3A novel method for purification of recombinant adeno-associated virus vectors on a large scale显示文摘A novel method for recombinant adeno-associ-ated virus (rAAV) purification on large scale is described. The method involves three steps, including chloroform treatment, PEG/NaCl precipitation and chloroform extraction. The whole procedure can be performed in four hours. Using this purification method, we can reproducibly obtain, from 4 × 109 of proviral cells cultured in roller bottles, purified rAAV-GFP stocks with titers of around 5×1013 particles/mL and purity greater than 95%. The infectious titers of the vector stocks were up to 2×1012 TU/mL, thus particle-to-infectivity rate was about 25. Under an electronic microscope, most rAAV particles appeared full and a few were in intermediate form. Empty particles were rarely seen. The purified rAAV-GFP stocks have been successfully used in in vitro and in vivo transfection experiments. 5Spectra response from macromolecular structure evolution of tectonically deformed coal of different deformation mechanisms显示文摘Coals with different deformation mechanisms(brittle deformation,brittle-ductile deformation,and ductile deformation) repre-sent different ways in macromolecular structure evolution based on the metamorphism.The evolution of coal structure could affect the occurrence condition of coalbed methane(CBM) because the nanopore structure affected by macromolecular struc-ture is the most important reservoir for CBM.This paper analyzes the evolutions and mechanisms of structure and functional group of tectonically deformed coals(TDCs) collected from Huainan-Huaibei coalfield using X-ray diffraction(XRD),Raman spectroscopy,and Fourier Transform Infrared(FTIR) spectroscopy methods.The results show that the macromolecular struc-ture evolutions of TDC are different from the primary structure coal as a result of the different metamorphic grade and defor-mation mechanisms.The different deformation mechanisms variously affect the process of functional group and polyconden-sation of macromolecular structure.Furthermore,the tectonic deformation leads to secondary structural defects and reduces the structure stability of TDC.The coupled evolution on stacking and extension caused by the changes of secondary structural de-fects results from different deformation mechanisms.We consider that the changes of chemical structure and secondary struc-tural defects are the primary reasons for the various structure evolutions of TDC compared with primary structure coal.LI XiaoShi JU YiWen HOU QuanLin LIN Hong 2012Science China Earth Sciences2012,55,8:18

7Daclatasvir plus asunaprevir in treatment-na?ve patients with hepatitis C virus genotype 1b infection显示文摘AIM To assess daclatasvir plus asunaprevir(d UAL) in treatment-na?ve patients from China's Mainland, Russia and South Korea with hepatitis C virus(HCV) genotype 1 b infection. METHODS Patients were randomly assigned(3:1) to receive 24 wk of treatment with d UAL(daclatasvir 60 mg once daily and asunaprevir 100 mg twice daily) beginning on day 1 of the treatment period(immediate treatment arm) or following 12 wk of matching placebo(placebodeferred treatment arm). The primary endpoint was a comparison of sustained virologic response at posttreatment week 12(SVR12) compared with the historical SVR rate for peg-interferon plus ribavirin(70%) among patients in the immediate treatment arm. The first 12 wk of the study were blinded. Safety was assessed in d UAL-treated patients compared with placebo patients during the first 12 wk(doubleblind phase), and during 24 wk of d UAL in both arms combined.RESULTS In total, 207 patients were randomly assigned to immediate(n = 155) or placebo-deferred(n = 52) treatment. Most patients were Asian(86%), female(59%) and aged < 65 years(90%). Among them, 13% had cirrhosis, 32% had IL28 B non-CC genotypes and 53% had baseline HCV RNA levels of ≥ 6 million IU/m L. Among patients in the immediate treatment arm, SVR12 was achieved by 92%(95% confidence interval: 87.2-96.0), which was significantly higher than the historical comparator rate(70%). SVR12 was largely unaffected by cirrhosis(89%), age ≥ 65 years(92%), male sex(90%), baseline HCV RNA ≥ 6 million(89%) or IL28 B non-CC genotypes(96%), although SVR12 was higher among patients without(96%) than among those with(53%) baseline NS5 A resistanceassociated polymorphisms(at L31 or Y93 H). during the double-blind phase, aminotransferase elevations were more common among placebo recipients than among patients receiving d UAL. during 24 wk of d UAL therapy(combined arms), the most common adverse events(≥ 10%) were elevated alanine aminotransferase and upper respiratory tract infection; emergent grade 3-4 laboratory abnormalities were infrequently observed, and all grade 3-4 aminotransferase abnormalities(alanine aminotransferase, n = 9; aspartate transaminase, n = 6) reversed within 8-11 d. Two patients discontinued d UAL treatment; one due to aminotransferase elevations, nausea, and jaundice and the other due to a fatal adverse event unrelated to treatment. 