1Efficacy and safety of metformin and sitagliptin based triple antihyperglycemic therapy(STRATEGY):a multicenter,randomized,controlled,non-inferiority clinical trial显示文摘Despite the current guideline's recommendation of a timely stepwise intensification therapy,the 'clinical inertia',termed as the delayed treatment intensification,commonly exists in the real world,which may be partly due to the relatively little substantial evidence and no clear consensus regarding the efficacy and safety of triple oral agents in patients inadequately controlled with dual therapy.In this clinical trial performed in 237 centers in China,5,535 type 2 diabetic patients inadequately controlled by previous therapies were treated with a stable metformin/sitagliptin dual therapy for 20 weeks.The patients who did not reach the glycated hemoglobin A1c(HbA1c) goal were then further randomized into glimepiride,gliclazide,repaglinide,or acarbose group for an additional 24-week triple therapy.A mean HbAlc reduction of 0.85%was observed when sitagliptin was added to the patients inadequately controlled with metformin in 16 weeks.Further HbAlc reductions in the 24-week triple therapy stage were 0.65%in glimepiride group,0.70%in gliclazide group,0.61%in repaglinide group,and 0.45%in acarbose group.The non-inferiority criterion for primary hypotheses was met for gliclazide and repaglinide,but not for acarbose,compared with glimepiride,when added to metformin/sitagliptin dual therapy.The incidences of adverse events(AEs) were 29.2%in the dual therapy stage and30.3%in the triple therapy stage.Metformin/sitagliptin as baseline therapy,with the addition of a third oral antihyperglycemic agent,including glimepiride,gliclazide,repaglinide,or acarbose,was effective,safe and well-tolerated for achieving an HbAlc<7.0%goal in type 2 diabetic patients inadequately controlled with previous therapies.The timely augmentation of up to three oral antihyperglycemic agents is valid and of important clinical benefit to prevent patients from exposure to unnecessarily prolonged hyperglycemia.Wen Xu Yiming Mu Jiajun Zhao Dalong Zhu Qiuhe Ji Zhiguang Zhou Bin Yao Anhua Mao Samuel S.Engel Bin Zhao Yan Bi Longyi Zeng Xingwu Ran Juming Lu Linong Ji Wenying Yang Weiping Jia Jianping Weng 2017Science China(Life Sciences)2017,60,3:19

3Development of Land Surface Model BCC_AVIM2.0 and Its Preliminary Performance in LS3MIP/CMIP6显示文摘The improvements and validation of several parameterization schemes in the second version of the Beijing Climate Center Atmosphere–Vegetation Interaction Model(BCC_AVIM2.0) are introduced in this study. The main updates include a replacement of the water-only lake module by the common land model lake module(Co LM-lake) with a more realistic snow–ice–water–soil framework, a parameterization scheme for rice paddies added in the vegetation module, renewed parameterizations of snow cover fraction and snow surface albedo to accommodate the varied snow aging effect during different stages of a snow season, a revised parameterization to calculate the threshold temperature to initiate freeze(thaw) of soil water(ice) rather than being fixed at 0°C in BCC_AVIM1.0, a prognostic phenology scheme for vegetation growth instead of empirically prescribed dates for leaf onset/fall, and a renewed scheme to depict solar radiation transfer through the vegetation canopy. The above updates have been implemented in BCC_AVIM2.0 to serve as the land component of the BCC Climate System Model(BCC_CSM). Preliminary results of BCC_AVIM in the ongoing Land Surface, Snow, and Soil Moisture Model Intercomparison Project(LS3 MIP) of the Coupled Model Intercomparison Project Phase 6(CMIP6) show that the overall performance of BCC_AVIM2.0 is better than that of BCC_AVIM1.0 in the simulation of surface energy budgets at the seasonal timescale. Comparing the simulations of annual global land average before and after the updates in BCC_AVIM2.0 reveals that the bias of net surface radiation is reduced from-12.0 to-11.7 W m-2 and the root mean square error(RMSE) is reduced from 20.6 to 19.0 W m-2;the bias and RMSE of latent heat flux are reduced from 2.3 to-0.1 W m-2 and from 15.4 to14.3 W m-2, respectively;the bias of sensible heat flux is increased from 2.5 to 5.1 W m-2 but the RMSE is reduced from 18.4 to 17.0 W m-2.Weiping LI Yanwu ZHANG Xueli SHI Wenyan ZHOU Anning HUANG Mingquan MU Bo QIU Jinjun JI 2019Journal of Meteorological Research2019,33,5:14
