Modified Shenlingbaizhu decoction reduces intestinal adenoma formation in adenomatous polyposis coli multiple intestinal neoplasia mice by suppression of hypoxia-inducible factor 1α-induced CD4+CD25+forkhead box P3 regulatory T cells显示文摘OBJECTIVE: To test the hypothesis that modified Shenlingbaizhu decoction(MSD) attenuates the formation of intestinal adenomas by regulating activation of CD4+CD25+ forkhead box P3(Fox P3) regulatory T cells(Tregs) by downregulation of hypoxia-inducible factor 1α(HIF-1α).METHODS: Chemical fingerprints of ginsenoside Rb1, ginsenoside Rc, paeoniflorin, and dioscin in standard extractions were used as material bases of MSD. Adenomatous polyposis coli multiple intestinal neoplasia(ApcMin/+) mice, which harbor a mutation in adenomatous polyposis coli, were used to host intestinal adenomas. Peripheral blood and spleen Tregs were analyzed by flow cytometry. Protein expression was analyzed by immunohistochemistry and Western blotting.RESULTS: The number and size of intestinal adenomas were significantly reduced by MSD treatment.Mucosal thickening and the spleen size were also substantially decreased by MSD. The carcinogenesis process in ApcMin/+mice resembled that of human colorectal cancer. Molecular markers of neoplasms, such as β-catenin, cyclooxygenase-2, proliferating cell nuclear antigen, and p53, were substan-tially ameliorated by MSD treatment. Moreover,MSD downregulated peripheral and spleen CD4 +CD25+Fox P3+ Tregs and reduced in situ expression of CD4, CD25, and Fox P3 in intestinal adenomas.MSD also suppressed HIF-1α expression in the intestinal adenomas, and HIF-1α inhibition decreased expression of Fox P3 in Jurkat T cells under hypoxic conditions.CONCLUSION: MSD is a valid prescription to control the formation of intestinal adenomas in ApcMin/+mice. It exerts anti-cancer effects partially through suppression of HIF-1α that induced activation of CD4+CD25+Fox P3+ Tregs in vivo and in vitro.Xu Wenjuan Han Qinrui Liang Shuntian Li Lu Shao Meng Yao Xueqing Sun Xuegang 2018Journal of Traditional Chinese Medicine2018,38,1:3

The Formation and Evolution of Uvarovite in UHP Serpentinite and Rodingite and its Constraints on Chromium Mobility in the Oceanic Subduction Zone显示文摘The uvarovite-andradite and uvarovite-andradite-grossular solid-solution series are rare in nature. The discovery of uvarovite-andradite in serpentinite and rodingite from the ultra-high pressure(UHP)metamorphic belt in southwestern Tianshan provided an opportunity to investigate its behavior in the subduction zone.Uvarovite(defined as chromiumgarnet)from serpentinite is homogeneous in a single grain, covering compositions in the uvarovite-andradite solid solution series of Adr_(58-66)Uv_(33-41),with few grossular components.Uvarovite from rodingites contain various Cr_(2)O_(3) contents(1.7-17.9 wt%)and mineral compositions being in the range of Adr_(21-31)Uv_(41-50)Grs_(22-37),Adr_(52-90)Uv_(5-25)Grs_(0-21) and Adr_(19-67)Uv_(3-63)Grs_(13-42).Discontinuous chemical variation of uvarovite from core to rim indicates that uvarovite formed by consuming andradite and chromite, which could provide Ca,Cr,Al and Fe.Raman signals of water were identified for uvarovite from both serpentinite and rodingite, with high water content in uvarovite from serpentinite. The high pressure mineral assemblage,as well as the association with perovskite, indicated that the studied uvarovite from serpentinite and rodingite was formed through high pressure metamorphism, during the subduction zone serpentinization and rodingitization.High alkaline and highly reduced fluids released from serpentinization or rodingitization in the oceanic subduction zone promote the mobility of chromium and enable its long-distance migration.ZHANG Cong SHEN Tingting ZHANG Lifei LIN Congcong ZHANG Zhongwei Qin Xueqing HU Han QIU Tian XIANG Zhenqun ZHANG Jianxin 2021Acta Geologica Sinica(English Edition)2021,95,5:1
Thermal-physiological Strategies Underlying the Sympatric Occurrence of Three Desert Lizard Species显示文摘Sympatric reptiles are the ideal system for investigating temperature-driven coexistence. Understanding thermally physiological responses of sympatric lizards is necessary to reveal the physiological mechanisms that underpin the sympatric occurrence of reptiles. In this study, we used three lizard species, Eremias argus, E. multiocellata, and Phrynocephalus przewalskii, which are sympatric in the Inner Mongolia desert steppe, as a study system. By comparing their resting metabolic rates(RMR) and locomotion at different body temperatures, we aimed to better understand their physiological responses to thermal environments, which