1Clinical significance of K-ras and BRAF mutations in Chinese colorectal cancer patients显示文摘AIM:To identify and assess mutations in the K-ras and BRAF genes in a cohort of Chinese patients with colorectal cancer (CRC) for their association with various clinicopathological parameters and prognosis.METHODS:Genomic DNA was isolated from frozen tissues.Pyrosequencing analysis was conducted to detect mutations in the K-ras (codons 12,13,and 61) and BRAF genes (codon 600).Statistical analysis was carried out using SPSS-15.0 software.RESULTS:Among the 118 colorectal cancer patients,we detected 41 (34.7%) mutations in the K-ras gene.Mutation frequencies at codon 12 and codon 13 were 23.7% (28/118) and 10.2% (12/118),respectively.Only one patient harbored a point mutation at codon 61 (0.8%,1/118).Gender was the only factor that showed an obvious relationship with K-ras gene mutation (female 44.7% vs male 28.2%,P=0.037).Other clinicopathological features,such as age,location of the tumor,tumor differentiation,Tumor,Node and Metastases classification,and the Union for International Cancer Control staging,showed no positive relationship with K-ras gene mutations.No significant correlation was observed between the presence of K-ras mutations (codons 12,13,and 61) and the survival of the patients.BRAF mutations were rare,and only two patients (1.7%) harbored a detectable mutation at codon 600.CONCLUSION:K-ras gene mutation is a common event in our 118 Chinese CRC patients,with an obvious relationship with gender.However,it seems not to be an independent prognostic factor in CRC patients.The BRAF gene is rarely mutated in Chinese CRC patients.Hong Shen Ying Yuan Han-Guang Hu Xiao-Xian Ye Mo- Dan Li Xian Zhong Wei-Jia Fang Shu Zheng 2011World Journal of Gastroenterology2011,17,6:29
2Furazolidone-based triple and quadruple eradication therapy for Helicobacter pylori infection显示文摘AIM: To evaluate the efficacy of furazolidone-based triple and quadruple therapy in eradicating Helicobacter pylori(H. pylori) in a multi-center randomized controlled trial.METHODS: A total of 720 H. pylori positive patients with duodenal ulcer disease were enrolled at 10 different hospitals in Jiangxi province in China. The patients were randomly assigned to four treatment groups as follows: patients in Groups 1 and 3 received rabeprazole(10 mg), amoxicillin(1000 mg) and furazolidone(100 mg) twice daily for 7 and 10 d, respectively; patients in Groups 2 and 4 received rabeprazole(10 mg), bismuth(220 mg), amoxicillin(1000 mg) and furazolidone(100 mg) twice daily for 7 and 10 d, respectively. The primary outcome measure was H. pylori eradication rate 4 wk after treatment by intention-to-treat and per protocol analysis, while the secondary outcome measures were symptom and sign changes at the end of treatment and 4 wk after the end of treatment, as well as the proportion of patients who developed adverse events.RESULTS: The demographic data of the four groups were not significantly different. Overall, 666 patients completed the scheme and were re-assessed with the 13C-urea breath test. The intention-to-treat analysis of the H. pylori eradication rates in Groups 1, 2, 3 and 4 were 74.44%, 82.78%, 78.89% and 86.11%, respectively. The H. pylori eradication rate in Group 4 was significantly higher than that in Group 1. According tothe per protocol analysis, the H. pylori eradication rates in Groups 1, 2, 3 and 4 were 81.21%, 89.22%, 85.54% and 92.26%, respectively. The H. pylori eradication rate in Group 4 was significantly higher than that in Group 1. The number of adverse events was 15(8.3%), 16(8.9%), 15(8.3%) and 17(9.4%) in Groups 1, 2, 3 and 4, respectively, including dizziness, vomiting, diarrhea, nausea, skin rash, itchy skin, and malaise. 4Large Scale Photochemical Synthesis of M@TiO_(2) Nanocomposites (M = Ag, Pd, Au, Pt) and Their Optical Properties, CO Oxidation Performance, and Antibacterial Effect显示文摘Well-dispersed M@TiO_(2)(M=Ag,Pd,Au,Pt)nanocomposite particles with a diameter of 200-400 nm can be synthesized on a large scale by a clean photochemical route which does not require any additives using spherical rutile nanoparticles as a support.The sizes of Pt,Au,and Pd nanoparticles formed on the surface of TiO_(2) particles are about 1 nm,5 nm,and 5 nm,respectively,and the diameter of Ag nanoparticles is in the range 2-20 nm.Moreover,the noble metal nanoparticles have good dispersity on the particles of the TiO_(2) support,resulting