 2022Cancer Communications2022,42,1:1

8Diel dynamics of multi-omics in elkhorn fern provide new insights into weak CAM photosynthesis显示文摘Crassulacean acid metabolism(CAM)has high water-use efficiency(WUE)and is widely recognized to have evolved from C3 photosynthesis.Different plant lineages have convergently evolved CAM,but the molecular mechanism that underlies C3-to-CAM evolution remains to be clarified.Platycerium bifurcatum(elkhorn fern)provides an opportunity to study the molecular changes underlying the transition from C3 to CAM photosynthesis because both modes of photosynthesis occur in this species,with sporotrophophyll leaves(SLs)and cover leaves(CLs)performing C3 and weak CAM photosynthesis,respectively.Here,we report that the physiological and biochemical attributes of CAM in weak CAM-performing CLs differed from those in strong CAM species.We investigated the diel dynamics of the metabolome,proteome,and transcriptome in these dimorphic leaves within the same genetic background and under identical environmental conditions.We found that multi-omic diel dynamics in P.bifurcatum exhibit both tissue and diel effects.Our analysis revealed temporal rewiring of biochemistry relevant to the energy-producing pathway(TCA cycle),CAM pathway,and stomatal movement in CLs compared with SLs.We also confirmed that PHOSPHOENOLPYRUVATE CARBOXYLASE KINASE(PPCK)exhibits convergence in gene expression among highly divergent CAM lineages.Gene regulatory network analysis identified candidate transcription factors regulating the CAM pathway and stomatal movement.Taken together,our results provide new insights into weak CAM photosynthesis and new avenues for CAM bioengineering.Cheng Li Wenjie Huang Xiaoxu Han Guohua Zhao Wenyang Zhang Weijun He Bao Nie Xufeng Chen Taijie Zhang Wenhui Bai Xiaopeng Zhang Jingjing He Cheng Zhao Alisdair RFernie Timothy JTschaplinski Xiaohan Yang Shijuan Yan Li Wang 2023Plant Communications2023,4,5:0

11Wood forming tissue-specific expression of PdSuSy and HCHL increases holocellulose content and improves saccharification in Populus显示文摘Development of strategies to deconstruct lignocellulosic biomass in tree species is essential for biofuels and biomaterials production.We applied a wood forming tissue-specific system in a hybrid poplar to express both PdSuSy(a sucrose synthase gene from Populus deltoides×P.euramericana that has not been functionally characterized)and HCHL(the hydroxycinnamoyl-CoA hydratase-lyase gene from Pseudomonas fluorescens,which inhibits lignin polymerization in Arabidopsis).The PdSuSy-HCHL overexpression poplars correspondingly driven by the promoters of Arabidopsis AtCesA7 and AtC4 H resulted in a significant increase in cellulose(>8%),xylan(>12%)and glucose(>29%)content,accompanying a reduction in galacturonic acid(>36%)content,compared to control plants.The saccharification efficiency of these overexpression poplars was dramatically increased by up to 27%,but total lignin content was unaffected.These transgenic poplars showed inhibited growth characteristics,including>16%reduced plant height,>10% reduced number of internodes,and>18% reduced fresh weight after growth of 4 months,possibly due to relatively low expression of HCHL in secondary xylem.Our results demonstrate the structural complexity and interaction of the cell wall polymers in wood tissue and outline a potential method to increase biomass saccharification in woody species.Yang Zhang Hua Xu Yingzhen Kong Jiawen Hua Xianfeng Tang Yamei Zhuang Yue Bai Gongke Zhou Guohua Chai 2021Journal of Forestry Research2021,32,4:0
12Alterations of gene expression profiles induced by sulfur dioxide in rat lungs显示文摘Sulfur dioxide(SO_(2))is a ubiquitous air pollutant,presents in low concentrations in urban air and in higher concentrations in working environment.Few data are avail-able on the effects of being exposed to this pollutant on the molecular mechanism,although some biochemical changes in lipid metabolism,intermediary metabolism and oxidative stress have been detected.The present investigation aimed at analyzing the gene expression profiles of the lungs of Wistar rats short-term(20 ppm,6 h/day,for seven days)and long-term(5 ppm,1 h/day,for 30 days)exposed to SO_(2)by Affymetrix GeneChip(RAE230A)analysis.It was found that 31 genes,containing 18 known genes and 13 novel genes,were up-regulated,and 31 genes,containing 20 known genes and 11 novel genes,were down-regulated in rats short-term exposed to SO_(2)compared with control rats.While there were 176 genes,containing 82 known genes and 94 novel genes,were up-regulated,and 85 genes,containing 46 known genes and 39 novel genes,were down-regulated in rats long-term exposed to SO_(2)compared with control rats.It is suggested that:(1)SO_(2)exerts its effects by different mechanisms in vivo at high-dose short-term inhalation and at low-dose long-term inhalation;(2)a notable feature of the gene expression profile was the decreased expression of genes related to oxidative phosphorylation in lungs of rats short-term exposed to SO_(2),which shows high-dose short-term exposed to SO_(2)may cause the deterioration of mitochondrial functions;(3)discriminating genes in lungs of rats long-term exposed to SO_(2)included those involved in fatty acid metabolism,immune,inflammatory,oxidative stress,oncogene,tumor suppresser and extracellular matrix.The mechanism of low-dose long-term exposed to SO_(2)is more complex.MENG Ziqiang QIN Guohua BAI Juli ZHANG Jianbiao ZHANG Xin YANG Zhenghua 2007Frontiers in Biology2007,2,4:0

14Synergistic effect of V_(2)O_(5)and Bi_(2)O_(3) on the grain boundary structure of high-frequency NiCuZn ferrite ceramics显示文摘High-frequency soft magnetic ferrite ceramics are desired in miniaturized and efficient power electronics but remain extremely challenging to deploy on account of the power loss(Pcv)at megahertz frequencies.Here,we prepared NiCuZn ferrite with superior high-frequency properties by V_(2)O_(5)and Bi_(2)O_(3)synergistic doping,which proves to be a potent pathway to reduce Pcv of the ferrite at megahertz frequencies.The sample doped with 800 ppm V_(2)O_(5)and 800 ppm Bi_(2)O_(3)yielded the most optimized magnetic properties with a Pcv of 113 kW/m^(3)(10 MHz,5 mT,25℃),an initial permeability(μi)of 89,and a saturation induction(Bs)of 340 mT,which is at the forefront of the reported results.These outstanding properties are closely related to the notable grain boundary structure,which features a new type of nano-Bi_(2)Fe_(4)O_(9)phase around ferrite grains and a Ca/Si/V/O amorphous layer.Our results indicate great strides in correlating the grain boundary structure with multiple-ion doping and set the scene for the developing high-frequency soft magnet ferrites.Xiuyuan FAN Guohua BAI Zhenhua ZHANG Qiming CHEN Jiaying JINa Jiafeng XUa Xuefeng ZHANG Mi YAN 2022Journal of Advanced Ceramics2022,11,6:0
