1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2GPS-constrained inversion of present-day slip rates along major faults of the Sichuan-Yunnan region, China显示文摘A linked-fault-element model is employed to invert for contemporary slip rates along major active faults in the Sichuan-Yunnan region (96°-108°E, 21°-35°N) using the least squares method. The model is based on known fault geometry, and constrained by a GPS-derived horizontal velocity field. Our results support a model attributing the eastward extrusion of the Tibetan Plateau driven mainly by the north-northeastward indentation of the Indian plate into Tibet and the gravitational collapse of the plateau. Resisted by a relatively stable south China block, materials of the Sichuan-Yunnan region rotate clockwise around the eastern Himalayan tectonic syntaxis. During the process the Garzê-Yushu, Xianshuihe, Anninghe, Zemuhe, Daliangshan, and Xiaojiang faults, the southwest extension of the Xiaojiang fault, and the Daluo-Jinghong and Mae Chan faults constitute the northeast and east boundaries of the eastward extrusion, with their left slip rates being 0.3-14.7, 8.9-17.1, 5.1 ± 2.5, 2.8 ± 2.3, 7.1 ± 2.1, 9.4 ± 1.2, 10.1 ± 2.0, 7.3 ± 2.6, and 4.9 ± 3.0 mm/a respectively. The southwestern boundary consists of a widely distributed dextral transpressional zone other than a single fault. Right slip rates of 4.2 ± 1.3, 4.3 ± 1.1, and 8.5 ± 1.7 mm/a are detected across the Nanhua-Chuxiong-Jianshui, Wuliangshan, and Longling-Lancang faults. Crustal deformation across the Longmenshan fault is weak, with short-ening rates of 1.4 ± 1.0 and 1.6 ± 1.3 mm/a across the Baoxing-Beichuan and Beichuan-Qingchuan segments. Northwest of the Longmenshan fault lies an active deformation zone (the Longriba fault) with 5.1±1.2 mm/a right slip across. Relatively large slip rates are detected across a few faults within the Sichuan-Yunnan block: 4.4±1.3 mm/a left slip and 2.7±1.1 mm/a shortening across the Litang fault, and 2.7±2.3 mm/a right-lateral shearing and 6.7±2.3 mm/a shortening across the Yunongxi fault and its surrounding regions. In conclusion, we find that the Sichuan-Yunnan region is divided into more than a dozen active micro-blocks by a large number of faults with relatively slow slip rates. 32018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. 9Relationship between Cognition Function and Hippocampus Structure after Long-term Microwave Exposure显示文摘Objective To analyze the effects of long-term microwave exposure on hippocampal structure and function in the rat.Methods Experiments were performed on 184 male Wistar rats(three exposure groups and a sham group).Microwaves were applied daily for 6 min over 1 month at average power densities of 2.5,5,and 10 mW/cm2.Learning and memory abilities were assessed by Morris water maze.High performance liquid chromatography was used to detect neurotransmitter concentrations in the hippocampus.Hippocampal structures were observed by histopathological analysis.Results Following long-term microwave exposure there was a significant decrease in learning and memory activity in the 7 d,14 d,and 1 m in all three microwave exposure groups.Neurotransmitter concentrations of four amino acids(glutamate,aspartic acid,glycine,and gamma-aminobutyric acid) in hippocampus were increased in the 2.5 and 5 mW/cm2 groups and decreased in the 10 mW/cm2 group.There was evidence of neuronal degeneration and enlarged perivascular spaces in the hippocampus in the microwave exposure groups.Further,mitochondria became swollen and cristae were disordered.The rough endoplasmic reticulum exhibited sacculated distension and there was a decrease in the quantity of synaptic vesicles.Conclusion These data suggest that the hippocampus can be injured by long-term microwave exposure,which might result in impairment of cognitive function due to neurotransmitter disruption.ZHAO Li PENG Rui Yun WANG Shui Ming WANG Li Feng GAO Ya Bing DONG Ji LI Xiang SU Zhen Tao 2012Biomedical and Environmental Sciences2012,25,2:27