in excellent catalytic activities.Complete conversion in catalytic CO oxidation is reached at temperatures as low as 333 and 363 K,respectively,for Pt@TiO_(2) and Pd@TiO_(2) catalysts.In addition,the antibacterial effects of the as-synthesized TiO_(2) nanoparticles,silver nanoparticles,and Au@TiO_(2) and Ag@TiO_(2) nanocomposites have been tested against Gram-negative Escherichia coli(E.coli)bacteria.The results demonstrate that the presence of the TiO_(2) matrix enhances the antibacterial effect of silver nanoparticles,and the growth of E.coli can be completely inhibited even if the concentration of Ag in Ag@TiO_(2) nanocomposite is very low(10μg/mL).Shao Feng Chen Jian Ping Li Kun Qian Wei Ping Xu Yang Lu Wei Xin Huang Shu Hong Yu 2010Nano Research2010,3,4:20
5Relative position determination of a lunar rover using high-accuracy multi-frequency same-beam VLBI显示文摘Multi-frequency same-beam VLBI means that two explorers with a small separation angle are simultaneously observed with the main beam of receiving antennas. In the same-beam VLBI, the differential phase delay between two explorers and two receiving telescopes can be obtained with a small error of several picoseconds. The differential phase delay, as the observable of the same-beam VLBI, gives the separation angular information of the two explorers in the celestial sphere. The two-dimensional relative position on the plane-of-sky can thus be precisely determined with an error of less than 1 m for a distance of 3.8×105 km far away from the earth, by using the differential phase delay obtained with the four Chinese VLBI stations. 6Association of Dietary Pattern during Pregnancy and Gestational Diabetes Mellitus: A Prospective Cohort Study in Northern China显示文摘Objective To examine the association of maternal dietary patterns during pregnancy with gestational diabetes mellitus(GDM) in northern China.Methods The dietary intakes of pregnant women were recorded twice by 24‐hour dietary recalls for three days prior to having been diagnosed with GDM, at 5‐15 and 24‐28 gestational weeks, respectively.GDM was diagnosed, and serum glycosylated hemoglobin(HbA1c) was measured at 24‐28 weeks.Dietary patterns were assessed by factor analysis. The association of the dietary pattern with GDM and HbA1c was examined by multiple logistic models.Results Of 753 participants, 64(8.5%) were diagnosed with GDM. Four dietary patterns were identified: Western pattern(dairy, baked/fried food and white meat), traditional pattern(light‐colored vegetables, fine grain, red meat and tubers), mixed pattern(edible fungi, shrimp/shellfish and red meat)and prudent pattern(dark‐colored vegetables and deep‐sea fish). Compared with the prudent pattern,both the Western pattern and the traditional pattern were associated with an increased risk of GDM(a OR = 4.40, 95% CI: 1.58‐12.22; a OR = 4.88, 95% CI: 1.79‐13.32) and a high level of HbA1c(a OR = 12.37,95% CI: 1.47‐103.91; a OR = 26.23, 95% CI: 2.54‐270.74). 7Functional CT for assessment of early vascular physiology in liver tumors显示文摘BACKGROUND:CT perfusion has been reported to have great advantages in detecting hepatic diseases.However, currently there are no studies showing the potential value of multi-slice CT perfusion with the deconvolution model method in diagnosing early hemodynamic changes caused by liver tumors.This study was undertaken to determine if early hemodynamic changes caused by liver tumors can be depicted with the perfusion imaging of multi-slice CT. METHODS:Ten New Zealand white rabbits before and after VX2 liver tumor inoculation served as the experimental animals.Ten normal rabbits served as controls.All underwent multi-slice CT perfusion for the measurement of hepatic blood flow(HBF),hepatic blood volume(HBV), mean transit time(MTT),permeability of capillary vessel surface(PS)and hepatic artery index(HAI). RESULTS:With the exception of MTT,which decreased significantly at the tumor periphery,HBF,HBV,PS and HAI increased significantly compared with the surrounding normal tissue.All these changes occurred at days 5-9 after tumor inoculation.Statistically significant changes in these values were detected with tumor growth. 