1Research Progress on Heat Stress of Rice at Flowering Stage显示文摘Global warming has caused frequent occurrence of heat stress at the flowering stage of single-season rice in the Yangtze River region of China, which results in declines of spikelet fertility and yield in rice. Rice flowering stage is the most sensitive period to high temperatures, and therefore, the key for heat stress happening is the flowering stage coinciding with high temperature, which causes spikelet fertility decreasing in heat-sensitive varieties, and is the major factor for heat injury differences among various rice planting regions. With the development of rice breeding, temperature indexes for heat stress has been converted from daily maximum temperature of 35 oC to 38 oC with the stress duration of more than 3 d. During the flowering stage, anther dehiscence inhibition and low pollen shedding onto the stigma are two main reasons for spikelet fertility reduction under high temperatures. At panicle initiation stage, high temperatures aggravate spikelet degeneration, and destroy floral organ development. Various types of rice varieties coexist in production, and indica-japonica hybrid rice demonstrates the highest heat resistance in general, followed by indica and japonica rice varieties. In production, avoiding high temperature is the main strategy of preventing heat stress, and planting suitable cultivars and adjustment of sowing date are the most effective measures. Irrigation is an effective real-time cultivation measure to decline the canopy temperature during the rice flowering stage. We suggested that further study should be focused on exploring heat injury differences among different rice variety types, and innovating rice-planting methods according to planting system changes in rice planting regions with extreme heat stress. 2Dual-targeting and microenvironment-responsive micelles as a gene delivery system to improve the sensitivity of glioma to radiotherapy显示文摘Dbait is a small double-stranded DNA molecule that has been utilized as a radiosensitizer to enhance the sensitivity of glioma to radiotherapy(RT). However, there is no effective drug delivery system to effectively overcome the blood–brain barrier(BBB). The aim of this study was to develop a gene delivery system by using the BBB and glioma dual-targeting and microenvironment-responsive micelles(ch-Kn(s-s)R8-An) to deliver Dbait into glioma for RT. Angiopep-2 can target the low-density lipoprotein receptor-related protein-1(LRP1) that is overexpressed on brain capillary endothelial cells(BCECs) and glioma cells. In particular, due to upregulated matrix metalloproteinase 2(MMP-2) in the tumor microenvironment, we utilized MMP-2-responsive peptides as the enzymatically degradable linkers to conjugate angiopep-2. The results showed that ch-Kn(s-s)R8-An micelles maintained a reasonable size(80–160 nm) with a moderate distribution and a decreased mean diameter from the cross-linking as well as exhibited low critical micelle concentration(CMC) with positive surface charge, ranging from 15 to40 mV. The ch-K5(s-s)R8-An/pEGFP showed high gene transfection efficiency in vitro, improved uptake in glioma cells and good biocompatibility in vitro and in vivo. In addition, the combination of ch-K5(s-s)R8-An/Dbait with RT significantly inhibited the growth of U251 cells in vitro. 4Divergent Projection Patterns Revealed by Reconstruction of Individual Neurons in Orbitofrontal Cortex显示文摘The orbitofrontal cortex(OFC)is involved in diverse brain functions via its extensive projections to multiple target regions.There is a growing understanding of the overall outputs of the OFC at the population level,but reports of the projection patterns of individual OFC neurons across different cortical layers remain rare.Here,by combining neuronal sparse and bright labeling with a whole-brain florescence imaging system(fMOST),we obtained an uninterrupted three-dimensional whole-brain dataset and achieved the full morphological reconstruction of 25 OFC pyramidal neurons.We compared the wholebrain projection targets of these individual OFC neurons in different cortical layers as well as in the same cortical layer.We found cortical layer-dependent projections characterized by divergent patterns for information delivery.Our study not only provides a structural basis for understanding the principles of laminar organizations in the OFC,but also provides clues for future functional and behavioral studies on OFC pyramidal neurons.Junjun Wang Pei Sun Xiaohua Lv Sen Jin Anan Li Jianxia Kuang Ning Li Yadong Gang Rui Guo Shaoqun Zeng Fuqiang Xu Yu-Hui Zhang 2021Neuroscience Bulletin2021,37,4:5
