1Age-related macular degeneration: Epidemiology, genetics, pathophysiology, diagnosis, and targeted therapy显示文摘Age-related macular degeneration (AMD) is a complex eye disorder and is the leading cause of incurable blindness worldwide in the elderly. Clinically, AMD initially affects the central area of retina known as the macula and it is classified as early stage to late stage (advanced AMD). The advanced AMD is classified into the nonexudative or atrophic form (dry AMD) and the exudative or neovascular form (wet AMD). More severe vision loss is typically associated with the wet form. Multiple genetic factors, lipid metabolism, oxidative stress and aging, play a role in the etiology of AMD. Dysregulation in genetic to AMD is established to 46%–71% of disease contribution, with CFH and ARMS2/HTRA1 to be the two most notable risk loci among the 103 identified AMD associated loci so far. Chronic cigarette smoking is the most proven consistently risk living habits for AMD. Deep learning algorithm has been developed based on image recognition to distinguish wet AMD and normal macula with high accuracy. Currently, anti-vascular endothelial growth factor (VEGF) therapy is highly effective at treating wet AMD. 2MicroRNA in vivo precipitation identifies miR-151-3p as a computational unpredictable miRNA to target Stat3 and inhibits innate IL-6 production显示文摘MicroRNAs(miRNAs)function as important regulators in the immune response and inflammation.Several approaches have been reported to computationally predict miRNAs and their potential targets.However,there are still many miRNA–target interactions that are unpredictable by using the current computational algorithms.We established a miRNA in vivo precipitation method(miRIP)to identify unpredictable miRNAs with definite targets in these cells.Because Stat3 is a well-known transcription factor involved in innate immunity and inflammation,we utilized the miRIP method to identify miRNAs that bind Stat3 mRNA in macrophages.Among the captured miRNAs,miR-151-3p was confirmed to interact with Stat3 mRNA 3′-UTR and downregulate the Stat3 protein levels.LPS stimulation decreased miR-151-3p expression,thereby increasing IL-6 production.Therefore,we found that miR-151-3p inhibited LPS-induced IL-6 production by targeting Stat3.These data further confirmed miRIP as an efficient method to identify unpredictable miRNAs and explore miRNAs-mediated regulation in innate immunity and inflammation.Xiang Liu Xiaoping Su Sheng Xu Huamin Wang Dan Han Jiangxue Li Mingyan Huang Xuetao Cao 2018Cellular & Molecular Immunology2018,15,2:8

4Effectiveness and safety of Suxiao Jiuxin pill in treating acute coronary syndrome:a systematic review and Meta-analysis显示文摘OBJECTIVE:To evaluate the effectiveness and safety of Suxiao Jiuxin pill(SX)in acute coronary syndrome(ACS)treatment.METHODS:An extensive search of four English databases(Medline/PubMed,Cochrane Library,Embase,and World Health Organization International Clinical Trials Registration Platform)and four Chinese databases(Chinese National Knowledge Infrastructure,Wanfang,China Science and Technology Journal,and Chinese Biomedical Literature Service System)was performed.Randomized,controlled trials(RCTs)involving SX combined with conventional therapy versus conventional therapy were included.The extracted data included populations,interventions,outcomes,and risk of bias.The cardiovascular events served as the primary outcome.Review Manager 5.3 software was used for data analysis.Relative risks(RRs)with 95%confidence intervals(CIs)were the effect measure.RESULTS:A total of eight RCTs with 979 patients were included.There were 559 patients with unstable angina(UA)in six RCTs and 420 patients with acute myocardial infarction(AMI)in two RCTs.Our review showed that SX plus conventional therapy might reduce the incidence of the total endpoint(RR:0.34,95%CI:0.17,0.68,P=0.002),with no obvious adverse events(RR:1.29,95%CI:0.60,2.77,P=0.52)compared