1Inhibition of signal transducer and activator of transcription 3 expression by RNA interference suppresses invasion through inducing anoikis in human colon cancer cells显示文摘AIM:To investigate the roles and mechanism of signal transducer and activator of transcription 3 (STAT3) in invasion of human colon cancer cells by RNA interference. METHODS: Small interfering RNA (siRNA) targeting Signal transducer and activator of transcription 3 (STAT3) was transfected into HT29 colon cancer cells. STAT3 protein level and DNA-binding activity of STAT3 was evaluated by western blotting and electrophoretic mobility shift assay (EMSA), respectively. We studied the anchorage-independent growth using colony formation in soft agar, and invasion using the boyden chamber model, anoikis using DNA fragmentation assay and terminal deoxynucleotidyltransferase-mediated dUTP nick-end labeling (TUNEL), respectively. Western blot assay was used to observe the protein expression of Bcl-xL and survivin in colon cancer HT29 cells. RESULTS: RNA interference (RNAi) mediated by siRNA leads to suppression of STAT3 expression in colon cancer cell lines. Suppression of STAT3 expression by siRNA could inhibit anchorage-independent growth, and invasion ability, and induces anoikis in the colon cancer cell line HT29. It has been shown that knockdown of STAT3 expression by siRNA results in a reduction in expression of Bcl-xL and survivin in HT29 cells. 22018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 3Programmable time-domain digital-coding metasurface for non-linear harmonic manipulation and new wireless communication systems显示文摘Optical non-linear phenomena are typically observed in natural materials interacting with light at high intensities, and they benefit a diverse range of applications from communication to sensing. However,controlling harmonic conversion with high efficiency and flexibility remains a major issue in modern optical and radio-frequency systems. Here, we introduce a dynamic time-domain digital-coding metasurface that enables efficient manipulation of spectral harmonic distribution. By dynamically modulating the local phase of the surface reflectivity, we achieve accurate control of different harmonics in a highly programmable and dynamic fashion, enabling unusual responses, such as velocity illusion. As a relevant application, we propose and realize a novel architecture for wireless communication systems based on the time-domain digital-coding metasurface, which largely simplifies the architecture of modern communication systems, at the same time yielding excellent performance for real-time signal transmission. 4Effects of large dose of dexamethasone on inflammatory mediators and pancreatic cell apoptosis of rats with severe acute pancreatitis显示文摘AIM: To investigate the influence of high dose of dexamethasone on inflammatory mediators and apoptosis of rats with severe acute pancreatitis (SAP). METHODS: SAP rats were randomly assigned to the model group and treatment group while the normal rats were assigned to the sham operation group. The mortality,ascite volumes,ascites/body weight ratio and pancreas pathological changes of all rats were observed at 3,6 and 12 h after operation. Their contents of amylase and endotoxin in plasma and contents of tumor necrosis factor (TNF-α),phospholipase A2 (PLA2) and IL-6 in serum were also determined. The microarray sections of their pancreatic tissues were prepared,terminal transferase dUTP nick end labeling (TUNEL) staining was performed and apoptotic indexes were calculated. RESULTS: There was no marked difference between treatment group and model group in survival. The contents of amylase and endotoxin in plasma and contents of TNF-α,PLA2 and IL-6 in serum,ascite volumes,ascites/body weight ratio and pancreas pathological scores were all lower in treatment group than in model group to different extents at different time points P < 0.05,58.3 (26.4) ng/L vs 77.535 (42.157) ng/L in TNF-α content,8.00 (2.00) points vs 9.00 (2.00) points in pathological score of pancreas respectively; P < 0.01,0.042 (0.018) EU/mL vs 0.056 (0.0195) EU/mL in endotoxin content,7791 (1863) U/L vs 9195 (1298) U/L in plasma amylase content,1.53 (0.79) vs 2.38 (1.10) in ascites/body weight ratio,8.00 (1.00) points vs 11.00 (1.50) points in pathological score of pancreas; P < 0.001,3.36 (1.56) ng/L vs 5.65 (1.08) ng/L in IL-6 content,4.50 (2.00) vs 7.20 (2.00),4.20 (1.60) vs 6.40 (2.30),3.40 (2.70) vs 7.90 (1.70) in ascite volumes,respectively. 