5Contamination trends of polybrominated diphenyl ethers, organochlorine pesticides and heavy metals in sediments from Dagu Drainage River estuary, Tianjin显示文摘Concentrations of polybrominated diphenyl ethers (PBDEs), organochlorine pesticides (OCPs) and heavy metals in two sediment cores, taken from the Dagu Drainage River, were determined. The contamination trends of these three kinds of compounds were obtained. The results showed that concentrations of PBDEs increased in recent years; there were new contamination sources for OCPs in this area; contamination levels of heavy metals had no significant change in the last several years. 6The combination of chidamide with the CHOEP regimen in previously untreated patients with peripheral T-cell lymphoma: a prospective, multicenter, single arm, phase 1b/2 study显示文摘Objective:To assess the efficacy and safety of the novel histone deacetylase inhibitor,chidamide,in combination with cyclophosphamide,doxorubicin,vincristine,etoposide,and prednisone(Chi-CHOEP)for untreated peripheral T-cell lymphoma(PTCL).Methods:A prospective,multicenter,single arm,phase 1 b/2 study was conducted.A total of 128 patients with untreated PTCL(18–70 years of age)were enrolled between March 2016 and November 2019,and treated with up to 6 cycles with the Chi-CHOEP regimen.In the phase 1 b study,3 dose levels of chidamide were evaluated and the primary endpoint was determination of the maximumtolerated dose and recommended phase 2 dose(RP2 D).The primary endpoint of the phase 2 study was 2-year progression-free survival(PFS).Results:Fifteen patients were enrolled in the phase 1 b study and the RP2 D for chidamide was determined to be 20 mg,twice a week.A total of 113 patients were treated at the RP2 D in the phase 2 study,and the overall response rate was 60.2%,with a complete response rate of 40.7%.At a median follow-up of 36 months,the median PFS was 10.7 months,with 1-,2-,and 3-year PFS rates of 49.9%,38.0%,and 32.8%,respectively.The Chi-CHOEP regimen was well-tolerated,with grade 3/4 neutropenia occurring in approximately two-thirds of the patients.No unexpected adverse events(AEs)were reported and the observed AEs were manageable.Conclusions:This large cohort phase 1 b/2 study showed that Chi-CHOEP was well-tolerated with modest efficacy in previously untreated PTCL patients.Wei Zhang Liping Su Lihong Liu Yuhuan Gao Quanshun Wang Hang Su Yuhuan Song Huilai Zhang Jing Shen Hongmei Jing Shuye Wang Xinan Cen Hui Liu Aichun Liu Zengjun Li Jianmin Luo Jianxia He Jingwen Wang O.A.O’Connor Daobin Zhou 2021Cancer Biology & Medicine2021,18,3:4
7Structural and magnetic properties of Sm_2Fe_(17-x)Nb_x (x = 0-4) alloys prepared by HDDR processes and their nitrides显示文摘Sm2Fe17-xNbxNy (x = 0-4) powder was synthesized by HDDR treatment and nitrogenation. The effects of partial Nb substitution for Fe on the structural and magnetic properties of Sm2Fe17?xNbx alloys and their nitrides were investigated. It was seen that Sm2(Fe,Nb)17 phase exists in both annealed and HDDR-treated Sm2Fe17?xNbx alloys. However, its content is decreased with the increase in Nb substitution. In annealed alloys, Sm2(Fe,Nb)17 phase becomes unstable and will dissociate into SmFe2 and Fe-rich phases when x > 1.5. With HDDR-treatment, the Nb concentration in recombined Sm2(Fe,Nb)17 phase is decreased, and the content of Fe-rich phases is increased. Sm2Fe17?xNbx powder exhibits dendritic cracks and fine particles with a size of less than 300 nm. In nitrogenated alloys, N atoms mainly enter 2:17-type phase to form Sm2(Fe,Nb)17Ny. Partial Nb atoms in Sm2(Fe,Nb)17Ny phase will be released or excluded by nitrogen atoms. Fe-rich phases increase, and are followed by the amorphous Sm2(Fe,Nb)17Ny phase. Nb substitution for Fe with x = 0.5 and 1.0 in Sm2Fe17?xNbxNy powders increases the coercivity and remanence. 