with conventional therapy for patients with UA.Additionally,SX plus conventional therapy also reduced the incidence of the total endpoint(RR:0.35,95%CI:0.18,0.68,P=0.002)compared with conventional therapy in patients with AMI.SX plus conventional therapy also reduced the incidence of ventricular fibrillation(RR:0.23,95%CI:0.10,0.57,P=0.001)compared with conventional therapy in patients with AMI.CONCLUSION:Our results suggest that SX is beneficial for treating patients with UA or AMI.However,our findings should be treated with caution because of the poor methodological quality of the included trials.Therefore,more multicenter,large-sample,high-quality RCTs are required to provide high-quality evidence.Liu Chao Duan Lian Huang Mingyan Li Jun Xiong Xingjiang Chen Guang He Haoqiang Zhang Zhenpeng Wang Jie 2020Journal of Traditional Chinese Medicine2020,40,4:1

6Concurrent silencing of TBCE and drug delivery to overcome platinum-based resistance in liver cancer显示文摘Platinum-based chemotherapy resistance is a key factor of poor prognosis and recurrence in hepatocellular carcinoma(HCC).Herein,RNAseq analysis revealed that elevated tubulin folding cofactor E(TBCE)expression is associated with platinum-based chemotherapy resistance.High expression of TBCE contributes to worse prognoses and earlier recurrence among liver cancer patients.Mechanistically,TBCE silencing significantly affects cytoskeleton rearrangement,which in turn increases cisplatin-induced cycle arrest and apoptosis.To develop these findings into potential therapeutic drugs,endosomal pH-responsive nanoparticles(NPs)were developed to simultaneously encapsulate TBCE siRNA and cisplatin(DDP)to reverse this phenomena.NPs(siTBCE+DDP)concurrently silenced TBCE expression,increased cell sensitivity to platinum treatment,and subsequently resulted in superior anti-tumor effects both in vitro and in vivo in orthotopic and patient-derived xenograft(PDX)models.Taken together,NP-mediated delivery and the co-treatment of siTBCE+DDP proved to be effective in reversing chemotherapy resistance of DDP in multiple tumor models.Senlin Li Siyu Chen Zhihui Dong Xingdong Song Xiuling Li Ziqi Huang Huiru Li Linzhuo Huang Ganyuan Zhuang Ran Lan Mingyan Guo Wende Li Phei Er Saw Lei Zhang 2023Acta Pharmaceutica Sinica B2023,13,3:1

9Malondialdehyde treatment reduced immunoreactivity of amandin and delayed its digestion显示文摘Objectives:The secondary products of lipid oxidation are one of the main factors inducing protein oxidation.The effects of oxidation treatment with malondialdehyde(MDA)on the immunoreactivity of amandin and its digestion were studied.Materials and Methods:The rabbit IgG binding ability of amandin was analyzed by western blotting,and the changes in amandin oxidation and immunoreactivity during digestion of amandin with different degrees of oxidation were investigated in combination with an almond allergen enzyme-linked immunosorbent assay kit.Alteration of linear epitopes of amandin by oxidation was investigated by liquid chromatography-tandemmass spectrometry(LC-MS/MS).Results:The results showed that the immunoreactivity of amandin was significantly reduced after 1 mmol/L MDA and 100 mmol/L MDA treat-ment.However,the 1 mmol/L MDA treatment was owing to cleavage of linear epitope peptide in amandin and oxidation of the active amino acid.The 100 mmol/L MDA treatment was due to aggregation of amandin and significant decrease in its solubility.Oxidation also reduced di-gestibility of amandin and significantly affected immunoreactivity during digestion.LC-MS/MS also identified four oxidation-prone methionine sites(aa 264-274,298-308,220-240,and 275-297)in gamma conglutinin 1.Conclusions:MDA treatment reduced the immunoreactivity of amandin.MDA treatment also led to protein aggregation,which slowed down the digestion of amandin and altered the immunoreactivity of amandin during digestion.Xin Luo Mingyan Ai Yongyan Wu Qia Wang Hongbo Song Qun Huang Jiankang Lu 2023Food Quality and Safety2023,7,2:0
