3FoxM1 overexpression promotes epithelial-mesenchymal transition and metastasis of hepatocellular carcinoma显示文摘AIM: To investigate the expression of forkhead box protein M1(Fox M1) in the process of epithelial mesenchymal transition in hepatocellular carcinoma(HCC) and its role in metastasis.METHODS: Fox M1 and E-cadherin expression in HCC tissue microarray specimens was evaluated by immunohistochemical staining,and statistical methods were applied to analyze the correlation between FoxM 1 and epithelial-mesenchymal transition(EMT).KaplanMeier analysis of the correlation between the Fox M1 expression level and recurrence or overall survival of HCC patients was performed.The expression of FoxM 1,E-cadherin and snail homologue 1(SNAI1) in HCC cell lines was evaluated by real-time reverse transcriptionpolymerase chain reaction and Western blot.Hepatocyte growth factor(HGF) was used to induce EMT and stimulate cell migration in HCC cells.The expression of Fox M1 and SNAI1 was regulated by transfection with plasmids pc DNA3.1 and si RNAs in vitro.The occurrence of EMT was evaluated by Transwell assay,morphologic analysis and detection of the expression of EMT markers(E-cadherin and vimentin).Luciferase and chromatin immunoprecipitation assays were used to evaluate whether SNAI1 is a direct transcriptional target of FoxM 1.RESULTS: FoxM 1 expression was increased significantly in HCC compared with para-carcinoma(10.7 ± 0.9 vs 8.2 ± 0.7,P < 0.05) and normal hepatic(10.7 ± 0.9 vs 2.7 ± 0.4,P < 0.05) tissues.Overexpression of Fox M1 was correlated with HCC tumor size,tumor number,macrovascular invasion and higher TNM stage,but was negatively correlated with E-cadherin expression in microarray specimens and in cell lines.Fox M1 overexpression was correlated significantly with HCC metastasis and EMT.In vitro,we found that FoxM 1 plays a key role in HGF-induced EMT,and overexpression of Fox M1 could suppress E-cadherin expression and induce EMT changes,which were associated with increased HCC cell invasiveness.Next,we confirmed that FOXM1 directly binds to and activates the SNAI1 promoter,and we identified SNAI1 as a direct transcriptional target of FOXM1.Moreover,inhibiting the expression of SNAI1 significantly inhibited FoxM 1-mediated EMT.CONCLUSION: Fox M1 overexpression promotes EMT and metastasis of HCC,and SNAI1 plays a critical role in FoxM 1-mediated EMT.Fan-Di Meng Ji-Chao Wei Kai Qu Zhi-Xin Wang Qi-Fei Wu Ming-Hui Tai Hao-Chen Liu Rui-Yao Zhang Chang Liu 2015World Journal of Gastroenterology2015,21,1:17
4Pyogenic liver abscesses associated with nonmetastatic colorectal cancers: An increasing problem in Eastern Asia显示文摘AIM: To elaborate the clinicopathologic features of colorectal cancer-related pyogenic liver abscess (PLA). METHODS: Reported cases of colorectal cancer-related PLAs were collected from the literature published up to October 2011 and evaluated for their clinicopathologic features. Data of collected cases included demographics, clinical presentation, microbial findings and treatment. Categorical variables were compared by 2 analysis and continuous variables were evaluated using Student's t test. RESULTS: A total 96 cases of colorectal cancer-related PLA were collected from the previous literature. Most patients (60%) were male and 40% cases occurred in the age group of 61-70 years. Apart from some special types of PLA, there were significant differences in the microbiological spectrum between Eastern Asia and non-Eastern Asian countries, which implied different risk factors and courses of the disease. Gram negative bacteria especially Klebsiella pneumoniae (K. pneu- moniae ) PLA was predominant in Eastern Asia (80.0%) in contrast to non-Eastern Asian countries (P < 0.01). Meanwhile, most of the Eastern Asian patients exhibited smaller size of liver abscess and atypical presentation. Sigmoid colon and rectum (72.73%) were the main sites of tumor in Eastern Asian patients, whereas tumor sites were uneven among most of the non-Easter Asian PLA patients. 5Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 6The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 8Overexpression of Sweet Pepper Glycerol-3-Phosphate Acyltransferase Gene Enhanced Thermotolerance of Photosynthetic Apparatus in Transgenic Tobacco显示文摘In order to investigate the relationship between the lipid composition in thylakoid membrane and thermostability of pho-tosynthetic apparatus,tobacco transformed with sweet pepper sense glycerol-3-phosphate acyltransferase (GPAT) gene were used to analyze the lipid composition in thylakoid membrane,the net photosynthetic rate and chlorophyll fluorescence parameters under high temperature stress. The results showed that the saturated extent of monogalactosyldiacylglycerol (MGDG),sulfoquinovosyldiacylglycerol,digalactosyldiacylglycerol and phosphatidylglycerol in thylakoid membrane of transgenic tobacco T1 lines increased generally. Particularly,the saturated extent in MGDG increased obviously by 16.2% and 12.0% in T1-2 and T1-1,respectively. With stress temperature elevating,the maximum efficiency of photosystem II (PSII) photochemistry (Fv/Fm),actual photochemical efficiency of PSII in the light ( PSII) and net photosynthetic rate (Pn) of the two lines and wild type tobacco plants decreased gradually,but those parameters decreased much less in transgenic plants. Even though the recovery process appeared differently in the donor and acceptor side of PSII in transgenic tobacco compared with wild-type plants,the entire capability of PSII recovered faster in transgenic tobacco,which was shown in the parameters of PI,Fv/Fm and PSII,as a result,the recovery of Pn was accelerated. 10Removal of Pb(Ⅱ) from aqueous solution by hydrous manganese dioxide:Adsorption behavior and mechanism显示文摘Hydrous manganese dioxide (HMO) synthesized by redox of potassium permanganate and hydrogen peroxide was used as an adsorbent for Pb(Ⅱ) removal.The specific surface area,pore volume and BJH pore diameter of the HMO were 79.31m2/g,0.07cm3/g and 3.38 nm,respectively.The adsorption equilibrium at 298K could be well described by the Langmuir isotherm equation with q max value of 352.55mg/g.The negative values of G and the positive values of H and S indicated the adsorption process was spontaneous and endothermic.The pseudo second-order equation could best fit the adsorption data.The value of the calculated activation energy for Pb(Ⅱ) adsorption onto the HMO was 38.23 kJ/mol.The uptake of Pb(Ⅱ) by HMO was correlated with increasing surface hydroxyl group content and the main adsorbed speciation was PbOH+.The final chemical state of Pb(Ⅱ) on the surface of HMO was similar to PbO.HMO was a promising candidate for Pb(Ⅱ) removal from aqueous solution.Meng Xu Hongjie Wang Di Lei Dan Qu Yujia Zhai Yili Wang 2013Journal of Environmental Sciences2013,25,3:9

13Design,methodology,and baseline of whole city‑million scale children and adolescents myopia survey(CAMS)in Wenzhou,China显示文摘Background:Myopia is the most common visual impairment in children and adolescents worldwide.This study described an economical and effective population-based screening pipeline and performed the project of a million scale children and adolescents myopia survey(CAMS),which will shed light on the further study of myopia from the level of epidemiology and precision medicine.Methods:We developed a novel population-based screening pattern,an intelligent screening process and internetbased information transmission and analysis system to carry out the survey consisting of school children in Wenzhou,China.The examination items include unaided distance visual acuity,presenting distance visual acuity,and noncycloplegic autorefraction.Myopia and high myopia were defined as spherical equivalent(SE)≤−1.00 diopters(D)and SE≤−6.00 D,respectively.Next,the reports of the vision checking were automatically sent to parents and the related departments.The CAMS project will be done two to four times annually with the support of the government.An online eyesight status information management system(OESIMS)was developed to construct comprehensive and efficient electronic vision health records(EVHRs)for myopia information inquiry,risk pre-warning,and further study.Results:The CAMS completed the first-round of screening within 30 days for 99.41%of Wenzhou students from districts and counties,in June 2019.A total of 1,060,925 participants were eligible for CAMS and 1,054,251(99.37%participation rate)were selected through data quality control,which comprised 1305 schools,and 580,609,251,050 and 170,967 elementary,middle,and high school students.The mean age of participants was 12.21±3.32 years(6-20 years),the female-to-male ratio was 0.82.The prevalence of myopia in elementary,middle,and high school students was 38.16%,77.52%,and 