1Special Section:SARS-CoV-2 Preliminary evidence from a multicenter prospective observational study of the safety and efficacy of chloroquine for the treatment of COVID-19显示文摘Effective therapies are urgently needed for the SARS-CoV-2 pandemic.Chloroquine has been proved to have antiviral effect against coronavirus in vitro.In this study,we aimed to assess the efficacy and safety of chloroquine with different doses in COVID-19.In this multicenter prospective observational study,we enrolled patients older than 18 years old with confirmed SARS-CoV-2 infection excluding critical cases from 12 hospitals in Guangdong and Hubei Provinces.Eligible patients received chloroquinephosphate s00 mg,orally,once(half dose)or twice(full dose)daily.Patients treated with non-chloroquine therapy were included as historical controls.The primary endpoint is the time to undetectable viral RNA.Secondary outcomes include the proportion of patients with undetectable viral RNA by day 10 and 14,hospitalization time,duration of fever,and adverse events.A total of 197 patients completed chloroquine treatment,and 176patients were included as historical controls.The median time to achieve an undetectable viral RNA was shorter in chloroquine than in non-chloroquine(absolute difference in medians-6.0 days;95%CI-6.0 to—4.0).The duration of fever is shorter in chloroquine(geometric mean ratio 0.6;95%CIO.5 to 0.8).No serious adverse events were observed in the chloroquine group.Patients treated with half dose experienced lower rate of adverse events than with full dose.Although randomized trials are needed for further evaluation,this study provides evidence for safety and efficacy of chloroquine in COVID-19 and suggests that chloroquine can be a cost-effective therapy for combating the COVID-19 pandemic.Mingxing Huang Man Li Fei Xiao Pengfei Pang Jiabi Liang Tiantian Tang Shaoxuan Liu Binghui Chen Jingxian Shu Yingying You Yang Li Meiwen Tang Jianhui Zhou Guanmin Jiang Jingfen Xiang Wenxin Hong Songmei He Zhaoqin Wang Jianhua Feng Changqing Lin Yinong Ye Zhilong Wu Yaocai Li Bei Zhong Ruilin Sun Zhongsi Hong Jing Liu Huili Chen Xiaohua Wang Zhonghe Li Duanqing Pei Lin Tian Jinyu Xia Shanping Jiang Nanshan Zhong Hong Shan 2020National Science Review2020,7,9:20
2Regional stress field in Yunnan revealed by the focal mechanisms of moderate and small earthquakes显示文摘We determined focal mechanism solutions of 627 earthquakes of magnitude M ≥ 3.0 in Yunnan from January 2008 to May 2018 by using broadband waveforms recorded by 287 permanent and temporary regional stations. The results clearly revealed predominantly strike-slip faulting characteristics for earthquakes in Yunnan, with focal depths concentrated in the top 10 km of the crust. The earthquake mechanisms obtained were combined with the global centroid moment tensor solutions of 80 additional earthquakes from 1976 to 2016 to invert for the regional variations of stress field orientation by using a damped regional-scale stress inversion scheme.Results of the stress field inversion confirmed that the Yunnan region is under a strike–slip stress regime, with both maximum and minimum stress axes being nearly horizontal. The maximum compressional axes are primarily oriented in a northwest-southeast direction, and they experience a clockwise rotation from north to south, whereas the maximum extensional axes are oriented largely northeast-southwest. The maximum compressional axes are in line with the global positioning system–inferred horizontal velocity field and the southeastward escape of the Sichuan–Yunnan Rhombic Block, whereas the maximum extensional axes are consistent with anisotropy derived from SKS splitting. Against the strike–slip background, normal faulting stress regimes can be seen in the Tengchong volcanic area as well as in other areas with complex crisscrossing fault zones.JianHui Tian Yan Luo Li Zhao 2019Earth and Planetary Physics2019,3,3:16
3Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects. 4Comparative analysis between endometrial proteomes of pregnant and non-pregnant ewes during the peri-implantation period显示文摘Background:Early pregnancy failure has a profound impact on both human reproductive health and animal production.2/3 pregnancy failures occur during the peri-implantation period;however,the underlying mechanism(s)remains unclear.Well-organized modification of the endometrium to a receptive state is critical to establish pregnancy.Aberrant endometrial modification during implantation is thought to be largely responsible for early pregnancy loss.Result:In this study,using well-managed recipient ewes that received embryo transfer as model,we compared the endometrial proteome between pregnant and non-pregnant ewes during implantation period.After embryo transfer,recipients were assigned as pregnant or non-pregnant ewes according to the presence or absence of an elongated conceptus at Day 17 of pregnancy.By comparing the endometrial proteomic profiles between pregnant and non-pregnant ewes,we identified 94 and 257 differentially expressed proteins(DEPs) in the endometrial caruncular and intercaruncular areas,respectively.Functional analysis showed that the DEPs were mainly associated with immune response,nutrient transport and utilization,as well as proteasome-mediated proteolysis.Conclusion:These analysis imply that dysfunction of these biological processes or pathways of DEP in the endometrium is highly associated with early pregnancy loss.In addition,many proteins that are essential for the establishment of pregnancy showed dysregulation in the endometrium of non-pregnant ewes.These proteins,as potential candidates,may contribute to early pregnancy loss.Haichao Zhao Linlin Sui Kai Miao Lei An Dong Wang Zhuocheng Hou Rui Wang Min Guo Zhilong Wang Jiqiang Xu Zhonghong Wu Jianhui Tian 2015Journal of Animal Science and Biotechnology2015,6,3:8

6Association of a missense mutation in the luteinizing hormone/choriogonadotropin receptor gene (LHCGR) with superovulation traits in Chinese Holstein heifers显示文摘Background: Upon binding luteinizing hormone in the ovary, the luteinizing hormone/choriogonadotropin receptor (LHCGR) is necessary for follicular maturation and ovulation, as well as luteal function. We detected mutations in the LHCGR gene and evaluated their association with superovulation. Methods: Using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and DNA sequencing, we examined polymorphisms in LHCGR and the genotypes associated with superovulation traits in 127 Chinese Holstein heifers. Results: A G/T polymorphism (ss52050737) in exon 11 was significantly associated with the total number of ova and the number of transferable embryos. 9Targeted therapy of desmoid-type fibromatosis: mechanism, current situation, and future prospects显示文摘Desmoid-type fibromatosis (DF) is a rare monoclonal fibroblastic proliferation that is characterized by locally infiltrative but rarely metastatic lesions. Tyrosine kinase and γ-secretase inhibitors are primarily used in the targeted therapy of DF. The use of these drugs, however, is mainly based on the recommendations of retrospective studies with small sample sizes. Previous studies that focused on the mechanism, efficacy, and safety of targeted therapy for DF were reviewed to provide references for clinical applications and research. The efficacy and safety of targeted therapy were compared with those of other systemic therapy options. Targeted therapy does not provide considerable advantages in efficacy and safety over other medical treatments and is usually applied after the failure of antihormonal therapies, nonsteroidal anti-inflammatory drugs, and chemotherapy. 10Phosphorylation of histone H3 on Ser10 by auto-phosphorylated PAK1 is not essential for chromatin condensation and meiotic progression in porcine oocytes显示文摘Background: The p21-activated kinase 1 (PAK1) is essential for mitosis and plays an important role in the regulation of microtubule assembly during oocyte meiotic maturation in mice; however, little is known about its role in porcine oocytes. Result: Total p21-activated kinase 1 (PAK1) and phosphorylated PAK1 at Thr423 (PAK1 Thr423 ) were consistently expressed in porcine oocytes from the germinal vesicle (GV) to the second metaphase (MII) stages, but phosphorylation of histone H3 at Ser10 (H3 Ser10 ) was only expressed after the GV stage. Immunofluorescence analysis revealed that PAK1 Thr423 and H3 Ser10 colocalized on chromosomes after the GV stage. Blocking of endogenous PAK1 Thr423 by injecting a specific antibody decreased the phosphorylation level of H3 Ser10 ; however, it had no impact on chromatin condensation, meiotic progression, cleavage rate of blastomeres or the rate of blastocyst formation. 