1Matrix stiffness modulates tip cell formation through the p-PXN-Rac1-YAP signaling axis显示文摘Endothelial tip cell outgrowth of blood-vessel sprouts marks the initiation of angiogenesis which is critical in physiological and pathophysiological procedures.However,how mechanical characteristics of extracellular matrix(ECM)modulates tip cell formation has been largely neglected.In this study,we found enhanced CD31 expression in the stiffening outer layer of hepatocellular carcinoma than in surrounding soft tissues.Stiffened matrix promoted sprouting from endothelial cell(EC)spheroids and upregulated expressions of tip cell-enriched genes in vitro.Moreover,tip cells showed increased cellular stiffness,more actin cytoskeleton organization and enhanced YAP nuclear transfer than stalk and phalanx ECs.We further uncovered that substrate stiffness regulates FAK and Paxillin phosphorylation in focal adhesion of ECs promoting Rac1 transition from inactive to active state.YAP is subsequently activated and translocated into nucleus,leading to increased tip cell specification.p-Paxillin can also loosen the intercellular connection which also facilitates tip cell specification.Collectively our present study shows that matrix stiffness modulates tip cell formation through p-PXN-Rac1-YAP signaling axis,shedding light on the role of mechanotransduction in tip cell formation.This is of special significance in biomaterial design and treatment of some pathological situations.Yaru Guo Feng Mei Ying Huang Siqin Ma Yan Wei Xuehui Zhang Mingming Xu Ying He Boon Chin Heng Lili Chen Xuliang Deng 2022Bioactive Materials2022,7,1:4
2A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 3Human mobility data in the COVID-19 pandemic:characteristics,applications,and challenges显示文摘The COVID-19 pandemic poses unprecedented challenges around the world.Many studies have applied mobility data to explore spatiotemporal trends over time,investigate associations with other variables,and predict or simulate the spread of COVID-19.Our objective was to provide a comprehensive overview of human mobility open data to guide researchers and policymakers in conducting data-driven evaluations and decision-making for the COVID-19 pandemic and other infectious disease outbreaks.We summarized the mobility data usage in COVID-19 studies by reviewing recent publications on COVID-19 and human mobility from a data-oriented perspective.We identified three major sources of mobility data:public transit systems,mobile operators,and mobile phone applications.Four approaches have been commonly used to estimate human mobility:public transit-based flow,social activity patterns,index-based mobility data,and social media-derived mobility data.We compared mobility datasets’characteristics by assessing data privacy,quality,space–time coverage,high-performance data storage and processing,and accessibility.We also present challenges and future directions of using mobility data.This review makes a pivotal contribution to understanding the use of and access to human mobility data in the COVID-19 pandemic and future disease outbreaks.Tao Hu Siqin Wang Bing She Mengxi Zhang Xiao Huang Yunhe Cui Jacob Khuri Yaxin Hu Xiaokang Fu Xiaoyue Wang Peixiao Wang Xinyan Zhu Shuming Bao Wendy Guan Zhenlong Li 2021International Journal of Digital Earth2021,14,9:1

5A GIS-based analytical framework for evaluating the effect of COVID-19 on the restaurant industry with big data显示文摘COVID-19 cripples the restaurant industry as a crucial socioeconomic sector that contributes immensely to the global economy.However,what the current literature less explored is to quantify the effect of COVID-19 on restaurant visitation and revenue at different spatial scales,as well as its relationship with the neighborhood character-istics of customers’origins.Based on the Point of Interest(POI)measures derived from SafeGraph data providing mobility records of 45 million cell phone users in the US,our study takes Lower Manhattan,New York City,as the pilot study,and aims to examine 1)the change of restaurant visitations and revenue in the period prior to and after the COVID-19 outbreak,2)the areas where restaurant customers live,and 3)the association between the neighborhood characteristics of these areas and lost customers.By doing so,we provide a geographic information system-based analytical frame-work integrating the big data mining,web crawling techniques,and spatial-economic modelling.Our analytical framework can be implemented to estimate the broader effect of COVID-19 on other industries and can be augmented in a financially monitoring manner in response to future pandemics or public emergencies.Siqin Wang Ruomei Wang Xiao Huang Zhenlong Li Shuming Bao 2023Big Earth Data2023,7,1:1