8Relationship of Inflammation and Endothelial Dysfunction with Risks to Cardiovascular Disease among People in Inner Mongolia of China显示文摘Objective To explore the relationship of inflammation and endothelial dysfunction with risks to cardiovascular disease(CVD).Methods Blood pressure,body weight,body height,waist circumference and lifestyle risk factors were measured and studied among 2589 participants in Inner Mongolia of China,and biomarkers of inflammation and endothelial dysfunction including high-sensitivity C-reactive protein(hsCRP),soluble inter-cellular adhesion molecule-1(sICAM-1),soluble E-selectin(sE-selectin),and angiotensin II were investigated.Results Subjects with metabolic risk factors for CVD had higher levels of hsCRP,sE-selectin and sICAM-1 than those without such risk factors(all P<0.05).Levels of all biomarkers positively and significantly increased with aggregation of the metabolic risk factors among the subjects(all P for trend<0.001).Data from the multivariate analysis showed that participants with high levels of hsCRP [odds ratio(OR): 1.96,95% confidence interval(CI): 1.52-2.53],sE-selectin(OR: 1.35,95% CI: 1.05-1.72),and angiotensin II(OR: 1.81,95% CI: 1.40-2.33) were more likely to develop hypertension; participants with high levels of hsCRP(OR: 2.33,95% CI: 1.85-2.94),sE-selectin(OR: 1.24,95% CI: 1.00-1.54),and sICAM-1(OR: 1.70,95% CI: 1.30-2.22) were more likely to develop dyslipidemia,and those with high levels of hsCRP(OR: 2.95,95% CI: 2.27-3.83) and sICAM-1(OR: 2.80,95% CI: 2.06-3.80) were more likely to develop hyperglycemia.Conclusion Biomarkers of inflammation and endothelial dysfunction were separately associated with relevant metabolic risk factors for CVD.And appropriate measures should be taken to control inflammation and improve endothelial function among individuals with different metabolic risk factors for CVD.PENG Hao HAN Shu Hai LIU Hai Ying Vasisht CHANDNI CAI Xiao Qing ZHANG Yong Hong 2013Biomedical and Environmental Sciences2013,26,10:15
9The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 10Induction, modulation and potential targets of miR-210 in pancreatic cancer cells显示文摘BACKGROUND: MiR-210 is induced by hypoxia and plays different roles in the development of certain cancers. However, little is known about its role in pancreatic cancer (PC). This study aimed to explore the induction and modulation of PC by miR-210 and its potential molecular targets. METHODS: PC cells were cultured under normoxic and hypoxic conditions. Expression of miR-210 and hypoxia-inducible factor (HIF)-1α was detected using quantitative reverse-transcription polymerase chain reaction. Cancer cells were transiently transfected with HIF-1α small interfering RNA (siRNA) and miR-210 mimics, and cell proliferation was measured using the CCK-8 assay. Potential targets for miR-210 were then identified using a dual luciferase reporter assay. RESULTS: Hypoxic conditions induced miR-210 expression in six PC cell lines (AsPC-1, BxPC-3, MIAPaCa-2, PANC-1, Su86.86 and SW1990), but not in Capan-1 or T3M4 cells. Transfection of HIF-1α siRNA into PANC-1 cells markedly inhibited HIF-1α expression, and subsequently down- regulated miR-210 expression under hypoxic conditions. MiR-210 had no observable impact on the proliferation of PANC-1 or Su86.86 cells and dual luciferase reporter assays showed significantly reduced luciferase activity in the wild- type E2F3, EFNA3, GIT2, MNT, ZNF462 and EGR3 constructs, compared to the corresponding mutants, but not in HOXA3. CONCLUSIONS: These results suggest that miR-210 expression in PC cells is induced by hypoxia through a HIF-1α-dependent pathway, but does not influence PC cell proliferation. 11Progress of regional oceanography study associated with western boundary current in the South China Sea显示文摘Recent progress of physical oceanography in the South China Sea (SCS) associated with the western boundary current (WBC) and eddies is reviewed in this paper. It includes Argo observations of the WBC, eddy detection in the WBC based on satellite images, cross-continental shelf exchange in the WBC, eddy-current interaction, interannual variability of the WBC, air-sea interaction, the SCS throughflow (SCSTF), among others. The WBC in the SCS is strong, and its structure, variability and dynamic processes on seasonal and interannual time scales are yet to be fully understood. In this paper, we summarize progresses on the variability of the WBC, eddy-current interaction, air-sea interaction, and the SCSTF achieved in the past few years. Firstly, using the drifting buoy observations, we point out that the WBC becomes stronger and narrower after it reaches the central Vietnam coast. The possible mechanisms influencing the ocean circulation