3Growth-associated protein 43 and neural cell adhesion molecule expression following bone marrow-derived mesenchymal stem cell transplantation in a rat model of ischemic brain injury显示文摘BACKGROUND:Transplantation of bone marrow-derived mesenchymal stem cells(BMSCs) improves motor functional recovery,but the mechanisms remain unclear.OBJECTIVE:To investigate expression of growth-associated protein 43(GAP-43) and neural cell adhesion molecule following BMSC transplantation to the lateral ventricle in rats with acute focal cerebral ischemic brain damage.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment using immunohistochemistry was performed at the laboratories of Department of Neurology,Renmin Hospital of Wuhan University and Doctoral Scientific Research Work Station of C-BONS PHARMA,Hubei Province,China,from January 2007 to December 2008.MATERIALS:Monoclonal mouse anti-rat 5-bromo-2-deoxyuridine and neural cell adhesion molecule antibodies were purchased from Sigma,USA;monoclonal mouse anti-rat GAP-43 antibody was purchased from Wuhan Boster,China.METHODS:Rat models of right middle cerebral artery occlusion were established using the thread method.At 1 day after middle cerebral artery occlusion,20 μL culture solution,containing 5 × 105 BMSCs,was transplanted to the left lateral ventricle using micro-injection.MAIN OUTCOME MEASURES:Scores of neurological impairment were measured to assess neural function.Expression of GAP-43 and neural cell adhesion molecule at the lesion areas was examined by immunohistochemistry.RESULTS:GAP-43 and neural cell adhesion molecule expression was low in brain tissues of the sham-operated group,but expression increased at the ischemic boundary(P < 0.05).Transplantation of BMSCs further enhanced expression of GAP-43 and neural cell adhesion molecule(P < 0.05) and remarkably improved neurological impairment of ischemic rats(P < 0.05).CONCLUSION:BMSC transplantation promoted neurological recovery in rats by upregulating expression of GAP-43 and neural cell adhesion molecule.Yu Peng Qimei Zhang Hui You Weihua Zhuang Ying Zhang Chengyan Li 2010Neural Regeneration Research2010,5,13:18

7Liver Enzymes Concentrations Are Closely Related to Pre-diabetes:Findings of the Shanghai Diabetes Study Ⅱ (SHDS Ⅱ)显示文摘Objective To investigate the relationship of liver enzymes with hyperglycemia in a large population in Shanghai and identify the association between liver enzymes and insulin resistance.Methods A total of 3 756 participants were enrolled.Each participant underwent an oral glucose tolerance test and completed a questionnaire.Anthropometric indices were recorded and serum samples were collected for measurement.Results Liver enzymes concentrations were independently associated with i-IGT,IFG+IGT,and diabetes.With the increase of ALT and GGT concentrations,ORs for i-IGT,IFG+IGT,and diabetes increased gradually.By comparing patients in the highest quartile of GGT concentrations or ALT concentrations with those in the lowest quartile (Q1),ORs for i-IGT,IFG+IGT,or diabetes was significant after adjustment.Both ALT and GGT concentrations were linearly correlated with HOMA-IR and independently associated with HOMA-IR [ALT OR (95% CI):2.56 (1.51-4.34) P=0.00;GGT OR (95% CI):2.66 (1.53-4.65) P=0.00].Conclusion Serum ALT and GGT concentrations were closely related to pre-diabetes and diabetes in the Shanghai population and positively associated with insulin resistance.GAO Fei PAN Jie Min HOU Xu Hong FANG Qi Chen LU Hui Juan TANG Jun Ling GU Hui Lin PAN Zhi Jian YAO You Hua SHEN Wei Zhen JIA Wei Ping 2012Biomedical and Environmental Sciences2012,25,1:15
8Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 10Grey GM(1,1) Model with Function-Transfer Method for Wear Trend Prediction and its Application显示文摘Trend forecasting is an important aspect in fault diagnosis and work state supervision. The principle, where Grey theory is applied in fault forecasting, is that the forecast system is considered as a Grey system; the existing known information is used to infer the unknown information’s character, state and development trend in a fault pattern, and to make possible forecasting and decisions for future development. It involves the whitenization of a Grey process. But the traditional equal time interval Grey GM (1,1) model requires equal interval data and needs to bring about accumulating addition generation and reversion calculations. Its calculation is very complex. However, the non equal interval Grey GM (1,1) model decreases the condition of the primitive data when establishing a model, but its requirement is still higher and the data were pre processed. The abrasion primitive data of plant could not always satisfy these modeling requirements. Therefore, it establishes a division method suited for general data modeling and estimating parameters of GM (1,1), the standard error coefficient that was applied to judge accuracy height of the model was put forward; further, the function transform to forecast plant abrasion trend and assess GM (1,1) parameter was established. These two models need not pre process the primitive data. It is not only suited for equal interval data modeling, but also for non equal interval data modeling. Its calculation is simple and convenient to use. The oil spectrum analysis acted as an example. The two GM (1,1) models put forward in this paper and the new information model and its comprehensive usage were investigated. 