| 2 | ACPAs promote IL-1β production in rheumatoid arthritis by activating the NLRP3 inflammasome显示文摘OBJECTIVES:Anti-citrullinated protein antibodies(ACPAs)are a group of autoantibodies targeted against citrullinated proteins/peptides and are informative rheumatoid arthritis(RA)biomarkers.ACPAs also play a crucial role in RA pathogenesis,and their underlying mechanism merits investigation.METHODS:Immunohistochemical(IHC)assays were carried out to determine IL-1βlevels in ACPA+and ACPA−RA patients.PBMCderived monocytes were differentiated into macrophages before stimulation with ACPAs purified from RA patients.The localization and interaction of molecules were analyzed by confocal microscopy,co-IP,and surface plasmon resonance.RESULTS:In our study,we found that IL-1βlevels were elevated in ACPA+RA patients and that ACPAs promoted IL-1βproduction by PBMC-derived macrophages.ACPAs interacted with CD147 to enhance the interaction between CD147 and integrinβ1 and,in turn,activate the Akt/NF-κB signaling pathway.The nuclear localization of p65 promoted the expression of NLRP3 and pro-IL-1β,resulting in priming.Moreover,ACPA stimulation activated pannexin channels,leading to ATP release.The accumulated ATP bound to the P2X7 receptor,leading to NLRP3 inflammasome activation.CONCLUSIONS:Our study suggests a new hypothesis regarding IL-1βproduction in RA involving ACPAs,which may be a potential therapeutic target in RA treatment. | Xiwen Dong Zhaohui Zheng Peng Lin Xianghui Fu Fanni Li Jianli Jiang Ping Zhu | 2020 | Cellular & Molecular Immunology2020,17,3: | 11 |
| 3 | Seasonal,diurnal and nocturnal variations of carbonyl compounds in the semi-urban environment of Orléans,France显示文摘Atmospheric carbonyls were measured at a semi-urban site in Orléans, France, from October 2010to July2011. Formaldehyde, acetaldehyde and acetone were found to be the most abundant carbonyls, with average concentrations of 3.1, 1.0, 2.0 ppb, respectively in summer, 2.3, 0.7, 2.2 ppb, respectively in autumn, 2.2, 1.0, 2.1 ppb, respectively in spring,and 1.5, 0.7, 1.1 ppb, respectively in winter. Photo-oxidation of volatile organic compounds(VOCs) was found to make a remarkable contribution to atmospheric carbonyls in the semi-urban site based on the distinct seasonal and diurnal variations of the carbonyls, as well as the significantly positive correlations between the carbonyls and ozone. | 8 | Flow Instability of a Centrifugal Pump Determined Using the Energy Gradient Method显示文摘The stability of the centrifugal pump has not been well revealed because of the complexity of internal flow. To analyze the flow characteristics of a centrifugal pump operating at low capacity, methods of numerical simulation and experimental research were adopted in this paper. Characteristics of the inner flow were obtained. Standard k–ε turbulence models were used to calculate the inner flow of the pump under off-design conditions. The distribution of the energy gradient function K was obtained by three-dimensional numerical simulation at different flow rates. The relative velocity component was acquired from the absolute velocity obtained in particle image velocimetry. By comparing with experimental results, it was found that flow instability occurs at the position of maximum K. The flow stability reduces with an increasing flow rate. | 9 | Gene Expression and Activity of Enzymes Involved in Sugar Metabolism and Accumulation During “Huangguan” and “Yali” Pear Fruit Development显示文摘Since the carbohydrate content affects pear flavor during the process of growth, it is necessary to determine the sugar components that accumulate in the fruit. We analyzed the fruit carbohydrate content, and the gene expression and activity ofacid invertase(AI), neutral invertase(NI), sucrose synthase(SS), and sucrose phosphate synthase(SPS) during the development of 'Huangguan' and 'Yali' pears. The results demonstrate that during development, the fruit sugar metabolism of the 'Huangguan' pear follows a typical sorbitol–starch-soluble sugars middle model, whereas the 'Yali' pear fruit follows a typical sorbitol–sucrose–starch-soluble sugars middle model. In the 'Huangguan' pear, we found the AI and NI gene expressions, as well as AI( P < 0.05) and NI( P < 0.01) enzyme activities, to be positively correlated, whereas we found the NI gene expression and NI enzyme activity of 'Yali' pear to be negatively correlated( P < 0.01). We observed the high levels oflate-stage AI and early-stage SS during development to roughly correspond with the gene expression found in the late and early stages, respectively, suggesting their potential regulatory roles in 'Huangguan' pear fruit development. | 16 | Calculation model of AC loss for CICC(cable-in-conduit conductor) based on strain显示文摘The CICC (cable-in-conduit conductor) in ITER (International Thermal-nuclear Experimental Reactor) will run in high-current, fast transient magnet field and complex environment. In response to the impact of magnet fields above 10 T, the Nb3Sn conductor has been introduced. However, the AC (alternating current) loss mechanism of Nb3Sn conductor on strain has not been explored. So, it is necessary to study the AC loss calculation method with transient electromagnetic field and wide range of strain, the coupling current in complex field and current signal of field is simplified to