in the northern SCS are discussed, and the dynamic mechanisms that induce the countercurrent in the region of northern branch of WBC in winter are also studied quantitatively using momentum balance. The geostropic component of the WBC was diagnosed using the ship observation along 18°N, and we found that the WBC changed significantly on interannual time scale. Secondly, using the ship observations, two anti-cyclonic eddies in the winter of 2003/2004 in the northern SCS, and three anti-cyclonic eddies in the summer of 2007 along 18°N were studied. The results show that the two anti-cyclonic eddies can propagate southwestward along the continental shelf at the speed of first Rossby wave (~0.1 ms-1 ) in winter, and the interaction between the three anti-cyclonic eddies in summer and the WBC in the SCS is preliminarily revealed. Eddies on the continental shelf of northern SCS propagated southeastward with a maximum speed of 0.09 ms-1 , and those to the east of Vietnam coast had the largest kinetic energy, both of which imply strong interaction between eddy activity and WBC in the SCS. Thirdly, strong intraseasonal variability (ISV) of sea surface temperature (SST) near the WBC regions was found, and the ISV signal of SST in winter weakens the ISV signal of latent heat flux by 20%. 13Gastric motility functional study based on electrical bioimpedance measurements and simultaneous electrogastrography显示文摘For some time now,the research on gastric motility and function has fallen behind in the amount of research on gastric endocrine,exocrine secretion,and gastric morphology.In this paper,a noninvasive method to study gastric motility was developed,taking bioimpedance measurements over the gastric area simultaneously with the electrogastrography(EGG).This is based on the concept of observing and analyzing simultaneously the intrinsic electrical gastric activity(basic electric rhythm) and the mechanical gastric activity.Additionally,preliminary clinical studies of healthy subjects and subjects with functional dyspepsia(FD) and gastritis were carried out.The impedance gastric motility(IGM) measurements of the healthy and FD subjects were compared,along with the studies of the FD subjects before treatment and after one week and three weeks of treatment.We also compared IGM measurements of healthy subjects and subjects with erosive gastritis,along with the studies of the subjects with erosive gastritis before treatment and after one week of treatment.Results show that FD subjects have poor gastric motility(P<0.01).After a week of treatment,the gastric motility of FD subjects was not yet improved although the EGG had returned to normal by this time.By three weeks of treatment,the regular IGM rhythm returned in FD subjects.There was a significant difference of IGM parameters between the gastritis and healthy subjects(P<0.05).The EGG rhythm of the gastritis subjects returned to normal at one week post-treatment,while IGM parameters showed a trend to improvement(P>0.05),These results suggest the possibility of clinic application of the proposed method.Zhang-yong LI Chao-shi REN Shu ZHAO Hong SHA Juan DENG 2011Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2011,12,12:9
14Plasma metabolomic and lipidomic alterations associated with COVID-19显示文摘The pandemic of the coronavirus disease 2019(COVID-19)has become a global public health crisis.The symptoms of COVID-19 range from mild to severe,but the physiological changes associated with COVID-19 are barely understood.In this study,we performed targeted metabolomic and lipidomic analyses of plasma from a cohort of patients with COVID-19 who had experienced different symptoms.We found that metabolite and lipid alterations exhibit apparent correlation with the course of disease in these patients,indicating that the development of COVID-19 affected their whole-body metabolism.In particular,malic acid of the TCA cycle and carbamoyl phosphate of the urea cycle result in altered energy metabolism and hepatic dysfunction,respectively.It should be noted that carbamoyl phosphate is profoundly down-regulated in patients who died compared with patients with mild symptoms.And,more importantly,guanosine monophosphate(GMP),which is mediated not only by GMP synthase but also by CD39 and CD73,is significantly changed between healthy subjects and patients with COVID-19,as well as between the mild and fatal cases.In addition,dyslipidemia was observed in patients with COVID-19.Overall,the disturbed metabolic patterns have been found to align with the progress and severity of COVID-19.This work provides valuable knowledge about plasma biomarkers associated with COVID-19 and potential therapeutic targets,as well as an important resource for further studies of the pathogenesis of COVID-19.Di Wu Ting Shu Xiaobo Yang Jian-Xin Song Mingliang Zhang Chengye Yao Wen Liu Muhan Huang Yuan Yu Qingyu Yang Tingju Zhu Jiqian Xu Jingfang Mu Yaxin Wang Hong Wang Tang Tang Yujie Ren Yongran Wu Shu-Hai Lin Yang Qiu Ding-Yu Zhang You Shang Xi Zhou 2020National Science Review2020,7,7:9