4The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 5A Quantized Kernel Least Mean Square Scheme with Entropy-Guided Learning for Intelligent Data Analysis显示文摘Quantized kernel least mean square(QKLMS) algorithm is an effective nonlinear adaptive online learning algorithm with good performance in constraining the growth of network size through the use of quantization for input space. It can serve as a powerful tool to perform complex computing for network service and application. With the purpose of compressing the input to further improve learning performance, this article proposes a novel QKLMS with entropy-guided learning, called EQ-KLMS. Under the consecutive square entropy learning framework, the basic idea of entropy-guided learning technique is to measure the uncertainty of the input vectors used for QKLMS, and delete those data with larger uncertainty, which are insignificant or easy to cause learning errors. Then, the dataset is compressed. Consequently, by using square entropy, the learning performance of proposed EQ-KLMS is improved with high precision and low computational cost. 6Controlled release of hydrogen by implantation of magnesium induces P53-mediated tumor cells apoptosis显示文摘Hydrogen has been used to suppress tumor growth with considerable efficacy.Inhalation of hydrogen gas and oral ingestion of hydrogen-rich saline are two common systemic routes of hydrogen administration.We have developed a topical delivery method of hydrogen at targeted sites through the degradation of magnesium-based biomaterials.However,the underlying mechanism of hydrogen’s role in cancer treatment remains ambiguous.Here,we investigate the mechanism of tumor cell apoptosis triggered by the hydrogen released from magnesium-based biomaterials.We find that the localized release of hydrogen increases the expression level of P53 tumor suppressor proteins,as demonstrated by the in vitro RNA sequencing and protein expression analysis.Then,the P53 proteins disrupt the membrane potential of mitochondria,activate autophagy,suppress the reactive oxygen species in cancer cells,and finally result in tumor suppression.The anti-tumor efficacy of magnesium-based biomaterials is further validated in vivo by inserting magnesium wire into the subcutaneous tumor in a mouse.We also discovered that the minimal hydrogen concentration from magnesium wires to trigger substantial tumor apoptosis is 91.2μL/mm^(3)per day,which is much lower than that required for hydrogen inhalation.Taken together,these findings reveal the release of H2 from magnesium-based biomaterial exerts its anti-tumoral activity by activating the P53-mediated lysosome-mitochondria apoptosis signaling pathway,which strengthens the therapeutic potential of this biomaterial as localized anti-tumor treatment.Rui Zan Hao Wang Weijie Cai Jiahua Ni Berengere J.C.Luthringer-Feyerabend Wenhui Wang Hongzhou Peng Weiping Ji Jun Yan Jiazeng Xia Yang Song Xiaonong Zhang 2022Bioactive Materials2022,7,3:3
7The pathology,phylogeny,and epidemiology of Echinococcus ortleppi(G5 genotype):a new case report of echinococcosis in China显示文摘Background:Cystic echinococcosis(CE),caused by the larval stage of the complex Echinococcus granulosus sensu lato(s.l.),is a zoonotic parasitic disease with a high social burden in China.E.ortleppi is a species(formerly genotype 5 of E.granulosus s.l.)with unique epidemic areas(tropical areas),transmission patterns(mainly cattle origin),and pathological characteristics(large and small hook lengths)compared to other species that cause CE.A 19-year-old female patient in an area with no history of echinococcosis in Guizhou Province,China,was diagnosed with E.ortleppi infection in 2019.This study is to understand the source of this human E.ortleppi infection.Methods:We performed computer tomography(CT)scans,surgical operation,morphological sectioning,molecular diagnosis,phylogenetic analyses,and epidemiological investigation in Anshun City,Guizhou Province,China in 2019.Results:The patient presented with intermittent distension and pain in the upper abdomen without other abnormal symptoms.Routine blood examination results were normal.However,abdominal CT revealed a fertile