84.00%,respectively,and the high myopia incidence was 0.95%,6.90%,and 12.98%.Conclusions:The CAMS standardized myopia screening model involves automating large-scale information collection,data transmission,data analysis and early warning,thereby supporting myopia prevention and control.The entire survey reduced 90%of staff,cost,and time consumption compared with previous surveys.This will provide new insights for decision support for public health intervention.Liangde Xu Youyuan Zhuang Guosi Zhang Yunlong Ma Jian Yuan Changseng Tu MiaoMiao Li Wencan Wang Yaru Zhang Xiaoyan Lu Jing Li Xinting Liu Zhengbo Xue Meng Zhou Jie Sun Jinhua Bao Ming Li Fan Lu Hong Wang Jianzhong Su Jia Qu 2021Eye and Vision2021,8,1:7
14Triassic deep-marine sedimentation in the western Qinling and Songpan terrane显示文摘Triassic sequences in both the western Qinling and Songpan terrane are composed mostly of deep-marine sediments. A detailed study was carried out on main sedimentary facies of Triassic successions, showing that they resulted from diverse sedimentary processes, such as submarine debris-flows, turbidity currents, bottom-flows, suspension fallout, and fluidized sediment flows. Debris-flows are dividable into two types, gravelly and sandy debris-flows, respectively, and the sandy debris-flow deposits make up considerable portion of the Triassic successions concerned. Turbidite is characterized by occurrence of normal grading, and the whole Bouma sequences, though widely developed, are not totally attributed to true turbidity currents. The non-graded Ta division is thought to originate from sandy debris flows, whereas the rest divisions result from low-density currents or from bottom-current modification if they contain sedimentary structures related to traction currents. Four types of facies associations are distinguished within Triassic deep-marine successions: massive and thick-bedded coarse-grained facies association, medium- and thick-bedded sandstone with interlayered fine-grained facies association, interlayered thin-bedded fine-grained facies association, and syn-sedimentary slump/breccia facies association. Spatial distribution of the different facies associations suggests that Lower Triassic sedimentation occurred primarily in continental slope, submarine channels, and base-of-slope aprons in the Hezuo-Jianzha region of the western Qinling, whereas the Middle Triassic consists mainly of sedimentary facies of base-of-slope aprons and submarine incised valleys. The counterparts in the Dangchang-Diebu region, in contrast, are characterized by platform carbonates. The shallow-marine carbonates evolved into deep-marine facies since the Ladinian, indicative of rapid drowning of the Carnian carbonate platform in Middle Triassic times. Depositional history of Lower Triassic and lower portion of Middle Triassic successions in the northern Songpan terrane is similar to that of the Dangchang-Diebu region of the western Qinling, as manifested by development of Lower-early Middle Triassic shallow-marine carbonate and a rapid shift to base-of-slope apron sediments since the Ladinian.MENG QingRen QU HongJie HU JianMin 2007Science China Earth Sciences2007,50,z2:7

17Intensive phototherapy vs.exchange transfusion for the treatment of neonatal hyperbilirubinemia:a multicenter retrospective cohort study显示文摘Background:Intensive phototherapy(IPT)and exchange transfusion(ET)are the main treatments for extreme hyperbilirubinemia.However,there is no reliable evidence on determining the thresholds for these treatments.This multicenter study compared the effectiveness and complications of IPT and ET in the treatment of extreme hyperbilirubinemia.Methods:This retrospective cohort study was conducted in seven centers from January 2015 to January 2018.Patients with extreme hyperbilirubinemia that met the criteria of ET were included.Patients were divided into three subgroups(low-,medium-,and high-risk)according to gestational week and risk factors.Propensity score matching(PSM)was performed to balance the data before treatment.Study outcomes included the development of bilirubin encephalopathy,duration of hospitalization,expenses,and complications.Mortality,auditory complications,seizures,enamel dysplasia,ocular motility disorders,athetosis,motor,and language development were evaluated during follow-up at age of 3 years.Results:A total of 1164 patients were included