1Qiguiyishen decoction reduced the accumulation of extracellular matrix in the kidneys of rats with adriamycin-induced nephropathy显示文摘OBJECTIVE: To investigate the effect of Qiguiyishen decoction(QGYS) on the severity of nephropathy.METHODS: Twenty-four rats were randomly divided into four groups(Ⅰ, Ⅱ, Ⅲ, Ⅳ) according to the random number table. Group Ⅰ as control group did not establish nephropathy model. Groups Ⅱ,Ⅲ, and Ⅳ were intravenously administered Adriamycin(7.5 mg/kg) through the tail vein to establish nephropathy model. QGYS was prepared with the extracts of Huangqi(Radix Astragali Mongolici),Danggui(Radix Angelicae Sinensis),Niuxi(RadixAchyran-this Bidentatae), and Chuanxiong(Rhizoma Chuanxiong).Group Ⅳ was administered QGYS (2mL·kg-1·d-1), group Ⅲ was administered benazepril(10 mL·kg-1·d-1), and group Ⅰ, Ⅱ was administered water(2mL·kg-1·d-1)once daily for eight weeks.RESULTS: QGYS reduced the excretion of urinary protein and N-acetyl-β-D-glucosaminidase and alleviated the accumulation of extracellular matrix(ECM) in renal tissue. 5A New Framework for Focused Web Crawling显示文摘Focused crawlers are important tools to support applications such as specialized Web portals, online searching, and Web search engines. A topic driven crawler chooses the best URLs and relevant pages to pursue during Web crawling. It is difficult to deal with irrelevant pages. This paper presents a novel focused crawler framework. In our focused crawler, we propose a method to overcome some of the limitations of dealing with the irrelevant pages. We also introduce the implementation of our focused crawler and present some important metrics and an evaluation function for ranking pages relevance. 13Bioceramic scaffolds with triply periodic minimal surface architectures guide early-stage bone regeneration显示文摘The pore architecture of porous scaffolds is a critical factor in osteogenesis,but it is a challenge to precisely configure strut-based scaffolds because of the inevitable filament corner and pore geometry deformation.This study provides a pore architecture tailoring strategy in which a series of Mg-doped wollastonite scaffolds with fully interconnected pore networks and curved pore architectures called triply periodic minimal surfaces(TPMS),which are similar to cancellous bone,are fabricated by a digital light processing technique.The sheet-TPMS pore geometries(s-Diamond,s-Gyroid)contribute to a 3‒4-fold higher initial compressive strength and 20%-40%faster Mg-ion-release rate compared to the other-TPMS scaffolds,including Diamond,Gyroid,and the Schoen’s I-graph-Wrapped Package(IWP)in vitro.However,we found that Gyroid and Diamond pore scaffolds can significantly induce osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs).Analyses of rabbit experiments in vivo show that the regeneration of bone tissue in the sheet-TPMS pore geometry is delayed;on the other hand,Diamond and Gyroid pore scaffolds show notable neo-bone tissue in the center pore regions during the early stages(3-5 weeks)and the bone tissue uniformly fills the whole porous network after 7 weeks.Collectively,the design methods in this study provide an important perspective for optimizing the pore architecture design of bioceramic scaffolds to accelerate the rate of osteogenesis and promote the clinical translation of bioceramic scaffolds in the repair of bone defects.Miaoda Shen Yifan Li Fengling Lu Yahui Gou Cheng Zhong Shukun He Chenchen Zhao Guojing Yang Lei Zhang Xianyan Yang Zhongru Gou Sanzhong Xu 2023Bioactive Materials2023,,7:1

16Single-cell landscape analysis reveals distinct regression trajectories and novel prognostic biomarkers in primary neuroblastoma显示文摘Neuroblastoma(NB),which is the most common pediatric extracranial solid tumor,varies widely in its clinical presentation and outcome.NB has a unique ability to spontaneously differentiate and regress,suggesting a potential direction for therapeutic intervention.However,the underlying mechanisms of regression remain largely unknown,and more reliable prognostic biomarkers are needed for predicting trajectories for NB.We performed scRNAseq analysis on 17 NB clinical samples and three peritumoral adrenal tissues.Primary NB displayed varied cell constitution,even among tumors of the same pathological subtype.Copy number variation patterns suggested that neuroendocrine cells represent the malignant cell type.Based on the differential expression of sets of related marker genes,a subgroup of neuroendocrine cells was identified and projected to differentiate into a subcluster of benign fibroblasts with highly expressed CCL2 and ZFP36,supporting a progressive pathway of spontaneous NB regression.We also identified prognostic markers(STMN2,TUBA1A,PAGE5,and ETV1)by evaluating intra-tumoral heterogeneity.Lastly,we determined that ITGB1 in M2-like macrophages was associated with favorable prognosis and may serve as a potential diagnostic marker and therapeutic target.In conclusion,our findings reveal novel mechanisms underlying regression and potential prognostic markers and therapeutic targets of NB.Qingqing Liu Zhenni Wang Yan Jiang Fengling Shao Yue Ma Mingzhao Zhu Qing Luo Yang Bi Lijian Cao Liang Peng Jianwu Zhou Zhenzhen Zhao Xiaobin Deng Tong-Chuan He Shan Wang 2022Genes & Diseases2022,9,6:0
