| 2 | Nascent Proteome and Glycoproteome Reveal the Inhibition Role of ALG1 in Hepatocellular Carcinoma Cell Migration显示文摘Asparagine-linked glycosylation protein 1 homolog(ALG1)participates in the initial stage of protein N-glycosylation and N-glycosylation has been implicated in the process of hepatocellular carcinoma(HCC)progression.However,whether ALG1 plays a role in human HCC remains unknown.In this study,the expression profile of ALG1 in tumorous and corresponding adjacent non-tumor tissues was analyzed.The relationship of ALG1 expression with clinical features and prognosis of HCC patients was also evaluated using immuno-histochemical method.Here we found ALG1 decreased in HCC tissues compared with adjacent normal liver tissues,which predicted an unfavorable prognosis.Combined with RNA interference,nascent proteome and glycoproteome were determined systematically in Huh7 cell line.Bioinformatics analysis indicated that the differentially expressed proteins participating in the response of ALG1 knockdown were most significantly associated with cell-cell adhesion.Functional studies confirmed that knockdown of ALG1 reduced cell adhesion capacity,and promoted cell migration.Furthermore,down-regulation of H8N2(on N-glycosite N651)and H5N4S2F1(on N-glycosite N692)from N-cadherin was identified as a feature of ALG1 knockdown.Our findings revealed that ALG1 controlled the expression of glycosylated N-cadherin and played a role in HCC migration,with implications for prognosis. | Xinyi Cao Yuyin Shao Peiyi Meng Zhao Cao Guoquan Yan Jun Yao Xinwen Zhou Chao Liu Lei Zhang Hong Shu Haojie Lu | 2022 | Phenomics2022,2,4: | 2 |

| 7 | Response of peptide intensity to concentration in ESI–MS-based proteome显示文摘High-throughput quantification with label-free methods has received considerable attention in electrospray ionization(ESI)-mass spectrometry(MS),but the manner by which MS signals respond to peptide concentration remains unclear in proteomics.We developed a new mathematical formula to describe the intrinsic log-log relationship between the MS intensity response and peptide concentration in an analytical ESI process.Experimental results showed that the calibration curve is fairly fit to the log-log formula with a linear dynamic range of approximate four to five orders of magnitude.However,we found that the ionization of analytical peptides can be severely suppressed by coexisting matrix peptides,such that the calibration curve can be poorly leveled off on both ends.Our study suggests that the interferences from coexisting matrix peptides should be reduced in the ESI process to use the log-log calibration curve successfully for the high-throughput quantification. | LU WenYuan YIN XueFei LIU XiaoHui YAN GuoQuan YANG PengYuan | 2014 | Science China Chemistry2014,57,5: | 1 |

| 11 | Novel high-PSRR high-order curvature-compensated bandgap voltage reference显示文摘This paper proposes a novel high-power supply rejection ratio(high-PSRR) high-order curvature-compensated CMOS bandgap voltage reference(BGR) in SMIC 0.18 μm CMOS process. Three kinds of current are added to a conventional BGR in order to improve the temperature drift within wider temperature range, which include a piecewise-curvaturecorrected current in high temperature range, a piecewise-curvature-corrected current in low temperature range and a proportional-to-absolute-temperature T^(1.5) current. The high-PSRR characteristic of the proposed BGR is achieved by adopting the technique of pre-regulator. Simulation results shows that the temperature coefficient of the proposed BGR with pre-regulator is 8.42x10^(-6)′ /℃ from - 55 ℃ to 125 ℃ with a 1.8 V power supply voltage. | 15 | Proteomics analysis provides novel biomarkers and therapeutic target candidates in the treatment of the Huang-Pu-Tong-Qiao formula in an AD rat mode显示文摘Background:Huang-Pu-Tong-Qiao formula(HPTQ),a traditional Chinese herbal formula,has a variety of pharmacological effects.It has been used to treat Alzheimer’s disease(AD)for decades.This study aimed to screen differentially expressed proteins in the hippocampus of AD model rats treated with HPTQ.Proteomic studies of the effects of