1Five-year follow-up of 23 asymmetrical Parkinson's disease patients treated with unilateral subthalamic nucleus stimulation显示文摘In this study, 23 asymmetrical Parkinson's disease patients were treated with unilateral deep brain stimulation of the subthalamic nucleus and followed up for 5 years. At 5 years after stimulation treatment, Unified Parkinson's Disease Rating Scale II, III and axial symptom scores in the off-drug condition were significantly increased compared those at baseline. However, total Unified Parkinson's Disease Rating Scale II, III and axial symptom scores were significantly lower with stimulation-on compared with the synchronous stimulation-off state in off-drug condition, and the motor symptoms of contralateral side limbs were effectively controlled. Only low Hoehn-Yahr stage was correlated with good long-term postoperative improvement in motor symptoms. The mean levodopa-equivalent daily dose after stimulation treatment was significantly lower than that before treatment, but dyskinesias became worse. 2Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART.Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu 2021Cell Research2021,31,8:2
3Combining immune checkpoint blockade with ATP-based immunogenic cell death amplifier for cancer chemo-immunotherapy显示文摘Amplifying“eat me signal”during tumor immunogenic cell death(ICD)cascade is crucial for tumor immunotherapy.Inspired by the indispensable role of adenosine triphosphate(ATP,a necessary“eat me signal”for ICD),a versatile ICD amplifier was developed for chemotherapy-sensitized immunotherapy.Doxorubicin(DOX),ATP and ferrous ions(Fe^(2+))were co-assembled into nanosized amplifier(ADO-Fe)throughπ‒πstacking and coordination effect.Meanwhile,phenylboric acid-polyethylene glycol-phenylboric acid(PBA-PEG-PBA)was modified on the surface of ADO-Fe(denoted as PADO-Fe)by the virtue of d-ribose unit of ATP.PADO-Fe could display active targetability against tumor cells via sialic acid/PBA interaction.In acidic microenvironment,PBA-PEG-PBA would dissociate from amplifier.Moreover,high H_(2)O_(2)concentration would induce hydroxyl radical(·OH)and oxygen(O_(2))generation through Fenton reaction by Fe^(2+).DOX and ATP would be released from the amplifier,which could induce ICD effect and“ICD adjuvant”to amplify this process.Together with programmed death ligands 1(PD-L1)checkpoint blockade immunotherapy,PADO-Fe could not only activate immune response against primary tumor,but also strong abscopal effect against distant tumor.Our simple and multifunctional ICD amplifier opens a new window for enhancing ICD effect and immune checkpoint blockade therapy.Jiulong Zhang Xiaoyan Sun Xiufeng Zhao Chunrong Yang Menghao Shi Benzhuo Zhang Haiyang Hu Mingxi Qiao Dawei Chen Xiuli Zhao 2022Acta Pharmaceutica Sinica B2022,12,9:2

10PCA-U-Net based breast cancer nest segmentation from microarray(jj)hyperspectral images显示文摘The incidence of breast cancer is tending younger globally,and tumor development,clinical treatment,and prognosis are largely influenced by histopathological diagnosis.For diagnosed patients,the distinction between the cancer nests and normal tissue is the basis of breast cancer treatment.Microscopic hyperspectral imaging technology has shown its potential in auxiliary pathological examinations due to the superior imaging modality and data characteristics.This paper presents a method for cancer nest segmentation from hyperspectral images of breast cancer tissue microarray samples.The scheme combines the strengths of the U-Net neural network and unsupervised principal component analysis,which reduces the amount of calculation and improves the recognition accuracy.The experimental accuracy of cancer nest segmentation reaches 87.14%.Furthermore,a set of quantitative pathological characteristic parameters reflects the degree of breast cancer lesions from multiple angles,providing a relatively comprehensive reference for the pathologist’s diagnosis.In-depth exploration of the combined development of deep learning and microscopic hyperspectral imaging technology is worthy to promote efficient diagnosis of breast tumors and concern for human health.Jiansheng Wang Yan Wang Xiang Tao Qingli Li Li Sun Jiangang Chen Mei Zhou Menghan Hu Xiufeng Zhou 2021Fundamental Research2021,1,5:1