6How does heterogeneity in dwelling type preferences relate to housing and built environment characteristics?显示文摘Understanding housing preferences is critical for successful compact city development.However,there is limited research on understanding preference heterogeneity in dwelling type choices.Using the Household Income and Labour Dynamics in Australia survey,this paper identifies the key housing and built environment characteristics associated with changes in dwelling type choice from detached houses to high-density.A latent class choice model captures the heterogeneity of dwelling type preferences within a traditionally low-density city,Brisbane,Australia.Findings reveal six household classes with distinct dwelling preferences:Class 1(senior households without children with other family members)and Class 2(couple families with children)in inner-city areas,Class 3(high-income young households)and Class 4(low-income households without children)in middle-city areas,Class 5(low-income families with children)and Class 6(middle-income young families without children)in outer-city areas.Residential environments with better access to educational facilities encourage Classes 3 and 6 to change to high-density living.Greater land use diversity encourages Classes 2,3,and 6 to move towards high-density living.Thefindings can be used to design and improve high-density housing for targeted population groups across inner-,middle-and outer-city areas.Yuanyuan Huang Scott N.Lieske Siqin Wang Yan Liu 2023International Journal of Digital Earth2023,16,1:0
7Studies on the Inversion Phenomenon of Physical Properties Observed in the Huagang Formation Reservoir in the Xihu Sag Based on the Water-Rock Reaction Experiments显示文摘In the Xihu Sag,the reservoirs of the Paleogene Huagang formation have entered the middle diagenetic stage A and the rock physical properties of the water layer are considerably more suitable for the gas migration and storage than those of the present gas layer,indicating the inversion of the physical properties.In this study,core samples were collected from the corresponding reservoir to conduct water-rock reaction experiments in acidic,alkaline,and neutral systems under the specific temperature and pressure.The reasons for the inversion of physical properties were investigated based on the experiment results in reservoir diagenetic environments.The inversion of physical properties can be attributed to the fact that the diagenetic environment around the gas-water interface controls the water-rock reaction effect.With different types of acidic substances,two different situations corresponding to inverted physical properties were analyzed along with the corresponding mechanisms.When the pore fluid is acidic,the physical properties make the overall water layer a better reservoir space than the gas layer,which can be referred to as the overall inversion of physical properties.When the fluid were generally neutral or weakly alkaline and the gas layer was rich in CO2,only the physical properties of the water layer adjacent to the gas-water interface were more favorable for the gas migration than those of the gas layer.This phenomenon can be referred to as the near-interface inversion of physical properties.HUANG Siqin XU Fanghao XU Guosheng CUI Hengyuan ZHANG Wu 2021Journal of Ocean University of China2021,20,4:0
8Kinetic network models to elucidate aggregation dynamics of aggregation-induced emission systems显示文摘Aggregation-induced emission(AIE)is a phenomenon where a molecule that is weakly or non-luminescent in a diluted solution becomes highly emissive when aggregated.AIE luminogens(AIEgens)hold promise in diverse applications like bioimaging,chemical sensing,and optoelectronics.Investigation in AIE luminescence is also critical for understanding aggregation kinetics as the aggregation process is an essential component of AIE emission.Experimental investigation of AIEgen aggregation is challenging due to the fast timescale of the aggregation and the amorphous aggregate structures.Computer simulations such as molecular dynamics(MD)simulation provide a valuable approach to complement experiments with atomic-level knowledge to study the fast dynamics of aggregation processes.However,individual simulations still struggle to systematically elucidate heterogeneous kinetics of the formation of