10Temperature effect and thermal impact in lithium-ion batteries: A review显示文摘Lithium-ion batteries, with high energy density(up to 705 Wh/L) and power density(up to 10,000 W/L), exhibit high capacity and great working performance. As rechargeable batteries, lithium-ion batteries serve as power sources in various application systems. Temperature, as a critical factor, significantly impacts on the performance of lithium-ion batteries and also limits the application of lithium-ion batteries. Moreover, different temperature conditions result in different adverse effects. Accurate measurement of temperature inside lithiumion batteries and understanding the temperature effects are important for the proper battery management. 11Magma mixing of granites at Lianghe:In-situ zircon analysis for trace elements, U-Pb ages and Hf isotopes显示文摘In this paper, we report an integrated study of trace element, U-Pb age and Hf isotopic composition of zircons from alkali feldspar granites, granodiorites and diorite enclaves in a recently discovered ring complex at Lianghe in western Yunnan, Chi na. The granitoids showed identical U-Pb ages of 127, 115 and 122 Ma, from felsic to mafic, but had different zircon trace el ements and Hf isotopic compositions. Trace element content decreased with a gradual increase in εHf(t) values of ?9.1 to ?5.4, ?4.5 to 0, and 3.6 to 6.2, respectively. Results indicate that changes in zircon trace elements generally correlate with changes in Hf isotope signatures within single samples and among various granitoids. These relationships reflect the mixing of felsic and mafic magmas. Evidence indicates that depleted mantle-derived mafic magma underplating caused ancient crustal melting, and then formed large-scale granites in Lianghe during the Early Cretaceous. 12Chinese Trauma Surgeon Association for management guidelines of vacuum sealing drainage application in abdominal surgeries-Update and systematic review显示文摘Vacuum sealing drainage (VSD) is frequently used in abdominal surgeries. However, relevant guidelines are rare. Chin ese Trauma Surge on Associati on orga nized a committee composed of 28 experts across China in July 2017, aiming to provide an evidence-based recommendation for the application of VSD in abdominal surgeries.Eleven questions regarding the use of VSD in abdominal surgeries were addressed:(1) which type of materials should be respectively chosen for the intraperitoneal cavity, retroperitoneal cavity and superficial incisions?(2) Can VSD be preventively used for a high-risk abdominal incision w让h primary suture?(3) Can VSD be used in severely contaminated/infected abdominal surgical sites?(4) Can VSD be used for temporary abdominal cavity closure under some special conditions such as severe abdominal trauma, infection, liver transplantation and intra-abdominal volume increment in abdominal compartment syndrome?(5) Can VSD be used in abdominal organ inflammation, injury, or postoperative drainage?(6) Can VSD be used in the treatment of intestinal fistula and pancreatic fistula?(7) Can VSD be used in the treatment of intra-abdominal and extra-peritoneal abscess?(8) Can VSD be used in the treatment of abdominal wall wounds, wound cavity, and defects?(9) Does VSD in crease the risk of bleeding?(10) Does VSD increase the risk of intestinal wail injury?(11) Does VSD increase the risk of peritoneal adhesion? Focusing on these questions, evidence-based recommendations were given accordingly. VSD was strongly recommended regarding the questions 2-4. Weak recommendations were made regarding questions 1 and 5-11. 13Evolution and Coarsening of Carbides in 2.25Cr-1Mo Steel Weld Metal During High Temperature Tempering显示文摘Transformation and coarsening of carbides in 2.25Cr-1Mo steel weld metal during tempering at 700 ℃ for different time intervals ranging from 1 to 150 h were examined by transmission electron microscopy and scanning electron microscopy. M3C carbides were observed in the as-welded specimens and when tempered, the precipitates were mainly composed of M3C, M7C3, and M23C6 carbides. A sequence for corresponding carbide transformation during tempering with initial precipitation of M3C and the subsequent precipitation of M7C3 and M23C6 was proposed. The precipitation of M7C3 with higher chromium content was the main factor contributing to the decrease in coarsening rate of precipitates after prolonged tempering. 