may explain the sympatric occurrence of these lizards. Our results showed that E. argus had significantly higher RMR and sprint speed than E. multiocellata, and higher RMR than P. przewalskii. In addition, the optimal temperature that maximized metabolic rates and locomotion for E. argus and E. multiocellata was 36°C, whereas for P. przewalskii it was 39°C. Our study revealed the physiological responses to temperatures that justify the sympatric occurrence of these lizards with different thermal and microhabitat preferences and active body temperatures. Eremias argus and E. multiocellata, which have lower body temperatures than P. przewalskii, depend on higher RMR and locomotion to compensate for their lower body temperatures in field conditions. Our study also highlights the importance of using an integrative approach, combining behavior and physiology, to explore the basis of sympatric occurrence in ectothermic species.Xueqing WANG Shuran LI Li LI Fushun ZHANG Xingzhi HAN Junhuai BI Baojun SUN 2019Asian Herpetological Research2019,10,3:1

Insight into sulfur and iron effect of binary nickel-iron sulfide on oxygen evolution reaction显示文摘Nickel-iron sulfide has shown attractive activity in electrocatalytic oxygen evolution reaction(OER).However, the effects of low valence sulfur(S^(2−))and metal species on OER in binary nickel-iron sulfide have rarely been systematically studied.Works based on post-catalysis characterization have led to the assumption that the real active species are nickel-iron oxyhydroxide, and that nickel-iron sulfide acts only as a precatalyst. Therefore,to study the role of S,Ni,and Fe for the development of nickel-iron sulfide catalyst is of self-evident importance.Herein,a facile solvothermal method is used to synthesize acetylene black coated with nickel-iron sulfide nanosheets.Electrochemical tests show that the presence of low valence S species makes the catalyst have faster OER kinetics, larger active area, and intermediate active species adsorption area. Therefore, the present study reveals the enhancing effect of low valence sulfur species(S^(2−))on OER in binary nickel-iron sulfide. In situ Raman spectroscopy shows that the generation ofγ-NiOOH intermediate is essential and Fe does not directly participate in the oxygen production.Density functional theory(DFT)calculation shows that Ni-OH deprotonation is a rate-determining step for both binary nickel-iron sulfide and nickel sulfide. The addition of Fe into NiSx lightly increases the charge transfer of Ni atom to O atom, which makes deprotonation easier and thereby improves the OER performance.Qikang Wu Songrui Wang Jiahui Guo Xueqing Feng Han Li Shanshan Lv Yan Zhou Zheng Chen 2022Nano Research2022,15,3:1

Obesity-induced inflammatory miR-133a mediates apoptosis of granulosa cells and causes abnormal folliculogenesis显示文摘Obesity has been reported to promote disordered folliculogenesis, but the exact molecular mechanisms are still not fully understood. In this study,we find that miR-133a is involved in obesity-induced follicular development disorder. After feeding with a high-fat diet(HFD)and fructose water for nine weeks, the mouse body weight is significantly increased, accompanied by an inflammatory state and increased expression of miR-133a in the adipose tissues and ovaries as well as accelerated follicle depletion.Although miR-133a is increased in the fat and ovaries of HFD mice, the increased miR-133a in the HFD ovaries is not derived from exosome transferred from obese adipose tissues but is synthesized by ovarian follicular cells in response to HFD-induced inflammation. In vivo experiments show that intrabursal injection of miR-133a agomir induces a decrease in primordial follicles and an increase in antral follicles and atretic follicles, which is similar to HFD-induced abnormal folliculogenesis.Overexpression of miR-133a modestly promotes granulosa cell apoptosis by balancing the expression of anti-apoptotic proteins such as C1QL1 and XIAP and pro-apoptotic proteins such as PTEN.Overall, this study reveals the function of miR-133a in obesityinduced ovarian folliculogenesis dysfunction and sheds light on the etiology of female reproductive disorders.Ruizhi Chen Xueqing Wu Han Qiu Baiming Yang Yao Chen Xiang Chen Yingshan Li Shaochun Yuan Dan Liu Luanjuan Xiao Yanhong Yu 2023Acta Biochimica et Biophysica Sinica2023,55,8:0