12A nomogram predicting clinical pregnancy in the first fresh embryo transfer for women undergoing in vitro fertilization and intracytoplasmic sperm injection(IVF/ICSI) treatments显示文摘The extent to which factors affect the probability of clinical pregnancy in the first fresh embryo transfer after assisted conception is unknown.In order to examine the predictors of clinical pregnancy,a retrospective cohort study was launched between January 1,2013 and December 31,2016 in four infertility clinics including 19837 in vitro fertilization and intracytoplasmic sperm injection(IVF/ICSI)fresh cycles with known outcomes and relevant records.A multivariable logistic regression was used to select the most significant predictors in the final nomogram for predicting clinical pregnancy.Furthermore,the model was validated by an independent validation set and the performance of the model was evaluated by the receiver operating characteristic(ROC)curves along with the area under the ROC curve(AUC)and calibration plots.In a training set including 17854 participants,we identified that female age,tubal factor,number of embryos transferred,endometrial thickness and number of good-quality embryos were independent predictors for clinical pregnancy.We developed a nomogram using these five factors and the predictive ability was 0.66 for AUC(95%CI=0.64−0.68),which was independently validated in the validation set(AUC=0.66,95%CI=0.65−0.68).Our results show that some specific factors can be used to provide infertile couples with an accurate assessment of clinical pregnancy following assisted conception and facilitate to guide couples and clinicians.Fang Wu Feng Liu Yichun Guan Jiangbo Du Jichun Tan Hong Lv Qun Lu Shiyao Tao Lei Huang Kun Zhou Yankai Xia Xinru Wang Hongbing Shen Xiufeng Ling Feiyang Diao Zhibin Hu Guangfu Jin 2019The Journal of Biomedical Research2019,33,6:1

15Extraction and Antioxidant Activity Analysis of Crude Polysaccharides from Wild Lactarius volemus Fr. in Yunnan Province显示文摘[Objective]This study aimed to investigate the optimal extraction process of crude polysaccharides from wild Lactarius volemus Fr. in Yunnan Province and preliminarily analyzed its antioxidant activity in vitro. [Method] With water extraction and alcohol precipitation method,the optimal conditions for extracting crude polysaccharides from wild L. volemus Fr. were screened by single-factor and orthogonal experiments. The antioxidant activity of the extracted crude polysaccharides was determined with DPPH assay. [Result] The optimal conditions for pigment removal with activated carbon were: activated carbon amount of 20 g / L,water bath time of 40 min,water bath temperature of 40 ℃; the optimal conditions for extracting crude polysaccharides from wild L. volemus Fr. with hot water extraction method were: hot water extraction time of 3 h,solid-liquid ratio of 1∶ 45,extraction frequency of twice. Under the optimized extraction conditions,the yield of crude polysaccharides was 21. 33 mg / g. In addition,the antioxidant activity of 0. 665 mg / ml crude polysaccharides was 52. 46%; the amount of crude polysaccharides was proportional to the antioxidant activity. [Conclusion]Hot water extraction method can be used as a high-efficiency extraction technology of crude polysaccharides from wild L. volemus Fr. with simple operation and low costs. 