HPTQ on AD are key to understanding the therapeutic mechanisms of HPTQ and identifying potential therapeutic targets.Methods:We hence used the isobaric tags for relative and absolute quantification(ITRAQ)approach to investigate the differentially expressed proteins in the hippocampus of AD model rats before and after HPTQ administration and to identify the potential therapeutic target proteins of HPTQ.In this study,the learning and memory abilities of AD rats were examined by the Morris water maze test.After HPTQ administration,the differentially expressed proteins in the hippocampus of AD rats were quantified and analyzed in silico.Furthermore,western blotting was used to verify the expression of related proteins.Results:The Morris water maze results showed that HPTQ could improve the learning and memory ability of AD model rats.The proteomics analysis results showed that 57 proteins were differentially expressed,of which 35 were up-regulated and 22 were down-regulated.Bioinformatics analysis indicated that proteins with altered expression after HPTQ treatment were involved in several biological processes that have the potential to exert neuroprotective effects.These included promoting the translation of ribosomes,improving the deposition of amyloid-beta(Aβ),regulating autophagy,regulating neuronal synaptic function and plasticity,and alleviating oxidative stress.Conclusion:In conclusion,we identified several potential therapeutic target proteins and related mechanistic pathways of HPTQ in the treatment of AD,laying the foundation for further investigation of the therapeutic effects of HPTQ. | QIAN CHEN XIN LEI GUANHUA HU YAN WANG ZHENGQING FANG GUOQUAN WANG HANG SONG SHU YE BIAO CAI | 2023 | BIOCELL2023,47,6: | 0 |
| 16 | Physiological and Biochemical Characteristics and Response Patterns of Salinity Stress Responsive Genes (SSRGs) in Wild Quinoa (Chenopodium quinoa L.)显示文摘Cultivating salt-tolerant crops is a feasible way to effectively utilize saline-alkali land and solve the problem of underutilization of saline soils.Quinoa,a protein-comprehensive cereal in the plant kingdom,is an exceptional crop in terms of salt stress tolerance level.It seems an excellent model for the exploration of salt-tolerance mechanisms and cultivation of salt-tolerant germplasms.In this study,the seeds and seedlings of the quinoa cultivar Shelly were treated with different concentrations of NaCl solution.The physiological,biochemical characteristics and agronomic traits were investigated,and the response patterns of three salt stress-responsive genes(SSRGs)in quinoa were determined by real-time PCR.The optimum level of stress tolerance of quinoa cultivar Shelly was found in the range of 250–350 mM concentration of NaCl.Salt stress significantly induced expression of superoxide dismutase(SOD),peroxidase(POD),and particularly betaine aldehyde dehydrogenase(BADH).BADH was discovered to be more sensitive to salt stress and played an important role in the salt stress tolerance of quinoa seedlings,particularly at high NaCl concentrations,as it displayed upregulation until 24 h under 100 mM salt treatment.Moreover,it showed upregulation until 12 h under 250 mM salt stress.Taken together,these results suggest that BADH played an essential role in the salt-tolerance mechanism of quinoa.Based on the expression level and prompt response induced by NaCl,we suggest that the BADH can be considered as a molecular marker for screening salt-tolerant quinoa germplasm at the early stages of crop development.Salt treatment at different plant ontogeny or at different concentrations had a significant impact on quinoa growth.Therefore,an appropriate treatment approach needs to be chosen rationally in the process of screening salt-tolerant quinoa germplasm,which is useful to the utilization of saline soils.Our study provides a fundamental information to deepen knowledge of the salt tolerance mechanism of quinoa for the development of salt-tolerant germplasm in crop breeding programs. | Yurong Jiang Muhammad Yasir Yuefen Cao Lejia Hu Tongli Yan Shuijin Zhu Guoquan Lu | 2023 | Phyton-International Journal of Experimental Botany2023,92,2: | 0 |