12The focal depths of the 2008 Panzhihua earthquake sequence and the stress field in the source region显示文摘Focal depths of the 2008 Ms6.1 Panzhihua earthquake sequence and tectonic stress field in the source area are investigated.Source depths of 24 earthquakes in Panzhihua earthquake sequence with a magnitude M≥3.0 were determined using the seismic depth phase sPL;additionally,the focal depths of 232 earthquakes were measured by fitting the threecomponent waveforms of the P and S waves.The source depth of the main shock is^12 km.The majority of the aftershocks with magnitude M≥3.0 occurred in the brittle upper crust at the depths range of 12-18 km.Further,the Source mechanisms of the 232events around the Panzhihua earthquake source area were determined,and the results show that the earthquakes have predominantly strike-slip mechanisms in the Dianzhong Block,but display complexity of the focal mechanisms outside and near the boundary of the Dianzhong block.The 232 earthquake mechanisms from this study are combined with the solutions from the Global Centroid Moment Tensor (GCMT) catalog to derive 2D stress field.The inversion results show that the Dianzhong block is predominantly under a strike slip faulting regime and the direction of the maximum principal compressionσ1 is northwestsoutheast (NW-SE)-trending.The distribution is coincide with GPS velocity field.However,orientations of principal stress axes as well as the faulting types change outside and near the Dianzhong block.The results show that the tectonic stress field in the study area is predominantly controlled by the southeast (SE)-trending horizontal movement and clockwise rotation of the Dianzhong block as a result of the eastward movement of eastern Tibetan meeting the old and rigid South China block (SCB).The Panzhihua earthquake ruptured at^12 km depth where the tectonic stress regime is under the SE-direction horizontal compression and the NE-direction horizontal extension.Yan LUO Li ZHAO Jianhui TIAN 2020Science China Earth Sciences2020,63,3:2
13Spatial homogenization of soil-surface pollen assemblages improves the reliability of pollen-climate calibration-set显示文摘To make a reliable reconstruction of past climate from soil-surface modern pollen,it is necessary to reduce the sources of error.In this paper,pollen percentages of the sub-continental scale modern pollen-climate dataset from China and Mongolia(with 68%soil-surface samples)are homogenized at various spatial scales.A tailored calibration-set is then applied to lake sediment-surface pollen assemblages from north-central China to evaluate their predictive power.Results indicate that spatial homogenization of modern pollen percentages can increase the proportion of inertia explained by climatic variables in CCA and improve the model performance of leave-one-out cross-validation using WA-PLS.Soil-surface pollen assemblages can thus be employed into a calibration-set for reliable climate estimation and they perform better when the calibration-set has been locally homogenized.Small-scale(e.g.,radii 2,5,or 10 km)homogenization reduces the local noise in soil-surface pollen assemblages and improves the cross-validated performance,while broader scale homogenization(more than 20 km radius)blurs the pollen-climate relationship.Lake sediment-surface pollen assemblages from close to the shore could contain pollen grains transported by rivers or from the shore vegetation and thus fail to represent regional climate well like the assemblages from the central part and deep-water area of lake.Fang TIAN Xianyong CAO Ran ZHANG Qinghai XU Wei DING Xingqi LIU Baolin PAN Jianhui CHEN 2020Science China Earth Sciences2020,63,11:1

