| 2 | Implication of snoRNA U50 in human breast cancer显示文摘Deletion of chromosome 6q is frequent in breast cancer,and the deletion often involves a region in 6q14-q16.At present,however,the underlying tumor suppressor gene has not been established.Based on a recent study identifying snoRNA U50 as a candidate for the 6q14-16 tumor suppressor gene in prostate cancer,we investigated whether U50 is also involved in breast cancer.PCR-based approaches showed that U50 underwent frequent genomic deletion and transcriptional downregulation in cell lines derived from breast cancer.Muta-tion screening identified the same 2-bp deletion of U50 as in prostate cancer in both cell lines and primary tumors from breast cancer,and the deletion was both somatic and in germline.Genotyping of a cohort of breast cancer cases and controls for the mutation demonstrated that,while homozygous genotype of the mutation was rare,its heterozygous genotype occurred more frequently in women with breast cancer.Functionally,re-expression of U50 resulted in the inhibition of colony formation in breast cancer cell lines.These results suggest that noncoding snoRNA U50 plays a role in the development and/or progression of breast cancer. | Xue-Yuan Dong Peng Guo Jeff Boyd Xiaodong Sun Qunna Li Wei Zhou Jin-Tang Dong | 2009 | Journal of Genetics and Genomics2009,36,8: | 11 |
| 3 | Knowledge extraction from Chinese wiki encyclopedias显示文摘The vision of the Semantic Web is to build a 'Web of data' that enables machines to understand the semantics of information on the Web.The Linked Open Data(LOD) project encourages people and organizations to publish various open data sets as Resource Description Framework(RDF) on the Web,which promotes the development of the Semantic Web.Among various LOD datasets,DBpedia has proved a successful structured knowledge base,and has become the central interlinking-hub of the Web of data in English.However,in the Chinese language,there is little linked data published and linked to DBpedia.This hinders the structured knowledge sharing of both Chinese and cross-lingual resources.This paper deals with an approach for building a large-scale Chinese structured knowledge base from Chinese wiki resources,including Hudong and Baidu Baike.The proposed approach first builds an ontology based on the wiki category system and infoboxes,and then extracts instances from wiki articles.Using Hudong as our source,our approach builds an ontology containing 19 542 concepts and 2381 properties.802 593 instances are extracted and described using the concepts and properties in the extracted ontology and 62 679 of them are linked to equivalent instances in DBpedia.As from Baidu Baike,our approach builds an ontology containing 299 concepts,37 object properties,and 5590 data type properties.1 319 703 instances are extracted from Baidu Baike,and 84 343 of them are linked to instances in DBpedia.We provide RDF dumps and SPARQL endpoint to access the established Chinese knowledge bases.The knowledge bases built using our approach can be used not only in Chinese linked data building,but also in many useful applications of large-scale knowledge bases,such as question-answering and semantic search. | Zhi-chun WANG Zhi-gang WANG Juan-zi LI Jeff Z.PAN | 2012 | Journal of Zhejiang University-Science C(Computers and Electronics)2012,13,4: | 11 |
| 4 | The Glutamatergic Postrhinal Cortex–Ventrolateral Orbitofrontal Cortex Pathway Regulates Spatial Memory Retrieval显示文摘A deficit in spatial memory has been taken as an early predictor of Alzheimer’s disease(AD) or mild cognitive impairment(MCI). The uncinate fasciculus(UF) is a long-range white-matter tract that connects the anterior temporal lobe with the orbitofrontal cortex(OFC)in primates. Previous studies have shown that the UF impairment associated with spatial memory deficits may be an important pathological change in aging and AD, but its exact role in spatial memory is not well understood. The pathway arising from the postrhinal cortex(POR) and projecting to the ventrolateral orbitofrontal cortex(vlOFC)performs most of the functions of the UF in rodents.Although the literature suggests an association between spatial memory and the regions connected by the POR–vlOFC pathway, the function of the pathway in spatialmemory is relatively unknown. To further illuminate the function of the UF in spatial memory, we dissected the POR–vlOFC pathway in mice. We determined that the POR–vlOFC pathway is a glutamatergic structure, and that glutamatergic neurons in the POR regulate spatial memory retrieval. We also demonstrated that the POR–vlOFC pathway specifically transmits spatial information to participate in memory retrieval. | 5 | Optical observations of LIGO source GW 170817 by the Antarctic Survey Telescopes at Dome A,Antarctica显示文摘The LIGO detection of gravitational waves(GW) from merging black holes in 2015 marked the beginning of a new era in observational astronomy. The detection of an electromagnetic signal from a GW source is the critical next step to explore in detail the physics involved. The Antarctic Survey Telescopes(AST3),located at Dome A, Antarctica, is uniquely situated for rapid response time-domain astronomy with its continuous night-time coverage during the austral winter. We report optical observations of the GW source(GW 170817) in the nearby galaxy NGC 4993 using AST3. The data show a rapidly fading transient at around 1 day after the GW trigger, with the i-band magnitude declining from 17:23 ± 