| 4 | Imputation from SNP chip to sequence: a case study in a Chinese indigenous chicken population显示文摘Background: Genome-wide association studies and genomic predictions are thought to be optimized by using whole-genome sequence(WGS) data. However, sequencing thousands of individuals of interest is expensive.Imputation from SNP panels to WGS data is an attractive and less expensive approach to obtain WGS data. The aims of this study were to investigate the accuracy of imputation and to provide insight into the design and execution of genotype imputation.Results: We genotyped 450 chickens with a 600 K SNP array, and sequenced 24 key individuals by whole genome re-sequencing. Accuracy of imputation from putative 60 K and 600 K array data to WGS data was 0.620 and 0.812 for Beagle, and 0.810 and 0.914 for FImpute, respectively. By increasing the sequencing cost from 24 X to 144 X, the imputation accuracy increased from 0.525 to 0.698 for Beagle and from 0.654 to 0.823 for FImpute. With fixed sequence depth(12 X), increasing the number of sequenced animals from 1 to 24, improved accuracy from 0.421 to0.897 for FImpute and from 0.396 to 0.777 for Beagle. Using optimally selected key individuals resulted in a higher imputation accuracy compared with using randomly selected individuals as a reference population for resequencing. With fixed reference population size(24), imputation accuracy increased from 0.654 to 0.875 for FImpute and from 0.512 to 0.762 for Beagle as the sequencing depth increased from 1 X to 12 X. With a given total cost of genotyping, accuracy increased with the size of the reference population for FImpute, but the pattern was not valid for Beagle, which showed the highest accuracy at six fold coverage for the scenarios used in this study.Conclusions: In conclusion, we comprehensively investigated the impacts of several key factors on genotype imputation. Generally, increasing sequencing cost gave a higher imputation accuracy. But with a fixed sequencing cost, the optimal imputation enhance the performance of WGP and GWAS. An optimal imputation strategy should take size of reference population, imputation algorithms, marker density, and population structure of the target population and methods to select key individuals into consideration comprehensively. | 6 | Attribution analysis based on Budyko hypothesis for land evapotranspiration change in the Loess Plateau, China显示文摘Land evapotranspiration(ET) is an important process connecting soil, vegetation and the atmosphere, especially in regions that experience shortage in precipitation.Since 1999, the implementation of a large-scale vegetation restoration project has significantly improved the ecological environment of the Loess Plateau in China.However, the quantitative assessment of the contribution of vegetation restoration projects to long-term ET is still in its infancy.In this study, we investigated changes in land ET and associated driving factors from 1982 to 2014 in the Loess Plateau using Budyko-based partial differential methods.Overall, annual ET slightly increased by 0.28 mm/a and there were no large fluctuations after project implementation.An attribution analysis showed that precipitation was the driving factor of inter-annual variability of land ET throughout the study period;the average impacts of precipitation, potential evapotranspiration, and vegetation restoration on ET change were 61.5%, 11.5% and 26.9%, respectively.These results provide an improved understanding of the relationship between vegetation condition change and climate variation on terrestrial ET in the study area and can support future decision-making regarding water resource availability. | HE Guohua ZHAO Yong WANG Jianhua GAO Xuerui HE Fan LI Haihong ZHAI Jiaqi WANG Qingming ZHU Yongnan | 2019 | Journal of Arid Land2019,11,6: | 5 |
| 7 | Size fraction of phytoplankton and the contribution of natural plankton to the carbon source of Zhikong scallop Chlamys farreri in mariculture ecosystem of the Sanggou Bay显示文摘The biomass and size fraction of phytoplankton in terms of chlorophyll a(Chl a) was measured during four cruises conducted in April, July, October 2013 and January 2014 in mariculture area, the Sanggou Bay, China.Results show that total Chl a levels in the surface seawater of the Sanggou Bay generally range from 0.10 to 20.46μg/L, with an average value of 2.13 μg/L. Nano-phytoplankton was the most important size-fraction and accounted for about 65.1% of total Chl a. In order to evaluate the importance of the 'protozoan trophic link' for energy transfer from the microbial loop to filter-feeding feeders, Zhikong scallop Chlamys farreri was then offered a natural planktonic community as potential prey. Results show that