15Revolutionize livestock breeding in the future: an animal embryo-stem cell breeding system in a dish显示文摘Meat and milk production needs to increase ~ 70–80% relative to its current levels for satisfying the human needs in 2050.However,it is impossible to achieve such genetic gain by conventional animal breeding systems.Based on recent advances with regard to in vitro induction of germ cell from pluripotent stem cells,herein we propose a novel embryo-stem cell breeding system.Distinct from the conventional breeding system in farm animals that involves selecting and mating individuals,the novel breeding system completes breeding cycles from parental to offspring embryos directly by selecting and mating embryos in a dish.In comparison to the conventional dairy breeding scheme,this system can rapidly achieve 30–40 times more genetic gain by significantly shortening generation interval and enhancing selection intensity.However,several major obstacles must be overcome before we can fully use this system in livestock breeding,which include derivation and mantaince of pluripotent stem cells in domestic animals,as well as in vitro induction of primordial germ cells,and subsequent haploid gametes.Thus,we also discuss the potential efforts needed in solving the obstacles for application this novel system,and elaborate on their groundbreaking potential in livestock breeding.This novel system would provide a revolutionary animal breeding system by offering an unprecedented opportunity for meeting the fast-growing meat and milk demand of humans.Zhuocheng Hou Lei An Jianyong Han Ye Yuan Dongbao Chen Jianhui Tian 2019Journal of Animal Science and Biotechnology2019,10,1:1

18TaGW2L,a GW2-like RING finger E3 ligase,positively regulates heading date in common wheat(Triticum aestivum L.)显示文摘RING finger E3 ligases play an important role in regulating plant growth and development by mediating substrate degradation.In this study,we identified TaGW2L,encoding a Grain width and weight2(GW2)-like RING finger E3 ligase,as a novel positive regulator of heading date in wheat(Triticum aestivum L.).TaGW2L exhibited high amino acid sequence similarities with TaGW2 homoeologs,particularly in the conserved RING finger domain.Expression analysis indicated that TaGW2L was constitutively expressed in various wheat tissues.TaGW2L showed transactivation activity in yeast and could interact with the ubiquitin-conjugating enzymes E2_(s).An in vitro ubiquitination assay verified that TaGW2L possessed a similar E3 ligase activity to TaGW2.Overexpression of the TaGW2L-7A homoeolog in wheat led to a significantly earlier heading date under both natural conditions and long-day conditions.Transcriptome analysis revealed that multiple known genes positively regulating wheat heading were significantly upregulated in the TaGW2L-7A-overexpression plants compared with the wild-type control.Together,our findings shed light on the role of TaGW2L in wheat heading date and provide potential applications of TaGW2L for the adaptation improvement of crops.Daijing Zhang Xiaoxu Zhang Wu Xu Tingting Hu Jianhui Ma Youfu Zhang Jian Hou Chenyang Hao Xueyong Zhang Tian Li 2022The Crop Journal2022,10,4:1

20TaTIP41 and TaTAP46 positively regulate drought tolerance in wheat by inhibiting PP2A activity显示文摘Drought is a major environmental stress limiting global wheat(Triticum aestivum)production.Exploring drought tolerance genes is important for improving drought adaptation in this crop.Here,we cloned and characterized TaTIP41,a novel drought tolerance gene in wheat.TaTIP41 is a putative conserved component of target of rapamycin(TOR)signaling,and the Ta TIP41 homoeologs were expressed in response to drought stress and abscisic acid(ABA).The overexpression of Ta TIP41 enhanced drought tolerance and the ABA response,including ABA-induced stomatal closure,while its downregulation using RNA interference(RNAi)had the opposite effect.Furthermore,Ta TIP41 physically interacted with TaTAP46,another conserved component of TOR signaling.Like TaTIP41,TaTAP46 positively regulated drought tolerance.Furthermore,TaTIP41 and TaTAP46 interacted with type-2A protein phosphatase(PP2A)catalytic subunits,such as TaPP2A-2,and inhibited their enzymatic activities.Silencing TaPP2A-2 improved drought tolerance in wheat.Together,our findings provide new insights into the roles of TaTIP41 and TaTAP46 in the drought tolerance and ABA response in wheat,and their potential application in improving wheat environmental adaptability.Jianhui Ma Yuke Geng Hong Liu Mengqi Zhang Shujuan Liu Chenyang Hao Jian Hou Youfu Zhang Daijing Zhang Weijun Zhang Xueyong Zhang Tian Li 2023Journal of Integrative Plant Biology2023,65,9:0