5SIMULATION OF FLUID-SOLID INTERACTION ON WATER DITCHING OF AN AIRPLANE BY ALE METHOD显示文摘Ditching is considered as one of the important aspects of safety performances of airplanes.It is related primarily with the fluid-solid interaction,whose studies mainly depend on experiments at the present time.Numerical and analytical methods for fluid-solid interaction by using 3-D full scale airplane's model will reduce the dependence on the expensive model tests.Numerical studies can be used to estimate the safety of ditching and provide a reference for the crashworthiness design.This article proposes a 3-D dynamical structural model after the real shape of an airplane and an Arbitrary Lagrange-Euler (ALE) fluid-field model,to simulate the fluid-solid interactions caused by low speed ditching.The simulation is based on interaction computational methods,within LS-DYNA nonlinear finite-element code.The results of pressure distributions and accelerating time histories of the airplane's subfloor are discussed in the context of the safety of ditching,and the simulation results and the analytical methods are verified.HUA Cheng FANG Chao CHENG Jin 2011Journal of Hydrodynamics2011,23,5:20

8Protective effects and mechanisms of Baicalin and octreotide on renal injury of rats with severe acute pancreatitis显示文摘AIM: To investigate the protective effects and mechanisms of Baicalin and octreotide on renal injury of rats with severe acute pancreatitis (SAP). METHODS: One hundred and eighty SD rats were randomly assigned to the model group, Baicalin-treated group, octreotide-treated group and sham operation group. The mortality, plasma endotoxin level, contents of blood urea nitrogen (BUN), creatinine (CREA), phospholipase A2 (PLA2), nitrogen monoxide (NO), tumor necrosis factor (TNF)-α, IL-6 and endothelin-1 (ET-1) in serum, expression levels of renal Bax and Bcl-2 protein, apoptotic indexes and pathological changes of kidney were observed at 3, 6 and 12 h after operation. RESULTS: The renal pathological changes were milder in treated group than in model group. The survival at 12 h and renal apoptotic indexes at 6 h were significantly (P < 0.05) higher in treated group than in model group [66.67% vs 100%; 0.00 (0.02)% and 0.00 (0.04)% vs 0.00 (0.00)%, respectively]. The serum CREA content was markedly lower in octreotide-treated group than in model group at 3 h and 6 h (P < 0.01, 29.200 ± 5.710 μmol/L vs 38.400 ± 11.344 μmol/L; P < 0.05, 33.533 ± 10.106 μmol/L vs 45.154 ± 17.435 μmol/L, respectively). The expression level of renal Bax protein was not significantly different between model group and treated groups at all time points. The expression level of renal Bcl-2 protein was lower in Baicalin-treated group than in model group at 6 h [P < 0.001, 0.00 (0.00) grade score vs 3.00 (3.00) grade score]. The Bcl-2 expression level was lower in octreotide-treated group than in model group at 6 h and 12 h [P < 0.05, 0.00 (0.00) grade score vs 3.00 (3.00) grade score; 0.00 (0.00) grade score vs 0.00 (1.25) grade score, respectively]. The serum NO contents were lower in treated groups than in model group at 3 h and 12 h [P < 0.05, 57.50 (22.50) and 52.50 (15.00) μmol/L vs 65.00 (7.50) μmol/L; P < 0.01, 57.50 (27.50) and 45.00 (12.50) μmol/L vs 74.10 (26.15) μmol/L, respectively]. The plasma endotoxin content and serum BUN content (at 6 h and 12 h) were lower in treated groups than in model group. 9Antigenic variation of the human influenza A(H3N2) virus during the 2014–2015 winter season显示文摘The human influenza A(H3N2) virus dominated the 2014–2015 winter season in many countries and caused massive morbidity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolat ed since 2010, we discov ered that two groups with obvious antigenic divergence, named SW13(A/Switzerland/9715293/2013-like strains) and HK14(A/Hong Kong/5738/2014-like strains), co-circulated during the 2014–2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an important role in the antigenic variation of both the groups. 