0:13 magnitude to 17:72 ± 0:09 magnitude in ~1:8 h. The brightness and time evolution of the optical transient associated with GW 170817 are broadly consistent with the predictions of models involving merging binary neutron stars. | 6 | Development and optimization of thermal contrast amplification lateral flow immunoassays for ultrasensitive HIV p24 protein detection显示文摘Detection of human immunodeficiency virus(HIV)p24 protein at a single pg/ml concentration in point-of-care(POC)settings is important because it can facilitate acute HIV infection diagnosis with a detection sensitivity approaching that of laboratory-based assays.However,the limit of detection(LOD)of lateral flow immunoassays(LFAs),the most prominent POC diagnostic platform,falls short of that of laboratory protein detection methods such as enzyme-linked immunosorbent assay(ELISA).Here,we report the development and optimization of a thermal contrast amplification(TCA)LFA that will allow ultrasensitive detection of 8 pg/ml p24 protein spiked into human serum at POC,approaching the LOD of a laboratory test.To achieve this aim,we pursued several innovations as follows:(a)defining a new quantitative figure of merit for LFA design based on the specific to nonspecific binding ratio(BR);(b)using different sizes and shapes of gold nanoparticles(GNPs)in the systematic optimization of TCA LFA designs;and(c)exploring new laser wavelengths and power regimes for TCA LFA designs.First,we optimized the blocking buffer for the membrane and running buffer by quantitatively measuring the BR using a TCA reader.The TCA reader interprets the thermal signal(i.e.,temperature)of GNPs within the membrane when irradiated by a laser at the plasmon resonance wavelength of the particle.This process results in higher detection and quantitation of GNPs than in traditional visual detection(i.e.,color intensity).Further,we investigated the effect of laser power(30,100,200 mW),GNP size and shape(30 and 100 nm gold spheres,150 nm gold-silica shells),and laser wavelength(532,800 nm).Applying these innovations to a new TCA LFA design,we demonstrated that 100nm spheres with a 100 mW 532 nm laser provided the best performance(i.e.,LOD=8pg/ml).This LOD is significantly better than that of the current colorimetric LFA and is in the range of the laboratory-based p24 ELISA.In summary,this TCA LFA for p24 protein shows promise for detecting acute HIV infection in POC settings. | Li Zhan Timothy Granade Yilin Liu Xierong Wei Ae Youngpairoj Vickie Sullivan Jeff Johnson John Bischof | 2020 | Microsystems & Nanoengineering2020,6,1: | 3 |
| 7 | The onset of deep recycling of supracrustal materials at the Paleo-Mesoarchean boundary显示文摘The recycling of supracrustal materials, and in particular hydrated rocks, has a profound impact on mantle composition and thus on the formation of continental crust, because water modifies the physical properties of lithological systems and the mechanisms of partial melting and fractional fractionation. On the modern Earth, plate tectonics offers an efficient mechanism for mass transport from the Earth’s surface to its interior, but how far this mechanism dates back in the Earth’s history is still uncertain. Here, we use zircon oxygen(O) isotopes to track recycling of supracrustal materials into the magma sources of early Archean igneous suites from the Kaapvaal Craton, southern Africa. The mean δ^(18)O values of zircon from TTG(tonalite–trondhjemite–granodiorite) rocks abruptly increase at the Paleo-Mesoarchean boundary(ca.3230 million years ago;Ma), from mantle zircon values of 5‰–6‰ to approaching 7.1‰, and this increase occurs in ≤3230 Ma rocks with elevated Dy/Yb ratios. The^(18)O enrichment is a unique signature of low-temperature water–rock interaction on the Earth’s surface. Because the later phase was emplaced into the same crustal level as the older one and TTG magmas would derive from melting processes in the garnet stability field(>40 km depth), we suggest that this evident shift in TTG zircon O isotopic compositions records the onset of recycling of the mafic oceanic crust that underwent seawater hydrothermal alteration at low temperature. | 9 | ^(99m)TC-Methylene diphosphonate uptake at injury site correlates with osteoblast differentiation and mineralization during bone healing in mice显示文摘99m Tc-Methylene diphosphonate(99m Tc-MDP) is widely used in clinical settings to detect bone abnormalities.However, the mechanism of99 m Tc-MDP uptake in bone is not well elucidated. In this study, we utilized a mouse tibia injury model, single-photon emission computed tomography(gamma scintigraphy or SPECT),ex vivo micro-computed tomography, and histology to monitor99 m Tc-MDP uptake in injury sites during skeletal healing. In an ex vivo culture system, calvarial cells were differentiated into osteoblasts with osteogenic medium, pulsed with99 m Tc-MDP at different time points, and quantitated for99 m Tc-MDP uptake with a gamma counter. We demonstrated that99 m Tc-MDP uptake in the injury sites corresponded to osteoblast generation in those sites throughout the healing process. The99 m Tc-MDP uptake within the injury sites peaked on day 7 post-injury, while the injury sites were occupied by mature osteoblasts also starting from day 7.99 m Tc-MDP uptake started to decrease 14 days post-surgery, when we observed the highest level of bony tissue in the injury sites. We also found that99 m Tc-MDP uptake was associated with osteoblast maturation and mineralization in vitro.