in this study.After PSM,296 patients in the IPT only group and 296 patients in the IPT plus ET group were further divided into the low-,medium-,and high-risk subgroups with 188,364,and 40 matched patients,respectively.No significant differences were found between the IPT only and IPT plus ET groups in terms of morbidity,complications,and sequelae.Hospitalization duration and expenses were lower in the low-and medium-risk subgroups in the IPT only group.Conclusions:In this study,our results suggest that IPT is a safe and effective treatment for extreme hyperbilirubinemia.The indication of ET for patients with hyperbilirubinemia could be stricter.However,it is necessary to have a contingency plan for emergency ET as soon as IPT is commenced especially for infants with risk factors.If IPT can be guaranteed and proved to be therapeutic,ET should be avoided as much as possible.Meng Zhang Yang He Jun Tang Wenbin Dong Yong Zhang Benjin Zhang Hong Wan Quanmin Deng Lirong Guan Bin Xia Zhong Chen Min Ge Jing Zhao Wenxing Li Jingjun Pei Yi Qu Dezhi Mu 2022Chinese Medical Journal2022,,5:7
18Diversity and redundancy of the ripening regulatory networks revealed by the fruitENCODE and the new CRISPR/Cas9 CNR and NOR mutants显示文摘Tomato is considered as the genetic model for climacteric fruits,in which three major players control the fruit ripening process:ethylene,ripening transcription factors,and DNA methylation.The fruitENCODE project has now shown that there are multiple transcriptional circuits regulating fruit ripening in different species,and H3K27me3,instead of DNA methylation,plays a conserved role in restricting these ripening pathways.In addition,the function of the core tomato ripening transcription factors is now being questioned.We have employed CRISPR/Cas9 genome editing to mutate the SBP-CNR and NAC-NOR transcription factors,both of which are considered as master regulators in the current tomato ripening model.These plants only displayed delayed or partial non-ripening phenotypes,distinct from the original mutant plants,which categorically failed to ripen,suggesting that they might be gain-of-function mutants.Besides increased DNA methylation genome-wide,the original mutants also have hyper-H3K27me3 in ripening gene loci such as ACS2,RIN,and TDR4.It is most likely that multiple genetic and epigenetic factors have contributed to their strong non-ripening phenotypes.Hence,we propose that the field should move beyond these linear and twodimensional models and embrace the fact that important biological processes such as ripening are often regulated by highly redundant network with inputs from multiple levels.Ying Gao Ning Zhu Xiaofang Zhu Meng Wu Cai-Zhong Jiang Donald Grierson Yunbo Luo Wei Shen Silin Zhong Da-Qi Fu Guiqin Qu 2019Horticulture Research2019,6,1:7

20Expression of circulating microRNA-20a and let-7a in esophageal squamous cell carcinoma显示文摘AIM: To investigate the expressions of micro RNA-20a(mi R-20a) and let-7a in esophageal squamous cell carcinoma(ESCC) and their diagnostic value. METHODS: Seventy patients with ESCC and 40 healthy subjects were enrolled to investigate the expression of mi R-20 a and let-7a using quantitative real-time PCR. The expression of mi R-20 a and let-7a was compared between ESCC patients and healthy subjects. The plasma levels of mi R-20 a and let-7a in relation to patient clinicopathologic parameters, the receiver operating characteristic(ROC) curve, and the sensitivity and specificity of mi R-20 a and let-7a in ESCC diagnosis were analyzed.RESULTS: Plasma levels of mi R-20 a were significantly higher in ESCC patients than in healthy controls, and plasma levels of let-7 were lower in ESCC patients than in healthy controls(both P < 0.05). The area under the ROC curve of mi R-20 a was 0.767(95%CI: 0.677-0.857; P < 0.001), when the cut-off value was set at 4.77, the sensitivity and specificity were 64.3% and 75.0%, respectively. The area under the ROC curve of let-7a was 0.829(95%CI: 0.754-0.904; P < 0.001), when the cut-off value was set at 6.22, the sensitivity and specificity were 74.3% and 85.0%, respectively. Thus, the sensitivity and specificity of let-7a were higher than those of mi R-20 a. The median relative plasma expression of let-7a in clinical stage Ⅲ/Ⅳ(0.24) was lower than that in stage Ⅰ/Ⅱ(0.42), while the expression of mi R-20 a according to stage was not statistically different.