scallops obtained carbon source from natural plankton with the rate of 11 033.05 μg/(g·d). Protists(nanoflagellates and ciliates) were the dominant source of carbon retained by scallop(48.78%). The microbial loop provided 58.45% of the carbon source for farmed scallops. | 8 | Bifunctional Oxygen Electrocatalyst of Mesoporous Ni/NiO Nanosheets for Flexible Rechargeable Zn–Air Batteries显示文摘One approach to accelerate the stagnant kinetics of both the oxygen reduction and evolution reactions(ORR/OER)is to develop a rationally designed multiphase nanocomposite,where the functions arising from each of the constituent phases,their interfaces,and the overall structure are properly controlled.Herein,we successfully synthesized an oxygen electrocatalyst consisting of Ni nanoparticles purposely interpenetrated into mesoporous NiO nanosheets(porous Ni/NiO).Benefiting from the contributions of the Ni and NiO phases,the well-established pore channels for charge transport at the interface between the phases,and the enhanced conductivity due to oxygen-deficiency at the pore edges,the porous Ni/NiO nanosheets show a potential of 1.49 V(10 mA cm^-2)for the OER and a half-wave potential of 0.76 V for the ORR,outperforming their noble metal counterparts.More significantly,a Zn-air battery employing the porous Ni/NiO nanosheets exhibits an initial charging-discharging voltage gap of 0.83 V(2 mA cm^-2),specific capacity of 853 mAh gZn^-1 at 20 mA cm^-2,and long-time cycling stability(120 h).In addition,the porous Ni/NiO-based solid-like Zn-air battery shows excellent electrochemical performance and flexibility,illustrating its great potential as a next-generation rechargeable power source for flexible electronics. | Peitao Liu Jiaqi Ran Baorui Xia Shibo Xi Daqiang Gao John Wang | 2020 | Nano-Micro Letters2020,12,5: | 4 |
| 9 | Effect of Qiangxin Huoli decoction on rats with adriamycin-induced chronic heart failure显示文摘OBJECTIVE: To investigate the effect of Qiangxin Huoli decoction on rats with chronic heart failure(CHF) induced by adriamycin(ADR), and to investigate the underlying mechanism of this effect.METHODS: Ninety-six healthy Wistar rats were divided into six groups: control, CHF model, CHF treated by Shenfu injection, and three CHF groups treated with Qiangxin Huoli decoction at high, medium, and low doses, respectively. Qiangxin Huoli decoction was administered orally to protect the stomach in the three Qiangxin Huoli decoction groups, while the control group and the CHF model group were administered the same volume of 0.9% physiological saline, and the Shenfu group wereadministered the same volume of Shenfu injection. Ten days later, the CHF model was then induced in all groups except the control group by intraperitoneal injection of ADR at gradient dose intervals. The bodyweights were recorded on days 10, 20, 30, and 40. Hemodynamic indices were recorded, including left ventricular systolic pressure(LVSP), left ventricular end-diastolic pressure(LVEDP), maximum increase in left ventricular pressure(+dp/dt_(max)), maximum decrease in left ventricular pressure(-dp/dt_(max)), heart rate(HR), and electrocardiogram using an eight-channel physiological recorder with LabChart software monitoring. The plasma brain natriuretic peptide(BNP) concentration was determined by enzyme-linked immunosorbent adsorption. The expressions of B-cell lymphoma-2(Bcl-2) and Bcl-2-associated X protein(Bax) were detected by immunohistochemical methods.RESULTS: The CHF model group were in poor condition, and the mean bodyweight was significantly decreased compared with the control group. Furthermore, compared with the control group, the CHF groups had significantly decreased LVSP, +dp/dt_(max), and-dp/dt_(max), and significantly increased LVEDP. The CHF groups also showed significant increases in HR, S-T segment elevation, and plasma BNP levels compared with the control group. Compared with the CHF model group, the treatment groups had significantly increased Bax expression(P < 0.05) and significantly decreased Bcl-2 expression(P < 0.01), indicating less apoptosis. | 10 | CO2 emission driving forces and corresponding mitigation strategies under low-carbon economy mode:evidence from China's Beijing-Tianjin-Hebei region显示文摘On account of the background of China's 'new normal' characterized by slower economic growth, this paper analyses the low-carbon economy status quo in the Beijing-Tianjin-Hebei region and empirically investigates the relationship between carbon dioxide(CO_2) emissions and its various factors for China's Beijing-Tianjin-Hebei region using panel data econometric technique. We find evidence of existence of Environmental Kuznets Curve. Results also show that economic scale, industrial structure, and