10Aqueous-soluble components of sporoderm-removed Ganoderma lucidum spore powder promote ferroptosis in oral squamous cell carcinoma显示文摘Objective: Ferroptosis is a novel cell death process which displays a promising role in cancer treatment.However, clinically available drugs targeting ferroptosis are rarely used, and yet there are no studies reporting on inducing ferroptosis via Chinese herbal extracts. Here we explored the tumor inhibition effects of Ganoderma lucidum(G. lucidum) on oral squamous cell carcinoma(OSCC). Specifically, we aimed to clarify the biological mechanism of components in the dietary, aqueous-soluble sporoderm-removed G. lucidum spore powder(A-GSP).Methods: Preliminary transcriptome analysis revealed the significant enrichment of the ferroptosis pathway.Cellular Fe2+, glutathione(GSH), malondialdehyde(MDA), reactive oxygen species(ROS) and lipid peroxide levels were measured to identify ferroptosis occurrence. Western blotting was used to measure ferroptosis-related proteins. Changes in mitochondria morphology and function were observed with transmission electron microscopy(TEM) and ATP detection assays. Ferroptosis inhibitor ferrostatin-1 was then used to verify the anti-tumor effects of A-GSP. Finally, nude mice xenograft models of oral cancer confirmed that A-GSP inhibited tumor growth.Results: A-GSP promoted ferroptosis in oral cancer cells by inducing Fe2+influx, GSH depletion, as well as lipid peroxide and ROS accumulation. Ferroptosis-related proteins exhibited corresponding changes, particularly Acylco A synthetase long chain family member 4(ACSL4) increase and glutathione peroxidase 4(GPX4) decrease. A-GSP considerably lowered mitochondrial volume and ridge number, while significantly decreasing ATP production. Ferrostatin-1 reversed all of these A-GSP-induced changes. 12Successive magnetic ordering in two Co^(Ⅱ)-ladder metal-organic frameworks显示文摘Metal-organic frameworks showing successive magnetic ordering are far less common than inorganic compounds.Here,we report two metal-organic frameworks[Co^Ⅱ(ox)(bphy)·0.1(DMF)·0.1(Me OH)]n(1)and[Co3^Ⅱ(ox)3(bphy)2(DMF)2]n(2)comprised of zigzag and necklace Co^Ⅱ-ladders,respectively.Together with a previously reported compound[Co^Ⅱ(ox)(bphy)·0.2(DMF)]n(3)consisting of spiraling zigzag Co^Ⅱ-ladders,these three compounds provide a good system for comparative structural and magnetic studies.Comprehensive magnetic analysis reveals that the three compounds undergo long-range magnetic ordering at^2.6 K but exhibit vastly different short-range magnetic correlations:compound 1 shows short-range spin-canted antiferromagnetism ordering at^14.0 K;compound 2 demonstrates successive short-range antiferromagnetism ordering at^15.5and^12.6 K;compound 3 shows slow magnetic relaxation with Tb≈4.6 K.These results demonstrate long-range magnetic ordering is readily accessible in the frameworks of ox^2-bridging Co^Ⅱ-ladders linked by bphy,where short-range magnetic correlations can be systematicly tuned by the Co^Ⅱ-ladder structures.Zhu Zhuo Guoling Li Chenglin Shu Wei Wang Caiping Liu Mingyan Wu Yougui Huang Maochun Hong 2021Science China Chemistry2021,64,1:0
13CD146 promotes malignant progression of breast phyllodes tumor through suppressing DCBLD2 degradation and activating the AKT pathway显示文摘Background As a rapid-progressing tumor,breast malignant phyllodes tumors(PTs)are challenged by the lack of effective therapeutic strategies and suitable prognostic markers.This study aimed to clarify the role and mechanism of CD146 on promoting PTs malignant progression,and to identify a novel prognosis marker and treatment target of breast malignant PTs.Methods The expression and prognostic significance of CD146 in PTs was detected through single-cell RNA-sequencing(scRNA-seq),immunostaining,real-time PCR and other methodologies.Functional experiments including proliferation assay,colony formation