16Genetic polymorphisms of 16 X-STR loci in the Hani population from Southwest China显示文摘X chromosomal short tandem repeats(X-STRs)have the characteristics of both autosomal and uniparental genetic markers and have been shown to be particularly useful in forensic casework.However,relevant research or reports have not focused on X-STRs in the Hani population.To investigate the genetic variation and forensic efficiency of 16 X-STR loci in the Hani ethnic minority,we calculated the allele frequencies and forensic parameters of 451(116 males and 335 females)unrelated healthy Hani individuals from Yunnan Province,Southwest China.All these loci are highly polymorphic in Hani individuals in Yunnan Province except DXS6800.The combined power of discrimination in males(PDM)and power of discrimination in females(PDF)were found to be 0.999999998433993 and 0.999999999999998,respectively.Furthermore,a population genetic structure investigation between the Yunnan Hani population and another 18 populations was performed using a principal component analysis,multidimensional scaling plot and neighbouring-joining phylogenetic tree and the findings illustrated that neighbouring populations and different nationalities in the same area appeared to have a closer evolutionary relationship.This study provides the first batch of X chromosome genetic polymorphism data of the Hani population in Yunnan Province,Southwest China and enriches the reference database of the Chinese minority population.Linlin Liu Jinyong Yao Yangzhi Huang Lei Gao Jiameng Dai Xiaokun Yuan Xiufeng Zhang Shengjie Nie Liping Hu 2022Forensic Sciences Research2022,7,2:0
17Effects of dendritic cell and subgroup changes on bone marrow transplantation treatment of multiple sclerosis显示文摘BACKGROUND:Bone marrow transplantation is an effective treatment for severe forms of various autoimmune disorders.Dendritic cell reconstitution is thought to be one factor contributing to host immune recovery and therapeutic efficacy. OBJECTIVE:To assess the effects of bone marrow transplantation on an animal model of experimental autoimmune encephalomyelitis(EAE),and to investigate changes in dendritic cells and subgroups following bone marrow transplantation. DESIGN,TIME AND SETTING:This experimental,neuroimmunological study was performed in Sun Yat-sen University between August 2006 and May 2007. MATERIALS:A total of 30 female C57BL/6 mice,aged 6-8 weeks,served as recipients,and 20 female adult C57BL/6 served as donors.Myelin oligodendrocyte glycoprotein 35- 55 amino acid peptide(MOG35-55) of murine origin was synthesized by Bio-Scientific(Xi'an,China,purity>95%). Complete Freund's adjuvant was purchased from Difco Laboratories,Detroit,Ml;pertussis vaccine was purchased from Alexis,San Diego,CA;radiation device and flow cytometry for FACS analysis were purchased from Theratron 780-C,Canada and Coulter,Fullerton,CA,respectively. METHODS:The C57BL/6 mice,aged 6-8 weeks,were immunized by subcutaneous injection of MOG35-55 peptide emulsified in complete Freund's adjuvant,which contained 500μg Mycobacterium tuberculosis H37RA.The mice were subsequently intravenously injected with pertussis vaccine to induce EAE.The mice were randomly assigned to transplantation and EAE model groups(n = 12 for each).Bone marrow cells[(5-10)×10~6]were transplanted into EAE mice via the tail vein 4-6 hours following total body irradiation,and the model group was not treated. MAIN OUTCOME MEASURES:Mouse behavioral changes following EAE were evaluated daily. Injured spinal cord tissue sections were stained with hematoxylin and eosin 20 days after the initial immunization to observe inflammatory infiltration using light microscopy.Flow cytometry was used to detect the ratio and absolute number of DC1(CD8a^-CD11c^+) and DC2(CD8a^+CD11c^+) in peripheral blood 36 days after bone marrow transplantation. RESULTS:Significant improvement in clinical signs was observed in EAE mice following bone marrow transplantation compared with the model group(P<0.01),as well as attenuated lymphocyte and macrophage infiltration.Compared with the model group,the absolute number of dendritic cells and DC1,as well as the DC1/DC2 ratio,was significantly greater following bone marrow transplantation(P<0.05 or P<0.01). 