cyst with a diameter of approximately 8 cm,uniform density,and a clear boundary,but without an evident cyst wall in the right lobe of the liver.The cyst was fertile,and phylogenetic analyses revealed that the isolates represented a new E.ortleppi genus haplotype.A result of 10‒14 years incubation period with indigenous infection was considered available for the case through the epidemiological survey.Conclusions:CE due to E.ortleppi infection can be confused with other diseases causing liver cysts,resulting in misdiagnosis.A transmission chain of E.ortleppi may exist or existed in the past in the previously considered nonendemic areas of echinococcosis in southwestern China.Xu Wang Aiya Zhu Hongying Cai Baixue Liu Gang Xie Rui Jiang Ji Zhang Nanzi Xie Yayi Guan Robert Bergquist Zhenghuan Wang Yang Li Weiping Wu 2021Infectious Diseases of Poverty2021,10,6:3
8Collective Entity Alignment for Knowledge Fusion of Power Grid Dispatching Knowledge Graphs显示文摘Knowledge graphs(KGs)have been widely accepted as powerful tools for modeling the complex relationships between concepts and developing knowledge-based services.In recent years,researchers in the field of power systems have explored KGs to develop intelligent dispatching systems for increasingly large power grids.With multiple power grid dispatching knowledge graphs(PDKGs)constructed by different agencies,the knowledge fusion of different PDKGs is useful for providing more accurate decision supports.To achieve this,entity alignment that aims at connecting different KGs by identifying equivalent entities is a critical step.Existing entity alignment methods cannot integrate useful structural,attribute,and relational information while calculating entities’similarities and are prone to making many-to-one alignments,thus can hardly achieve the best performance.To address these issues,this paper proposes a collective entity alignment model that integrates three kinds of available information and makes collective counterpart assignments.This model proposes a novel knowledge graph attention network(KGAT)to learn the embeddings of entities and relations explicitly and calculates entities’similarities by adaptively incorporating the structural,attribute,and relational similarities.Then,we formulate the counterpart assignment task as an integer programming(IP)problem to obtain one-to-one alignments.We not only conduct experiments on a pair of PDKGs but also evaluate o ur model on three commonly used cross-lingual KGs.Experimental comparisons indicate that our model outperforms other methods and provides an effective tool for the knowledge fusion of PDKGs.Linyao Yang Chen Lv Xiao Wang Ji Qiao Weiping Ding Jun Zhang Fei-Yue Wang 2022IEEE/CAA Journal of Automatica Sinica2022,9,11:3

10Increased osteoblast adhesion on nanophase Ti6Al4V显示文摘The objective of the present study was to prepare a novel nanostructured surface of Ti6Al4V alloy by the severe plastic deformation (SPD) and the chemical treatment process and to evaluate the adhesion of osteoblast on the nanophase titanium alloy. In the in vitro study, the primary cultured osteoblasts of neonatal rat calvaria were cultured on the nanophase and the as-received smooth Ti6Al4V substrates. Then osteoblasts adhesion behaviors on different substrates were observed by the fluorescence mi-croscopy, scanning electron microscopy (SEM) and RT-PCR analysis. The results of our research showed increased osteoblast adhesion on the nanophase titanium alloy compared with the as-received case. On the nanophase substrate, the presence of extensive filopodia, strong cellular adhesion and early cellular confluency could be observed. In addition, the expression of the adhesion-related integrin β1 mRNA was also higher on the nanophase substrate. 11Composite hydrophilic coating for conditioner aluminum fins显示文摘To solve the so called “white rust” and ’water bridge' problems of the aluminum fins for heat exchanger of automobile air conditioner, aimed at nationalizing the art of hydrophilic coating technology, the choice of coating forming and curing materials was investigated. By measuring the water contact angle, SEM surface scanning and ingredients analysis of the coating, optimal parameters and composition are acquired. The coating forming mechanisms of the composition was also expatiated.