15Lithium–matrix composite anode protected by a solid electrolyte layer for stable lithium metal batteries显示文摘Lithium (Li) metal with an ultrahigh specific theoretical capacity and the lowest reduction potential is strongly considered as a promising anode for high-energy-density batteries. However, uncontrolled lithium dendrites and infinite volume change during repeated plating/stripping cycles hinder its practical applications immensely. Herein, a house-like Li anode (housed Li) was designed to circumvent the above issues. The house matrix was composed of carbon fiber matrix and affords a stable structure to relieve the volume change. An artificial solid electrolyte layer was formed on composite Li metal, just like the roof of a house, which facilitates uniform Li ions diffusion and serves as a physical barrier against electrolyte corrosion. With the combination of solid electrolyte layer and matrix in the composite Li metal anode, both dendrite growth and volume expansion are remarkably inhibited. The housed Li|LiFePO4 batteries exhibited over 95% capacity retention after 500 cycles at 1.0 C in coin cell and 85% capacity retention after 80 cycles at 0.5 C in pouch cell. 16The recommendations of Chinese Parkinson’s disease and movement disorder society consensus on therapeutic management of Parkinson’s disease显示文摘Background:Parkinson’s disease(PD)is a chronic,progressive and debilitating disease,which affects over 2.5 million people in China.PD is characterized clinically by resting tremor,muscular rigidity,bradykinesia and postural instability.As the disease progresses,additional complications can arise such as non-motor and neurobehavioral symptoms.Pharmacological treatment and surgical intervention for PD have been implemented in China.Until 10 years ago,there was lack of standardization for the management of PD in different regions and among different physicians,leading to different treatment levels in different regions and different physicians.Since then,the Chinese Parkinson’s Disease and Movement Disorder Society have published three versions of guidelines for the management of PD in China,in 2006,2009 and 2014,respectively.Correspondingly,the overall level of treatment for PD in China improved.Objectives:To update the treatment guidelines based on current foreign and domestic practice guidelines and clinical evidence,and to improve the treatment options available to physicians in the management of PD.Summary:A variety of treatment recommendations in the treatment guidelines have been proposed,including physical activity and disease-modifying medication,which should be initiated at the early-stage of the disease.The principles of dosage titration should be followed to avoid acute adverse reactions to the drugs,to achieve a satisfactory clinical effect with a low dose and to reduce the incidence of long-term motor complications.Moreover,different treatment strategies should be considered at different stages of the disease.Importantly,treatment guidelines and personalized treatments should be valued equally.A set of treatment recommendations has been developed to assist physicians to improve and optimize clinical outcomes for patients with PD in China.Shengdi Chen Piu Chan Shenggang Sun Haibo Chen Baorong Zhang Weidong Le Chunfeng Liu Guoguang Peng Beisha Tang Lijuan Wang Yan Cheng Ming Shao Zhenguo Liu Zhenfu Wang Xiaochun Chen Mingwei Wang Xinhua Wan Huifang Shang Yiming Liu Pingyi Xu Jian Wang Tao Feng Xianwen Chen Xingyue Hu Anmu Xie Qin Xiao 2016Translational Neurodegeneration2016,5,1:23
17Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment.Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao 2021Acta Pharmaceutica Sinica B2021,11,12:23
18The Chromosome-Level Genome Sequence of the Autotetraploid Alfalfa and Resequencing of Core Germplasms Provide Genomic Resources for Alfalfa Research显示文摘Alfalfa(Medicago sativa)is one of the most important forage crops in the world;however,its molecular genetics and breeding research are hindered due to the lack of a high-quality reference genome.Here,we report a de novo assembled 816-Mb high-quality,chromosome-level haploid genome sequence for‘Zhongmu No.1’alfalfa,a heterozygous autotetraploid.The contig N50 is 3.92 Mb,and 49165 genes are annotated in the genome.The alfalfa genome is estimated to have diverged from M.truncatula approximately 8 million years ago.Genomic population analysis of 162 alfalfa accessions revealed high genetic diversity,weak population structure,and extensive gene flow from wild to cultivated alfalfa.Genome-wide association studies identified many candidate genes associated with important agronomic traits.Furthermore,we showed that MsFTa2,a Flowering Locus T homolog,whose expression is upregulated in salt-resistant germplasms,may be associated with fall dormancy and salt resistance.Taken together,these genomic resources will facilitate alfalfa genetic research and agronomic improvement.Chen Shen Huilong Du Zhuo Chen Hongwei Lu Fugui Zhu Hong Chen Xiangzhao Meng Qianwen Liu Peng Liu Lihua Zheng Xiuxiu Li Jiangli Dong Chengzhi Liang Tao Wang 2020Molecular Plant2020,13,9:21