8Alterations of peripheral CD4^(+)CD25^(+)Foxp3^(+)T regulatory cells in mice with STZ-induced diabetes显示文摘Complications arising from abnormal immune responses are the major causes of mortality and morbidity in diabetic patients.CD4^(+)CD25^(+)T regulatory cells(Tregs)play pivotal roles in controlling immune homeostasis,immunity and tolerance.The effect of hyperglycemia on CD4^(+)CD25^(+)Tregs has not yet been addressed.Here we used streptozotocin(STZ)-induced diabetic mice to study the effects of long-term hyperglycemia on CD4^(+)CD25^(+)Tregs in vivo.Four months after the onset of diabetes,the frequency of CD4^(+)CD25^(+)Foxp3^(+)T regulatory cells was significantly elevated in the spleen,peripheral blood lymphocytes(PBLs),peripheral lymph nodes(pLNs)and mesenteric LNs(mLNs).CD4^(+)CD25^(+)Tregs obtained from mice with diabetes displayed defective immunosuppressive functions and an activated/memory phenotype.Insulin administration rescued these changes in the CD4^(+)CD25^(+)Tregs of diabetic mice.The percentage of thymic CD4^(+)CD25^(+)naturally occurring Tregs(nTregs)and peripheral CD41Helios1Foxp31 nTregs were markedly enhanced in diabetic mice,indicating that thymic output contributed to the increased frequency of peripheral CD4^(+)CD25^(+)Tregs in diabetic mice.In an in vitro assay in which Tregs were induced fromCD4^(+)CD25^(+)T cells by transforming growth factor(TGF)-b,high glucose enhanced the efficiency of CD4^(+)CD25^(+)Foxp3^(+)T inducible Tregs(iTregs)induction.In addition,CD4^(+)CD25^(+)T cells from diabetic mice were more susceptible to CD4^(+)CD25^(+)Foxp3^(+)TiTreg differentiation than those cells from control mice.These data,together with the enhanced frequency of CD4^(+)CD25^(+)Foxp3^(+)T iTregs in the periphery of mice with diabetes,indicate that enhanced CD4^(+)CD25^(+)Foxp3^(+)T iTreg induction also contributes to a peripheral increase in CD4^(+)CD25^(+)Tregs in diabetic mice.Our data show that hyperglycemia may alter the frequency of CD4^(+)CD25^(+)Foxp3^(+)T Tregs in mice,which may result in late-state immune dysfunction in patients with diabetes.Yu Zhen Lina Sun He Liu Kaizhong Duan Chun Zeng Lianjun Zhang Di Jin Jianxia Peng Wenjun Ding Yong Zhao 2012Cellular & Molecular Immunology2012,9,1:4

12Altered serous levels of monoamine neurotransmitter metabolites in patients with refractory and non-refractory depression显示文摘The study examined plasma metabolite changes of monoamine neurotransmitters in patients with treatment-resistant depression (TRD) and non-TRD before and after therapy. All 30 TRD and 30 non-TRD patients met the diagnostic criteria for a depressive episode in accordance with the International Classification of Diseases, Tenth Revision. Before treatment, and at 4, 6, and 8 weeks after treatment, the plasma metabolite products of monoamine neurotransmitters in TRD group, including 5-hydroxyindoleacetic acid, 3-methoxy-4-hydroxyphenyl ethylene glycol and homovanillic acid, were significantly lower than those in the non-TRD group. After two types of anti-depressive therapy with 5-serotonin and norepinephrine reuptake inhibitor, combined with psychotherapy, the Hamilton Depression Rating Scale scores were significantly reduced in both groups of patients, and the serous levels of 5-hydroxyindoleacetic acid and 3-methoxy-4-hydroxyphenyl ethylene glycol were significantly increased. In contrast, the homovanillic acid level exhibited no significant change. 13VqMYB154 promotes polygene expression and enhances resistance to pathogens in Chinese wild grapevine显示文摘Resveratrol plays a crucial phytoalexin role in the grapevine and is beneficial to human health.However,the molecular mechanism of resveratrol accumulation in the enhancement of disease resistance is unclear.Here,we report that the transcription factor VqMYB154 from Vitis quinquangularis accession Danfeng-2 is strongly expressed under artificial inoculation with Uncinula necator and regulates resveratrol accumulation.Unlike its homolog,VqMYB154 has a pathogen-induced promoter and responds to stimulation by U.necator,Pseudomonas syringae,and other treatments.Yeast one-hybrid and GUS activity assays confirmed that VqMYB154 can activate the stilbene synthase genes VqSTS9,VqSTS32,and VqSTS42 by directly binding to their promoters.Overexpression of VqMYB154 in grape leaves resulted in activation of the stilbene pathway,upregulation of STS genes,and accumulation of stilbenoids.In addition,heterologous overexpression of VqMYB154 in Arabidopsis activated resistance-related genes and resulted in greater programmed cell death and accumulation of reactive oxygen species,which led to resistance against P.syringae.These results suggest that the transcription factor VqMYB154 from V.quinquangularis accession Danfeng-2 participates in the regulatory mechanism that improves the biosynthesis and accumulation of stilbenes and enhances resistance to disease in grapevine.Changyue Jiang Dan Wang Jie Zhang Yan Xu Chaohong Zhang Jianxia Zhang Xiping Wang Yuejin Wang 2021Horticulture Research2021,8,1:2