12SSR Analysis of Genetic Relationship and Classification in Chrysanthemum Germplasm Collection显示文摘To further test the accuracy and rationality of the Chinese classification system for chrysanthemum(Chrysanthemum morifolium), 10 simple sequence repeat(SSR) markers were used to identify a collection of 88 chrysanthemum and its related genera accessions. In total, 42 effective alleles across 88 accessions were detected; 3 429 bands were obtained by PCR amplification, including 2 630 polymorphic bands. The similarity coefficient ranged from 0.53 to 0.88. Cluster analysis based on UPGMA illustrated that the wild species and large-flower cultivars were first divided into two clusters, then the large-flower cultivars formed five distinct groups according to petal type, indicated that petal type can be a classification criterion. In the wild species cluster, C. vestitum and C. zawadaskii grouped with A. trilobata, suggested that the Ajania genera was closely related to the Chrysanthemum genera. ‘Hangbaiju’, ‘Gongju’ and ‘Chuju’ were grouped together, and ‘Boju’, O. longilobus and C. mongolicum constituted another branch, showed a correlation with geographic region of origin. Population structure analysis was subsequently performed with K values ranging from 2 to 10, and the most likely estimate for the population structure is five subpopulations, which is nearly consistent with the clustering results. Principle component analysis was further performed to verify the classification results. 13Intense Pulsed Light Therapy for Patients with Meibomian Gland Dysfunction and Ocular Demodex Infestation显示文摘To observe the clinical changes of meibomian gland dysfunctipn(MGD)and ocular Demodex infestation after intense pulsed light(IPL)treatment to further examine the mechanism of IPL treating patients with MGD and ocular Demodex infestation.The medical records of 25 patients(49 eyes)with MGD treated with IPL,were retrospectively examined to determine outcomes.Associated ocular-surface parameters(ocular surface disease index,OSD1;lipid layer thickness,LLT;noninvasive first breakup time,NIF-BUT;noninvasive average breakup time,NIAvg?BUT;tear film breakup area,TBUA;Schirmer I Test,SIT;corneal fluorescein staining,CFS),eyelid margin abnormalities,meibum quality and expressibility,MG morphological parameters(macrostructure and microstructure),and the number of Demodex infestation were examined before and after treatment.The MG microstructure and the Demodex infestation were examined via in vivo confocal microscopy(IVCM).The results showed that there were statistically significant differences in associated ocular-surface parameters(all P<0.05)before and after IPL treatment,except SIT(P=0.065).Eyelid margin abnormalities,meibum quality and expressibility obviously improved in upper and lower eyelid after IPL treatment(all P<0.0001).MG macrostructure(MG dropouts)decreased in upper(P=0.002)and lower eyelid(P=0.001)after IPL treatment.The nine parameters of MG microstructure in upper and lower eyelid all distinctly improved after IPL treatment(all P<0.0001).The mean number of Demodex mites on the upper lid margin(6.59±7.16 to 3.12±3.81/9 eyelashes)and lower lid margin(2.55±2.11 to 1.29±1.53/9 eyelashes)significantly reduced after IPL treatment(all P<0.0001).The Demodex eradication rate was 20%(8/40)in upper lid margin and 34.15%(14/41)in lower lid margin.These findings indicate that IPL shows great therapeutic potential for patients of MGD and ocular Demodex infestation.Sheng-nan CHENG Fa-gang JIANG Hua CHEN Hui GAO Yu-kan HUANG 2019Current Medical Science2019,39,5:15

15Hepatitis B surface antigen clearance in inactive hepatitis B surface antigen carriers treated with peginterferon alfa-2a显示文摘AIM: To examine the association between interferon(IFN) therapy and loss of hepatitis B surface antigen(HBs Ag) in inactive HBs Ag carriers. METHODS: This was a retrospective cohort study in inactive HBs Ag carriers, who were treatment-naive, with a serum HBs Ag level < 100 IU/m L and an undetectable hepatitis B virus(HBV) DNA level(< 100 IU/m L). All the 20 treated patients received subcutaneous PEG-IFN alfa-2a 180 μg/wk for 72 wk and were then followed for 24 wk. There were 40 untreated controls matched with 96 wk of observation. Serum HBs Ag, HBV DNA, and alanine aminotransferases were monitored every 3 mo in the treatment group and every 3-6 mo in the control group. RESULTS: Thirteen(65.0%) of 20 treated patients achieved HBs Ag loss, 12 of whom achieved HBs Ag seroconversion. Mean HBs Ag level in treated patients decreased to 6.69 ± 13.04 IU/m L after 24 wk of treatment from a baseline level of 26.22 ± 33.00 IU/m L. Serum HBV DNA level remained undetectable(< 100 IU/m L) in all treated patients during the study. HBs Ag level of the control group decreased from 25.72 ± 25.58 IU/m L at baseline to 17.11 ± 21.62 IU/m L at week 96(P = 0.108). 17Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 18Xa1 Allelic R Genes Activate Rice Blight Resistance Suppressed by Interfering TAL Effectors显示文摘Xanthomonas oryzae pathovar oryzae(Xoo)uses transcription activator-like effectors(TALEs)to cause bacterial blight(BB)in rice.In turn,rice has evolved several mechanisms to resist BB by targeting TALEs.One mechanism involves the nucleotide-binding leucine-rich repeat(NLR)resistance gene Xa1 and TALEs.Reciprocally,Xoo has evolved TALE variants,C-terminally truncated versions(interfering TALEs or iTALEs),to overcome Xa1 resistance.However,it remains unknown to what extent the two co-adaptive mechanisms mediate Xoo–rice interactions.In this study,we cloned and characterized five additional Xa1 allelic R genes,Xa2,Xa31(t),Xa14,CGS-Xo111,and Xa45(t)from a collection of rice accessions.Sequence analysis revealed that Xa2 and Xa31(t)from different rice cultivars are identical.These genes and their predicted proteins were found to be highly conserved,forming a group of Xa1 alleles.The XA1 alleles could be distinguished by the number of C-terminal tandemrepeats consisting of 93 amino acid residues and ranged from four in XA14 to seven in XA45(t).Xa1 allelic genes were identified in the 3000 rice genomes surveyed.On the other hand,iTALEs could suppress the resistance mediated by Xa1 allelic R genes,and iTALE genes were prevalent(95%)in Asian,but not in African Xoo strains.Our findings demonstrate the prominence of a defense mechanism in which rice depends on Xa1 alleles and a counteracting mechanismin which Xoo relies on iTALEs for BB.Chonghui Ji Zhiyuan Ji Bo Liu He Cheng Hua Liu Sanzhen Liu Bing Yang Gongyou Chen 2020Plant Communications2020,1,4:14
19The behavior of a rectangular closed diaphragm wall when used as a bridge foundation显示文摘The rectangular closed diaphragm(RCD)wall is a new type of bridge foundation.Compared to barrette foundation,measuring the performance of RCD walls is relatively complicated because of their incorporation of a soil core.Using the FLAC3D software,this paper investigates the deformation properties,soil resistance and skin friction of a laterally loaded RCD wall as well as the settlement,axial force and load-sharing ratio of a vertically loaded RCD wall.Special attention is given to the analysis of factors that influence the performance of the soil core.It was found that under lateral loading,the RCD wall behaves as an end-bearing friction wall during the entire loading process.The relative displacement between the wall body and the soil core primarily occurs below the rotation point,and the horizontal displacement of the soil core is greater than that of the wall body.Under vertical loading,the degree of inner skin friction around the bottom of the soil core and the proportion of the loading supported by the soil core increase with increased cross-section size.The wall depth is directly proportional to the loading supported by the outer skin friction and the tip resistance of the wall body and is inversely proportional to the loading borne by the soil core.Qiangong CHENG Jiujiang WU Zhang SONG Hua WEN 2012Frontiers of Structural and Civil Engineering2012,6,4:13
20The WRKY transcription factor HpWRKY44 regulates CytP450-like1 expression in red pitaya fruit(Hylocereus polyrhizus)显示文摘Red pitaya(Hylocereus polyrhizus)fruit is a high-value,functional food,containing a high level of betalains.Several genes potentially related to betalain biosynthesis,such as cytochrome P450-like(CytP450-like),have been identified in pitaya fruit,while their transcriptional regulation remains unclear.In this work,the potential involvement of a WRKY transcription factor,HpWRKY44,in regulating CytP450-like1 expression in pitaya fruit was examined.HpWRKY44,a member of the Group 1 WRKY family,contains two conserved WRKY motifs and is localized in the nucleus.HpWRKY44 also exhibits trans-activation ability.Gene expression analysis showed that the expression of HpCytP450-like1 and HpWRKY44 increased steadily during pitaya fruit coloration,which corresponded with the production of elevated betalain levels in the fruit.HpWRKY44 was also demonstrated to directly bind to and activate the HpCytP450-like1 promoter via the recognition of the W-box element present in the promoter.Collectively,our findings indicate that HpWRKY44 transcriptionally activates HpCytP450-like1,which perhaps,at least in part,contributes to betalain biosynthesis in pitaya fruit.The information provided in the current study provides novel insights into the regulatory network associated with betalain biosynthesis during pitaya fruit coloration.Mei-nv Cheng Zi-juan Huang Qing-zhu Hua Wei Shan Jian-fei Kuang Wang-jin Lu Yong-hua Qin Jian-ye Chen 2017Horticulture Research2017,4,1:13