urbanization rate are crucial factors to promote CO_2emissions. However, technological progress, especially the domestic independent research and development, plays a key role in C0_2 emissions abatement. Next, we further analyze the correlation between each subregion and various factors according to Grey Relation Analysis. | 15 | Theoretical modeling and experimental verifications of the single-compressor-driven three-stage Stirling-type pulse tube cryocooler显示文摘This paper establishes a theoretical model of the single-compressor-driven (SCD) three-stage Stirlingtype pulse tube cryocooler (SPTC) and conducts experimental verifications. The main differences between the SCD type and the multi-compressor-driven (MCD) crycooler are analyzed, such as the distribution of the input acoustic power in each stage and the optimization of the operating parameters, in which both advantages and difficulties of the former are stressed. The effects of the dynamic temperatures are considered to improve the accuracy of the simulation at very low temperatures, and a specific simulation example aiming at 10 K is given in which quantitative analyses are provided. A SCD threestage SPTC is developed based on the theoretical analyses and with a total input acoustic power of 371.58 W, which reaches a no-load temperature of 8.82 K and can simultaneously achieve the cooling capacities of 2.4 W at 70 K, 0.17 W at 25 K, and 0.05 W at 10 K. | 19 | Dynamic shielding of bacterial outer membrane vesicles for safe and efficient chemo-immunotherapy against tumors显示文摘Bacterial outer membrane vesicles(OMVs)are potent immunostimulants of regulating the tumor microenvironment(TME)for immunotherapy,and can be used to deliver drugs.However,the severe systemic inflammatory response triggered by OMVs upon intravenous(i.v.)injection has limited their application.Here,we developed a safe and effective strategy by conjugating doxorubicin-loaded serum albumin(SA-DOX,AD)onto the surface of OMVs using a matrix metalloproteinase(MMP)-cleavable peptide linker(cL).This approach enabled the dynamic shielding of OMVs to reduce the systemic side effects while simultaneously enhancing the anti-tumor effects through chemo-immunotherapy.Specifically,the resulting OMV-cL-AD formulation exhibited significantly enhanced accumulation at the tumor site after i.v.administration,facilitated by the SA decoration on the OMVs surface.Subsequently,the shield on the OMV-cL-AD was cleaved by the over-expressed MMP in the TME,leading to the release of both OMVs and AD.This process provided OMV-induced immunotherapy and DOX-induced chemotherapy,resulting in synergistic tumor inhibition.In conclusion,our work demonstrated the potential of OMV-cL-AD as an effective immunochemotherapy strategy that can prolong the survival time of mice without inducing side effects. | Shuping Li Xin Li Jiaqi Meng Weier Bao Shuang Wang Peng Ye Xiao-Dong Gao Wei Wei | 2024 | Nano Research2024,17,2: | 1 |
| 20 | Oxygen vacancy-mediated WO_(3) phase junction to steering photogenerated charge separation for enhanced water splitting显示文摘Effective charge separation and transfer is deemed to be the contributing factor to achieve high photoelectrochemical(PEC)water splitting performance on photoelectrodes.Building a phase junction structure with controllable phase transition of WO_(3) can further improve the photocatalytic performance.In this work,we realized the transition from orthorhombic to monoclinic by regulating the annealing temperatures,and constructed an orthorhombic–monoclinic WO_(3)(o-WO_(3)/m-WO_(3))phase junction.The formation of oxygen vacancies causes an imbalance of the charge distribution in the crystal structure,which changes the W–O bond length and bond angle,accelerating the phase transition.As expected,an optimum PEC activity was achieved over the o-WO_(3)/m-WO_(3) phase junction in WO_(3)-450 photoelectrode,yielding the maximum O_(2) evolution rate roughly 32 times higher than that of pure WO_(3)-250 without any sacrificial agents under visible light irradiation.The enhancement of catalytic activity is attributed to the atomically smooth interface with a highly matched lattice and robust built-in electric field around the phase junction,which leads to a less-defective and abrupt interface and provides a smooth interfacial charge separation and transfer path,leading to improved charge separation and transfer efficiency and a great enhancement in photocatalytic activity.This work strikes out on new paths in the formation of an oxygen vacancy-induced phase transition and provides new ideas for the design of catalysts. | Huimin LI Qianqian SHEN Han ZHANG Jiaqi GAO Husheng JIA Xuguang LIU Qi LI Jinbo XUE | 2022 | Journal of Advanced Ceramics2022,11,12: | 1 |