19Overexpression of cyclooxygenase-2 in human HepG2, Bel-7402 and SMMC-7721 hepatoma cell lines and mechanism of cyclooxygenase-2 selective inhibitor celecoxib-induced cell growth inhibition and apoptosis显示文摘AIM: To investigate the cyclooxygenase-2 (COX-2)expression level in human HepG2, Bel-7402 and SMMC-7721hepatoma cell lines and the molecular mechanism of COX-2 selective inhibitor celecoxib-induced cell growth inhibition and cell apoptosis.METHODS: Hepatoma cells were cultured and treated with celecoxib. Cell in situ hybridization (ISH) and immunocytochemistry were used to detect COX-2 mRNA and protein expression. Proliferating cell nuclear antigen and phosphorylated Akt were also detected by immunocytochemistry assay. Cell growth rates were assessed by 3-(4, 5-dimethylthiazol-2-yl-2, 5-diphenyltetrazolium (MTT) bromide colorimetric assay. Celecoxibinduced cell apoptosis was measured by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) and flow cytometry (FCM). The phosphorylated Akt and activated fragments of caspase-9, caspase-3 were examined by Western blotting analysis.RESULTS: Increased COX-2 mRNA and protein expression were detected in all three hepatoma cell lines. Celecoxib could significantly inhibit cell growth and the inhibitory effect was in a dose- and time-dependent manner evidenced by MTr assays and morphological changes.The apoptotic index measured by TUNEL increased correspondingly with the increased concentration of celecoxib and the reaction time. With 50 μmol/L celecoxib treatment for 24 h, the apoptotic index of HepG2, BEL-7402and SMMC-7721 cells was 25.01±3.08%, 26.40±3.05%,and 30.60±2.89%, respectively. Western blotting analysis showed remarkable activation of caspase-9, caspase-3and dephosphorylation of Akt (Thr308). Immunocytochemistry also showed the reduction of PCNA expression and phosphorylation Akt (Thr308) after treatment with celecoxib.CONCLUSION: COX-2 mRNA and protein overexpression in HepG2, Bel-7402 and SMMC-7721 cell lines correlate with the increased cell growth rate. 20Subsoiling and Ridge Tillage Alleviate the High Temperature Stress in Spring Maize in the North China Plain显示文摘High temperature stress(HTS)on spring maize(Zea mays L.)during the filling stage is the key factor that limits the yield increase in the North China Plain(NCP).Subsoiling(SS)and ridge tillage(R)were introduced to enhance the ability of spring maize to resist HTS during the filling stage.The field experiments were conducted during the 2011 and 2012 maize growing seasons at Wuqiao County,Hebei Province,China.Compared with rotary tillage(RT),the net photosynthetic rate,stomatal conductance,transpiration rate,and chlorophyll relative content(SPAD)of maize leaves was increased by 40.0,42.6,12.8,and 29.7%under SS,and increased by 20.4,20.0,5.4,and 14.2%under R,repectively.However,the treatments reduce the intercellular CO2concentration under HTS.The SS and R treatments increased the relative water content(RWC)by 11.9 and6.2%,and the water use efficiency(WUE)by 24.3 and 14.3%,respectively,compared with RT.The SS treatment increased the root length density and soil moisture in the 0-80 cm soil profile,whereas the R treatment increased the root length density and soil moisture in the 0-40 cm soil profile compared with the RT treatment.Compared with 2011,the number of days with temperatures 33°C was more 2 d and the mean day temperature was higher 0.9°C than that in 2012,whereas the plant yield decreased by 2.5,8.5 and 10.9%,the net photosynthetic rate reduced by 7.5,10.5 and 18.0%,the RWC reduced by 3.9,5.6 and6.2%,and the WUE at leaf level reduced by 1.8,5.2 and 13.1%in the SS,R and RT treatments,respectively.Both the root length density and the soil moisture also decreased at different levels.The yield,photosynthetic rate,plant water status,root length density,and soil moisture under the SS and R treatments declined less than that under the RT treatment.The results indicated that SS and R can enhance the HTS resistance of spring maize during the filling stage,and led to higher yield by directly improving soil moisture and root growth and indirectly improving plant water status,photosynthesis and grain filling.The study can provide a theoretical basis for improving yield of maize by adjusting soil tillage in the NCP.TAO Zhi-qiang SUI Peng CHEN Yuan-quan LI Chao NIE Zi-jin YUAN Shu-fen SHI Jiang-tao GAO Wang-sheng 2013Journal of Integrative Agriculture2013,12,12:20