18Modifications in aerosol physical, optical and radiative properties during heavy aerosol events over Dushanbe, Central Asia显示文摘The location of Central Asia,almost at the center of the global dust belt region,makes it susceptible for dust events.The studies on atmospheric impact of dust over the region are very limited despite the large area occupied by the region and its proximity to the mountain regions (Tianshan,Hindu Kush-KarakoramHimalayas,and Tibetan Plateau).In this study,we analyse and explain the modification in aerosols’physical,optical and radiative properties during various levels of aerosol loading observed over Central Asia utilizing the data collected during 2010–2018 at the AERONET station in Dushanbe,Tajikistan.Aerosol episodes were classified as strong anthropogenic,strong dust and extreme dust.The mean aerosol optical depth (AOD) during these three types of events was observed a factor of ~3,3.5 and 6.6,respectively,higher than the mean AOD for the period 2010–2018.The corresponding mean fine-mode fraction was 0.94,0.20 and 0.16,respectively,clearly indicating the dominance of fine-mode anthropogenic aerosol during the first type of events,whereas coarse-mode dust aerosol dominated during the other two types of events.This was corroborated by the relationships among various aerosol parameters (AOD vs.AE,and EAE vs.AAE,SSA and RRI).The mean aerosol radiative forcing (ARF) at the top of the atmosphere (ARF_(TOA)),the bottom of the atmosphere (ARF_(BOA)),and in the atmosphere (ARFATM) were -35±7,-73±16,and38±17 Wm^(-2)during strong anthropogenic events,-48±12,-85±24,and 37±15 Wm^(-2)during strong dust event,and -68±19,-117±38,and 49±21 Wm-2during extreme dust events.Increase in aerosol loading enhanced the aerosol-induced atmospheric heating rate to 0.5–1.6 K day^(-1)(strong anthropogenic events),0.4–1.9 K day^(-1)(strong dust events) and 0.8–2.7 K day^(-1)(extreme dust events).The source regions of air masses to Dushanbe during the onset of such events are also identified.Our study contributes to the understanding of dust and anthropogenic aerosols,in particular the extreme events and their disproportionally high radiative impacts over Central Asia.Dipesh Rupakheti Maheswar Rupakheti Xiufeng Yin Julian Hofer Mukesh Rai Yuling Hu Sabur F.Abdullaev Shichang Kang 2021Geoscience Frontiers2021,12,6:0

20Carbonaceous aerosol transport from the Indo-Gangetic Plain to the Himalayas:Carbon isotope evidence and light absorption characteristics显示文摘The Indo-Gangetic Plain(IGP)is a major regional and global emitter of atmospheric pollutants,which adversely affect surrounding areas such as the Himalayas.We present a comprehensive dataset on carbonaceous aerosol(CA)composition,radiocarbon(D14C)-based source apportionment,and light absorption of total suspended particle(TSP)samples collected over a 3-year period from high-altitude Jomsom in the central Himalayas.The 3-year mean TSP,organic carbon(OC),and elemental carbon(EC)concentrations were 92.0±28.6,9.74±6.31,and 2.02±1.35 lg m^(3),respectively,with the highest concentrations observed during the pre-monsoon season,followed by the post-monsoon,winter,and monsoon seasons.The △^(14)C analysis revealed that the contribution of fossil fuel combustion(ffossil)to EC was 47.9%±11.5%,which is consistent with observations in urban and remote regions in South Asia and attests that EC likely arrives in Jomsom from upwind IGP sources via long-range transport.In addition,the lowest f_(fossil)(38.7%±13.3%)was observed in winter,indicating large contributions in this season from local biomass burning.The mass absorption cross-section of EC(MACEC:8.27±1.76 m^(2)/g)and watersoluble organic carbon(MACWSOC:0.98±0.45 m^(2)/g)were slightly higher and lower than those reported in urban regions,respectively,indicating that CA undergo an aging process.Organic aerosol coating during transport and variation of biomass burning probably led to the seasonal variation in MAC of two components.Overall,WSOC contributed considerably to the light absorption(11.1%±4.23%)of EC.The findings suggest that to protect glaciers of the Himalayas from pollution-related melting,it is essential to mitigate emissions from the IGP.Pengfei Chen Shichang Kang Chaoliu Li Zhaofu Hu Lekhendra Tripathee Mukesh Rai Tao Pu Xiufeng Yin Örjan Gustafsson 2023Geoscience Frontiers2023,14,2:0