Biotoxicity dynamic change and key toxic organics identification of coal chemical wastewater along a novel full-scale treatment process显示文摘It is particularly important to comprehensively assess the biotoxicity variation of industrial wastewater along the treatment process for ensuring the water environment security. However, intensive studies on the biotoxicity reduction of industrial wastewater are still limited. In this study, the toxic organics removal and biotoxicity reduction of coal chemical wastewater(CCW)along a novel full-scale treatment process based on the pretreatment process-anaerobic process-biological enhanced(BE)process-anoxic/oxic(A/O)process-advanced treatment process was evaluated. This process performed great removal efficiency of COD,total phenol,NH_(4)^(+)-N and total nitrogen. And the biotoxicity variation along the treatment units was analyzed from the perspective of acute biotoxicity, genotixicity and oxidative damage. The results indicated that the effluent of pretreatment process presented relatively high acute biotoxicity to Tetrahymena thermophila. But the acute biotoxicity was significantly reduced in BE-A/O process. And the genotoxicity and oxidative damage to Tetrahymena thermophila were significantly decreased after advanced treatment. The polar organics in CCW were identified as the main biotoxicity contributors.Phenols were positively correlated with acute biotoxicity, while the nitrogenous heterocyclic compounds and polycyclic aromatic hydrocarbons were positively correlated with genotoxicity.Although the biotoxicity was effectively reduced in the novel full-scale treatment process, the effluent still performed potential biotoxicity, which need to be further explored in order to reduce environmental risk.Weiwei Ma Xiaoqi Zhang Hongjun Han Xueqing Shi Qiaoping Kong Tong Yu Fei Zhao 2024Journal of Environmental Sciences2024,,4:0

A New Route for the Rapid Synthesis of Metal–Organic Frameworks at Room Temperature显示文摘Rapid synthesis of metal–organic frameworks(MOFs),especially high-valence MOFs at roomtemperature without external energy,is a challenging topic. In this work,a stable radical solution has been discovered.Various MOFs with versatile metal nodes and ligands were rapidly synthesized at room temperature in the absence of external energy.Especially,MOFs with conjugated ligands achieved instantaneous architecture(in less than 1 s)and quantitative yield.Radicals in the solution play a crucial role in the accelerated kinetics, and the new radical route paves a cyclic pathway for the MOF synthesis. The mechanism has been thoroughly investigated by electron paramagnetic resonance,in situ proton nuclear magnetic resonance,X-ray absorption spectra,in situ small-angle X-ray scattering-wide-angle X-ray scattering, and density functional theory calculations.Pei Zhang Xinchen Kang Liming Tao Lirong Zheng Junfeng Xiang Ran Duan Jikun Li Peng Chen Xueqing Xing Guang Mo Zhonghua Wu Buxing Han 2023CCS Chemistry2023,5,6:0
The Effects of Turbulence Intensity and Tip Speed Ratio on the Coherent Structure of Horizontal-Axis Wind Turbine Wake:A Wind Tunnel Experiment显示文摘The evolution laws of the large-eddy coherent structure of the wind turbine wake have been evaluated via wind tunnel experiments under uniform and turbulent inflow conditions. The spatial correlation coefficients, the turbulence integral scales and power spectrum are obtained at different tip speed ratios(TSRs)based on the time-resolved particle image velocity(TR-PIV)technique.The results indicate that the large-eddy coherent structures are more likely to dissipate with an increase in turbulence intensity and TSR.Furthermore, the spatial correlation of the longitudinal pulsation velocity is greater than its axial counterpart, resulting into a wake turbulence dominated by the longitudinal pulsation. With an increase of turbulence intensity, the integral scale of the axial turbulence increases, meanwhile, its longitudinal counterpart decreases.Owing to an increase in TSR,the integral scale of axial turbulence decreases, whereas, that of the longitudinal turbulence increases.By analyzing the wake power spectrum,it is found that the turbulent pulsation kinetic energy of the wake structure is mainly concentrated in the low-frequency vortex region. The dissipation rate of turbulent kinetic energy increases with an increase of turbulence intensity and the turbulence is transported and dissipated on a smaller scale vortex, thus promoting the recovery of wake.Yuxia Han Jianwen Wang Xin Li Xueqing Dong Caifeng Wen 2022Energy Engineering2022,119,6:0