9Ultrasound enhanced heterogeneous activation of peroxydisulfate by bimetallic Fe-Co/GAC catalyst for the degradation of Acid Orange 7 in water显示文摘Bimetallic Fe-Co/GAC(granular activated carbon) was prepared and used as heterogeneous catalyst in the ultrasound enhanced heterogeneous activation of peroxydisulfate(PS, S2O2-8) process. The effect of initial pH, PS concentration, catalyst addition and stirring rate on the decolorization of Acid Orange 7(AO7) was investigated. The results showed that the decolorization efficiency increased with an increase in PS concentration from 0.3 to 0.5 g/L and an increase in catalyst amount from 0.5 to 0.8g/L. But further increase in PS concentration and catalyst addition would result in an unpronounced increase in decolorization efficiency. In the range of 300 to 900 r/min, stirring rate had little effect on AO7 decolorization. 12Clinical Observation on Electroacupuncture for Post-stroke Flaccid Paralysis显示文摘Objective:To observe the clinical efficacy of electroacupuncture (EA) in treating post-stroke flaccid paralysis. Methods:Forty patients with post-stroke flaccid paralysis were randomized by the random number table into a treatment group and a control group, 20 cases in each. The treatment group was intervened by acupuncture at Jiquan (HT 1), Tianquan (PC 2), Ququan (LR 8), Yinlingquan (SP 9), and Yongquan (KI 1), and the control group was treated by acupuncture with conventional acupoint selection. Barthel index (BI) was adopted for evaluating the activities of daily living (ADL), and therapeutic efficacy was analyzed. Results:The two groups both had marked increases of BI score after treatment. Compared to the control group after 1 treatment course and 2 treatment courses respectively, the treatment group had significant differences in BI score (P<0.01). The total effective rate was 100.0% in the treatment group versus 90.0% in the control group, and the difference was statistically significant (P<0.01). 14Management of granulomatous lobular mastitis: an international multidisciplinary consensus(2021 edition)显示文摘Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. With the de-velopment of old-aging society, the incidence of neurodegenerative diseases is on the increase. How-ever, it is difficult to diagnose for most of neurodegenerative diseases. At present, there are too few effective therapies. Advances in stem cell biology have raised the hope and possibility for the therapy of neurodegenerative diseases. 19The diurnal and seasonal characteristics of urban heat island variation in Beijing city and surrounding areas and impact factors based on remote sensing satellite data显示文摘Based on the land surface temperature (LST), the land cover classification map,vegetation coverage, and surface evapotranspiration derived from EOS-MODIS satellite data, and by the use of GIS spatial analytic technique and multivariate statistical analysis method, the urban heat island (UHI) spatial distribution of the diurnal and seasonal variabilities and its driving forces are studied in Beijing city and surrounding areas in 2001. The relationships among UHI distribution and landcover categories, topographic factor, vegetation greenness, and surface evapotranspiration are analyzed. The results indicate that: (i) The significant UHI occur in Beijing city areas in the four seasons due to high heat capacity and multi-reflection of compression building, as well as with special topographic features of its three sides surrounded by mountains,especially in the summer. The UHI spatial distribution is corresponding with the urban geometry structure profile. The LST difference is approximately 4-6℃ between Beijing city and suburb areas, comparatively is 8- 10℃ between Beijing city area and outer suburb area in northwestern regions. (ii) The UHI distribution and intensity in daytime are different from nighttime in Beijing city area, the nighttime UHI is obvious. However, in the daytime, the significant UHI mainly appears in the summer, the autumn takes second place, and the UHI in the winter and the spring seem not obvious. The surface evapotranspiration in suburb areas is larger than that in urban areas in the summer, and high latent heat exchange is evident, which leads to LST difference between city area and suburb area.