the spectrum effects of coil excitation, and calculation technology of AC loss, which contains the frequency, magnet field, coil characteristics and other parameters, is constructed to meet the discrete Fourier transform (DFT). By comparative analysis of simulation, it is found that the AC loss calculation of the conductor with spectrum algorithm is closer to the actual project value than the traditional algorithm. For the rapid excitation, in particular plasma discharge and burst, spectrum algorithm and the traditional algorithm are consistent. For the relative error calculation of hysteresis loss and coupling loss, it is found that the coupling loss is cumulative linearly, where the hysteresis loss is not so. As a function of the amplitude, frequency and phase angle, the relative error is less than 40%. By the non-contact optical measuring technology, an image acquisition and data analysis system is designed to explore the spilled mechanism and characteristic. The oil trajectory, velocity and the rising time to the surface are obtained through this system. The influence of the water flow and the spilled discharge on the behavior of the spilled oil are analyzed from both qualitative and quantitative perspectives. The sensitivity study of the characteristic physical quantities to various factors are presented afterward. The spilled oil under water is mainly distributed in the form of the scattered particles with different sizes. The rising process of the oil can be divided into three stages: full, dispersion and aggregation period. The spilled discharge is the primary factor affecting the rising time of the oil particles. In the rising process of the oil particles, the vertical velocity of the oil is mainly affected by the spilled discharge, and the transverse velocity is more dependent on the water velocity. The deviation of the transverse oil velocity is much larger than that of the rising time and the vertical oil velocity. | 20 | Neuroprotective effect of heat shock protein 70 Inhibition of Tau protein expression显示文摘BACKGROUND: Previous studies have shown that heat shock protein 70 (HSP70) hasneuroprotective effects by decreasing phosphorylation of Tau protein, thereby reducing theexpression of Tau protein and proper aggregation.OBJECTIVE: To observe and verify expressional changes of HSP70 and Tau in retinal ganglioncells following stretch injury to the right optic nerve in rats, and to determine the effect of heat stresspretreatment on HSP70 and Tau protein expressions.DESIGN, TIME AND SETTING: A randomized, controlled, animal experiment was performed at theLaboratory of Neurology Research Institute of the First Affiliated Hospital of Chongqing MedicalUniversity from March to June 2006.MATERIALS: Instant SABC immunohistochemistry kit, as well as mouse anti-HSP70 and rabbitanti-Tau polyclonal antibodies, were purchased from Wuhan Boster Bioengineering Limited, China.METHODS: A total of 57 male, Wistar rats were randomly assigned to 4 groups: control (n = 3);150-180 g stretch force was induced in the right optic nerves in stretch-only group (n = 18) toestablish optic nerve stretch injury model; heat stress was applied to 18 animals in heat-stresstreatment group; 18 rats in the heat-stress pretreatment plus stretch group were subjected toidentical stretch injury as stretch-only group after 24-hour heat-stress pretreatment. According tosacrifice time, the groups were assigned to 6 subgroups at different time points of 4, 8, and 16 hours,and 1,3, and 5 days, with 3 rats in each subgroup. No treatment was performed in the control groupexcept anesthesia.MAIN OUTCOME MEASURES: Morphological changes of optic nerves and retinal ganglion cellsfollowing stretch injury were observed by light microscopy following hematoxylin-eosin staining.HSP70 and Tau protein expression levels were observed in retinal ganglial cells from each groupusing im munohistochemistry.RESULTS: (1) Compared with the control group, morphological axonal and retinal ganglial cellchanges, as well as a decreased number of retinal ganglial cells, were identified in the stretch-onlygroup (P< 0.01). Pathological damage in optical nerve and retinal ganglial cells were not remarkablein the heat-stress pretreatment plus stretch group, with no statistical difference in the number ofretinal ganglial cells compared with the control group (P > 0.05). (2) Compared with the control groupsignificantly increased HSP70 expression in retinal ganglial cells occurred in the stretch-only,heat-stress treatment, and heat-stress pretreatment plus stretch groups (P < 0.05 or P < 0.01). Thepeak of HSP70 expression was earlier in the heat-stress pretreatment plus stretch group comparedwith the stretch-only and heat-stress treatment groups, and was expressed over a longer period oftime compared with the heat-stress treatment group. Compared with the control group, Tauexpression in the retinal ganglial cells rapidly increased 4-16 hours following stretch injury in thestretch-only group (P < 0.05 or P < 0.01), and obviously decreased in the heat-stress pretreatmentplus stretch group (P < 0.05 or P < 0.01).CONCLUSION: Tau expression increased following stretch injury, with an earlier expression peakthan HSP70, which indicated that stretch injury-induced HSP70 expression was not strong or quickenough to sufficiently protect the nerve.