Molecular Mechanisms of Intracellular Delivery of Nanoparticles Monitored by an Enzyme‑Induced Proximity Labeling显示文摘Achieving increasingly finely targeted drug delivery to organs, tissues, cells, and even to intracellular biomacromolecules is one of the core goals of nanomedicines. As the delivery destination is refined to cellular and subcellular targets,it is essential to explore the delivery of nanomedicines at the molecular level. However, due to the lack of technical methods, the molecular mechanism of the intracellular delivery of nanomedicines remains unclear to date. Here,we develop an enzyme-induced proximity labeling technology in nanoparticles(nano-EPL)for the real-time monitoring of proteins that interact with intracellular nanomedicines.Poly(lactic-co-glycolic acid)nanoparticles coupled with horseradish peroxidase(HRP)were fabricated as a model(HRP(+)-PNPs)to evaluate the molecular mechanism of nano delivery in macrophages.By adding the labeling probe biotin-phenol and the catalytic substrate H_(2)O_(2)at different time points in cellular delivery,nano-EPL technology was validated for the real-time in situ labeling of proteins interacting with nanoparticles.Nano-EPL achieves the dynamic molecular profiling of 740 proteins to map the intracellular delivery of HRP(+)-PNPs in macrophages over time.Based on dynamic clustering analysis of these proteins,we further discovered that different organelles, including endosomes, lysosomes, the endoplasmic reticulum, and the Golgi apparatus, are involved in delivery with distinct participation timelines.More importantly, the engagement of these organelles differentially affects the drug delivery efficiency, reflecting the spatial–temporal heterogeneity of nano delivery in cells. In summary, these findings highlight a significant methodological advance toward understanding the molecular mechanisms involved in the intracellular delivery of nanomedicines.Junji Ren Zibin Zhang Shuo Geng Yuxi Cheng Huize Han Zhipu Fan Wenbing Dai Hua Zhang Xueqing Wang Qiang Zhang Bing He 2024Nano-Micro Letters2024,16,6:0
Satellite observed shape-based overall rotation—A new aspect in eddy kinematics显示文摘With the development of satellite altimetry technology, the resolution of sea-level anomaly(SLA)datasets is constantly improving.Current spatial resolution levels can reach a grid size of(1/4)°×(1/4)°,with daily measurements that span from 1993 to 2018,allowing for the precise identification and tracking of individual eddies. In the current study,in addition to the internal circulation and migration of eddies,a new aspect in eddy kinematics is revealed and investigated for the first time:shape-based overall eddy rotation(SOER),based on the intrinsic elliptical shape of eddies identified from a high-resolution SLA dataset. We found that eddies can maintain an elliptical shape and a slow and stable SOER during their migration process. The SOER speed was observed to be negatively correlated to eddy lifetime, and exhibited a dependence on latitude, decreasing from low-and high-to mid-latitudes.The SOER direction tended to be consistent with the direction of internal circulation, particularly for long-lived eddies. In addition,we identified a negative relationship between internal circulation speed and SOER speed while the migration speed was positively related to SOER speed. These findings further expand and improve eddy kinematics, which is of great significance for the future study of eddy dynamics.Xueqing Yang Guiyan Han Chunyong Ma Chuanchuan Cao Jie Yang Ge Chen 2022Acta Oceanologica Sinica2022,41,5:0
Identification of tick-borne pathogens using metagenomic analyses in H.longicornis feeding on humans in downtown Beijing显示文摘On August 14th,2018,a Beijing resident living in Xicheng District found a female H.longicornis tick attached to the skin at the front of his upper shin.On examination, the patient was afebrile and appeared well. The species of the tick was identified through morphological characteristics and phylogenetic analysis based on cytochrome C oxidase subunit I.This H.longicornis tick was screened for tick-borne pathogens such as viruses, bacteria and parasites.RNA pathogens were screened by PCR and sequencing, while DNA pathogens were screened by metagenomic analyses. It was found that the tick was positive for the DNA sequences of zoonotic and animal pathogens such as A phagocytophilum,Ehrlichia minasensis and C.burnetii.Considering the good health condition of the patient,we hypothesized that the pathogens originated from the tick specimen itself rather than host blood meal. For the first time, our study reveals the possible risk of transmission of tick-borne pathogens to human beings through tick bit in downtown Beijing.Further research is needed to screen for tick-borne pathogens among unfed ticks collected from central Beijing.Jizhou Lv Huiyu Wang Xueqing Han Lin Mei Xiangfen Yuan Yufang Kong Junhua Deng Zhen F.Fu Shaoqiang Wu Xiangmei Lin 2021Animal Diseases2021,1,3:0