19Toripalimab plus chemotherapy as second-line treatment in previously EGFR-TKI treated patients with EGFR-mutant-advanced NSCLC:a multicenter phase-II trial显示文摘This multicenter phase-II trial aimed to investigate the efficacy,safety,and predictive biomarkers of toripalimab plus chemotherapy as second-line treatment in patients with EGFR-mutant-advanced NSCLC.Patients who failed from first-line EGFR-TKIs and did not harbor T790M mutation were enrolled.Toripalimab plus carboplatin and pemetrexed were administrated every three weeks for up to six cycles,followed by the maintenance of toripalimab and pemetrexed.The primary endpoint was objective-response rate(ORR).Integrated biomarker analysis of PD-L1 expression,tumor mutational burden(TMB),CD8+tumor-infiltrating lymphocyte(TIL)density,whole-exome,and transcriptome sequencing on tumor biopsies were also conducted.Forty patients were enrolled with an overall ORR of 50.0%and disease-control rate(DCR)of 87.5%.The median progression free survival(PFS)and overall survival were 7.0 and 23.5 months,respectively.The most common treatment-related adverse effects were leukopenia,neutropenia,anemia,ALT/AST elevation,and nausea.Biomarker analysis showed that none of PD-L1 expression,TMB level,and CD8+TIL density could serve as a predictive biomarker.Integrated analysis of whole-exome and transcriptome sequencing data revealed that patients with DSPP mutation had a decreased M2 macrophage infiltration and associated with longer PFS than those of wild type.Toripalimab plus chemotherapy showed a promising anti-tumor activity with acceptable safety profiles as the second-line setting in patients with EGFR-mutant NSCLC.DSPP mutation might serve as a potential biomarker for this combination.A phase-III trial to compare toripalimab versus placebo in combination with chemotherapy in this setting is ongoing(NCT03924050).Tao Jiang Pingyang Wang Jie Zhang Yanqiu Zhao Jianying Zhou Yun Fan Yongqian Shu Xiaoqing Liu Helong Zhang Jianxing He Guanghui Gao Xiaoqian Mu Zhang Bao Yanjun Xu Renhua Guo Hong Wang Lin Deng Ningqiang Ma Yalei Zhang Hui Feng Sheng Yao Jiarui Wu Luonan Chen Caicun Zhou Shengxiang Ren 2021Signal Transduction and Targeted Therapy2021,6,11:8
20Relationship between Maternal PBMC HBV cccDNA and HBV Serological Markers and its Effect on HBV Intrauterine Transmission显示文摘Objective To investigate the relationship between maternal peripheral blood mononuclear cells (PBMC) hepatitis B virus(HBV) covalenty closed circular deoxyribonucleic acid(cccDNA) and other HBV serological markers and its effects on HBV intrauterine transmission. Methods We enrolled 290 newborns and their hepatitis B surface antigen(HBsAg) positive mothers. HBV cccDNA in PBMC and HBV DNA in serum were detected by a real‐time PCR‐TaqM an probe while HBV serological markers were detected with an electrochemiluminescence immunoassay. Results There was a positive correlation between the levels of PBMC HBV cccD NA and serum HBV DNA and HBeA g(r = 0.436 and 0.403, P < 0.001). The detection rate of pattern A [‘HBsA g(+), HBeA g(+), and anti‐HBc(+)’] was significantly higher in the PBMC HBV cccD NA positive group than in the control group(χ^2 = 48.48, P < 0.001). There was a significant association between HBV intrauterine transmission and PBMC HBV cccD NA(χ^2 = 9.28, P = 0.002). In the presence of serum HBV DNA, HBeA g, and PBMC HBV cccD NA, the risk of HBV intrauterine transmission was three times higher(OR = 3.69, 95% CI: 1.30‐10.42) than that observed in their absence. The risk of HBV intrauterine transmission was the greatest(OR = 5.89, 95% CI: 2.35‐14.72) when both PBMC HBV cccD NA and pattern A were present. A Bayesian network model showed that maternal PBMC HBV cccD NA was directly related to HBV intrauterine transmission. Conclusion PBMC HBV cccDNA may be a direct risk factor for HBV intrauterine transmission.