12Effective adsorption of sulfamethoxazole, bisphenol A and methyl orange on nanoporous carbon derived from metal-organic frameworks显示文摘Nanoporous carbons(NPCs) derived from metal–organic frameworks(MOFs) are attracting increasing attention in many areas by virtue of their high specific surface area, large pore volume and unique porosity. The present work reports the preparation of an NPC with high surface area(1731 m^2/g) and pore volume(1.68 cm^3/g) by direct carbonization of MOF-5. We examined the adsorption of three typical contaminants from aqueous solutions, i.e., sulfamethoxazole(SMX),bisphenol A(BPA) and methyl orange(MO), by using the as-prepared NPC. The results demonstrated that NPC could adsorb the contaminants effectively, with adsorption capacity(qm) of 625 mg/g(SMX), 757 mg/g(BPA) and 872 mg/g(MO), respectively. These values were approximately 1.0-3.2 times higher than those obtained for single-walled carbon nanotubes(SWCNTs) and commercial powder active carbon(PAC) under the same conditions. With its high surface area and unique meso/macropore structure, the enhanced adsorption of NPC most likely originates from the cooperative interaction of a pore-filling mechanism, electrostatic interaction,and hydrogen bonding. In particular, the p H value has a crucial impact on adsorption, suggesting the significant contribution of electrostatic interaction between NPC and the contaminants. 15Screening of Serum Biomarkers for Distinguishing between Latent and Active Tuberculosis Using Proteome Microarray显示文摘Objective To identify potential serum biomarkers for distinguishing between latent tuberculosis infection(LTBI) and active tuberculosis(TB). Methods A proteome microarray containing 4,262 antigens was used for screening serum biomarkers of 40 serum samples from patients with LTBI and active TB at the systems level. The interaction network and functional classification of differentially expressed antigens were analyzed using STRING 10.0 and the TB database, respectively. Enzyme-linked immunosorbent assays(ELISA) were used to validate candidate antigens further using 279 samples. The diagnostic performances of candidate antigens were evaluated by receiver operating characteristic curve(ROC) analysis. Both antigen combination and logistic regression analysis were used to improve diagnostic ability. Results Microarray results showed that levels of 152 Mycobacterium tuberculosis(Mtb)-antigenspecific IgG were significantly higher in active TB patients than in LTBI patients(P < 0.05), and these differentially expressed antigens showed stronger associations with each other and were involved in various biological processes. Eleven candidate antigens were further validated using ELISA and showed consistent results in microarray analysis. ROC analysis showed that antigens Rv2031 c, Rv1408, and Rv2421 c had higher areas under the curve(AUCs) of 0.8520, 0.8152, and 0.7970, respectively. In addition, both antigen combination and logistic regression analysis improved the diagnostic ability. Through the Grey forecast of reliability parameters and the reliability forecast of parts and systems, decisions were made in the real operative state of equipment in real time. It replaced the old method that required mathematics and physical statistics in a large base of test data to obtain a pre check, and it was used in a practical problem. 19Recent Progress and Development in Extrusion of Rare Earth Free Mg Alloys: A Review显示文摘Mg and its alloys are the lightest structural metals available and are extremely attractive for applications as lightweight components, particularly in the automobile, electronic, and aerospace industries. The global market for wrought Mg alloys has steadily expanded over the past decade. And numerous studies have been carried out to meet this increasing demand of high-performance Mg alloys. However, Mg extrusion alloys have had a very limited usage so far. To overcome existing industrial challenges, one desirable approach is the development of low-cost rare earth(RE) free Mg extrusion alloys with superior mechanical properties. This review will introduce the recent research highlights in the extrusion of Mg alloys, specifi cally focusing on low-cost RE-free Mg alloy. The results from both the literature and our previous study are summarized and critically reviewed. Several aspects of RE-free Mg extrusion alloys are described in detail:(1) novel alloying designs including Mg–Al-, Mg–Zn-, Mg–Ca-, Mg–Sn-, and Mg–Bi-based alloys,(2) advanced extrusion techniques, and(3) extrusion-related severe plastic deformation(SPD) processing. Accordingly, considering the large gap in mechanical properties between the current RE-free Mg alloys and high-performance aluminum alloys, new alloy design, processing route control, and recommendations for future research on RE-free Mg extrusion alloys are also proposed. 20The effects of four transplantation methods on five coral species at the Sanya Bay显示文摘Coral transplantation is considered as one of the major tools to increase coral abundance for degraded coral reefs.To investigate the effects of various methods and coral species in transplantation, coral fragments(n=902) of five coral species were transplanted by four methods at Luhuitou, the Sanya Bay, Hainan Province, China, where the reef has been over-exploited and is still threatened by human activities and natural disasters. Ten months after the transplant, the average survivorship of the transplanted corals was 45.5%. Methodologies had different effects on the transplanted corals, but none of them was efficacious for all coral species. Methodology could not change the decreasing trend for Montipora foliosa and Acropora hyacinthus, although it did slow down their decline. All transplants of A. hyacinthus and M. foliosa had high mortalities and significant decrease on survival area, while Porites andrewsi and Galaxea fascicularis had lower mortalities and partial mortalities. Only one method had significant effect on increasing survival area of G. fascicularis, same as P. andrewsi. Out of the five transplanted coral species, Pocillopora damicornis was the only species that had living tissue area increase in all applied methods, while the others had decreased live tissue area in one or more methods. The results of this study suggested that performing coral transplantation in a highly threatened area was not efficient unless the threats were diminished or erased. Moreover, proper species selection for coral transplantation is crucial, especially in a disturbed environment. Methodology, although having limited effects on improving results of coral transplantation, cannot compensate the maladjustment of vulnerable species to the stresses on the Luhuitou Reef.