amorphous AIEgen aggregates.Kinetic network models(KNMs),constructed from an ensemble of MD simulations,hold great potential in addressing this challenge.In these models,dynamic processes are modeled as a series ofMarkovian transitions occurring among metastable conformational states at discrete time intervals.In this perspective article,we first review previous studies to characterize the AIEgen aggregation kinetics and their limitations.We then introduce KNMs as a promising approach to elucidate the complex kinetics of aggregations to address these limitations.More importantly,we discuss our perspective on linking the output of KNMs to experimental observations of time-resolved AIE luminescence.We expect that this approach can validate the computational predictions and provide great insights into the aggregation kinetics for AIEgen aggregates.These insights will facilitate the rational design of improved AIEgens in their applications in biology and materials sciences.Zige Liu Michae L.Kalin Bojun Liu Siqin Cao Xuhui Huang 2024Aggregate2024,5,1:0
9Hollow copper sulfide nanoparticles carrying ISRIB for the sensitized photothermal therapy of breast cancer and brain metastases through inhibiting stress granule formation and reprogramming tumor-associated macrophages显示文摘As known,the benefits of photothermal therapy(PTT)are greatly limited by the heat tolerance of cancer cells resulting from overexpressed heat shock proteins(HSPs).Then HSPs further trigger the formation of stress granules(SGs)that regulate protein expression and cell viability under various stress conditions.Inhibition of SG formation can sensitize tumor cells to PTT.Herein,we developed PEGylated pH(low)insertion peptide(PEG-pHLIP)-modified hollow copper sulfide nanoparticles(HCuS NPs)encapsulating the SG inhibitor ISRIB,with the phase-change material lauric acid(LA)as a gatekeeper,to construct a pH-driven and NIR photo-responsive controlled smart drug delivery system(IL@H-PP).The nano medicine could specifically target slightly acidic tumor sites.Upon irradiation,IL@H-PP realized PTT,and the light-controlled release of ISRIB could effectively inhibit the formation of PTT-induced SG to sensitize tumor cells to PTT,thereby increasing the antitumor effect and inducing potent immunogenic cell death(ICD).Moreover,IL@H-PP could promote the production of reactive oxygen species(ROS)by tumor-associated macrophages(TAMs),repolarizing them towards the M1 phenotype and remodeling the immunosuppressive microenvironment.In vitro/vivo results revealed the potential of PTT combined with SG inhibitors,which provides a new paradigm for antitumor and antimetastases.Fan Tong Haili Hu Yanyan Xu Yang Zhou Rou Xie Ting Lei Yufan Du Wenqin Yang Siqin He Yuan Huang Tao Gong Huile Gao 2023Acta Pharmaceutica Sinica B2023,13,8:0
10Sequential delivery of PD-1/PD-L1 blockade peptide and IDO inhibitor for immunosuppressive microenvironment remodeling via an MMP-2 responsive dual-targeting liposome显示文摘Intelligent responsive drug delivery system opens up new avenues for realizing safer and more effective combination immunotherapy.Herein,a kind of tumor cascade-targeted responsive liposome(NLG919@Lip-pep1)is developed by conjugating polypeptide inhibitor of PD-1 signal pathway(AUNP-12),which is also a targeted peptide that conjugated with liposome carrier through matrix metalloproteinase-2(MMP-2)cleavable peptide(GPLGVRGD).This targeted liposome is prepared through a mature preparation process,and indoleamine-2,3-dioxygenase(IDO)inhibitor NLG919 was encapsulated into it.Moreover,mediated by the enhanced permeability and retention effect(EPR effect)and AUNP-12,NLG919@Lip-pep1 first targets the cells that highly express PD-L1 in tumor tissues.At the same time,the over-expressed MMP-2 in the tumor site triggers the dissociation of AUNP-12,thus realizing the precise block of PD-1 signal pathway,and restoring the activity of T cells.The exposure of secondary targeting moduleⅡVRGDC-NLG919@Lip mediated tumor cells targeting,and further relieved the immunosuppressive microenvironment.Overall,this study offers a potentially appealing paradigm of a high efficiency,low toxicity,and simple intelligent responsive drug delivery system for targeted drug delivery in breast cancer,which can effectively rescue and activate the body's anti-tumor immune response and furthermore achieve effective treatment of metastatic breast cancer.Chuan Hu Yujun Song Yiwei Zhang Siqin He Xueying Liu Xiaotong Yang Tao Gong Yuan Huang Huile Gao 2023Acta Pharmaceutica Sinica B2023,13,5:0