assay,transwell assay,and collagen contraction assay were conducted to validate the role of CD146 in malignant progression of PTs.The efficacy of anti-CD146 monoclonal antibody AA98 against malignant PTs was corroborated by a malignant PT organoid model and a PT patient-derived xenograft(PDX)model.Transcriptome sequencing,proteomic analysis,co-immunoprecipitation,and pull-down assay was employed to identify the modulating pathway and additional molecular mechanism.Results In this study,the scRNA-seq analysis of PTs disclosed a CD146-positive characteristic in theα-SMA+fibroblast subset.Furthermore,a progressive elevation in the level of CD146 was observed with the malignant progression of PTs.More importantly,CD146 was found to serve as an independent predictor for recurrence in PT patients.Furthermore,CD146 was found to augment the viability and invasion of PTs.Mechanistically,CD146 acted as a protective“shield”to prevent the degradation of Discoidin,CUB,and LCCL domain-containing protein 2(DCBLD2),thereby activating the phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT)signaling pathway and enhancing malignant behaviors of PT cells.In the malignant PT organoid and PDX model,a significant suppression of malignant PT growth was observed after the application of AA98.Conclusions These findings suggested that CD146 served as an efficacious marker for predicting PT malignant progression and showed promise as a prognosis marker and treatment target of breast malignant PTs.The study further unveiled the essential role of the CD146-DCBLD2/PI3K/AKT axis in the malignant progression of PTs.Jiewen Chen Qingji Xu Dan Liu Xun Li Mingyan Guo Xuehui Chen Jianyou Liao Rong Lei Wende Li Hongyan Huang Phei Er Saw Erwei Song Xiyun Yan Yan Nie 2023Cancer Communications2023,43,11:0
14Prevalence and Severity of HBV-Associated Acute-on-Chronic Liver Failure Due to Irregular Medication of Nucleos(t)ide Analogs显示文摘Hepatitis B virus(HBV)represents the commonest etiologic agent of acute-on-chronic liver failure(ACLF)in most Asian countries.Nucleos(t)ide analogs(NAs)are effective in the treatment of chronic HBV infections,but may also exacerbate the disease and stimulate its development into HBV-associated ACLF if not used appropriately.The current study aimed to assess the prevalence and severity of HBV-associated ACLF as a result from irregular medication of NAs(IMNA).A total of 1134 individuals with HBV-associated ACLF in nine hospitals in Heilongjiang Province were enrolled in this study between 2005 and 2015.Among these,777 chronic hepatitis B(CHB)and 357 HBV-associated liver cirrhosis cases were classified based on various predisposing factors,including IMNA,HBV reactivation(HBVR),infections,treatment drugs,alcohol use and others(hepatitis C virus,hepatitis E virus,gastrointestinal bleeding and unknown reasons).The percentage and improvement rate were examined.Among individuals with HBV-associated ACLF and CHB,IMNA was found in 9.01%,HBVR in 46.20%,infections in 9.52%,treatment drugs in 14.67%,alcohol in 11.71%,and others in 24.58%as predisposing factors.Improvement rates in cases with IMNA,HBVR,infections,treatment drugs,alcohol and others were 41.43%,58.50%,58.11%,56.14%,53.85%,and 65.97%,respectively.Multivariable analysis showed that IMNA,others,infections,hepatic encephalopathy and hepatorenal syndrome were associated with prognosis.Only IMNA independently predicted HBV-associated ACLF prognosis.Overall,our study demonstrated that the percentage of IMNAinduced HBV-associated ACLF was 12.61%,and worse disease conditions resulted from IMNA compared with other factors.Thus,the suitability of treatment with NAs should be thoroughly evaluated.Ying Zheng Shu Chen Yanxin Huang Lisheng Jiang Yongguo Li Yinghua Lan Shuchen Li Yuqin Xu Xiaodong Li Hongwei Zhao Yanbo Wang Ying Shen Chao Wei Honglin Zhou Rongshan Fan Xiqiu Zeng Mingxia Jiang Shupeng Song Mingyan Xu 2021Infectious Microbes & Diseases2021,3,4:0