17Modulation of Antiviral Immunity and Therapeutic Efficacy by 25-Hydroxycholesterol in Chronically SIV-Infected, ART-Treated Rhesus Macaques显示文摘Cholesterol-25-hydroxylase(CH25 H)and its enzymatic product 25-hydroxy cholesterol(25 HC)exert broadly antiviral activity including inhibiting HIV-1 infection.However,their antiviral immunity and therapeutic efficacy in a nonhuman primate model are unknown.Here,we report that the regimen of 25 HC combined with antiretroviral therapy(ART),provides profound immunological modulation towards inhibiting viral replication in chronically SIVmac239-infected rhesus macaques(RMs).Compared to the ART alone,this regimen more effectively controlled SIV replication,enhanced SIVspecific cellular immune responses,restored the ratio of CD4/CD8 cells,reversed the hyperactivation state of CD4^(+)T cells,and inhibited the secretion of proinflammatory cytokines by CD4^(^(+))and CD8^(+)T lymphocytes in chronically SIVinfected RMs.Furthermore,the in vivo safety and the preliminary pharmacokinetics of the 25 HC compound were assessed in this RM model.Taken together,these assessments help explain the profound relationship between cholesterol metabolism,immune modulation,and antiviral activities by 25 HC.These results provide insight for developing novel therapeutic drug candidates against HIV-1 infection and other related diseases.Chunxiu Wu Jin Zhao Ruiting Li Fengling Feng Yizi He Yanjun Li Runhan Huang Guangye Li Heng Yang Genhong Cheng Ling Chen Feng Ma Pingchao Li Caijun Sun 2021Virologica Sinica2021,36,5:0
18Temporal and Spatial Distribution of SARS-CoV-2 Aerosols in a Large-Scale Fangcang Shelter Hospital in Shanghai,China显示文摘The coronavirus disease 2019(COVID-19)pandemic caused by frequently mutating severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has had a worldwide impact.However,detailed data on the potential aerosol transmission of SARS-CoV-2 in real-world and controlled laboratory settings remain sparse.During the COVID-19 pandemic in Shanghai,China in 2022,samples were collected in a Fangcang shelter hospital,a large-scale temporary hospital rapidly built by converting the existing National Exhibition and Convention Center(Shanghai)into a health care facility.Aerosol samples at different sites and intervals around patients and in public areas,surface samples,and pharyngeal swab samples from corresponding patients were included.Samples were tested for SARS-CoV-2 using real-time quantitative polymerase chain reaction(RT-qPCR)assays,followed by sequencing if the cycle threshold(Ct)value was<30.The positivity rate for SARS-CoV-2 in aerosol samples was high in contaminated zones(37.5%,104/277),especially around the bed(41.2%,68/165)and near ventilation inlets(45.2%,14/31).The prevalence of SARS-CoV-2 around the bed,public areas,and air inlets of exhaust vents fluctuated and was closely related to the positivity rate among patients at corresponding sampling sites.Some surface samples of different personal protective equipment from medical staff had high positivity rates.Sixty sequences of joined ORF1ab and spike genes obtained from sixty samples represented two main clusters of Omicron SARS-CoV-2.There was consistency in virus sequences from the same patient and their environment,and the detected virus sequences matched those of virus strains in circulation during the collection periods,which indicated a high likelihood of cross-contamination in the Fangcang shelter hospital.In summary,the results provide a quantitative and real landscape of the aerosol transmission of SARS-CoV-2 and a patient-centered view of contamination in large and enclosed spaces and offer a useful guide for taking targeted measures to avoid nosocomial infections during the management of SARS-CoV-2 or other respiratory virus diseases in a Fangcang shelter hospital.Jiafu Jiang Zhe Yin Jing Li Leili Jia Rulin He Wenhui Yang Jihu Yang Hang Fan Sen Zhang Yunfei Wang Zengming Zhao Haoran Peng Lizhong Li Yi Yang Shi-Yong Fan Rong Xiang Jianshu Guo Jinjin Wang Juanning Wei Fengling Zhou Ding Liu Ping Zhao Yujun Cui Yunxi Liu Dongsheng Zhou Gang Dong 2023Engineering2023,,9:0
